Forage Protivity of Corn and Sorghum Hybrids in Rice Black-Streaked Dwarf Virus Prevalent Area (흑조위축병이 심한 지역에서 옥수수와 수수 품종의 사료생산성)

  • Silage productivity of 7 corn and 3 sorghum hybrids was determined at two planting dates differing in rice black-streaked dwarf virus (RBSDV) infection rates in the southern part of Korea where RBSDV is prevalent. 1. Percent stand was similar between two planting dates, but that of corn hybrids (85.0-99.5 %) was higher compared with sorghum (59.8-82.3 %). Culm length of corn hybrids at April 23 planting was longer compared with May 20 planting, but that of sorghum hybrids was similar between two planting dates. 2. RBSDV infection rate of corn hybrids at April 23 planting was lower compared with May 20 planting, but sorghum was not infected at both planting dates. Among corn hybrids Suweon 19 was most severely infected, but Suweon 99 was least infected at both planting dates. 3. Percent ear bearing plants and earltotal dry matter (TDM) ratio of corn hybrids at April 23 planting was higher than those of May 20 planting. Among the corn hygrids percent ear beraring plants, ear/TDM ratio of Suweon 19 was lowest at both planting dates due to severe RBSDV infection. Ear/TDM ratio of sorghum hybrids was below 17.8 % due to sterility and partially bud damages. 4. Silage yield at April 23 planting was higher than that at May 20 planting and sorghum hybrids outyielded corn hybrids. Silage yield of Suweon 19 and Jinjuok was lowest due to severe RBSDV infection. 5. Crude protein was similar regardless planting dates, species, and varieties except the lowest P 931. Ether extract at April 23 planting was lower then that at May 20 and corn was lower compared with sorghum hybrids. Crude fiber and nitrogen free extracts(NFE) was similar between planting dates. Crude fiber of corn was lower, but NEF was higher compared with sorghum, but similar among hybrids in a species. 6. In vitro dry matter digestibility (IVDMD) at May 20 planting was higher than that of April 23 planting and corn was higher than sorghum hybrids. Digestible dry matter (DDM) of both corn and sorghum at April 23 planting was higher than that of May 20 planting. At April 23 planting DDM of all corn and sorghum hybrids was similar, but DDM of Suweon 19 and Jinjuok was lower than other corn and sorghum hybrids.

Effects of Brewery Meal-Based Fermented Feedstuff Supplemented with Aspergillus Oryzae or Saccharomyces Cerevisiae on Ruminal Microorganism (Aspergilus Oyzae 혹은 Saccharomyces Cerevisiae를 첨가하여 제조한 맥주박 위주 발효사료가 반추위 미생물에 미치는 영향)

  • This study was conducted to examine effects of brewery meal-based fermented feedstuff supplemented with Aspergillus oryzae(AO) or Saccharomyces cerevisiae(SC) on luminal micro-organism of Korean native cattle. Two cows equipped with luminal cannulas were used as experimental animals. Experiment was done with three treatment groups: $71.5\%$ of commercial feed and $28.5\%$ of com silage(control): $45.0\%$ of commercial feed, $26.5\%$ of fermented feedstuff supplemented with AO and $28.5\%$ of corn silage(TAO): $45.0\%$ of commercial feed, $26.5\%$ of fermented ffedstuff supplemented with SC and $28.5\%$ of corn silage(TSC). The number of total viable bacteria (p<0.05), anaerobic fungi and protozoa(p<0.05) was higher in TAO and TSC than in control. The number of proteolytic bacteria(p<0.05), cellulolytic bacteria and xylan fermenters tended to be higher in TAO and TSC than in control. The dry matter recovery (DMR) of protozoa was higher in TAO and TSC than in control(p<0.05). The crude protein (CP) content of total microbes and protozoa was higher in TSC than in control and TAO (p<0.05). The CP content of bacteria was higher in TAO and TSC than in control(p<0.05). The ether extract(EE) content of the total microbes was higher in TAO than in control and TSC(p<0.05), and the EE of protozoa and bacteria were higher in TSC than in control and TAO(p<0.05). The ratio of essential amino acids of total microbe was higher in control than in TAO and TSC(p<0.05). The ratio of methionine and alanine of bacteria was higher in TAO and TSC than in control(p<0.05). The results suggested that the feeding of fermented feedstuff supplemented with AO or SC had an influence on the numbers of ruminal microorganism and the changes of microbial body composition.

  • This study was conducted to compare and evaluate fermentable and nutritional characteristics of brewery meal-based fermented feedstuffs supplemented with Aspergillus oryzae (AO) and Saccharomyces cerevisiae (SC). Experiments were divided into three treatment groups; fermented feedstuff supplemented with $1\%$ of AO(FFAO), fermented feedstuff supplemented with $1\%$ of SC(FFSC), and fermented feedstuff supplemented with $0.5\%$ of AO and $0.5\%$ of SC(FFAS). For changes of crude protein contents by 48 h fermentation, there were no significant differences among treatments. Ether extract(EE) contents were significantly increased by 48 h fermentation (p<0.05). Neutral detergent fiber(NDF) contents of FFAO, FFSC and FFAS were significantly decreased by 48 h fermentation(p<0.05), but acid detergent fiber(ADF) and acid detergent lignin (ADL) contents were not different. The pH of FFAO and FFAS was decreased more rapidly than that of FFSC(p<0.05), reaching a plateau after 24 h. Alcohol content was increased rapidly until 18 h in FFAO and was increased rapidly until 12 h in FFSC and FFAS, and alcohol content of FFAO, FFSC and FFAS was maintained constantly after 24 h. The ammonia N content of FFAO, FFSC and FFAS was 0.022, 0.073 and $0.040\%$ at 48 h, respectively, and then ammonia N was over twice higher in FFSC than in FFAO and FFAS(p<0.05). Dextrose content was increased until 6 h in FFAO but was rapidly decreased in FFSC and FFAS until 6 h(p<0.05). Lactate content was higher in FFAO and FFAS than in FFSC(p<0.05). Consequently, when we added AO in formulation of fermented feedstuff with brewery meal which moisture content was high, EE, alcohol, and lactate contents were increased, but NDF and ammonia N contents were reduced. Therefore, it is expected that AO will be effective to increase the feed value and the preservation of fermented feedstuff with a high moisture content.

  • The barley (Hordeum vulgare L.) is now widely grown as a whole crop silage in Korea, but the quality of that silage does not examined from farms. Therefore, this experiment was conducted to evaluate the forage quality of whole crop barley that was participated in Silage Quality Contest in 2008. These data were classified by region, dry matter (DM) yield, with or not additive, planting and harvest date. Difference on the lactic acid content of barley silage was detected in the region, DM yield, additive and harvest date (p<0.05), however, there were partially significant differences in chemical composition. There were significant differences among moisture content in DM yield, pH in additive, and crude ash content in additive and planting date of barley silage. Crude protein content was significant difference in the region, and ether extract content was in region, DM yield and harvest date, however, non-fiber carbohydrate was significant difference in planting date of barley silage. Neutral detergent fiber, acid detergent fiber and relative feed value were significant difference only in the cultivation region of whole crop barley. According to this experiment, differences in the silage quality were observed among whole crop barley silages. Therefore, nutritive value as well as moisture and pH are important in silage quality evaluation of whole crop barley. Especially, lactic acid is an important factor for the evaluation of whole crop barley silage.

  • This experiment was performed to evaluate the morphological responses of the gastric epithelial cells of the mouse, inoculated with Ehrlich carcinoma cells in the inguinal area, following administration of 5-fluorouracil, mitomycin C or Acriflavine-Guanosine compound (AG60). In this study, each mouse was inoculated with $1{\times}10^7$ Ehrlich carcinoma cells subcutaneously in the inguinal area. From next day after inoculations, 0.2 mL of saline, 5-fluorouracil (30 mg/kg), mitomycin C ($400{\mu}g/kg$) or AG60 (30 mg/kg) were injected to the animals every other day, respectively. Each animals were sacrificed after 7th injection and tissue were taken from the gastric mucosa. Thereafter, the ultrathin sections were stained with uranyl acetate and lead citrate. In the 5-fluorouracil-, mitomycin C- or AG60-treated mice, myelin figures and multivesicular bodies within the gastric mucous epithelial cells were observed more frequently than those of the normal control. In the 5-fluorouracil-treated mice, membrane structures containing a few mucous granules in the luminal space were observed. Indeed, bulging cytoplasmic process containing mucous granules protruding into the gastric lumen were observed in the mitomycin Ctreated mice. Therefore, this study suggested that AG60 as compared with 5-fluorourail and mitomycin C may effective medicine without damage to the secretion ability of gastric mucous epithelial cells.

Effects of Planting Density on Growth, Forage Yield and Chemical Composition of Jeju Native Sorghum(Sorghum bicolor L.) (제주 재래수수의 재식밀도에 따른 생육, 수량 및 사료가치에 미치는 영향)

  • Jeju native sorghums were grown with six different planting densities ($5{\times}5cm,\;10{\times}10cm,\;15{\times}15cm,\;20{\times}20cm,\;25{\times}25cm\;and\;30{\times}30cm$) from April 3 to September 6 in 2000 in Jeju island to determine the influence to growth, yield and feed value by the planting densities. Days to heading were delayed from 66 days to 69 days as increasing of plant density. Plant height was 195.1cm at $30{\times}30cm$ plot, whereas that was 216.5cm at $10{\times}10cm$ plot, indicating that the plant was higher by the decrease of the planting density. As plant density increased, the number of leaves and steam diameter were decreased. Fresh forage yield increased from 25.4 to 61.3 MT/ha, dry matter (DM) yield from 5.8 to 16.5 MT/ha, crude protein (CP) yield from 0.4 to 1.5 MT/ha and total digestible nutrients (TDN) yield from 2.7 to 8.8 MT/ha as planting density from $30{\times}30cm\;to\;10{\times}10cm$ plot. Crude protein content increased from 7.0 to $9.0\%$, ether extract content from 1.7 to $4.4\%$, nitrogen free extract content from 36.9 to $38.4\%$ and TDN content from 46.1 to $55.5\%$ as planting density was decreased. whereas crude fiber content decreased from 43.9 to $37.1\%$ and crude ash content from 8.4 to $6.8\%$.

Effects of Seeding Date on Ecological Response, Forage Yield Potential and Chemical Composition in Jeju Native Sorghum(Sorhgum bicolor L.) (제주 재래수수의 파종기에 따른 생태반응, 사료수량성 및 조성분 분석)

  • In order to identify the growth characteristics, the yield and chemical composition of Jeju native sorghum(Sorghum bicolor L.) based on seeding date in Jeju region. Seeding carried out the 10-day intervals from March 27 to May 27 in 2000, respectively. Plant height was 222.9 cm, Showing the highest on seeding date, April 7 among that of any other seeding date. On the other hand, those of early and late seeding gradually decreased. The results of stem diameter, number of leaves were similar to those of the plant height. The yield of fresh, dry matter forage, crude protein and TDN found the highest on April 7 seeding, 48.0 MT/ha, 12.0 MT/ha, 1.0 MT/ha, 5.3 MT/ha, respectively. In early and late seeding, the yield was gradually decreased. According to delaying the seeding date, March 27 to May 27, the contents of crude protein (from 7.8 to $11.4\%$) ether extract(from 1.7 to $3.3\%$), nitrogen free extract(from 34.1 to $40.0\%$) and TDN(from 42.9 to $52.5\%$) were gradually increased, respectively. On the other hand, those of crude fiber (from 40.6 to $32.3\%$) and crude ash (from 10.3 to $7.6\%$) were decreased. These results showed that April 7 was the optimum seeding date with the sole object of feed production of Jeju native sorghum.

  • Metabolomics aims at the comprehensive, qualitative and quantitative analysis of wide arrays of endogenous metabolites in biological samples. It has shown particular promise in the area of toxicology and drug development, functional genomics, system biology and clinical diagnosis. In this study, analytical technique of MS instrument with high resolution mass measurement, such as time-of-flight (TOF) was validated for the purpose of investigation of amino acids, sugars and fatty acids. Rat urine and serum samples were extracted by selected each solvent (50% acetonitrile, 100% acetonitrile, acetone, methanol, water, ether) extraction method. We determined the optimized liquid chromatography/time-of-flight mass spectrometry (LC/TOFMS) system and selected appropriated columns, mobile phases, fragment energy and collision energy, which could search 17 metabolites. The spectral data collected from LC/TOFMS were tested by ANOVA. Obtained with the use of LC/TOFMS technique, our results indicated that (1) MS and MS/MS parameters were optimized and most abundant product ion of each metabolite were selected to be monitorized; (2) with design of experiment analysis, methanol yielded the optimal extraction efficiency. Therefore, the results of this study are expected to be useful in the endogenous metabolite fields according to validated SOP for endogenous amino acids, sugars and fatty acids.

Analysis of Volatile Flavor Compounds from the Leaves of Eucommia ulmoides (두충잎의 휘발성 향기성분 분석)

  • This study was performed to analyze the volatile flavor compounds of Eucommia ulmoides leaves as influenced by harvesting time and drying method. Essential oils of fresh, air-dried and freeze-dried leaves of Eucommia ulmoides were extracted by SDE(simultaneous steam distillation and extraction) method using pentane and diethyl ether(1:1), and their volatile flavor compounds were analyzed by GC and GC-MS. Total 51 components, including 10 hydrocarbons, 15 alcohols, 12 aldehydes, 4 ketones, 4 esters and 6 acids were identified in fresh Eucommia ulmoides harvested in July. In fresh samples harvested in September, 15 hydrocarbons, 10 alcohols, 5 aldehydes, 4 ketones, 4 esters and 3 acids were identified. In fresh Eucommia ulmoides, aldehydes(8.25ppm) were the most abundant compounds in July samples and alcohols(18.87ppm) in September ones. Seventy one components, including 21 hydrocarbons, 12 alcohols, 12 aldehydes, 9 ketones, 5 esters, 8 acids and 4 miscellaneous ones were identified in air-dried samples harvested in July. In air-dried samples harvested in September, 10 hydrocarbons, 9 alcohols, 3 aldehydes, 3 ketones, 4 esters, 4 acids and 1 miscellaneous one were identified, and the most abundant compounds in July and September samples were hydrocarbons at 5.06ppm and 15.11ppm, respectively. A total of 41 components, including 13 hydrocarbons, 9 alcohols, 5 aldehydes, 3 ketones, 6 esters and 5 acids were identified in freeze-dried samples harvested in July. Freeze-dried samples harvested in August also contained 41 components but with different types, and the ones of September 26 compounds. In freeze-dried ones, hydrocarbons were the most abundant compounds in July sample and esters in August and September samples.

Bioavailability and Feed Value of Starfish with Various Treatments (처리방법에 따른 불가사리의 이용율 및 사료적 가치)

  • To evaluate the feed value of starfish, antimicrobial effects of its extract, nutrients contents, concentration of amino acids and its bioavailability were tested. Steaming and ether processes were applied to obtain the extract from starfish for antimicrobial effects examination. The starfish was dried at $60^{\circ}C$ for 3 days before grinding for processing and fermentation. Ground starfish(GS), extruded starfish(ES), fermented starfish(EFS) were added with enzyme and without enzyme(Non enzyme fermented starfish : NEFS). Then the nutrient composition and bioavailability of those were analyzed. The extract from starfish showed no inhibition of the growth of lactobacillus and pathogenic bacteria. Protein content showed significantly higher 62.86% and 52.82%, respectively in EFS and NEFS than GS and EGS(p<0.05). The Ca content of GS, EGS, EFS and NEFS was 17.26%, 18.26%, 5.37% and 8.55%, respectively. It was low in EFS and NEFS due to measure the Ca content after fermentation. Total amino acid was 17.17 mg/g, 20.28 mg/g, 36.30 mg/g and 29.96 mg/g in GS, EGS, EFS and NEFS, respectively. The ratio of total amino acid to protein tended to show the similar tendency as total amino acid. Both total amino acid and its ratio to protein were increased by the fermentation. Bioavailability of the protein and Ca showed more 80% in EFS and NEFS. The nutrients availability of EFS were significantly higher in laying hens than other treatments. The results of these experiments indicate that starfish would be applied as a feed ingredients if it was properly treated.
