Interferon-γ-mediated secretion of tryptophanyl-tRNA synthetases has a role in protection of human umbilical cord blood-derived mesenchymal stem cells against experimental colitis

  • Mesenchymal stem cells (MSCs) are multipotent adult stem cells that present immunosuppressive effects in experimental and clinical trials targeting various rare diseases including inflammatory bowel disease (IBD). In addition, recent studies have reported tryptophanyl-tRNA synthetase (WRS) possesses uncanonical roles such as angiostatic and anti-inflammatory effects. However, little is known about the function of WRS in MSC-based therapy. In this study, we investigated if a novel factor, WRS, secreted from MSCs has a role in amelioration of IBD symptoms and determined a specific mechanism underlying MSC therapy. Experimental colitis was induced by administration of 3% DSS solution to 8-week-old mice and human umbilical cord blood-derived MSCs (hUCB-MSCs) were injected intraperitoneally. Secretion of WRS from hUCB-MSCs and direct effect of WRS on isolated $CD4^+$ T cells was determined via in vitro experiments and hUCB-MSCs showed significant therapeutic rescue against experimental colitis. Importantly, WRS level in serum of colitis induced mice decreased and recovered by administration of MSCs. Through in vitro examination, WRS expression of hUCB-MSCs increased when cells were treated with interferon-${\gamma}$ ($IFN-{\gamma}$). WRS was evaluated and revealed to have a role in inhibiting activated T cells by inducing apoptosis. In summary, $IFN-{\gamma}$-mediated secretion of WRS from MSCs has a role in suppressive effect on excessive inflammation and disease progression of IBD and brings new highlights in the immunomodulatory potency of hUCB-MSCs.

Effects of Korean Radish on DSS-Induced Ulcerative Colitis in Mice

  • The present study aimed to investigate the comparative evaluation of pharmacological efficacy between sulfasalazine alone and combination with herbal medicine on dextran sodium sulfate (DSS)-induced UC in mice. Balb/c mice received 5% DSS in drinking water for 7 days to induce colitis. Animals were divided into five groups (n = 9): group I-normal group, group II-DSS control group, group III-DSS + sulfasalazine (30 mg/kg), group IV-DSS + sulfasalazine (60 mg/kg), group V-DSS + sulfasalazine (30 mg/kg) + Radish Extract mixture (30 mg /kg) (SRE). DSS-treated mice developed symptoms similar to those of human UC, such as severe bloody diarrhea and weight loss. SRE supplementation, as well as sulfasalazine, suppressed colonic length and mucosal inflammatory infiltration. In addition, SRE treatment significantly reduced the expression of pro-inflammatory signaling moleculesthrough suppression both mitogen-activated protein kinases(MAPK) and nuclear factor-kappa B ($NF-{\kappa}B$) signaling pathways, and prevented the apoptosis of colon. Moreover, SRE administration significantly led to the up-regulation of anti-oxidant enzyme including SOD and Catalase. This is the first report that Radish extract mixture combined with sulfasalazine protects against experimental UC via the inhibition of both inflammation and apoptosis, very similar to the standard-of-care sulfasalazine.

  • 자가면역성 간염, 원발성 담관성 간경화증, 원발성 경화성 담관염은 대표적인 자가 면역성 간질환이다. 이 각 질환의 임상적, 조직학적, 혈청학적 특징이 혼재되어 어느 한 질환으로 진단하기 어려운 경우를 중복증후군(overlap syndrome)이라 하고, 최근 소아에게서도 드물게 보고되고 있다. 한편 경화성 담관염은, 염증성 장질환에서 가장 흔히 동반되는 간담도계질환으로 알려져 있다. 저자들은 궤양성 대장염으로 진단된 환아에게서 간조직 검사, 혈청 검사, 방사선 검사 등을 통해 자가면역성 간염-원발성 경화성 담관염의 중복증후군을 진단하였기에 보고하는 바이다. 염증성 장질환 환아에게서 자가 면역성 간질환 또는 중복증후군 동반여부를 염두에 두어 임상적, 조직학적, 혈청학적 검사를 통해 적절한 진단과 치료가 필요하겠다.

Treatment with Extracellular Vesicles from Giardia lamblia Alleviates Dextran Sulfate Sodium-Induced Colitis in C57BL/6 Mice

  • Inflammatory bowel disease (IBD) is a chronic and recurrent illness of the gastrointestinal tract. Treatment of IBD traditionally involves the use of aminosalicylic acid and steroids, while these drugs has been associated with untoward effects and refractoriness. The absence of effective treatment regimen against IBD has led to the exploration of new targets. Parasites are promising as an alternative therapy for IBD. Recent studies have highlighted the use of parasite-derived substances, such as excretory secretory products, extracellular vesicles (EVs), and exosomes, for the treatment of IBD. In this report, we examined whether EVs secreted by Giardia lamblia could prevent colitis in a mouse model. G. lamblia EVs (GlEVs) were prepared from in vitro cultures of Giardia trophozoites. Clinical signs, microscopic colon tissue inflammation, and cytokine expression levels were detected to assess the effect of GlEV treatment on dextran sulfate sodium (DSS)-induced experimental murine colitis. The administration of GlEVs prior to DSS challenge reduced the expression levels of pro-inflammatory cytokines, including tumor necrosis factor alpha, interleukin 1 beta, and interferon gamma. Our results indicate that GlEV can exert preventive effects and possess therapeutic properties against DSS-induced colitis.

  • Glycyrrhizae Radix (GR), commomly known as liquorice, is a medicinal and edible plant widely used in East Asia with its pharmacological properties. Currently, Glycyrrhiza uralensis, G. glabra and G. inflata are used for pharmaceutical purposes in Korea and then the improved Glycyrrhiza varieties, WON-GAM (WG) has been developed by Korea Rural Development Administration. To evaluate equivalence of efficacy, several comparative studies between already-registered species and new cultivars have been conducted. The aim of this study was to evaluate the effect of WG on fecal microbiota in DSS-induced colitis model. Fecal microbiota was analyzed by terminal restriction fragment length polymorphism (T-RFLP). The composition of the fecal microbiota did not show a specific pattern based on experimental groups; however, a tendency toward an increase in the proportion of Lactobacillales was observed. Glycyrrhiza varieties could change composition of fecal microbiota in DSS-induced colitis model. 효소처리를 이용한 마늘 추출물이 Lipopolysaccharide 유발 급성 장염 모델에 미치는 효과 및 기전 연구 (Study of the Effect and Underlying Mechanism of Enzyme-Treated Garlic Extract on a Lipopolysaccharide-Induced Acute Colitis Model)

  • Objective: This study aims to explore the pharmacological effects and mechanisms of enzyme (Viscozyme)-treated garlic extract (EG) in an animal model of acute enteritis induced by lipopolysaccharide (LPS). Methods: The experiment included four subgroups: normal, control, EG200 (treated with 200 mg/kg EG), and EG400 (treated with 400 mg/kg EG). Drug administration lasted 3 days, followed by the induction of acute enteritis in all groups (except normal) through the intraperitoneal administration of 20 mg/kg of LPS 1 h after the last oral dose. Autopsy was conducted 24 h later to collect serum and colon tissue. Serum was analyzed for reactive oxygen species (ROS) and C-reactive protein (CRP), while Western blotting was performed on the colon tissue. Results: After analyzing the ROS and CRP levels in serum, the EG treatment group exhibited a significant decrease compared with the control group. The EG treatment group exhibited a significant decrease in the activation of the mitogen-activated protein kinases (MAPKs)/nuclear factor-kappa B p65 (NF-κB) pathway compared with the control group. EG administration significantly regulated apoptosis-related factors, including B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X, cysteine aspartyl-specific protease-3, and cytochrome C. Conclusions: EG treatment in mice with LPS-induced acute colitis reduced the ROS and CRP levels, suppressed the MAPKs/NF-κB pathway in the colon, and effectively alleviated acute enteritis by modulating apoptosis-related factors. Based on these findings, EG emerges as a promising candidate for the prevention and treatment of acute colitis, showing its potential therapeutic efficacy in this experimental model.

  • Objectives : The objective of this study was to investigate the improvement effect of Sprout of Coix lacryma-jobi var. mayuen Stapf water extract (SC) on the dextran sulfate sodium (DSS)-induced ulcerative colitis mice. Methods : The antioxidant activity of SC was measured through total polyphenol and total flavonoid content in vitro. The experiment was conducted with seven-week-old male Balb/c mice. After 1 week adaptation, acute colitis was induced by oral administration of 5% DSS dissolved in drinking water, for 7 days. And normal mice received drinking water without DSS throughout the entire experimental period. For each experiment, the mice were divided into 4 groups and 24 colitis mice were arbitrarily allocated into 3 groups (n = 8/group); Normal group, Control group, SC 100 mg/kg treated group (SCL), SC 200 mg/kg treated group (SCH). Serum and colon tissues were collected after one weeks of drug administration. Results : ROS levels, ONOO- levels, AST, and ALT in serum were decreased in SC treated groups compared to the control group. Western blotting measurements of Nrf2, HO-1, SOD, catalase, GPx-1/2, IL-4, IL-10, and Bcl2 showed that the SC treated groups was increased compared to the Control group. Also, western blot measurements of NF-κBp65, p-IκBα, COX-2, iNOS, TNF-α, IL-1β, Bax, and Caspase-3 showed that the SC treated groups was reduced compared to the Control group. Conclusion : Taken together, these results suggest that SC treatment can attenuate the DSS-induced colitis though inhibiting NF-κB pathway and enhancing Nrf2 pathway. Therefore, SC was the potential to be used as a natural therapeutic drug.

천추(天樞)(ST25) 고삼약침(苦蔘藥鍼)이 DSS로 유발된 대장염 백서에 미치는 영향 (The Effects of Sophorae Radix Pharmacopuncture Therapy on the Chunchu(ST25) for DSS-induced Colitis in Rats)

  • Objectives : The purpose of this study is to investigate the effects of Sophorae radix pharmacopuncture(SRP) therapy on the Chunchu($ST_{25}$) on intestinal disease in rats with dextran sulfate sodium(DSS)-induced colitis. Methods : The experimental model of colitis was induced by an infection of DSS for fifteen days. After colitis was induced, SRP therapy was practiced on the Chunchu($ST_{25}$) once every three days for a total of five times. Afterward, colon length, damage to the colonic mucosa, complete blood count, aspartate aminotransferase(AST), alanine transferase(ALT), blood urea nitrogen(BUN), creatinine, TNF-${\alpha}$ and IL-6 were measured. Results : The results were obtained as below. 1. Colon length was significantly increased in the SRP group 2 compared to the control group. 2. Damage of colonic mucosa was observed less in the SRP groups than in the other groups. 3. The mean rates of white blood cell(WBC), neutrophils and lymphocytes were significantly increased in SRP group 2 compared to the control group. 4. The mean rates of HGB and hematocrit(HCT) were significantly increased in the SRP compared to the control group. 5. No significant differences were shown in AST, ALT, BUN and Creatinine between the SRP groups and the control group. 6. The mean rate of TNF-${\alpha}$ was significantly decreased in the SRP groups compared to the control group. 7. The mean of IL-6 was significantly decreased in the SRP groups compared to the control group. Conclusions : These results indicated that SRP therapy on Chunchu($ST_{25}$) functions as a treatment to the dextran sulfate sodium-induced colitis.

Fecal Microbiota Transplantation (FMT) Alleviates Experimental Colitis in Mice by Gut Microbiota Regulation

  • Inflammatory bowel disease (IBD) is an increasing global burden and a predisposing factor to colorectal cancer. Although a number of treatment options are available, the side effects could be considerable. Studies on fecal microbiota transplantation (FMT) as an IBD intervention protocol require further validation as the underlying mechanisms for its attenuating effects remain unclear. This study aims to demonstrate the ameliorative role of FMT in an ulcerative colitis (UC) model induced by dextran sulfate sodium (DSS) and elucidate its relative mechanisms in a mouse model. It was shown that FMT intervention decreased disease activity index (DAI) levels and increased the body weight, colon weight and colon length of experimental animals. It also alleviated histopathological changes, reduced key cytokine expression and oxidative status in the colon. A down-regulated expression level of genes associated with NF-κB signaling pathway was also observed. The results of 16S rRNA gene sequencing showed that FMT intervention restored the gut microbiota to the pattern of the control group by increasing the relative abundance of Firmicutes and decreasing the abundances of Bacteroidetes and Proteobacteria. The relative abundances of the genera Lactobacillus, Butyricicoccus, Lachnoclostridium, Olsenella and Odoribacter were upregulated but Helicobacter, Bacteroides and Clostridium were reduced after FMT administration. Furthermore, FMT administration elevated the concentrations of SCFAs in the colon. In conclusion, FMT intervention could be suitable for UC control, but further validations via clinical trials are recommended.

염증성 장 질환 동물 모델에서 황금탕과 감초사심탕의 효능 연구 (A Study of the Effectiveness of Hwanggeum-tang and Gamchosasim-tang in the Mice Model of Inflammatory Bowel Disease)

  • Objectives: This study investigates the mechanism of Hwanggeum-tang (HGT) and Gamchosasim-tang (GST) on inflammatory bowel disease (IBD). Methods: The mice (C57BL/6N) were treated with distilled water and 3% dextran sulfate sodium (DSS) to experimentally induce ulcerative colitis. The mice were divided into 7 groups of (6 mice: normal, negative control, positive control (with sulfasalazine), 4 experimental groups (with HGT and GST, respectively). RAW 264.7 cells were used for cell experiments. The experiment was conducted in two ways: in vitro and in vivo. Results: In the experimental group (HGT, GST) of in vitro experiments, NO production decreased, and significant changes in gene expression and protein activation were observed. The length of the colon recovered in the experimental groups (HGT, GST) of the in vivo experiment was longer than that of the negative control group, and the mucosal barrier was recovered. Sone significant changes in the amount of mRNA expression were partially observed, and significant changes in protein activation also were confirmed. Conclusions: HGT and GST are effective in treating IBD caused by DSS. In the same herbal preparation group, the higher the concentration, the better the experimental effect, and when the same concentration was tested, HGT was more effective than GST. Herbal medicine has a higher antioxidant effect than sulfasalazine, so it is also excellent for cell protection.
