Optimization of Submerged Culture Conditions for Exo-biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica was studied. Maltose, yeast extract and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in flask culture were pH 5.0, $25^{\circ}C$ and 150 rpm in a meidum containing of 30 g maltose, 6 g yeast extract, 2 g polypeptone, 0.5 g $K_2HPO_4$, 0.2 g $KH_2PO_4$, 0.2 g $MnSo_4\;{\cdot}\;5H_2O$, 0.2 g $MgSO_4\;{\cdot}\;7H_2O$ in 1-L distilled water. Exo-biopolymer production and mycelial growth in the suggested medium were significantly increased in a 2.5-L jar fermentor, where the maximum biopolymer concentration was 8 g/1.

  • The effect of agitation speed on the production of mycelial growth and exo-biopolymer in bioreaetor culture was investigated. The maximum production of mycelia and exo-biopolymer was obtained at 350 rpm, where the maximum concentrations of mycelial growth and exo-biopolymer were 62 g/L and 23 g/L, respectively. The effect of agitation speed on the hyphal length and the number of tips was examined. The hyphal length and the number of tips of Cordyceps sinensis 16 in bioreaetor culture at 350 rpm were increased up to 660 ${\mu}m$ and 14/ml, respectively.

Label-Free Rapid and Simple Detection of Exonuclease III Activity with DNA-Templated Copper Nanoclusters

  • In this study, DNA-templated copper nanoclusters (DNA-CuNCs) were used to detect exonuclease III (Exo III) activity, which is important for the diagnosis and therapy of several diseases. The results of this study showed that Exo III was affected by the concentrations of magnesium ions and sodium ions, and its optimal conditions for cleavage were $5mM\;Mg^{2+}$ and less than $25mM\;Na^+$. With a blunt-end DNA, more than 98% of DNA was digested by Exo III. As expected, with two or four cytosines in the terminal position of a 4-base overhanging DNA such as 5'-GGCC-3' and 5'-CCCC-3', there was little cleavage by Exo III compared with a blunt-end DNA.

  • Amniotic membrane stem cells are considered as a good alternative to embryonic stem cells, but their use in clinical studies is still not common. Here, exosomes from canine amniotic membrane mesenchymal stem cells (cAmMSC-exo) were used for dog sperm cryopreservation. Upon cryopreserved straws using cryoprotectant containing 0, 0.5, 1, or 2 ㎍/mL of cAmMSC-exo were thawed, motility and membrane integrity were analyzed. However, results showed no significant differences between the groups. We concluded that cAmMSC-exo with lower than 2 ㎍/mL have no effects on sperm cryopreservation, and further studies to get higher concentrations of cAmMSC-exo should be conducted for clinical application.

  • 노보르나디엔의 이량화반응에 의해 제조되는 다중고리 탄화수소화합물 제조에 대한 연구결과를 설명하였다. NBD 이량체 중에서 pentacyclic exe-t-exo, hexacyclic exe-eodo, hexacyclic endo-endo 이성질체를 선택적으로 제조하기위하여 이량화 반응을 실시하였다. 각각의 이량화반응 촉매, 반응절차 그리고 분석방법이 개발되었다. 본 실험을 통하여 적당한 반응조건, 비교적 높은 이량체수율을 얻었으며, 이 반응기술은 고에너지밀도 연료 개발에 유용하게 사용될 수 있을 것이다.

  • The potential physiological role and technological application of the premature termination of DNA polymerization through the off-switch of exo+ polymerases were studied using 3' phosphorothioate-modified or unmodified primers with single base mismatch distal to the 3' terminus. With exonuclease-digestible unmodified primers, a gradient premature termination of DNA polymerization was observed when amplified with exo+ polymerases. With 3' allele specific phosphorothioate-modified primers, an efficient off-switch effect occurred in the discrimination of a single nucleotide polymorphism when directly using genomic DNA. Clearly, the off-switch of exo+ polymerases is useful in biomedical research.

Exo-inulinase 생산 균주의 분리ㆍ동정 및 효소 생산의 최적화 (Isolation and identification of Exo-Inulinase Producing Bacterium and Optimization of the Enzyme Production)

  • A bacterium producing exo-inulinase was isolated from soil and identified Pseudomonas sp. and named as Pseudomonas sp. NO5. The optimal culture conditions for the efficient production of exo-inulinase from Pseudomonas sp. NO5 were obtained by cultivating with the medium 1$\%$ sucrose, 0.5$\%$ yeast extract, 0.5$\%$ $(NH_4)_2$$HPO_4$, 0.05$\%$ $MgSO_4$$7H_2$0, 0.001$\%$ and $FeSO_4$$7H_2$0 at $37^{\circ}C$ in initial pH 7.0 for 20 hours. The enzyme was induced maximally in the presence of sucrose or inulin at early stationary phase about 20 hour after cultivation.

Exo-xylanase 생산균의 분리 및 동정 (Isolation and Identification of Exo-xylanase Producing Microorganism)

  • 부패한 나무, 퇴비, 제지공장의 폐지 및 폐수 등으로부터 분리한 300여 종류의 섬유소 분해균 중 xylanase 활성이 다른 균주에 비해 비교적 높았던 33번 균주를 균의 형태학적, 생화학적 특성과, 균체 지방산 조성에 의하여 Pseudomonas sp.로 동정하였다. Xylanase 활성의 최적 온도외 최적 pH는 각각 $50^{\circ}C$와 5.5이었고, 효소의 안정성은 $45^{\circ}C$ 이하의 온도와 pH 5.0에서 7.0 사이에서 유지되었다. 이 균주가 생산하는 xylanase는 효소반응분해물의 paper chromatography에 의하여 주로 exo-type의 xylanase임이 밝혀졌다.

  • Pseudomonas stutzeri IAM 12097이 생산(生産)하는 Exo-maltotetraohydrolase의 gel filtration에 의하여 추정(推定)된 분자량(分子量)은 60,000이었고, SDS-polyacrylamide gel electrophoresis에 의하여 추정(推定)된 분자량(分子量)은 63,000이었으며 등전점(等電點)은 PH 4.8이었다. 최적(最適)pH는 6.6, PH안정(安定)범위는 $6.0{\sim}10.5$, 최적온도(最適溫度)는 $45{\sim}50^{\circ}C,\;40^{\circ}C$이하(以下)에서는 안정(安定)하였으며, $55^{\circ}C$이상(以上)에서는 급격하게 불활성화(不活性化)되었다. 본효소(本酵素)는 $Ag^+,\; Hg^{++},\;I_2,\;{\beta}-cycoldextrin$에 의하여 완전(完全)히 저해(沮害)되었고 EDTA, ${\rho}-CMB$, IAA에 의하여 약간 저해(沮害)되었다. soluble starch, amylopectin, amylose에 대(對)한 Michaelis constant(Km)는 각각(各各) 7.70mg/ml, 6.17mg/ml, 5.56mg/ml이었다.

  • Pseudomonas stutzeri IAM 12097이 생산(生産)하는 Exo-maltotetraohydrolase는 soluble starch, amylose, amylopectin, oyster glycogen, corn, potato, glutinous rice, green banana, arrow root 의 호화전분(糊化澱粉), maltopentaose, maltohexaose, maltoheptaose, maltooctaose를 가수분해(加水分解)하였으나, ${\alpha},{\beta},{\gamma}-cyclodextin$, sucrose, raffinose, pullulan, maltose, maltotriose, maltotetraose는 분해(分解)하지 못하였다. 소당류중(少糖類中) maltohexaose를 가장 잘 분해(分解)하였으며, 각종기질(各種基質)의 주분해산물(主分解産物)은 maltotetraose였다. pullulanase의 혼용(混用)으로 호화전분(糊化澱粉)의 분해율(分解率)은 증가(增加)되었으나 생전분(生澱粉)에 대(對)하여는 혼용효과(混用效果)를 인정(認定)할 수 없었고 생전분중(生澱粉中)에서 corn starch가 가장 잘 분해(分解)되며 raw potato starch, raw arrow root starch, raw high amylose corn starch의 분해(分解)는 미약(微弱)하였다.
