Quality Changes of 'Yeobong' Strawberry with CA Storage Conditions (CA저장조건에 따른 '여봉' 딸기의 품질변화)

  • 'Yeobong' strawberries were stored at $2^{\circ}C$ under controlled atmosphere (CA) containing the combinations of $O_2{\;}(3.8%)$ and $CO_2{\;}(10,{\;}15,{\;}20%)$ and air as control. Atmospheres of $3%{\;}O_2+15%{\;}CO_2$ and $8%{\;}O_2+15%{\;}CO_2$ reduced respiration and ethylene production rates. Fruits kept under CA conditions of 10% and 15% $CO_2$ were harder than those stored under 20% $CO_2$ and air. The CA conditions of 10% or 15% $CO_2$ maintained approximately 80% of vitamin C for 24 days. Redness were increased and then decreased, but the changing trends were not clear among the storage conditions. Anthocyanin contents in $3%{\;}O_2+15%{\;}CO_2$ and $8%{\;}O_2+15%{\;}CO_2$ were slightly increased for 16 days and then decreased thereafter, while anthocyanin content in air was rapidly increased for 8 days. After 16 days of storage, off-flavor were perceived in all CA storage. And strawberries stored in 20% $CO_2$ conditions were unacceptable after 20 days. The 20% $CO_2$ significantly affected off-flavor of strawberries, but there was no significant difference between 10% $CO_2$ and 15% $CO_2$ conditions. Ethanol which is in relation to off-flavor was higher with elevated $CO_2$ levels. Although CA conditions under $3%{\;}O_2+15%{\;}CO_2$ was effective in delaying the quality changes, there was high ethanol content compared to $8%{\;}O_2+15%{\;}CO_2$ condition. Strawberry kept under $8%{\;}O_2+15%{\;}CO_2$ was maintained the shelf-life for 24 days and the condition prolonged more $4{\sim}8$ days and 12 days than the other CA conditions and air, resectively.

Current Research Trend of Postharvest Technology for Chrysanthemum (국화 수확 후 관리기술의 최근 연구 동향)

  • Chrysanthemum is a cut flower species that normally lasts for 1 to 2 weeks, in some cases 3-4 weeks. This has been attributed to low ethylene production during senescence. Reduction in cut flower quality has been attributed to the formation of air embolisms that partially or completely blocks the water transport from the vase solution to the rest of the cut flower stem, increasing hydraulic resistance which may cause severe water stress, yellowing, wilting of leaf, and chlorophyll degradation. Standard type chrysanthemum can be harvested when buds were still tightly closed and then fully opened with the simple bud-opening solution. Standard type chrysanthemum can also be harvested when the minimum size of the inflorescence is about 5-6 cm bud which opened into the first flower full-sized flower. While spray varieties can be harvested when 2-4 most mature flowers have opened (40% opening). Cut flowers are sorted by stem length, weight, condition, and so on. Standard chrysanthemum is 80 cm length for standard type and 70cm for spray type. Pre-treatment with a STS, plant regulator such as GA, BA, 1-MCP, chrysal, germicide, and sucrose, significantly improved the vase life and quality of cut flowers. It is well established that vase solutions containing sugar can improve the vase life of cut chrysanthemum. Chrysanthemum is normally packed in standard horizontal fiberboard boxes. Chrysanthemum should normally be stored at $5{\sim}7^{\circ}C$. Precooling resulted in reduction in respiration, decomposition, and transpiration activities as well as decoloration retardation. There was significant difference between "wet" storage in 3 weeks and "dry" storage in 2 weeks. In separate pulsing solution trials, various germicides were tested, as well as PGRs to maintain the green color of leaves and turgidity. Prolonging vase life was attained with the application of optimal solution such as HQS, $AgNO_3$, GA, BA and sucrose. This also retarded senescence in leaves of cut flower stems. Fresh cut chrysanthemum can be transported using a refrigerated van with $5{\sim}7^{\circ}C$. Increasing consumption and usage of cut chrysanthemum of various cultivars would require efficient transport system, and effective information exchange among producer, wholesaler, and consumer.

Investigation of Viscoelastic Properties of EPDM/PP Thermoplastic Vulcanizates for Reducing Innerbelt Weatherstrip Squeak Noise of Electric Vehicles (전기차 인너벨트 웨더스트립용 EPDM/PP Thermoplastic Vulcanizates 재료설계인자에 따른 점탄성과 글라스 마찰 소음 상관관계 연구)

  • Due to enormous market growing of electric vehicles without combustion engine, reducing unwanted BSR (buzz, squeak, and rattle) noise is highly demanded for vehicle quality and performance. Particularly, innerbelt weatherstrips which not only block wind noise, rain, and dust from outside, but also reduce noise and vibration of door glass and vehicle are required to exhibit high damping properties for improved BSR performance of the vehicle. Thermoplastic elastomers (TPEs), which can be recycled and have lighter weight than thermoset elastomers, are receiving much attention for weatherstrip material, but TPEs exhibit low material damping and compression set causing frictional noise and vibration between the door glass and the weatherstrip. In this study, high damping EPDM (ethylene-propylene-diene monomer)/PP (polypropylene) thermoplastic vulcanizates (TPV) were investigated by varying EPDM/PP ratio and ENB (ethylidene norbornene) fraction in EPDM. Viscoelastic properties of TPV materials were characterized by assuming that the material damping is directly related to the viscoelasticity. The optimum material damping factor (tanδ peak 0.611) was achieved with low PP ratio (14 wt%) and high ENB fraction (8.9 wt%), which was increased by 140% compared to the reference (tanδ 0.254). The improved damping is believed due to high fraction of flexible EPDM chains and higher interfacial slippage area of EPDM particles generated by increasing ENB fraction in EPDM. The stick-slip test was conducted to characterize frictional noise and vibration of the TPV weatherstrip. With improved TPV material damping, the acceleration peak of frictional vibration decreased by about 57.9%. This finding can not only improve BSR performance of electric vehicles by designing material damping of weatherstrips but also contribute to various structural applications such as urban air mobility or aircrafts, which require lightweight and high damping properties.

Optimization of One-Step Dilution Method of Vitrified Bovine IVM/IVF/IVC Blastocysts (초자화 동결된 체외생산 소 배반포기배의 1 단계 융해 방법의 적정화)

  • This study was to establish an effective dilution technique in a vitrification of bovine blastocysts for the field trial. For vitrification, blastocysts were exposed in glycerol (G) and ethylene glycol (EG) mixture in m-DPBS supplemented with 10% FBS. Blastocysts were first exposed to 10% (v/v) G for 5 min, and subsequently were transferred to 10% G plus 20% EG (v/v) for 5 min. Finally, embryos were transferred to 25% G plus 25% EG (v/v) for 30 see and were placed in nitrogen vapor for 3 min, and then were plunged into L$N_2$. At thawing, the straw containing blastocysts was placed in air for 10 sec, and then plunged into a water bath at $25^{\circ}C$ until all ice had disappeared. They were placed in $25^{\circ}C$ and 36$^{\circ}C$ water according to treatment group for different time. Also, in vitro survival was assessed by the re-expansion and hatched rates at 24 hand 48 h postwarming, respectively. The results obtained in these experiments were summarized as follows; 1) In the survival rates of vitrified bovine blastocysts according to different dilution time at thawing, the data of 1 min group (86.6, 56.6%) were higher than those of other treatment groups (2 min; 93.5, 35.4%, 2.5 min; 76.9, 30.7%, 3 min; 88.8, 36.1% and 3.5 min; 83.7, 8.1%). 2) When the in vitro survival of vitrified groups according to different developmental stage was examined at 48 h after thawing using 1 min dilution method, the hatching rates of fast developed embryos (expanded blastocyst: 81.3%: early hatching blastocyst: 86.2%) were higher than that of delayed developed one (early blastocyst: 46.6%). 3) In addition, when the in vitro survival of vitrified groups according to different embryo age was compared, the hatched rates at 48 h after thawing of Day 7 (66.6%) and Day 8 embryos (60.0%) were significantly higher than that of Day 9 embryos (22.7%) (P<0.05). These results demonstrate that vitrified bovine IVM/IVF/IVC blastocysts can be successfully survived in vitro using one-step dilution (1 min) method.

Influence of Elevated CO2 and Air Temperature on Photosynthesis, Shoot Growth, and Fruit Quality of 'Fuji'/M.9 Apple Tree (CO2 및 기온 상승이 '후지'/M.9 사과나무의 광합성, 신초생장 및 과실품질에 미치는 영향)

  • This study was conducted to find out the influence of elevated atmospheric $CO_2$ concentrations and air temperature on photosynthesis and fruit quality of 'Fuji'/M.9 apple trees and to investigate these to the effects of climate change during the last four years (2009-2012). The treatments employed were: 'Ambient' (ambient temperature + ambient $CO_2$ concentration); 'High $CO_2$' (ambient temperature + elevated $CO_2$ concentration); 'High Temp'. (elevated temperature + ambient $CO_2$ concentration); and 'High $CO_2$ + High Temp'. (elevated temperature + elevated $CO_2$ concentration). The elevated temperature plots were maintained at $4^{\circ}C$ higher than ambient air temperature, while the elevated $CO_2$ plots were maintained at 700 ${\mu}mol{\cdot}mol^{-1}$. Annual treatment period was applied from end of April to beginning of November for four years. Results showed that elevated $CO_2$ decreased stomatal conductance and leaf SPAD value, but increased photosynthetic rate, intercellular $CO_2$ concentration (Ci), and starch content of mesophyll tissue. In the vegetative growth, elevated temperature increased total number of shoot and total shoot growth per tree, but elevated $CO_2$ decreased average shoot length. In the fruit quality, elevated $CO_2$ increased soluble solid content, fruit red color, and ethylene production. In conclusion, elevated $CO_2$ increased photosynthetic rate of apples during the early growth, but effect of increased photosynthetic rate due to elevated $CO_2$ was decreased during latter growth stage. Elevated temperature, on the other hand, tended to decrease photosynthetic rate of apples during the early growth, but that tended to increase during latter growth stage. Both elevated $CO_2$ and temperature tended to decrease the degree of decreased photosynthetic rate due to each factor.

Comparison of Developmental Efficiency Following Cryopreservation of Hanwoo Embryos (한우 수정란의 동결보존 후 발달 효율 비교)

  • The cryopreservation of Hanwoo embryos has become an integral part of assisted reproduction in animal. The objective of this study was to assess the effect of The objectives of this study were: (1) to evaluate the influence of bovine embryo developmental stage on in vitro embryo development after freezing, (2) to study the efficiency compared with conventional freezed embryos at different embryo source. For conventional slow-freezing, day 7 or 8 expanded blastocysts were collected. The standard freezing medium was 1.8 M ethylene glycol (EG). Embryos were equilibrated in 1.8 Methylene glycol(EG) with 0.1 M sucrose in Dulbecco's phosphate-buffered saline (D-PBS) supplemented with 0.5% bovine serum albumin. Embryos were then loaded individually into 0.25 ml-straw and placed directly into cooling chamber of programmable freezer precooled to $-7^{\circ}C$, after 2 min, the straw was seeded, maintained at $-7^{\circ}C$ for 8 min, and then cooled to $-35^{\circ}C$ at $0.3^{\circ}C$/min, plunged and stored in liquid nitrogen for at least 3 days. For thawing, the straw containing embryos were warmed in air for 10 see and exposed to $37^{\circ}C$ water for 20 sec. Straws were then removed from $37^{\circ}C$ water. Rates of blastocyst survive and hatched were evaluated at 12 to 48h post-warming. The re-expansion and hatched rates of morula embryos were significantly lower than those obtained for blastocysts and expansion blastocysts (31.6%, 10.5% vs, 68.9%, 22.2% vs, 73.7%, 53.6%, respectively). No differences in re-expansion rates were found between in vivo and in vitro blastocysts. whereas hatched rates was significantly higher (51.2%) in vivo compared with in vitro embryos (18.6%). in conclusion, demonstrate that conventional freezing can be used successfully in cryopreservation of in vitro and in vivo bovine embryos, and that it might be considered for use in commercial programs and embryo preservation.

Study on Development of Canine Oocytes Treated by In Vitro Fertilization and ICSI

  • These study was to investigate the in vitro fertilization and viability of fresh and vitrified oocytes. Also, the developmental capacity of IVF and intracytoplasmic sperm injection (ICSI) oocytes were investigated. Then vitrification was performed with the use of 20% ethylene glycol + 20% DMSO + 0.5 M sucrose + 10% FCS + TCM-199 medium. Vitrification immature oocytes are cultured in vitrification solution for 10 min afterwards transferred to expose at room temperature for 5 min. and transferred to the ice water for 5 min. The oocytes were sealed in a 1.0 mm straw and placed in a $LN_2$ container. Frozen oocytes were rapidly thawed in a water bath at $30{\sim}35^{\circ}C$, and then placed in TCM-199 medium containing 0.5 M sucrose for 5 min each, respectively, at $38^{\circ}C$. After being washed for 2~3 times, using fresh medium the oocytes were cultured in TCM-l99 medium supplemented with 5% FCS at $38^{\circ}C$ in 5% $CO_2$ and air. The normal morphology of fresh and vitrified-thawed oocytes were $87.1{\pm}2.1%$ and $54.8{\pm}2.5%$, respectively. The viability rates of fresh and vitrified-thawed oocytes were $70.0{\pm}2.2%$ and $41.9{\pm}2.6%$, respectively. Viability rates of vitrified-thawed oocytes were lower than that of fresh follicular oocytes (p<0.05). The in vitro maturation rates of fresh and vitrified oocytes were $45.1{\pm}3.6%$ and $28.9{\pm}4.4%$, respectively. The IVF rates of fresh follicular and vitrified-thawed oocytes were 34.00.2% and $20.2{\pm}2.6%$, respectively. The in vitro maturation and fertilization rates of vitrified-thawed oocytes were lower than those of the fresh follicular oocytes (p<0.05). A total of 350 oocytes were fixed and stained after co-incubation with spermatozoa, of which 88 had identifiable nuclear material. After IVF for 20 hrs, $25.1{\pm}3.4%$ of the oocytes found to have been penetrated by spermatozoas. Oocytes were fixed and stained after ICSI, and 105 oocytes contained identifiable nuclear material. After IVF and ICSI for 20 hrs, $34.3{\pm}3.4%$ and $59.0{\pm}2.0%$ of the oocytes were found to have been penetrated by spermatozoas. The developmental rates upon ICSI were significantly higher than those of the IVF method (p<0.05).

Physical Properties and Flame Retardency of Polyhydroxyamides (PHAs) Having Pendant Groups in the Main Chain (주사슬에 곁사슬기를 갖는 폴리히드록시아미드의 물성 및 난연특성)

  • Physical properties and flammability of polyhydroxyamides (PHAs) haying poly (ethylene-glycol) methyl ether (MPEG) and/or dimethylphenoxy pendants were studied by using DSC, TGA, FTIR, pyrolysis combustion flow calorimeter (PCFC), and X-ray diffractometer. The degradation temperatures of the polymers were recorded in the ranges of $276{\sim}396^{\circ}C$ in air. PCFC results showed that the heat release (HR) capacity and total heat release (total HR) values of the PHAs were increased with in-creasing molecular weight of MPEG. In case of M-PHA 2 annealed at $290^{\circ}C$, the values of HR capacity were siginificantly decreased from 253 to 42 J/gK, and 60% weight loss temperatures increased from 408 to $856^{\circ}C$ with an annealing temperature. The activation energy for the decomposition reaction of the PHAs showed in the range of $129.3{\sim}235.1kJ/mol$, which increased with increasing conversion. Tensile modulus of PHAs were decreased as increasing chain of MPEG, and showed an increase more than initial modulus after converted to PBOs.

Microclimate and Crop Growth in the Greenhouses Covered with Spectrum Conversion Films using Different Phosphor Particle Sizes (광전환재 크기가 다른 광전환 필름 피복 온실 내 미기상 및 작물 생육)

  • The objective of this study was to analyze the microclimate and the growth of tomato and lettuce in the greenhouses covered with spectrum conversion films using different phosphor particles sizes. Two spectrum conversion films using phosphor particles larger than $10{\mu}m$ (Micro-film) and smaller than 500 nm (Nano-film) in radius, and poly-ethylene (PE) film were used in double-layered greenhouses as outer coverings. PE films were used as inner coverings in all the greenhouses. Thickness of the films for inner and outer coverings was 0.06 mm. Tensile strength, elongation, and tearing resistance of the Micro- and Nano-films were not different from those of the PE film. Transmittances at a wavelength of 300-1100 nm were a little higher at the Micro-film and lower at the Nano-film than that of the PE film, respectively. Air temperatures at the Micro- and Nano-films were over $2^{\circ}C$ higher than at the PE film, but no significant difference was observed between the two light conversion films. The soil temperature at the Nano-film was $1.5^{\circ}C$ and $3^{\circ}C$ higher than at the Micro- and PE films, respectively. The yields of tomato at the Micro- and Nano-films were 12% and 14% higher than at the PE film, but no significant difference was observed between the two spectrum conversion films. The total soluble solid showed no significant differences among all the films. The yields of lettuces at the Micro- and Nano-films were 27% and 59% higher than at the PE film. Hunter's red (a) value of the lettuce leaf was the highest at the Nano-film. In this experiment, tomatoes requiring high irradiation were better at the Nano film, while lettuce requiring low irradiation better at the Micro film.
