  • Ethinylestradiol (EE)-containing matrix was fabricated with ethylene-vinyl acetate(EVA) copolymer to control the release of the drug, Effect of addition of PEG 400 as receptor solution, the stripping of skin and Azone pretreatment on skin on the permeation of EE through the excised mouse skin was also studied. The permeation rate of EE through the excised mouse skin was affected by the PEG 400 volume fraction. The Azone pretreatment on skin didn't affect on the steady state flux, however, the lag time was shortened. The permeation rate of EE through the stripped skin was much larger than that through the whole skin. It showed that the stratum corneum acts as a barrier of skin permeation. The fact that there is little difference in EE permeation between the intact skin and the stripped skin with EVA membrane shows the permeation of EE through the mouse skin is mainly controlled by the membrane.

의약품의 Solid Lipid Nanoparticle의 제조 및 용출특성 (Preparation and Drug Release Profiles of Solid Lipid Nanoparticles(SLN))

  • Solid lipid nanoparticles(SLN) are particulate systems for parenteral drug administration and suitable for controlled release. SLN were prepared by homogenization process. Dispersion at increased temperature (molten lipid) was performed to yield SLN loaded with lipophilic drugs. Tetracaine base, lidocaine base, prednisolone, methyltestosterone and ethinylestradiol were used as model drugs to access the loading capacity and to study the release behavior. To investigate production parameters(lipids, surfactant concentration, homogenizing rpm) in the formation of SLN, particle size was performed by laser diffraction analysis. The mean particle size of SLN with stearic acid or trilaurin was below 1 micron. By decreasing the particle size and increasing the surfactant concentration, the release rate was increased especially in the case of highly lipophilic drug loaded SLN. Methyltestosterone or ethinylestradiol loaded SLN showed a distinctly prolonged release over a few days.

Importance of culture history on 17α-ethinylestradiol cometabolism by nitrifying sludge

  • $17{\alpha}-ethinylestradiol$ (EE2), a synthetic estrogen which interfere the endocrine and reproductive function in living organisms, has been found extensively to be deposited into municipal wastewater treatment plants and the environment via human excretion. EE2 has long been known to be efficiently cometabolized by ammonia-oxidizing bacteria (AOB) during ammonia ($NH_3$) oxidation. Current study aims to investigate the effect of culture history on the biotransformation of EE2 by nitrifying sludge which was enriched under different ammonia loading rates in continuous flow reactors. Result showed that past growth condition largely affected not only the metabolic rate of $NH_3$ oxidation but also EE2 cometabolism. Sludge previously acclimated with higher $NH_3$ loads as well as sludge dominated with AOB belong to high growth cluster (Nitrosomonas europaea-Nitrosococcus mobilis) showed higher rate of EE2 biotransformation than those one being acclimated with lower $NH_3$ loads because of its ability to provide more reducing power from $NH_3$ oxidation. Moreover, the correlation between the degradation rates of $NH_3$ and EE2 was higher in sludge being acclimated with higher load of $NH_3$ in comparison with other sludge. Implication of the findings emphasized the role of volumetric $NH_3$ loading rate in determining EE2 removal in wastewater treatment system.

Hypericum Perforatum Decreased Hippocampus TNF-${\alpha}$ and Corticosterone Levels with No Effect on Kynurenine/Tryptophan Ratio in Bilateral Ovariectomized Rats

  • The present study was designed to investigate the effect Hypericum Perforatum (HP), on behavioral changes, corticosterone, TNF-${\alpha}$ levels and tryptophan metabolism and disposition in bilateral ovariectomized rats compared to $17{\alpha}$-ethinylestradiol. Behavioral analysis by measuring immobility time in forced swimming test and open field test, serum and hippocampal corticosterone and TNF-${\alpha}$ along with hippocampal kynurenine/tryptophan ratio were determined in mature ovariectomized rats treated orally either by HP at three different doses 125, 250, and 500 mg/kg/day or by $17{\alpha}$-ethinylestradiol $30{\mu}g/kg/day$ for 30 days. Ovariectomized rats showed significant increase in immobility time in the forced swimming test. Along with elevation in serum and hippocampal TNF-${\alpha}$ and corticosterone levels associated with significant increase in hippocampal kynurenine/tryptophan ratio. Immobility time in the forced swimming test was decreased in rats treated by different doses of HP in a dose dependent manner and $17{\alpha}$-ethinylestradiol with no concomitant changes in the open field test. Only Rats treated with HP exhibited significant decrease in the elevated serum and hippocampal TNF-${\alpha}$ and corticosterone, which couldn't explain the associated insignificant effect on hippocampaus kynurenine/tryptophan ratio in comparison to ovariectomized untreated rats. It is concluded that increased tryptophan metabolism toward kynurenine secondary to elevated corticosterone and TNF-${\alpha}$ might be one of the pathohphysiological mechanisms that could explain depression like state observed in this rat model. Further, the observed attenuating effect of HP on TNF-${\alpha}$ and corticosterone could contribute in its antidepressant effect in this animal model by other ways than their effects on tryptophan-kynurenine metabolism pathway.

하수처리장의 내분비계장애물질에 대한 Yeast Two-hybrid Assay와 Enzyme-linked Immunosorbent Assay에 의한 에스트로겐활성도 평가 (Evaluation of the Estrogenic Activity by Yeast Two-hybrid Assay and Enzyme-linked Immunosorbent Assay in Sewage Treatment Plant)

  • 가정계열과 공단계열로 분리하여 처리되는 하수처리장에서 에스트로겐 활성을 평가하기 위하여 yeast two-hybrid assay와 enzyme-linked immunosorbent assay(ELISA)를 이용하여 내분비계장애물질의 농도와 활성도를 측정하였다. 그 결과 가정계열 유입수 중에서 estrone (E1), 17$\beta$-estradiol(E2), 17$\alpha$-ethinylestradiol(EE2) 그리고 APE의 농도는 각각 최대 167.1, 39.7, 7.3, 145.4 ng/L까지 검출되었다. 활성슬러지법에 의한 처리로, 17$\beta$-estradiol의 평균제거율은 77.5%, 고도처리 공정인 모래여과와 오존산화를 거친 후에는 80.8%까지 제거되는 것으로 나타났다. 동시에 Yeast two-hybrid assay로 각 내분비계장애물질의 농도-반응곡선으로부터 반응식을 구하여, 에스트로겐 활성에 미치는 각 물질의 기여도를 분석한 결과, 가정계의 활성슬러지법에 의한 처리수에서 estrone, 17$\beta$-estradiol 17$\alpha$-ethinylestradiol, APE 가 각각 70.7, 23.3, 3.7, 2.32%로 나타났다. 즉, 생물학적 처리공정을 통해 배출된 처리수의 에스트로겐 활성에 영향을 미치는 주된 기여물질은 estrone과 17$\beta$-estradiol인 것으로 나타났다.

Expression Patterns of the chgH:rfp Transgene in Response to 17α-Ethinylestradiol (EE2) Exposure in Marine Medaka Oryzias dancena

  • The functional utility of a transgenic marine medaka Oryzias dancena strain carrying the red fluorescent protein (RFP) gene driven by an endogenous choriogenin H (chgH) promoter was evaluated for its ability to detect waterborne $17{\alpha}$-ethinylestradiol (EE2), a synthetic estrogen derivative. The chgH:rfp transgenic marine medaka larvae showed an age-dependent tendency in the efficiency of EE2-mediated transgene expression, in which transgenic larvae older than 6 days post-hatching displayed a more effective response in their transgene expression to EE2 than did younger hatchlings. During experimental exposures to high concentrations of EE2 (200 to 1,000 ng/L), the transgenic responses in the hatchlings were broadly dose- and duration-dependent. With exposures using lower doses of EE2 (25, 50 and 100 ng/L), EE2-induced transgenic RFP was also observed in the transgenic larvae, although the lower doses required exposure of longer duration. Under the EE2 exposure and microscope assay conditions used in our study, transgenic marine medaka larvae exhibited a similar degree of EE2-mediated RFP phenotype expression at various salinity levels (0, 15 and 30 ppt).

  • Vitellogenin은 난황단백질의 전구체로서 생체내 발현은 에스트로젠과 에스트로젠 수용체가 반응함으로써 활성화되는 것으로 알려져 있다. 어류의 경우 간에서의 vitellogenin의 합성 및 분비는 성숙된 암컷에서만 일어날 수 있는 현상이나, 과거 여러 실험 결과를 바탕으로 수컷이나 미성숙한 암컷에서도 에스트로젠성 물질에 의해 합성이 유도되는 것으로 밝혀졌다. 따라서 수컷 또는 치어 plasma에서 vitellogenin의 측정은 외인성에스트로젠에 대한 노출을 평가할 수 있는 좋은 수단이 될 수 있다. (중략)

단기 발암성 검색법을 이용한 축산식품에 잔류할 수 있는 합성홀몬의 발암촉진효과에 대한 연구 (Promotive Effects of Several Synthetic Hormones Using Short-Term Carcinogen Screening Method)

  • 이 연구는 수입 축산식품중에 잔류해서 사람에게 해를 줄 수 있는 여러 합성 홀몬의 발암성을 알아보기 위하여 수행한 연구이다. 6주령의 F344 랫드를 5군으로 나누어 발암유발물질인 diethylstilbesterol(DENA)를 체중 kg당 200 mg을 복강 내 투여하고, 시험게시 제2주에서 시험종료시까지 제 1, 2, 3,군에 각각 diethylstilbesterol(DES), ethynylestradio($EE_2$), bovine somatotrophin(BST)을 투여하고, 제 4군에는 발암촉진이 잘 알려진 phenobabital을 투여하였으며, 제 5군은 대조군으로 기초사료만을 급여하였다. 동시에 모든군의 랫드는 D-galactosamine을 체중 kg 당 300 mg을 복강내로 1회 투여하였으며, 간변화의 증폭을 위하여 2/3 간부분 절제를 시험개시 제 5주에 실시하였다. 시험종료시 부검후 랫드의 간장은 GST-P에 대한 면역조직화학적 염색을 실시하여 발암성을 검색하였다. 두개의 합성 에스트로젠 홀몬($EE_2$, DES)은 대조군과 비교하였을 때 발암성이 있는 것으로 생각되어지고, BST는 발암성이 없는 것으로 생각된다.

내분비 장애물질 측정을 위한 붕어 비탈로제닌 정량 분석 ELISA 시스템 개발 (Development of Quantitative Vitellogenin ELISAs for Goldfish(Carassius auratus) used in Endocrine Disruptor Screening)

  • 난황단백질인 비탈로제닌을 성숙한 암컷 붕어 혈청으로부터 음이온 교환 크로마토그래피를 이용하여 정제 하였다. 정제한 비탈로제닌을 BALB/c mice를 이용하여 폴리크로날 항체를 생산하였고 이를 protein A column을 사용하여 정제하였다. 또한 이렇게 정제된 폴리크로날 항체를 이용한 붕어 비탈로제닌 측정용 효소면역측정법을 개발하였으며 그 측정 범위는 2∼l,000ng/mL이고 recovery 변동 범위는 88∼112%였다. 또한 이 효소면역측정법을 평가하기 위해 성숙한 수컷 붕어를 1,000ng/L ethinylestradiol(EE$_2$) 에 4주 동안 노출시켜 유도되어지는 비텔로제닌을 측정하였다. 그 결과 성숙한 수컷 붕어의 경우 비탈로제닌이 3주 만에 암컷 붕어의 평균수치만큼 유도됨을 알 수 있었다.

Transgene chgH-rfp expression at developmental stages and reproductive status in marine medaka (Oryzias dancena)

  • Background: The transgenic approach using estrogen-responsive regulator in fish has been given much attention as a potential means to detect and/or address estrogen-related aquatic pollutions. In order to address the development stage- and reproduction status-dependent expression patterns of the chgH-rfp transgene (red fluorescent protein transgene driven by choriogenin H promoter) in marine medaka Oryzias dancena, naturally occurring red fluorescent protein (RFP) signals under non-exposed conditions as well as the transgenically induced RFP signals under estrogen-exposed conditions were assayed. Results: Female transgenics begun to show naturally occurring RFP signals from the age of 7 weeks post hatching (WPH) without experimental estrogen exposure. Afterward, these RFP signals in female transgenics became robust with the progress of ovarian maturation. On the other hand, male transgenics did not show any naturally occurring RFP signal under non-exposed conditions irrespective of developmental stages and maturation statue. Upon exposures using estradiol-$17{\beta}$ (E2) and $17{\alpha}$-ethinylestradiol (EE2), RFP signals were significantly induced specifically in the livers of transgenic males. Conclusions: Male chgH-rfp transgenics were able to keep the "off" state of RFP expression during their entire life cycle unless exposed to exogenous estrogens. Owing to their tight regulation capability of estrogen-responsive transgene, transgenesis of chgH-rfp in male marine medaka could offer a useful model system for future ecotoxicogenomic studies regarding estrogenicity-related issues in aquatic and marine environments.
