Induction of FSTL1 Gene in Rat Myometrium by Exogenous Estrogen

  • The aim of this study was carried to the induction of follistatin-like 1 (FSTL1) in rat myometrium tissue by exogenous estrogen. PCR was performed by using estrogen treatment after ovariectomy of myometrium mRNA and only ovariectomy of myometrium mRNA and on ovariectomy of myometrium mRNA in rat normal uterus tissue. The PCR products were visualized on agarose gel (1.2%) electrophoresis. FSTL1 mRNA expression level was significantly higher (**P<0.001 or *P<0.05) in estrogen treatment after ovariectomy than in only ovariectomy, and was significantly higher (**P<0.001) in no ovariectomy than in estrogen with treatment after ovariectomy. In conclusion, Although the mechanisms of FSTL1 gene were uncertain, It also might be inducted by exogenous estrogen. And also it is thought to be related to the estorgen mechanism.

  • Changes of plasma vitellogenin (VTG) and glutamate pyruvate transaminase (GPT) were examined for determining whether hepatocyte was damaged during the process of VTG induction in the juvenile rockfish, Sebastes schlegeli exposed to exogenous estrogen (estradiol-l7$\beta$, E$_2$). Rockfishes were intraperitoneally injected with E$_2$(5 mg/kg B.W.) in 70% ethanol and plasma sampling were extracted at 0, 1, 3, 6, 9, 12, 15 days af-ter E$_2$administration. VTG and GPT were then analyzed by SDS -PAGE and Reitman -Frankel method, respectively. VTG band was detected at a molecular weight position of 175 kDa on Day 3 after E$_2$administration. This band became more distinct at 6 days, but its was gradually thinned with time -course, and not detected at 15 days. GPT was suddenly increased at 1 days after 22 administration and highest GPT was detected at 3 days. However. GPT was gradually decreased with time -course as the change of VTG. These results suggest that the process of VTG induction by exogenous E$_2$damage to hepatocyte, and plasma GPT was temporarily increased in the juvenile rockfish.

Exogenous Estrogen Toxicity in a Dog (개의 외인성 에스트로겐 중독 증례)

  • A 2-year-old female mongrel dog was referred to the Veterinary Medical Center of Chungbuk National University due to anorexia, depression and weight loss after the administration of exogenous estrogen for contraception in the local hospital. Although the dog was medicated with antibiotics and fluid for two weeks in the local hospital, there was a deterioration in the patient's general condition. Therefore the dog was referred to our Veterinary Medical Center. Hematological and biochemical analysis showed non-regenerative anemia, thrombocytopenia, severely immature leukocytosis, hyperbilirubinemia, hyperglycemia and elevated serum alkaline phosphatase, gamma-glutamyltransferase, and lactate dehydrogenase. Abdominal ultrasonography showed double line of gallbladder and irregular hypertrophic wall of uterus. On the basis of these examination's results and history taking, final diagnosis was exogenous estrogen-induced myelotoxicosis with cholecystitis and endometritis. The symptomatic therapy for estrogen toxicosis was performed for 26 days after the final diagnosis. However, the symptom had no response to therapy except endometritis and anemia. On day 135 after administration of estrogen, platelet and neutrophil count were normalized, but monocytic leukocytosis was occurred.

The Effect of Endogenous and Exogenous Estrogen on Risk Factors of Coronary Heart Disease -Part II : On Platelet Aggregation- (내인성 및 외인성 Estogen이 관상동맥질환 위험인자에 미치는 영향 -제2보: 내인성 및 외인성 Estrogen이 혈소판 응집에 미치는 영향-)

  • In the atherosclerotic subjects, arterial endothelial cell injury and plaque formation are suspected to be strong causable factors in developing acute coronary syndrome, and it was revealed that platelets have a very important role in this case. Women are exposed to atherosclerosis at a different degree after menopause or oral contraception. The purpose of this study was to determine the effects of endogenous and exogenous estrogen on the degree of platelet aggregation in platelet rich plasma(PRP) in twenty nonsmoking healthy Korean women for 12 weeks. The subjects were assigned to three groups: (1) eight women aged 49 to 60(yr) for the postmenopausel(Pst) group, (2) eight, aged 22 to 30(yr) for the premenopausa(Pre) group, (3) four, aged 23 to 30(yr) for the oral contraceptive (OC) group which used triphasic OC formulation. Fasting blood sample were obtained from the subjects, (1) once per 6 weeks in the Pst group, (2) every phase of the menstrual cycle in the Pre group, (3) each once during and after OC administration in the OC group. ADP, collagen and epinephrine were used as aggregating reagents, and platelet aggregation and time(Δt: time reaching to the maximum point of aggregation) in PRP were measured at the maximum point of aggregation in five minutes. All the data were adjusted for dietary effects, personality type and body mass index(BMI) by using analysis of covariation(ANCOVA). Platelet aggregation to ADP and collagen(MADP and MCOLL) were not significantly different among the three groups, and Δt to ADP and collagen(TADP and TCOLL) were not either. But maximum platelet aggregability and Δt to epinephrine(MEPIN and TEPIN) were significantly different among the three groups, and the OC group showed the lowest value (p<0.01). Maxtimum platelet aggregability and Δt during the menstrual cycle were not significantly different in the Pre group. Any other significant differences in the maximum platelet aggregability and Δt were found between oral contraception phase and washing out phase(menstruation) in the OC group. In results, maximum platelet aggregability and aggregation time to ADP and collagen seemed not to be affected by endogenous and exogenous estrogen, even though MEPIN and TEPIN showed significantly low in the OC group among the three groups.

  • Estrogens are known as the steroid hormones and essential regulators of developments, differentiations, and fertility in animals including humans. Recently, classic focus on estrogens which are considered as female hormone is changing in the whole field of reproductive endocrinology. Especially, interest in estrogen functions in male reproduction is increasing more and more, as numerous studies about the endocrine disruptors, interrupting the endocrine system, are being carried out. To understand exactly the function of estrogen in a male reproductive system, a summary for estrogen receptors upon developmental distributions in testis will be useful. In addition to the regulatory roles of estrogen in male, unexpected exposure to exogenous estrogens causes defects of differentiation of male reproductive system and an injury of spermatogenesis. Also, this review highlights the indicator of exogenous estrogens to perturb male fertility. These approaches would give tile practical information about estrogen roles in male development and reproduction.

  • To determine the effects of endogenous and exogenous strogen on serum lipid levels, twenty nonsmoking healthy Korean women were participated in this experiment for 12 weeks. They were assigned to three groups : (1) eight women aged 22 to 30(yr) for the premenopausal(Pre) group, (2) eight, aged 49 to 60(yr) for the postmenoparusal(Pst) group, (3) four, aged 23 to 30(yr) for the oral contraceptive(OC) group which used triphasic OC formulation. Fasting blood samples representing every phase of the hormonal levels were obtained from the subjects of the Pre and the OC group. From the subjects of the Pst group, fasting blood samples were obtained once per three weeks for 12 weeks. All the serum data were adjusted for dietary effects, exercise, personality type and body mass index(BMI) by using analysis of covariation(ANCOVA). Serum lipid levels of the three groups were significantly different. While serum levels of triglycerides(TG)(p<0.0001), low density lipoprotein-chloesterol(LDL-C)/high density liporotein-cholesterol(HDL-C) ratio (LDC-C/HDL-C)(P<0.01) and total cholesterol (TC)/HDL-C ratio (TC/HDL-C)(P<0.001) were significatnly high in the Pst group, serum HDL-C(P0.001) level was significantly high in the Pre group. The OC group showed significantly low serum TC(P<0.0001) and LDL-C(P<0.0001) levels. There was no signidicant difference in the fluictuation of serum lipid levels during the menstrual cycle of the Pre group. However, in the OC group, serum TG level was significantly increased at phase 2(P<0.05) where exogenous estrogen administration was highest. Even though other serum lipid levels of the OC group were not significantly fluctuated according to the exogenous estrogen administration, there was a trend of increased levels of serum TC, LDL-C, LDL-C/HDL-C and TC/HDL-C and decreased level of HDL-C during the menstruation period. Also, serum TC level was high(P<0.005) and serum TG level was low (P<0.005) at the baseline of the OC group compared with the periods of OC administration. When screening and counseling the female population at risk for coronary heart disease(CHD), the result of this study suggest that in may be desirable to divide the population into several groups according to their personal physiological characteristics, such as age, OC administration, menstrual cycle and menopause, as well as general risk factors for CHD.

  • Isoflavones are naturally occurring plant chemicals belonging to the 'phytoestrogen' class. The isoflavones are strikingly similar in chemical structure to natural estrogens. The phenolic ring is a key structural element of most compounds that bind to estrogen receptors. Dietary components that recently have received attention for their action as phytoestrogens are soy isoflavones. Soy products are the most significant dietary sources of isoflavones. Recently It is concerned clinical nutrition of isoflavone that is driven by reason of alternative sources of exogenous estrogen are constantly being needed. Estrogen therapy after the menopause offers protection from cardiovascular disease, reduces the extent of osteoporosis and relieves menopausal symptoms. Exogenous estrogen treatment is a fear of possible increased risk of developing breast cancer and because of side effects. Daily intake of soybean or soy food can affirmative effect to disease occurrence, that is based on mechanical investigation, experimental results of animals and human. Research into isoflavone is going on various field to relieve hormone - dependent disease such as cancer, menopausal symptom, cardiovascular disease and osteoporosis. Isoflavone is plenty in soybean meal, soy by-product, but only limited information is available on isoflavone efficacy into animal husbandry. Thus we conducted three experiments to investigate the effects of dietary isoflavone on productivities, antioxidative responses and bone metabolism in poultry. Dietary supplementation of isoflavone resulted in preventing the lipid oxidation of plasma and egg yolk. Dietary isoflavone improved bone development in egg-type growing chicks and broilers in terms of tibial strength. It was suggested that the proper use of feed additives such as isoflavone might provide means of improving antioxidative effect, skeletal strength, egg and eggshell quality.

Effect of Exogenous Estrogen on Growth and Mammary Gland Development in Korean Native Heifers (외인성 Estrogen이 한우 암송아지의 성장과 유선 및 유방의 발달에 미치는 영향)

  • The effects of exogenous estrogen on growth, mammary development, and the age at which the first estrous symptoms are detected were investigated for the first time in Korean native heifers at 6 months of age. At 90 days after treatment, weight gains of heifers implanted with 10 mg of estradiol were not different from those of the control heifers. However, weight gams of heifers implanted with 20 mg of estradiol were significantly increased at 90 days by 18%, relative to control values. At 180 days, cumulative weight gains of heifers implanted with either 10 mg or 20 mg of estradiol averaged 6.7% or 17.8%, respectively, greater than controls. But these values were not statistically significant. Regardless of dosage, teat diameter gains in the treatment groups were significantly increased at 90 days about 159%, relative to controls. Cumulative teat diameter gains in the heifers implanted with either 10 mg or 20 mg of estradiol were also significantly increased at 180 days by 100% or 128%, respectively, compared to controls. Teat length gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly increased at 90 days by 200% and 295%, respectively, compared to controls. Cumulative teat length gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly 265% and 325%, respectively, higher than controls. The heifers implanted with either 10 mg or 20 mg of estradiol showed a significant increase in teat volume gains at 90 days of 282% or 246%, respectively, over those of the control heifers. Cumulative teat volume gains in the heifers implanted with either 10 mg or 20 mg of estradiol were significantly increased at 180 days by 251% and 244%, respectively, compared to control values. However, there were no significant differences m gams of teat diameter, length and volume between heifers implanted with 10 mg and 20 mg of estradiol regardless of 90 days or 180 days. Heifers implanted with 10 mg or 20 mg of estradiol showed the first estrus 20 or 124 days, respectively, faster than controls. Overall, These data strongly suggest that estradiol implantation into heifers stimulates growth and mammary development and hasten the age at which the first estrus in Korean native heifers is observed.

Effect of cell-penetrating peptide-conjugated estrogen-related receptor ${\beta}$ on the development of mouse embryos cultured in vitro

  • Objective: Estrogen related receptor ${\beta}$ (Esrrb) is a member of the orphan nuclear receptors and may regulate the expression of pluripotencyrelated genes, such as Oct4 and Nanog. Therefore, in the present study, we have developed a method for delivering exogenous ESRRB recombinant protein into embryos by using cell-penetrating peptide (CPP) conjugation and have analyzed their effect on embryonic development. Methods: Mouse oocytes and embryos were obtained from superovulated mice. The expression of Oct4 mRNA and the cell number of inner cell mass (ICM) in the in vitro-derived and in vivo-derived blastocysts were first analyzed by real time-reverse transcription-polymerase chain reaction and differential staining. Then 8-cell embryos were cultured in KSOM media with or without $2{\mu}g/mL$ CPP-ESRRB protein for 24 to 48 hours, followed by checking their integration into embryos during in vitro culture by Western blot and immunocytochemistry. Results: Expression of Oct4 and the cell number of ICM were lower in the in vitro-derived blastocysts than in the in vivo-derived ones (p<0.05). In the blastocysts derived from the CPP-ESRRB-treated group, expression of Oct4 was greater than in the non-treated groups (p<0.05). Although no difference in embryonic development was observed between the treated and non-treated groups, the cell number of ICM was greater in the CPP-ESRRB-treated group. Conclusion: Treatment of CPP-ESRRB during cultivation could increase embryos' expression of Oct4 and the formation rate of the ICM in the blastocyst. Additionally, an exogenous delivery system of CPP-conjugated protein would be a useful tool for improving embryo culture systems.
