Effect of Growth Inhibition in Hep3B cell and HeLa cell by treatment of Euonymus alatus(Thunb.) Sieb extracts (귀전우(鬼箭羽)(Euonymus alatus(Thunb.) Sieb)가 간암세포(肝癌細胞)(Hep3B)와 자궁암세포(子宮癌細胞)(HeLa)의 성장억제(成長抑制)에 미치는 영향(影響)에 관한 연구(硏究))

  • Euonymus alatus(Thunb.) Sieb has been used for cancer theraphy such as Liver cancer, esophagus cancer, stomach cancer, and uterine cancer in folk. Eupolyphaga simemsis(ES) extracted from Euonymus alatus(Thunb.) Sieb increases HDL-C/TC ratio and LCAT activity. TGF-${\beta}$ and Insulin has been Knoen as factors to induce the supression of proliferation of Hep3B, Human hepatoblastoma cell. Plasma HDL3-C involved in carbohydrate methabolism in the liver was maintained in low level by Euonymus alatus(Thunb.) Sieb. The low level of plasma HDL3-C suppresses the transition from normal liver to Fat liver. Finally, this resuly in suppression of cancer in liver. In this study, Euonymus alatus(Thunb.) Sieb showed higher cell toxicity in hepatoblastoma cell(Hep3B) compared to the uterine cancer cell(HeLa). This means Euonymus alatus(Thunb.) Sieb seems not to affect in the all kinds of cancer. In future, we will study whether Euonymus alatus(Thunb.) Sleb have an effect on the liver cancer induced by virus and hepatocellular carcinoma. Also, the mechanims of suppression of cell proliferation in HCC need to investigated.

  • Euonymus alatus (EAE) and Euonymus alatus for. ciliatodentatus (Franch. & Sav.) Hiyama (EACHE) belong to the Celastraceae family and are one of the medicinal herbs used in traditional medicine as a therapeutic agent for blood circulation diseases. In this study, we prepared leaves methanolic extract of EAE (L-EAE), twigs methanolic extract of EAE (T-EAE), leaves methanolic extract of EACHE (L-EACHE) and twigs methanolic extract of EACHE (T-EACHE), and compared their anti-inflammatory activities. To analyze the anti-inflammatory activities, Raw 264.7 cells were used, since they are immature-macrophages and easily matured by lipopolysaccharide (LPS) stimulation. All extracts showed anti-inflammatory activities in the activated Raw 264.7 cells. That is, we showed that L-EACHE and T-EACHE are potent inhibitors of the production of nitric oxide and pro-inflammatory cytokines. These results are expected to serve as a guide for future studies on the ability of Celastraceae family to inhibit acute and chronic inflammatory diseases.

Vegetation Structure of Mountain Ridge from Miwangjae to Cheonwhangbong in Weolchulsan National Park (월출산국립공원 미왕재-천황봉 구간의 능선부 식생구조)

  • To investigate the vegetation structure of mountain ridge from Miwangjae to Cheonwhangbong, 11 plots($2000m^2$) set up with random sampling method were surveyed. Three groups of Quercus mongolica community, Quercus mongolica-Styrax japonica community, Q. mongolica-Pinus densiflora community were classified by cluster analysis. Quercus mongolica was a major woody plant species in the ridge area from Miwangjae to Cheonwhangbong, and Styrax japonica and Pinus densiflora was partly occupied. High positive correlations was proved between Prunus sargentii and Euonymus alatus for. ciliato-dentatus; Quercus serrata and Euonymus alatus; Sorbus alnifolia and Euonymus alatus, Magnolia sieboldii; Acer pseudo-sieboldianum and Euonymus alatus for. ciliato-dentatus; Rhododendron schlippenbachii and Euonymus alatus for. ciliato-dentatandus; Euonymus alatus and Lindera obtusiloba, Castanea crenata, Lindera erythrocarpa; Lespedeza bicolor and Rhododendron schlippenbachii; Castanea crenata and Magnolia sieboldii; Magnolia sieboldii and Lindera erythrocarpa, and relatively high negative correlations was proved between Rhododendron schlippenbachii and Euonymus alatus; Lespedeza bicolor and Castanea crenata; Stephanandra incisa and Magnolia sieboldii; Rhododendron yedoense var. poukhanense and Lindera erythrocarpa; Euonymus alatus for. ciliato-dentatus and Magnolia sieboldii; Fraxinus sieboldiana and Sapium japonicum; Prunus sargentii and Lespedeza maximowiczii; Lespedeza maximowiczii and Lindera erythrocarpa. Species diversity(H') of investigated groups was ranged $2.3225{\sim}2.7721$, and it was relatively low value compared to that of mountain ridge area of other national parks.

Quality Characteristics and Optimization of Rice Pound Cake prepared with Euonymus alatus by Using Response Surface Methodology (반응표면분석법을 이용한 혼잎나물 첨가 쌀 파운드케이크의 품질 특성 및 최적화)

  • This research was studied to optimize the recipe of rice pound cake with two concentrations of Euonymus alatus and grape seed oil, using central composite design (CCD). The mixing condition of rice pound cake was optimized by subjecting it to sensory evaluation and mechanical and physicochemical analysis, using response surface methodology (RSM). The results of the mechanical and physicochemical analysis showed significant values for color (lightness, redness, yellowness), texture (hardness, springiness, chewiness, gumminess, cohesiveness), loss rate, volume, specific volume, sweetness, saltiness, moisture content and pH (p<0.05). The results of the sensory evaluation showed significant values for color, flavor, taste, softness, appearance and overall quality (p<0.05). As a result, the optimized compounding ratio was found to be 4.28 g of Euonymus alatus and 33.18 g of grape seed oil.

A study of antiproliferative effect by Euonymus alatus (Thunb.) Sieb water-extract on SKBR3 human breast cancer cell line (귀전우(鬼箭羽) 물 추출물에 의한 유방암 세포주 증식억제에 관한 연구)

  • Purpose : 이 연구는 SKBR3 인간 유방암 세포주에 대한 Euonymus alatus (Thunb.) Sieb 추출물 (CWE) 의 증식억제, 항산화 작용 및 세포사 유발 효과를 검토하기 위해 이루어 졌다. Methods : SKBR3 세포주는 48시간 동안 다양한 농도 ($0-40\;{\mu}g/ml$)의 CWE를 첨가하면서 배양되었고, 세포의 생존 비율은 MTT 배양을 통해서 평가하였다. 또한 CWE의 증식억제 효과는 유방암 세포주의 세포사와 관련되어 있음을 형태학적인 변화와 올리고뉴클레오솜 DNA 분절을 통해 확인하였다. Results : CWE의 50%에서 효과를 나타내게 하는 약물농도인 $ED_{50}$ (effective dose 50%)은 $9.3+2.2{\mu}g/ml$이며, 약물의 농도에 의존하여 세포의 증식을 억제시켰다. 아울러, 다양한 농도와 배양시간에서 CWE가 ROS 생산을 억제하는 것을 밝힐 수 있었다. 따라서 이러한 작용과 항암예방효과는 농도와 노출 시간에 의존하였다. Conclusion : 이러한 관찰을 통해 Euonymus alatus (Thunb.) Sieb의 열수 추출물은 SKBR3 인간 유방암 세포주에 대해 강한 증식억제 효과와 강력한 항산화효과 및 세포사의 유발 효과를 가지는 것으로 인식할 수 있다.

Isolation and Characterization of α-glucosidase Inhibitors from Euonymus alatus (화살나무(Euonymus alatus)로 부터 α-glucosidase 저해 물질의 분리 및 동정)

  • ${\alpha}$-glucosidase inhibitory compounds (1-4) in a 50% methanol extract of Euonymus alatus were isolated by activity-based fractionations and the structures determined on the basis of chemical and spectral characterization techniques such as $^1H$ and $^{13}C$ nuclear magnetic resonance spectroscopy, $^1H-^1H$ correlation spectroscopy (COSY), and heteronuclear multiple bond correlation (HMBC). The compounds 1-4 belong to flavonols and exhibited potent inhibitory activities against ${\alpha}$-glucosidase, with $IC_{50}$ values of 25.3, 17.1, 47.3, and $35.1{\mu}M$, respectively. All the isolated compounds were more potent than the positive control acarbose. This is the first report describing the potential hypoglycemic effect of Euonymus alatus through ${\alpha}$-glucosidase inhibition and identification of its active components.

Inhibitory Effect of Rosmarinic acid Extrcted from Euonymus Alatus on Cyclooxygenase-2

  • Objectives and methods : Previous mechanistic studies suggest the cyclooxygenase-2 (COX-2) inhibitors represent the good candidates against tumor progression. MeOH extract of the stem barks of Euonymus alatus induced the strong inhibition of COX-2. A phenolic compound responsible for the anti- COX-2 known to involve in tumor adhesion and invasion has been studied through the methanol extracts. The compound, rosmarinic acid (ROS-A) was an ester of caffeic acid and 3,4-dihydroxyphenyllactic acid. ROS-A showed a strong inhibitory effect of COX-2 activity in a concentration-dependent manner. Then we have measured the IL-1${\beta}$, IL-6 and TNF-${\alpha}$ production related the immune regulation, induction of inflammatory related genes. Results and Conclusions :Hep3B cells produce proinflammatory cytokines of IL-1${\beta}$, IL-6 and TNF-${\alpha}$ while ROS A inhibited the cytokines production. Since IL-1${\beta}$, IL-6 and TNF-${\alpha}$ need the transcription factors such as nuclear factor- ${\kappa}$B (NF-${\kappa}$B) and activated protein-1 (AP-1), we measured the transcription factors. ROS-A inhibited the activation of p65, p50, c-Rel subunits of NF-${\kappa}$B and AP-1 transcription factors. These findings indicate that ROS A from the stem bark of E. alatus inhibits proliferation in metastatic cancer cells. It was suggested that stem barks of E. alatus could be suitable for anti-cancer drugs.

Dihydrobenzofuran Neolignans Isolated from Euonymus alatus Leaves and Twigs Attenuated Inflammatory Responses in the Activated RAW264.7 Macrophage Cells

  • Anti-inflammatory effects of dihydrobenzofuran neolignans isolated from Euonymus alatus leaves and twigs were evaluated in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophage cells. Six neolignans, (+)-simulanol (1), (+)-dehydrodiconiferyl alcohol (2), (-)-simulanol (3), (-)-dehydrodiconiferyl alcohol (4), (+)-dihydrodehyrodiconiferyl alcohol (5), threo-buddlenol B (6) effectively inhibited the production of nitric oxide (NO) induced by LPS, and the activity of iNOS. (-)-dehydrodiconiferyl alcohol (4), which showed the most potent inhibitory activity, attenuated the activity of iNOS enzyme and also the expression of iNOS and COX-2 proteins. The subsequent production of pro-inflammatory cytokines, interleukin-$1{\beta}$, interleukin-6, tumor necrosis factor-${\alpha}$ and prostaglandin E2 were also inhibited by the pretreatment of RAW264.7 cells with (-)-dehydrodiconiferyl alcohol (4). These neolignans are thought to contribute to anti-inflammatory effects of E. alatus, and expected to be potential candidates to prevent/treat inflammation-related diseases.

  • Objective : Euonymus alatus (Thunb.) Sieb (EA) is a traditional Korean herbal medicine, commonly used to treat tumors in Korea and China for centuries. Several earlier studies have indicated that EA exhibits anti-tumor properties, but its mechanism remains to be elucidated. In this study, we evaluated the molecular mechanism of EA in a human uterine leiomyomal smooth muscle cell (ULSMC) line. Methods : This study was evaluated by: (a), morphological changes by using acridine orange/ethidium bromide staining; (b), DNA fragmentation by TdT-mediated dUTP nick end labeling (TUNEL); and (c), sub-G1 cell analysis. Results : This study observed that EA treatment caused apoptotic cell death and depletion of intracellular glutathione (GSH) and that reduction of mitochondrial membrane potential was found to be involved in the initiation of apoptosis by EA. Conclusion : This results show that EA exerted clear cytotoxic effects and strongly inhibited the proliferation of ULSMC.

Inhibitory activity of Euonymus alatus against alpha-glucosidase in vitro and in vivo

  • The major goal in the treatment of diabetes mellitus is to achieve near-normal glycemic control. To optimize both fasting blood glucose and postprandial glucose levels is important in keeping blood glucose levels as close to normal as possible. ${\alpha}-Glucosidase$ is the enzyme that digests dietary carbohydrate, and inhibition of this enzyme could suppress postprandial hyperglycemia. The purpose of this study was to test the inhibitory activity of methanol extract of Euonymus alatus on ${\alpha}-glucosidase$ in vitro and in vivo to evaluate its possible use as an anti-diabetic agent. Yeast ${\alpha}-glucosidase$ inhibitory activities of methanol extract of E. alatus were measured at concentrations of 0.50, 0.25, 0.10, and 0.05 mg/ml. The ability of E. alatus to lower postprandial glucose was studied in streptozotocin-induced diabetic rats. A starch solution (1 g/kg) with and without E. alatus extract (500 mg/kg) was administered to diabetic rats by gastric intubation after an overnight fast. Plasma glucose levels were measured at 30, 60, 90, 120, 180, and 240 min. Plasma glucose levels were expressed in increments from baseline, and incremental areas under the response curve were calculated. Extract of E. alatus, which had an $IC_{50}$ value of 0.272 mg/ml, inhibited yeast ${\alpha}-glucosidase$ activity in a concentration-dependent manner. A single oral dose of E. alatus extract significantly inhibited increases in blood glucose levels at 60 and 90 min (p<0.05) and significantly decreased incremental response areas under the glycemic response curve (p<0.05). These results suggest that E. alatus has an antihyperglycemic effect by inhibiting ${\alpha}-glucosidase$ activity in this animal model of diabetes mellitus.
