Changes in Cytochrome c Oxidase and NO in Rat Lung Mitochondria Following Iron Overload

  • In this study, the effects of iron on cytochrome c oxidase (CcO) in rat lung mitochondria were examined. Similar to liver mitochondria, iron accumulated considerably in lung mitochondria (more than 2-fold). Likewise, the reactive oxygen species and nitric oxide (NO) content of mitochondria were increased by more than 50% and 100%, respectively. NO might be produced by nitric oxide synthase (NOS), eNOS and iNOS type, with particular contribution by NOS in mitochondria. The respiratory control ratio of iron overloaded lung mitochondria dropped to nearly 50% due to increased state 4. Likewise, cytochrome c oxidase activity was lowered significantly to approximately 50% due to excess iron. Real-time PCR revealed that the expression of isoforms 1 and 2 of subunit IV of CeO was enhanced greatly under excess iron conditions. Taken together, these results show that oxidative phosphorylation within lung mitochondria may be influenced by iron overload through changes in cytochrome c oxidase and NO.

BaTiO3 타겟의 R.F. 방전 중 변수에 따른 광반사분광 특성 (Optical Emission Spectroscopy with Parameters During R.F. Discharge of BaTiO3 Target)

  • In this study, optical emission spectroscopy was used to monitor the plasma produced during the RF magnetron sputtering of a $BaTiO_3$ target. The intensities of chemical species were measured by real time monitoring with various discharge parameters such as RF power, pressure, and discharge gas. The emission lines of elemental and ionized species from $BaTiO_3$ and Ti targets were analyzed to evaluate the film composition and the optimized growth conditions for $BaTiO_3$ films. The emissions from Ar(I, II), Ba(I, II) and Ti(I) were found during sputtering of the $BaTiO_3$ target in Ar atmosphere. With increasing RF power, all the line intensities increased because the electron density increased with increasing RF power. When the Ar pressure increased, the Ba(II) and Ti(I) line intensity increased, but the $Ar^+$ line intensity decreased with increasing pressure. This result shows that high pressure is of greater benefit for the ionization of Ba than for that of Ar. Oxygen depressed the intensity of the plasma more than Ar did. When the Ar/$O_2$ ratio decreased, the intensity of Ba decreased more sharply than that of Ti. This result indicates that the plasma composition strongly depends on the discharge gas atmosphere. When the oxygen increased, the Ba/Ti ratio and the thickness of the films decreased. The emission spectra showed consistent variation with applied power to the Ti target during co-sputtering of the $BaTiO_3$ and Ti targets. The co-sputtered films showed a Ba/Ti ratio of 1.05 to 0.73 with applied power to the Ti target. The films with different Ba/Ti ratios showed changes in grain size. Ti excess films annealed at $600^{\circ}C$ did not show the second phase such as $BaTi_2O_5$ and $TiO_2$.

금속산화물(Cu-ferrite)를 이용한 수소제조 연구 (Study on the hydrogen production using the metal oxide (Cu-ferrite))

  • Redox characteristics of metal oxide for hydrogen production by thermochemical water-splitting were investigated. $CuFe_2O_4$ as a redox pair that had a different molar ratio of Cu and Fe were prepared by co-precipitation method. Hydrogen production consisted of water-splitting step and thermal reduction step was performed below 1200K. Redox characteristics of Cu-ferrites were studied using the thermal gravimetric analysis technique. Also, structure change of Cu-ferrite during thermal reduction was investigated using the high temperature controlled XRD. In results, oxygen release of Cu-ferrite during the thermal reduction was initiated at oxygen site combined with Cu. Consequently, oxygen release amount of Cu-ferrite was increased with increase of Cu molar ratio of Cu-ferrite. It was found that thermal reduction of Cu-ferrite was begun at $875^\circ{C}$. It was confirmed that structure of Cu-ferrite was changed to metal and cation excess metal oxide during the thermal reduction step.

BaO 과잉량에 따른 BaTiO3의 전기전도도 (Electrical Conductivity Revisited in Excess BaO into BaTiO3)

  • In this study the electrical conductivity of excess BaO in $BaTiO_3$ was measured to investigate the relationship between defects and solubility in the temperature range of $900^{\circ}C$ to $1300^{\circ}C$ under various oxygen partial pressure. First of all, quenched $BaTiO_3$ powders of various Ba/Ti ratios were analysed by X-ray diffraction to confirm whether second phase is formed or not. As the results, we observed the solubility of BaO in the temperature range of $1200^{\circ}C$ to $1400^{\circ}C$, and it was also found that the conductivity minima move to lower $PO_2$ with increasing excess BaO within solubility limit.

$CeO_2$박막의 결정성 및 전기적 특성에 미치는 sputtering시 산소분압비의 영향 (Effects of oxygen partial pressure during sputtering on texture and electrical properties of $CeO_2$ thin films)

  • MFISFET(Metal-ferroelectric-insulator-semiconductor-field effect transistor)에의 적용을 위한 절연체로서 CeO$_2$ 박막을 r.f. magnetron sputtering법에 의해 제조하였다. 스퍼터링시 증착개스는 Ar과 $O_2$를 사용하였으며 산소분압비에 따른 $CeO_2$박막의 결정성 및 전기적 특성에 미치는 영향을 평가하였다. p형-Si(100)기판 위에 $600^{\circ}C$에서 증착된 $CeO_2$ 박막들은(200)방향으로 우선방향성을 가지고 성장하였으며 Ar만으로 증착된 박막의 우선방향성은 증가하였으나 상대적으로 많은 하전입자와 표면 거칠기로 인해 C-V특성에서 큰 이력특성을 보였고 산소분압비가 증가함에 따라 양호한 특성을 보였다. 이것은 이동가능한 이온전하의 감소에 기인한다고 할 수 있다. Ce:O의 비는 모든 박막에서 1:2.22~2.42를 보여 산소과잉의 조성을 나타냈으며 산소분압비에 따라 제조된 박막들의 누설전류값은 100 kV/cm의 전계에서 $10^{-7}$~$10^{-8}$A의 차수를 보였다.

Neuroprotective effects of L-carnitine against oxygen-glucose deprivation in rat primary cortical neurons

  • Purpose: Hypoxic-ischemic encephalopathy is an important cause of neonatal mortality, as this brain injury disrupts normal mitochondrial respiratory activity. Carnitine plays an essential role in mitochondrial fatty acid transport and modulates excess acyl coenzyme A levels. In this study, we investigated whether treatment of primary cultures of rat cortical neurons with L-carnitine was able to prevent neurotoxicity resulting from oxygen-glucose deprivation (OGD). Methods: Cortical neurons were prepared from Sprague-Dawley rat embryos. L-Carnitine was applied to cultures just prior to OGD and subsequent reoxygenation. The numbers of cells that stained with acridine orange (AO) and propidium iodide (PI) were counted, and lactate dehydrogenase (LDH) activity and reactive oxygen species (ROS) levels were measured. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and the terminal uridine deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling assay were performed to evaluate the effect of L-carnitine (1 ${\mu}M$, 10 ${\mu}M$, and 100 ${\mu}M$) on OGD-induced neurotoxicity. Results: Treatment of primary cultures of rat cortical neurons with L-carnitine significantly reduced cell necrosis and prevented apoptosis after OGD. L-Carnitine application significantly reduced the number of cells that died, as assessed by the PI/AO ratio, and also reduced ROS release in the OGD groups treated with 10 ${\mu}M$ and 100 ${\mu}M$ of L-carnitine compared with the untreated OGD group (P<0.05). The application of L-carnitine at 100 ${\mu}M$ significantly decreased cytotoxicity, LDH release, and inhibited apoptosis compared to the untreated OGD group (P<0.05). Conclusion: L-Carnitine has neuroprotective benefits against OGD in rat primary cortical neurons in vitro.

서태평양 해저산 해역에서 수괴와 무기영양염 거동에 기초한 동적 수층환경 특성 (Dynamic Characteristics of Water Column Properties based on the Behavior of Water Mass and Inorganic Nutrients in the Western Pacific Seamount Area)

  • 서태평양에 위치한 해저산 해역($150.2^{\circ}E$, $20^{\circ}N$ 부근)에서 수층 환경의 동적 특성을 파악하고자 수괴 및 용존산소, 무기영양염(질소, 인), 엽록소-a 등과 같은 수층 환경 인자의 거동을 살펴보았다. 2014년 10월에 해저산(OSM14-2)을 중심으로 동-서 및 남-북 방향으로 총 9개의 정점에서 CTD system을 이용하여 물리 화학적 자료를 획득하였다. 수온-염분 도표로부터 연구 해역에서 파악된 수괴는 표층에서 북태평양 열대수와 수온약층수, 중층에서 북태평양 중층수 그리고 저층에서 북태평양 심층수로 구분되었다. 용존산소 농도가 낮은 최소층(평균 $73.26{\mu}M$)은 산소 결핍 환경(dysoxic<$90{\mu}M$)으로 연구 해역 전반에 걸쳐 수심 700~1,200 m 사이에 분포하였다. 아질산염+질산염과 인산염으로 대표되는 무기영양염은 표면혼합층 내에서 빈영양 환경을 보인 후 수심 증가에 따라 점차적으로 증가해 용존산소 최소층에서 최대 농도를 나타냈으며, N:P ratio(13.7) 결과로부터 연구 해역은 식물플랑크톤이 성장하기에 질소 성분이 제한된 환경으로 파악되었다. 해저산을 중심으로 동-서 및 남-북 정점 라인에서 환경 인자의 수직 분포는 서쪽과 남쪽 해역에서 저층수 유입에 의한 영향으로 수심 500 m 부근에서 그리고 수심 2,500 m 이하의 저층 내에서도 서쪽 해역과 남쪽 해역에서 반대 해역과 비교해 환경 인자의 농도가 다르게 분포하였다. Redfield ratio(N:P=16:1)을 이용하여 구해진 Excess N 값은 연구 해역 전반에 걸쳐 음의 값을 보여 질소 제거 기작이 우세한 환경임을 나타냈으며, 서쪽 해역과 남쪽 해역 저층에서 상대적으로 높은 값이 관측되었다. 이러한 결과들은 해저산의 지형적인 특성이 저층 해류 순환에 영향을 미치고 이는 수층 환경 인자들의 거동을 결정하는데 중요하게 작용함을 지시한다.

시설원예용 온풍난방기내의 온도분포에 관한 연구 (I) - 난방유 사용시 온도특성 - (Temperature Distributions inside a Space Heater for Greenhouse (I) - Temperature Characteristics with Heating Oil -)

  • Air and flue gas temperature distributions in the space heater for greenhouse were measured to develop a thermal design technology for the space heater. Also, the characteristics of the fan supplying air to the space heater were investigated. The temperature of the flue gas inside the flue gas tube was linearly decreased as the lenght of than those of the flue gas with the oxygen concentration of 8.25% at the last exit of the second flue gas tube. Thus, the operating efficiency of the space heater could be increased with low air ratio decreased exhausting gas temperature and saved the energy consumption with decreased excess air flow. The temperature of the air supplied by fan was increased slowly around the first flue gas tube, meanwhile, increased sharply around the second flue gas tube due to large LMTD (Logarithmic Mean Temperature Difference) at the first flue gas tube than which of the second flue gas tube.

부직포 여과막 생물반응조에서의 폐활성슬러지 감량화 (Minimization of Excess Activated Sludge in Nonwoven Fabric Filter Bioreactor)

  • 생물학적 폐수처리과정에서 폐활성슬러지가 적게 발생하도록 하는 방법 중 현재 그 효과가 가장 확실한 것은 반응조 내의 F/M비를 낮게 유지하는 방법이다. 본 연구에서는 반응조 내의 미생물 농도를 높게 유지함으로써 F/M비를 낮게 유지할 수 있는 부직포 여과막 생물반응조에 COD농도가 약 300 mg/L이고 SS를 함유하지 않는 합성폐수를 단속적으로 주입하는 실험을 반복하여 폐활성 슬러지의 감량화 가능성을 파악하였다. 실험 결과, 반응조내의 MLSS농도가 최고 31,010 mg/L까지 증가함으로써 F/M비는 최저 0.02 g COD/g MLSS-day까지 감소하였다. 그러나 MLSS의 내생분해계수 및 산소섭취율을 측정한 결과 MLSS농도가 증가함에 따라 MLSS의 활성도는 감소하는 것으로 나타났다. 폐수주입기간 중 반응조내의 MLSS증가 및 전체 실험기간의 물질수지에 근거한 평균 미생물 성장계수 값이 각각 0.148 및 0.139 g MLSS/g COD의 낮은 값을 보임으로써 본 연구에서 채택된 부직포 여과막 생물반응조가 잉여슬러지 발생량을 감량화시키는 데 효과적임을 알 수 있었다.

Vitrification of Simulated Combustible Dry Active Wastes in a Pilot Facility

  • In order to evaluate and finally optimize the vitrification condition for combustible dry active waste (DAW), dust and gas generation characteristics were investigated for PE, cellulose, and mixed waste Tests were conducted by varying the operation variables such as melter configuration, excess oxygen amount, and waste feeding rate. Results showed that dust generation characteristics were affected by the operation parameters and the melter's configuration is the dominant one. For all tested DAWs, dust generation was reduced by increasing the waste feeding rate and the excessive oxygen amount in the melter. Among waste types, dust amount was decreased by the order of mixed wastes, PE, and cellulose. Other parameters such as temperature variation and operation time have also affected the dust generation. The optimum condition for the DAW vitrification was determined as the melter's configuration equipped for minimizing the waste dispersion with 20 kg/h of waste feeding rate and 100% of excessive oxygen supply. CO gas concentration in the off-gas was immediately influenced by the combustion state in the melter, but showed similar trend as the dust generation. For the NOx production during the vitrification process, thermal NOx, which is generated from the Post Combustion Chamber (PCC), rather than fuel NOx was assumed to be dominant. The gas cleaning of efficiencies of the PCC, wet scrubber, and Selective Catalytic Reduction system (SCR) were found to be high enough to keep the concentration of pollutants (CO, NOx, SOx, HCI) in the stack below their relevant emission limits.
