  • The LDPE films were photolyzed for 100 hours in UV apparatus with 400-W medium pressure mercury lamp radiating wavelengths longer than 254nm. A simple kinetic model was introduced and was applied to interpret the FT-IR absorbance of carbonyl and hydroperoxide bonds as a function of UV exposure time. From these data, the reaction mechanism and the rate determining step of photodegradation were determined.

Larvicidal potency of selected xerophytic plant extracts on Culex pipiens (Diptera: Culicidae)

  • Chemical insecticides released into the environment may have adverse biological effects. Therefore, there is a need for ecofriendly insecticides for mosquito control. Xerophytic plant extracts that may provide more ecofriendly active component were evaluated against Culex pipiens 4th instars. Plant extracts prepared using different solvents with a Soxhlet apparatus and different concentrations were tested against Culex pipiens larvae. The effects were observed at 24 h and 72 h intervals and $LD_{50}$ and $LD_{90}$ values determined. Chloroform ($CHCl_3$) and ethyl acetate (EtOAc) extracts of Althaea ludwigii were the most effective against Cx. pipiens $4^{th}$ instars, but were highly dependent on extract concentrations and exposure time. Results suggest that A. ludwigii extracts contain bioactive compounds, such as phenols and saponins, that may provide effective Cx. pipienslarval control. However, the extract was found to be toxic to zebrafish larvae, and may be toxic to other aquatic fauna. Further studies to determine the active components and toxicity to other fauna are needed.

Effects and Efficacy of Natural Product on Infectious Diseases of pseudomonas aeruginosa (천연물 유래 물질이 감염성 질환에 미치는 영향과 효능)

  • Pseudomonas aeruginosa is a ubiquitous gram-negative bacterium causing serious infections. The P. aeruginosa T3SS is a syringe-like apparatus on the bacterial surface, with 4 effector toxins: ExoS, ExoT, ExoY, and ExoU. Here, we investigated the effect of ExoS and ExoT of the T3SS of P. aeruginosa K strain (PAK). The type three secretion system (T3SS) is a major virulence system of Pseudomonas aeruginosa (P. aeruginosa). The effector protein Exotoxin S (ExoS) produced by P. aeruginosa is secreted into the host cells via the T3SS. For the purpose of screening the inhibitors with regard to ExoS secretion, we developed the sandwich-type enzyme-linked immunosorbent assay (ELISA) system. PAK clinical strains induce proinflammatory cytokine production through the T3SS, and this involves NF-κB activation in pneumonia mouse models. We tried to confirm the role of the NF-κB transcription factor in ExoS- and ExoT-induced pneumonia mouse models. pro-inflammatory cytokines induction in response to ExoS and ExoT infection relied on NF-κB activation. Our findings highlight the roles of natural poduct in inhibiting proinflammatory cytokine expression during ExoS and ExoT exposure in PAK infections, paving the way for a novel therapeutic approach for the treatment of pulmonary infections.

The Increased Expression of Gelatinolytic Proteases Due to Cigarette Smoking Exposure in the Lung of Guinea Pig (기니픽에서 흡연 노출에 의한 젤라틴 분해 단백 효소의 발현 양상에 관한 연구)

  • Background : Chronic obstructive pulmonary disease(COPD) is one of the major contributors to morbidity and mortality among the adult population. Cigarette smoking(CS) is undoubtedly the single most important factor in the pathogenesis of COPD. However, its mechanism is unclear. The current hypothesis regarding the pathogenesis of COPD postulates that an imbalance between proteases and antiproteases leads to the destructive changes in the lung parenchyma. This study had two aims. First, to evaluate the effect of CS exposure on histologic changes of the lung parenchyme, and second, to evaluate the effect of CS exposure on the expression of the gelatinolytic enzymes in BAL fluid cells in guinea pigs. Methods : Two groups of five guinea pigs were exposed to the whole smoke of 20 commercial cigarettes per day, 5 hours/day, 5 days/week, for 6weeks, and 12 weeks, respectively, using a smoking apparatus. Five age-matched guinea pigs exposed to room air were used as controls. Five or more sections were microscopically extamined(${\times}400$) and the number of cellular infiltration of the alveolar wall was measured in order to evaluate the effect of CS exposure on the histologic changes of lung parenchyme. The statistical significance was analyzed by a linear regression method. To evaluate the expression of the gelatinolytic enzymes in intraalveolar cells, BAL fluid was obtained and the intraalveolar cells were separated by centrifugation (500 g for 10 min at $4^{\circ}C$). Two sets of culture plates were loaded with $1{\times}10^6$ intraalveolar cells. One plate, contained O.1mM EDTA, a inhibitor of matrix metalloproteases(MMPs), and the other plate had no EDTA. Both plates were incubated for 48 hours at $37^{\circ}C$. After incubation, gelatinolytic protease expression in the supernatants was analyzed by gelatin zymography. Results : At the end of CS exposure, the level of blood carboxy Hb had increased significantly(4.1g/dl in control group, 24g/dl immediately after CS exposure, 18g/dl 30 min after CS exposure, 15g/dl 1 hour after CS exposure). Alveolar inflammatory cells were identified in the CS exposed guinea pigs. The number of alveolar cellular cells observed in a microscopic field ($400{\times}$) was $121.4{\pm}7.2$, $158.0{\pm}20.2$, $196.8{\pm}32.8$, in the control, the 6 weeks, and the 12 weeks group, respectively. The increased extent of inflammatory cellular infiltration of the lung parenchema showed a statistically significant linear relationship with the duration of CS exposure(p=0.001, $r^2=0.675$). Several types of gelatinolytic enzymes in the intraalveolar cells of CS exposed guinea pigs were expressed, of which some were inhibited by EDT A. However, the gelatinolytic enzymes were not expressed in the control groups. Conclusion : CS exposure increases inflammatory cellular infiltration of the alveolar wall and the expression of gelatinolytic proteases in guinea pigs. EDTA inhibits some of the gelatinolytic proteases. These findings suggest a possibility that CS exposure may increase MMP expression in the lungs of guinea pigs.

Median Modified Wiener Filter for Noise Reduction in Computed Tomographic Image using Simulated Male Adult Human Phantom (시뮬레이션된 성인 남성 인체모형 팬텀을 이용한 전산화단층촬영 에서의 노이즈 제거를 위한 Median Modified Wiener 필터)

  • Computed tomography (CT) has the problem of having more radiation exposure compared to other radiographic apparatus. There is a low-dose imaging technique for reducing exposure, but it has a disadvantage of increasing noise in the image. To compensate for this, various noise reduction algorithms have been developed that improve image quality while reducing the exposure dose of patients, of which the median modified Wiener filter (MMWF) algorithm that can be effectively applied to CT devices with excellent time resolution has been presented. The purpose of this study is to optimize the mask size of MMWF algorithm and to see the excellence of noise reduction of MMWF algorithm for existing algorithms. After applying the MMWF algorithm with each mask sizes set from the MASH phantom abdominal images acquired using the MATLAB program, which includes Gaussian noise added, and compared the values of root mean square error (RMSE), peak signal-to-noise ratio (PSNR), coefficient correlation (CC), and universal image quality index (UQI). The results showed that RMSE value was the lowest and PSNR, CC and UQI values were the highest in the 5 x 5 mask size. In addition, comparing Gaussian filter, median filter, Wiener filter, and MMWF with RMSE, PSNR, CC, and UQI by applying the optimized mask size. As a result, the most improved RMSE, PSNR, CC, and UQI values were showed in MMWF algorithms.

Radioactivation Analysis of Concrete Shielding Wall of Cyclotron Room Using Monte Carlo Simulation (PET 사이클로트론 가동에 따른 콘크리트 차폐벽의 방사화)

  • Cyclotron is a device that accelerates positrons or neutrons, and is used as a facility for making radioactive drugs having short half-lives. Such radioactive drugs are used for positron emission tomography (PET), which is a medical apparatus. In order to make radioactive drugs from a cyclotron, a nuclear reaction must occur between accelerated positrons and a target. After the reaction, unncessary neutrons are produced. In the present study, radioactivation generated from the collisions between the concrete shielding wall and the positrons and neutrons produced from the cyclotron is investigated. We tracked radioactivated radioactive isotopes by conducting experiments using FLUKA, a type of Monte Carlo simulation. The properties of the concrete shielding wall were comparatively analyzed using materials containing impurities at ppm level and materials that do not contain impurities. The generated radioactivated nuclear species were comparatively analyzed based on the exposure dose affecting human body as a criterion, through RESRAD-Build. The results of experiments showed that the material containing impurities produced a total of 14 radioactive isotopes, and $^{60}Co$(72.50%), $^{134}Cs$(16.75%), $^{54}Mn$(5.60%), $^{152}Eu$(4.08%), $^{154}Eu$(1.07%) accounted for 99.9% of the total dose according to the analysis having the exposure dose affecting human body as criterion. The $^{60}Co$ nuclear species showed the greatest risk of radiation exposure. The material that did not contain impurities produced a total of five nuclear species. Among the five nuclear species, 54Mn accounted for 99.9% of the exposure dose. There is a possibility that Cobalt can be generated by inducive nuclear reaction of positrons through the radioactivation process of $^{56}Fe$ instead of impurities. However, there was no radioactivation because only few positrons reached the concrete wall. The results of comparative analysis on exposure dose with respect to the presence of impurities indicated that the presence of impurities caused approximately 98% higher exposure dose. From this result, the main cause of radioactivation was identified as the small ppm-level amount of impurities.

Measurement of Retention, Repellency and Penetration of Pesticide for Protective Clothing (방제복에 대한 농약의 체류성, 발수성 및 침투성 측정)

  • BACKGROUND: The personal protective equipments (PPEs) is the most important factor for reducing the pesticide exposure during the preparation and spraying of pesticides. This study was to investigate the retention, repellence and penetration of protective clothing of PPEs according to the ISO 22608 'Liquid penetration resistance-pipette test. Protective clothing for agriculture worker is very important for their health. However, test method for measurement of protective clothing is not presented in Korea. METHODS AND RESULTS: In order to measure the retention, repellence and penetration of protective clothing, a apparatus for pipette method in accordance with the ISO Guideline was prepared. The test was conducted at $25{\pm}5^{\circ}C$, $60{\pm}10%$ of relative humidity and pipette applied to a apparatus was validated to take exact amount(0.2 mL). The retention, repellence and penetration of five types of protective clothing and one type of shirt were analyzed by GC/MS. Pendimethalin(5% a.i, emulsion) was used as a test pesticide to measure above factors. The retention were less than 11.0% with the exclusion of two types(F4 and shirt) and the repellency was more than 67.0% with the exclusion of shirt material. The penetration was less than 5.4%, however, that of shirt was 66.7%. CONCLUSION: This results indicated that all protective clothing were suitable to use as PPEs according to the criteria specified by ISO Guide 22608. However, shirt was not suitable due to high penetration. This test method established for measurement the retention, repellence and penetration of protective clothing will help to establish the test notice of pipette method.

A Study on Hydrophobic Surface Treatment for Microfluidic System Fabrication Based on SLA 3D Printing Method (SLA 3D 프린팅 방식 기반의 미세 유체 시스템 제작을 위한 소수성 표면 처리 연구)

  • The SLA (Stereolithography Apparatus) method is a type of 3D printing technique predicated on the transformation of liquid photocurable resin into a solid form through UV laser exposure, and its application is increasing in various fields. In this study, we conducted research to enhance the hydrophobicity and transparency of SLA 3D printing surfaces for microfluidic system production. The enhancement of surface hydrophobicity in SLA outputs was attainable through the application of hydrophobic coating methods, but the coating durability under different conditions varied depending on the type of hydrophobic coating. Additionally, to simultaneously achieve the required transparency and hydrophobic properties for the fabrication of microfluidic systems, we applied hydrophobic coatings to the proposed transparency enhancement method from prior research and compared the changes in contact angles. Teflon coating was proposed as a suitable hydrophobic coating method for the fabrication of microfluidic systems, given its excellent transparency and high coating durability in various environmental conditions, in comparison to titanium dioxide coating. Finally, we produced an Electrophoresis of Charged Droplet (ECD) chip, one of the digital microfluidics systems, using SLA 3D printing with the proposed Teflon coating method (Fluoropel 800). Droplet manipulation was successfully demonstrated with the fabricated chip, confirming the potential application of SLA 3D printing technology in the production of microfluidic systems.

Spectrometry Analysis of Fumes of Mixed Nuclear Fuel (U0.8Pu0.2)O2 Samples Heated up to 2,000℃ and Evaluation of Accidental Irradiation of Living Organisms by Plutonium as the Most Radiotoxic Fission Product of Mixed Nuclear Fuel

  • Purpose: The purpose of this work is to describe the spectrometric analysis of gaseous cloud formation over reactor mixed uranium-and-plutonium (UP) fuel $(U_{0.8}Pu_{0.2})O_2$ samples heated to a temperature $>2,000^{\circ}C$, and thus forecast and evaluate radiation hazards threatening humans who cope with the consequences of any accident at a fission reactor loaded by UP mixed oxide $(U_{0.8}Pu_{0.2})O_2$, such as a mixture of 80% U and 20% Pu in weight. Materials and methods: The UP nuclear fuel samples were heated up to a temperature of over $2,000^{\circ}C$ in a suitable assembly (apparatus) at out-of-pile experiments' implementation, the experimental in-depth study of metabolism of active materials in living organisms by means of artificial irradiation of pigs by plutonium. Spectrometric measurements were carried out on the different exposed organs and tissues of pigs for the further estimation of human internal exposure by nuclear materials released from the core of a fission reactor fueled with UP mixed oxide. Results: The main results of the research described are the following: (1) following the research on the influence of mixed fuel fission products (radioactive isotopes being formed during reactor operation as a result of nuclear decay of elements included into the fuel composition) on living organisms, the authors determined the quantities of plutonium dioxide ($PuO_2$) that penetrated into blood and lay in the pulmonary region, liver, skeleton and other tissues; and (2) experiments confirmed that the output speed of plutonium out of the basic precipitation locations is very small. On the strength of the experimental evidence, the authors suggest that the biological output of plutonium can be disregarded in the process of evaluation of the internal irradiation doses.

Effect of the Structure of the Smallest Poresize Layer on the Permeability of PES Microfiltration Membranes (최소 기공층 구조가 PES계 정밀여과막 투과 성능에 미치는 영향)

  • PES (polyethersulfone) membranes with highly enhanced their asymmetry were prepared by phase inversion process. The membranes were prepared by using PES/DMF (N,N-dimethylformamide)/TSA (p-toluenesulfonic acid)/PVP (polyvinylpyrrolidone) casting solution and water coagulant. The pre-coagulation of membrane surface which was induced by an addition of TSA as a demixing agent and PVP as a swelling polymer in the PES solution and humid exposure time, played a crucial role in determining morphological properties and the PWP (pure water permeation) performance. The PES solution was coated on polyester film under condition of 80% humidity for a while ($72{\sim}144$ sec) before immersing in a coagulation bath. The characterization of membranes was carried out by a capillary flow porometer, a FE-SEM and a permeation test apparatus. As the thickness of the smallest pore size layer (SPL) decreased, the asymmetry of membrane increased under conditions of 20 wt% of TSA and 10 wt% of PVP in 11 wt% of PES solution during longer humid contact time. As a result, the membranes showed a remarkable increase of PWP.
