Effects of the Dietary Mixture of Eucommia ulmoides Oliver on Osteoporosis (두충[Eucommia ulmoides Oliver] 조성물이 골다공증에 미치는 효과)

  • Inhibitory effects of Eucommia ulmoides Oliver against osteoporosis were studied with ovaryectomized rats. The diet mixture A of calcium, vitamin D$_3$, and vitamin K$_1$and the additional mixture B with extract of Eucommia ulmoides Oliver were fed for 7 months to ovaryectomized rats. The urine content of pyridinoline and deoxypyridinoline was used as bone biochemical markers. Concentrations of deoxypyridinoline of ovaryectomized rats fed with the mixture A and B decreased 9.0% and 26.9%, respectively, when they were compared to the ovaryectomized control. The trabecular bone areas of proximal tibias of ovaryectomized rats were measured and compared to the ovaryectomized control. The areas fed with the mixture A and B increased 9.9% and 42.0%, respectively, that proved the inhibitory effect of Eucommia ulmoides Oliver against osteoporosis.

Effects of Eucommia ulmoides Oliver Extract on Inhibition of β-hexosaminidase and Keratinocyte Differentiation (β-hexosaminidase 분비 억제 및 각질형성세포 분화에 대한 두충(Eucommia ulmoides Oliver) 추출물의 효과)

  • In this study, Eucommia ulmoides Oliver extracts was studied in order to see any effects on the ${\beta}$-hexosaminidase release suppression of RBL-2H3 cells and on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) related to the recovery of HaCaT keratinocyte skin barrier. Results showed that Eucommia ulmoides Oliver extracts reduced ${\beta}$-hexosaminidase release in RBL-2H3 cells and increased the effects of Eucommia ulmoides Oliver extract on the expression of filaggrin, transglutaminase-1 (TGase-1) and cornified cell envelope (CE) in HaCaT keratinocytes. Taken together, these results suggested that Eucommia ulmoides Oliver extract may be applicable for keratinocyte differentiation.

Effects of Eucommia Ulmoides Oliver and Treadmill Exercise on the Osteoporosis of Rats Caused by Glucocorticoid Induction (두충 추출물 투여와 트레드밀 운동이 골다공증 유발 백서의 골대사에 미치는 영향)

  • This study classified 40 of 10 weeks-old male Sprague Dawley rats which have osteoporosis caused by glucocorticoid(GC) induction into four groups of 10 rats and made the subjects medicated them Eucommia ulmoides Oliver and participate in treadmill exercise. Group I was non-treatment after GC induction; Group II was treadmill exercise after GC induction; Group III was Eucommia ulmoides Oliver feeding after GC induction; Group IV was treadmill exercise and Eucommia ulmoides Oliver feeding after GC induction. In the bone mineral density and bone strength test, there was a statistically significant difference between the group I, II, III compared to group IV(p<0.05). In the osteocalcin level as one of bone formation indexes, there was a statistically significant difference between the group I, II, III compared to group IV(p<0.05). In respect to the calcium there was a statistically significant difference between group I, II, III compared to group IV(p<0.05). In respect to histological findings, the group IV showed mild femur disruption and lytic change on femur section. The above results suggests that Eucommia ulmoides Oliver medicated and treadmill exercise is effective to prevention and treatment of osteoporosis.

Inhibitory Effects of Total Extract and Flavonols from Hardy Rubber Tree (Eucommia ulmoides Oliver) Leaves on the Glycation of Hemoglobin

  • Our previous study demonstrated that aqueous ethanolic extract of hardy rubber tree (HRT; Eucommia ulmoides Oliver) leaves potently inhibited glycation, and that two known flavonols and one new flavonol were the active compounds in protein glycation in vitro using the model system of bovine serum albumin and fructose. The aim of this study was to examine the effectiveness of HRT extract and its flavonols against the glycation of hemoglobin (Hb) in primary cultured rat erythrocytes. The extract at $40-400\;{\mu}g/mL$ and the flavonols at $0.2-20\;{\mu}g/mL$ significantly inhibited Hb glycation in a concentration-dependent manner. They were more effective than aminoguanidine, a well-known inhibitor of glycation. Three flavonols seemed to be responsible for the inhibitory activity of the extract, furthermore, the extract of HRT leaves showed advantages over single isolated flavonols in the inhibition of Hb glycation.

Studies on the General Pharmacological Activities of Eucommia ulmoides Oliver (두충나무의 일반약리활성(一般藥理活性) 연구(硏究))

  • The pharmacological activities on water extracts of Cortex, Ramulus and Folium from the Eucommia ulmoides Oliver were studied, and the following results were observed. The vasodilative action on the rabbit ear blood vessel was recognized in the Cortex, Ramulus and Folium. The spontaneous motility and the contraction induced by Ach., $BaCl_2$ and histamine in the isolated ileum of mice were suppressed and inhibited in the Ramulus only. On the contrary, the analgesic and anti-inflammatory activities were noted in the Cortex and Folium. Furthermore, the hypotensive effect was recognized in the Folium. The diuretic, bile secretory and anti-fatigue effects were shown in the Cortex and Folium.

  • The anti-diabetic effects of the methanol extracts of Cortex, Ramulus and Folium part of Eucommia ulmoides Oliver were investigated by the experimental diabetic model in rats. The increased blood glucose level in the diabetic rats induced by alloxan and streptozotocin was significantly lowered and a marked increase in body weight was shown with the treatments of the extracts of Cortex and Folium. The blood insulin level lowered by streptozotocin pretreatment was significantly recovered with the treatment of the extract of Folium. In glucose tolerance test, the extracts of Cortex and Folium displayed a significant inhibitory effect on the glucose level. The extracts of Cortex and Folium also caused a marked inhibition in increased blood pressure in the diabetic rats induced by alloxan.

Effects of Dietarv Supplementation of Eucommia ulmoides Oliver Leaves on Performance and Meat Quality in Broiler Chicks (두충잎의 첨가가 육계의 성장 및 육질에 미치는 영향)

  • The present study was conducted to investigate the effects of dietary Eucommia ulmoides Oliver leaves(EUOL) on the growth performance and meat quality of broiler chicks. The dietary treatments consisted of the control diet(corn-soybean meal basal), and EUOL 2, 4, and 6% diets. The latter three diets were prepared by adding 2, 4, and 6% EUOL to the control diet. Ninety-six Arbor Acre male broiler chicks were randomly allotted to 16 pens; four pens per treatment and 6 birds per pen. The growth performance and carcass weight of broiler chicks fed EUOL diets were not significantly different(P>O.05) from the control birds. Also, the muscle color degrees(L*a*b *)in brolier chicks were similar to each other. The blood concentrations of protein, sugar and glutamic-pyruvic transarninase(GPT) were not different(P>O.05) between control and treatment groups. The blood total cholestrol content was significantly(P<0.05) decreased by the addition of EUOL. However, the addition of EUOL did not change the muscle components significantly. The bone hardness of broiler chicks fed EUOL was significantly(P<0.05) higher than that of control chicks.

Antioxidative activities of ethanolic extracts of Du-zhong (Eucommia ulmoides Oliver) leaf and bark (두충나무(Eucommia ulmoides Oliver) 잎과 껍질의 에탄올 추출물의 항산화 활성)

  • Antioxidative activities of 50% ethanolic extracts from Du-zhong (Eucommia ulmoides Oliver) leaf and bark were investigated. Yields of the leaf and bark extract were 8.1±0.31 and 17.4±0.89%, respectively. Polyphenol contents of the leaf and bark extract were 64.1±3.35 and 42.4±2.38 ㎍ gallic acid equivalents/mg, respectively. Flavonoid contents of the leaf and bark extract were 24.0±3.15 and 36.7±3.18 ㎍ quercetin equivalents/mg, respectively. As concentration of the leaf and bark extract increased, their antioxidative activities proportionally increased. EC50 values of the leaf and bark extract for cation radical scavenging were 560.6±17.65 and 1,357.4±8.45 ㎍/mL, respectively. EC50 values of the leaf and bark extract for free radical scavenging were 574.2±14.70 and 2,103.1±108.59 ㎍/mL, respectively. EC50 values of the leaf and bark extract for ferric reducing antioxidant power were 319.9±13.42 and 705.9±26.08 ㎍/mL, respectively. EC50 values of the leaf and bark extract for nitrite scavenging were 2,329.2±35.11 and 5,467.6±243.92 ㎍/mL, respectively. In the presence of 74.8 ㎍/mL of the leaf extract and 177.2 ㎍/mL of the bark extract, linoleic acid peroxidation was inhibited by 70.0 and 79.1%, respectively. The Du-zhong leaf extract possessed higher antioxidative activities than its bark extract.

ACE Inhibitory Lignan Glycosides Isolated from Eucommia ulmoides Oliver (두충으로부터 분리한 Iignan glycoside 의 ACE 활성 억제)

  • To evaluate a potential possibility of Eucommia ulmoides Oliver as a functional food, ACE (angiotensin converting enzyme) inhibitory activities of leaf, bark, stem and 4 compounds isolated from E. ulmoides were tested. The 4 compounds were isolated and purified by silica gel column chromatography, thin layer chromatography and reverse phase column chromatography. Compound I was pinoresinol-4,4'di-O-${\beta}$-D-glucoside (PG) and compound II was dehydrodiconiferyl alcohol 4,${\gamma}$'-di-O-${\beta}$-D-glucopyranoside (DAG) originating from Eucommial Cortex. The highest amount of PC was present at raw and roasted bark as 135.13 mg% and 163.67 mg%, and the highest amount of DAG was present at raw and roasted leaf as 117.93 mg% and 133.93 mg% respectively. In an ACE inhibition test, 10 mg/ml of roasted leaf, raw and roasted bark extracts of E. ulmoides Oliver were 77.49%, 75.72% and 75.36% respectively, and 10mg/ml of PC and DAG were shown to be 78.51 and 81.20% respectively. $IC_{50}$ values of PG and DAG were 0.6${\pm}$0.2 and 0.5${\pm}$0.2 mg/ml respectively.
