에탄올과 식이지방량이 흰쥐의 혈액성상과 간조직에 미치는 영향 (Effects of Ethanol Consumption and Fat Content in Diet on Biochemical Indices of Blood and Liver in the Rat)

  • To investigate effects of ethanol and dietary fat on growth and bichemical indices of liver tissue and blood in rats 40 male rats of Sprague-Dawley wtrain weighing about 160g were divided into 5 groups (low-fat diet group ethanol-administered low-fat diet group high-fat diet group ethanol-administered high-fat diet group and commercial diet group) and fed expe-rimental diets for 8 weeks. Ethanol-administered groups consumed ethanol corresponding to 22 cal% which was considered as moderate drinking. Neither the ethanol intake nor the dietary fat level affected calorie intake. Nonetheless the low-fat diet group with ethanol had the lowest growth rate and 2-fold increase in the concentration of plasma triglyceride. There was no effect of ethanol and dietary fat level on contents of protein lipid and lipid composition of liver tissue. The level of lipid peroxide of liver tissue tended to be increased by ethanol intake but the increase was statistically insignificatnt. The low-fat ethanol group had lowered hepatic mitochondrial respiration rate and deformed structure of mitochondria of hepatocytes.

$\beta$-Carotene 첨가식이가 알코올성지방간 쥐의 지질과산화물 수준과 항산화효소 활성에 미치는 영향 (Effects of $\beta$-Carotene Supplementation on Lipid Peroxide Levels and Antioxidative Enzyme Activities in Alcoholic Fatty Liver Rats)

  • This study was performed to investigate the effect of dietary $\beta$-carotene supplementation on lipid peroxide levels and antioxidant enzyme activities in alcoholic fatty liver rats. Forty five Sprague-Dawley male rats aging 8 weeks were used as experimental animals, which were divided into the control diet (CD) and the ethanol diet (ED) and the ethanol + $0.02\%$ $\beta$-carotene diet (EPD) groups and fed the experimental diet respectively for 5 weeks. After the feeding, rats were sacrificed to get blood and liver to analyze lipid and lipid peroxide levels and antioxidant enzyme activities. The mean body weight and food intake of the ethanol diet group was significantly lower than that of the control diet. The liver index (LI) of the ethanol diet group was significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Serum levels of total lipid, triglyceride of the ethanol diet group were significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Total cholesterol levels were not significantly different among all groups. HDL-cholesterol of the ethanol diet group was significantly lower than those of the control diet and the $\beta$-carotene supplementation group. Liver TBARS of the ethanol diet group was significantly higher than those of the control diet and the $\beta$-carotene supplementation group. Liver lipofuscin and conjugated diene levels were not significantly different among all groups. The superoxide dismutase activity of the ethanol diet group was significantly lower than those of the control diet and the $\beta$-carotene supplementation group. Catalase and glutathione peroxidase activities were not significantly different among all groups. Because v-carotene supplementation significantly decrease the serum total lipid, triglyceride, liver TBARS revels and increase the superoxide dismutase activity in alcoholic ratty liver rats, $\beta$-carotene supplementation seems to give beneficial effect for the alcoholics.

만성적인 에탄올 섭취가 흰쥐의 엽산대사 및 혈장 호모시스테인 농도에 미치는 영향 (Effects of Chronic Ethanol Administration on Folate Metabolism and Plasma Homocysteine Concentration in the Rats)

  • The present study was conducted to investigate the effect of different levels of ethanol ingestion on 131ate metabolism and plasma homocysteine concentration in Sprague-Dawley male rats receiving 0, 10, 30% of their caloric intake as ethanol for S weeks. Diets containing 10% ethanol had no effect on plasma and red blood cell(rbc) 131a1e. However, in rats fed a 30% ethanol diet, rbc folate increased and plasma 131ate decreased significantly, In the rats maintained first on a 30% ethanol diet for S weeks and then on a control diet for 2 weeks, the levels of plasma and rbc f31ate were normalized by withdrawal of ethanol. Urinary fo1ate excretion increased markedly in rats fed 10% and 30% ethanol diets and decreased to 51% of controls by withdrawal of ethanol. Plasma homocysteine concentration increased significantly in rats fed a 30% ethanol diet. The results suggest that chronic ingestion of ethanol increased urinary 131ate excretion markedly, which may decrease plasma 131ate and deplete liver folate.

Effects of Normal Diet with or without Naringin Supplement Following Ethanol Diet on Changes in Lipid Profiles and Antioxidant Enzyme Activities in Rats

  • This study was performed to investigate the effect of normal diet with or without naringin supplement on the lipid and antioxidant metabolism in ethanol-treated rats for a short tenn. Male Sprague-Dawley rats were divided into three groups (n=10), which were assigned to one of three dietary categories : $E_8$ : ethanol diet for 8 wks, $E_4N_4$ : ethanol diet for the first 4 wks and normal diet for the last 4 wks, $E_4Nna_4$ : ethanol diet for the first 4 wks and normal diet with naringin supplement for the last 4 wks. Plasma total cholesterol concentrations were significantly higher in ethanol fed rats for 8 weeks. The HDL-C/total-C ratios of the $E_4N_4$ and the $E_4Nna_4$ groups were significantly higher than that of the $E_8$ group, while the atherogenic index was lower in the $E_4N_4$ and the $E_4Nna_4$ groups than in the $E_8$ group. The $E_4N_4$ and $E_4Nna_4$ diets significantly lowered both the hepatic cholesterol and triglyceride levels compared to the $E_8$ group. Accumulation of hepatic lipid droplets was observed to be the highest in the $E_8$ group. In the current study, the naringin supplement to normal diet significantly lowered both the hepatic HMG-CoA reductase and ACAT activities in ethanol pre-treated rats for 4 weeks. Antioxidant enzyme activities were also upregulated when ethanol feeding was ceased. Naringin supplement given for 4 weeks after ethanol cessation resulted in a significant decrease in the plasma cholesterol and hepatic lipids and plasma TBARS as well as the hepatic HMG-CoA reductase and ACAT activities compared to the rats given ethanol diet for the entire 8 weeks. Replacement of normal diet following a short tenn ethanol feeding was effective for the recovery of ethanol-induced fatty liver and for normalizing plasma and hepatic lipid profiles and antioxidant enzyme activities, regardless of an additional phytochemical supplement, naringin. The effect of naringin could seemingly be more evident if its supplementation period had been extended longer than 4 weeks after ethanol cessation.

당귀의 첨가 식이가 흰쥐의 지방대사와 알콜대사 및 간기능에 미치는 영향 (Effects of Angelica gigas Nakai diet on lipid metabolism, alcohol metabolism and liver function of rats administered with chronic ethanol)

  • 당귀(Angelica gigas Nakai)의 첨가 식이가 만성적인 알콜 투여시 흰쥐의 지방대사, 알콜대사 및 간기능에 미치는 영향을 조사하기 위해 Sprague-Dawley 수컷 쥐를 대조군(증류수 투여), 에탄올 투여군, 당귀 첨가 식이군, 에탄올 투여 와 당귀 첨가 식이군의 4그룹으로 나누어 30일간 에탄올 및 증류수는 경구 투여하였고, 당귀는 AIN-76 식이에 첨가하여 섭취하도록 하였다. 그룹간 혈중 알콜대사 변화를 조사하기 위해 21째 날에 13% 알콜을 모든 군의 쥐에 경구 투여한 후 혈중 알콜농도 변화를 5시간 동안 조사하였으며, 그 결과는 다음과 같다: 1) 당귀 첨가 식이군의 혈중 알콜농도가 정상식이군에 비하여 알콜투여 후 2시간부터 유의적으로 감소하였다; 2) 만성적인 알콜섭취와 동시 당귀 첨가 식이군의 알콜 투여 직후의 혈중 알콜 산화 속도는 정상식이군 혹은 당귀 첨가 식이군에 비하여 전반적으로 빠르게 진행되었다. 또한 30일 후에는 각 그룹의 쥐를 희생시켜 혈중 및 간장 중의 지방성분 및 혈중의 간장 관련 효소의 활성을 측정하였으며, 그 결과는 다음과 같다: 1) 당귀 첨가 식이는 혈중 LDL-콜레스테롤 함량을 비첨가 식이군에 비하여 유의적으로 낮추었다; 2) 당귀 첨가 식이는 알콜투여로 증가된 간장 중의 중성지방 및 총지방 함량과 혈중 ${\gamma}-GTP$의 수준을 유의적으로 낮추었다. 이상의 결과로부터 당귀 첨가식이가 만성적인 알콜 섭취로 야기될 수 있는 질병에 대한 개선 효과가 있음을 제안할 수 있겠다.

Effects of vitamin C and E supplementation on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet

  • We compared the preventive capacity of high intakes of vitamin C (VC) and vitamin E (VE) on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet. Thirty-two Wistar rats received the low fat (10% of total calories) Lieber-DeCarli liquid diet as follows: either ethanol alone (Alc group, 36% of total calories) or ethanol in combination with VC (Alc + VC group, 40 mg VC/100 g body weight) or VE (Alc + VE group, 0.8 mg VE/100 g body weight). Control rats were pair-fed a liquid diet with the Alc group. Ethanol administration induced a modest increase in alanine aminotransferase (ALT), aspartate aminotransferase (AST), conjugated dienes (CD), and triglycerides but decreased total radical-trapping antioxidant potential (TRAP) in plasma. VE supplementation to alcohol-fed rats restored the plasma levels of AST, CD, and TRAP to control levels. However, VC supplementation did not significantly influence plasma ALT, AST, or CD. In addition, a significant increase in plasma aminothiols such as homocysteine and cysteine was observed in the Alc group, but cysteinylglycine and glutathione (GSH) did not change by ethanol feeding. Supplementing alcohol-fed rats with VC increased plasma GSH and hepatic S-adenosylmethionine, but plasma levels of aminothiols, except GSH, were not influenced by either VC or VE supplementation in ethanol-fed rats. These results indicate that a low-fat ethanol diet induces oxidative stress and consequent liver toxicity similar to a high-fat ethanol diet and that VE supplementation has a protective effect on ethanol-induced oxidative stress and liver toxicity.

Effect of Hesperidin Supplementation on Lipid and Antioxidant Metabolism in Ethanol-fed Rats

  • This study examined the effect of hesperidin supplementation with an ethanol diet on lipid and antioxidant metabolism in rats. Male Sprague-Dawley rats were divided into two groups (n=10), and were assigned to one of two dietary categories: $E_8$, ethanol diet (50 g/L) for 8 wks; $E_8H_4$, ethanol diet for the first 4 wks and hesperidin (0.02%, w/w) supplemented ethanol diet for the last 4 wks. The plasma and hepatic lipids, hepatic cholesterol regulating enzyme activity, hepatic antioxidant enzyme activity and lipid peroxidation were determined. Supplementation with hesperidin for the last 4 wks during the 8 wks period of the ethanol diet, significantly increased the ADH activity. In conjunction with the chronic administration of ethanol, hesperidin supplementation resulted in a significant decrease in the hepatic cholesterol and triglyceride concentrations compared to the $E_8$ group. The hepatic HMG-CoA reductase and ACAT activities were significantly lower in the hesperidin-supplemented group. When comparing hepatic antioxidant enzyme activities, SOD, GSH-Px, and G6PD activities and GSH level were significantly higher in the $E_8H_4$ group than in the E8 group. Plasma TBARS levels were significantly lower in rats fed ethanol with hesperidin compared to the rats fed only ethanol; however, the hepatic TBARS levels were not significantly different between the groups. Accordingly, the additional hesperidin supplement with an ethanol diet might be effective for improving the hepatic lipid metabolism and antioxidant defense system.

고들빼기 첨가 식이가 알콜투여 흰쥐의 지방대사와 간기능에 미치는 영향 (Effects of Ixeris Sonchifolia H. Fiet on Lipid Metabolism and Liver Function of Rats Administered with Ethanol)

  • To investigate the effects of Ixeris sonchifolia Hance diets on serum and hepatic lipid levles and enzyme activities in rats administered with ethanol chronically, Sprague-Dawley male rats were AIN-76 diet(control), control diet plus ethanol, control plus Ixeris sonchifolia Hance diet, or control plus Ixeris sonchifolia Hance diet plus ethanol for 30 days. Ixeris sonchifolia Hance diets significantly decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and GOT levels that were increased due to the chronic ethanol administration. In addition, Ixeris sonchifolia Hance diets significantly decreased the liver triglyceride and total lipid levels that were increased due to the ethanol administration. The present findings, combined with previous data showing differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the serum Y-GPT activity of rats(Cha and OH[2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505) raise the possibility that GABA in plants could have a nytraceutical role in the recovery of chronic alcohol-related diseases. Effect of ethanol extracts from red pepper seeds on antioxidative defense system and oxidative stress in rats fed high-fat.high-cholesterol diet

  • The purpose of the present study was to investigate the effect of ethanol extracts from red pepper seeds on the antioxidative defense system and oxidative stress in rats fed a high fat high cholesterol diet. Rats were divided into four experimental groups which were composed of high fat high cholesterol diet group (HF), high fat high cholesterol diet with 0.1% ethanol extracts from red pepper seeds supplemented group (HEA), high fat high cholesterol diet with 0.2% ethanol extracts from red pepper seeds supplemented group (HEB) and high fat high cholesterol diet with 0.5% ethanol extracts from red pepper seeds supplemented group (HEC). Supplementation of ethanol extracts from red pepper seeds groups (HEA, HEB and HEC) resulted in significantly increased activities of hepatic glutathione peroxidase and catalase. Hepatic superoxide radical contents in microsome and mitochondria were significantly reduced in the groups supplemented with red pepper seeds ethanol extracts. Hepatic hydrogen peroxide content in the mitochondria was reduced in ethanol extracts from red pepper seeds supplemented groups. TBARS values in the liver were reduced in red pepper seeds ethanol extracts supplemented groups. Especially, HEB and HEC groups were significantly decreased compared to the HF group. Hepatic carbonyl values were significantly reduced in mitochondria in these supplemented groups. These results suggest that red pepper seeds ethanol extracts may reduce oxidative damage, by activation of antioxidative defense system in rats fed high fat high cholesterol diets.

갓(Brassica juncea) 첨가식이가 만성적인 알콜 투여시 흰쥐의 지방대사에 미치는 영향 (Effect of Leaf Mustard (Brassica juncea) on Lipid Metabolism of Rat with Chroethanol Administration)

  • This study was undertaken to investigate the effects of mustard leaf diet on the lipid metabolism under chronic alcohol administration. Sprague-Dawley male rats were fed either AIN-76 diet(control), control with ethanol, AIN-76 diet plus 5% mustard leaf(mustard leaf), or mustard leaf with ethanol lot 30 days. On the 21st day, all of the rats were given an oral dose of ethanol and blood-ethanol concentration were monitored for the next 5 hours. Lipid and enzyme determinations in serum and liver were carried out after 30 days. The results obtained were summarized as following: 1) Supplementing 5% of mustard leaf did not recover the body weight loss due to chronic alcohol administration. 2) There were no significant differences in blood ethanol concentrations among the experimental groups. 3) Mustard leaf diet decreased the plasma total cholesterol, triglyceride levels increased due to the chronic alcohol administration, but not HDL-, LDL-cholesterol, and liver lipids. 4) Mustard leaf diet decreased ${\gamma}$ -GTP level increased by chronic alcohol administration. Overall, these data suggest that mustard lear can have a recovery function, which was not via ethanol metabolism on the symptoms of alcohol related diseases.(Korean J Human Ecology 1(1) : 94~102, 1998)
