Carrageenan Content and its Chemical Composition of Eucheuma cottonii from the Coast of Philippine (Philippine산(産) Eucheuma cottonii의 carrageenan 함량(含星)과 그 성상(性狀)에 대하여)

  • One species of Rhodophyceae namely Eucheuma cottonii from the coast of Philippine was analyzed with respect to the content of carrageenan and such chemical characteristics as the content of sulphate and 3,6-anhydro-D-galactose and its solubility in potassium chloride solution. In addition, the same chemical properties were tested in the fractions separated by the different concentrations of potassium chloride. In comparison of the results of carrageenan in Eucheuma cottonii samples from Philippine and Chondrus ocellatus samples from Korea, carrageenan content in Eucheuma cottonii was higher than that of Chondrus ocellatus. Both samples showed more than forty-five percent carrageenan content. The Eucheuma cottonii carrageenan showed a higher 3,6-anhydro-D-galactose than the Chondrus ocellatus carrageenan. The sulphate content was higher in Chondrus ocellatus than Eucheuma cottonii. In fractionation of carrageenan by the solubility methods using potassium chloride solution, the yield of Eucheuma cottonii was highest in fraction I, fraction III was next and fraction II was the lowest.

Production of Biosugar from Red Macro-algae Eucheuma cottonii using Acid-hydrolysis (Eucheuma cottonii로부터 산 가수분해를 통한 biosugar 생산)

  • In this study, biosugar production by the red-algae Eucheuma cottonii was investigated using dilute sulfuric acid-catalyzed hydrolysis and data analysis by response surface methodology. This approach yielded 25.8 g/l total reducing sugar under the conditions of $160.1^{\circ}C$, 1% (v/v) sulfuric acid, and 13.1 min. The sugar concentration showed a linear inverse correlation with the combined severity factor (CSF) of the reaction conditions. CSF was calculated as $log(t{\cdot}e{xp}[(T_H-T_R)/14.75])-pH$, where t is the coupling reaction time, $T_H$ is the target temperature, and $T_R$ is the reference temperature ($100^{\circ}C$). In addition, levulinic acid production showed a linear positive correlation with CSF. E. cottonii may represent a useful feedstock for sugar production in the field of bioenergy.

Extract from Eucheuma cottonii Induces Apoptotic Cell Death on Human Osteosarcoma Saos-2 Cells via Caspase Cascade Apoptosis Pathway (Eucheuma cottonii 추출물에 의한 인체 골육종암 Saos-2 세포의 자가사멸 유도)

  • Osteosarcoma (OS) is the most common and malignant bone tumors. Although many types of resection surgery and experimental agents were developed, median survival and clinical prognosis are poorly investigated. Recently, several researches have reported that Eucheuma cottonii has potent as protective effects of coal dust-induced lung damage via inhibition of malondialdehyde (MDA) and oxidative stress in bronchoalveolar lavage fluids (BALF). However, anti-cancer effects and specific molecular mechanism of extract from Eucheuma cottonii (EE) has not been clearly studied yet. This study evaluated that anti-cancer potential of EE in human osteosarcoma Saos-2 cells. EE indicated cytotoxicity on Saos-2 cells in a dose-dependent manner. Morphological degradation and nucleic condensation were also observed under the EE treatment. However, it did not significantly affect on non-cancerous kidney HEK-293 cells under the same concentration which is shown cytotoxicity on Saos-2 cells. The phosphorylation of Fas-Associated Death Domain (FADD) and expression of cleaved caspase-8, -7 and -3 were upregulated in a dose-dependent manner. In immunofluorescence staining, expression level of Fas and cleaved PARP were upregulated by EE treatment. Furthermore, treatment of EE induces upregulation of sub G1 phase by flow cytometry analysis. The results demonstrated that EE has a therapeutic potential against osteosarcoma via FADD mediated caspase cascade apoptosis signal pathway.
