New Technology: The Ethanol Stress Response and Ethanol Tolerance of Saccharomyces cerevisiae (해외 기술: 효모 Saccharomyces cerevisiae의 에탄올 스트레스 반응과 에탄올 내성)

  • Saccharomyces cerevisiae는 전통적으로 알코올 음료와 bioethanol 생산에 이용되지만, 발효가 진행되는 동안 효모의 에탄올 생성은 에탄올의 축적에 의한 충격으로 세포활성에 손상을 초래한다. 본 연구는 S. cerevisiae의 에탄올 스트레스 반응과 에탄올 내성의 분자적 기초에 관해 수행되었으며, 에탄올 스트레스가 진행되는 동안 효모의 에탄올 생성 향상을 위한 유전 공학 전략의 수립에 활용될 수 있다. 이전의 연구들은 유전자 발현에 대한 에탄올 스트레스의 충격이 환경적 영향을 받기 때문에 다양한 균주와 조건들에 관해 이루어졌다. 그러나 에탄올 공격에 의해 영향을 받은 gene ontology 범주에서의 일부 공통점은 S. cerevisiae의 에탄올 스트레스 반응이 해당과정 및 미토콘드리아 기능과 관련된 유전자 발현의 증가와 에너지가 요구되는 성장과정과 관련된 유전자의 발현 감소에 따라 에너지 생산에 제약 받음을 의미한다. Genomewide screens를 이용한 연구는 vacuole function의 유지가 에탄올 내성에 대해 중요함을 암시한다. 아마도 단백질 turnover와 이온 항상성 유지에 이 세포기관의 역할이 중요하기 때문인 것으로 사료된다. 특히 에탄올 스트레스가 일어날 때 핵 내 Asr1과 Rat8의 축적은 비록 이 가설이 논란이 많은 주제로 남아있지만 S. cerevisiae가 에탄올 스트레스에 대한 특별한 반응을 가지고 있음을 의미한다.

Rescue of Oxidative Stress by Molecular Chaperones in Yeast

  • Heat shock proteins (HSPs) are induced in most living cells by mild heat treatment, ethanol, heavy metal ions and hypoxia. In yeast Saccharomyces cerevisiae, mild heat pretreatment strongly induces Hsp104 and thus provide acquired thermotolerance. The ability of hsp104 deleted mutant $({\triangle}hsp104)$ to acquire tolerance to extreme temperature is severely impaired. In providing thermotolerance, two ATP binding domains are indispensible, as demonstrated in ClpA and ClpB proteases of E. coli. The mechanisms by which Hsp104 protects cells from severe heat stress are not yet completely elucidated. We have investigated regulation of mitochondrial metabolic pathways controlled by the functional Hsp104 protein using $^{13}C_NMR$ spectroscopy and observed that the turnover rate of TCA cycle was enhanced in the absence of Hsp104. Production of ROS, which are toxic to kill cells radiply via oxidative stress, was also examined by fluorescence assay. Mitochondrial dysfunction was manifested in increased ROS levels and higher sensitivity for oxidative stress in the absence of Hsp104 protein expressed. Finally, we have identified mitochondrial complex I and Ferritin as binding protein(s) of Hsp104 by yeast two hybrid experiment. Based on these observations, we suggest that Hsp104 protein functions as a protector of oxidative stress via either keeping mitochondrial integrity, direct binding to mitochonrial components or regulating metal-catalyzed redox chemistry.

Antioxidant Effects of Elsholtzia splendens Extract on DMBA-induced Oxidative Stress in Mice

  • The present study was conducted to investigate the effects of flowers ethanol extract of Elsholtzia splendens (ESE) on the antioxidant defence system in mice with 7,12-dimethylbenz(a)anthracene (DMBA)-induced oxidative stress. The ESE was pre-administered orally to 2 groups of mice at 10 and 50mg/kg body weight (BW) for 5 weeks. Subsequently, mice with pretreatment of ESE received DMBA intragastrically at a dose of 34 mg/kg BW twice a week for 2 weeks. In DMBA alone group, biomarkers of oxidative stress (TBARS value, carbonyl content, and serum 8-OH-dG) were significantly increased. Also, the antioxidant enzymes were down-regulated. ESE significantly restored the TBARS value and carbonyl content at both doses, while a decrease in the elevated serum 8-OH-dG content was observed only at the higher dose. The DMBA-induced decreases in catalase and superoxide dismutase (SOD) activities were restored to nearly control levels by ESE. Glutathione peroxidase (GSH-px) and glutathione reductase (GR) activities, as well as the reduced glutathione (GSH)/oxidized GSH (GSSG) ratio, were significantly affected by ESE at the higher dose. These results suggest that ESE possesses antioxidant activity, which plays a protective role against DMBA-induced oxidative stress.

Study of the Cytotoxic Protective Effects of Jinsimyusaeng-hwan (Modified Ojayeonjong-hwan) Against Oxidative Stress (산화 스트레스에 대한 진심유생환(오자연종환 가감방) 추출물의 세포 독성 보호 효과에 관한 연구)

  • Objectives : This study was conducted to evaluate the cytotoxic protective effects of Jinsimyusaeng-hwan (Modified Ojayeonjong-hwan) against oxidative stress. Methods : 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azinobis-(3-ethyl-benzothiazoline-6-sulfonic acid)(ABTS) radical scavenging activity and ferric reducing ability of plasma (FRAP) method were used to estimate the antioxidant activity of Jinsimyusaeng-hwan. C2C12 myoblasts were used to reevaluate the antioxidant effects. And apoptosis analysis, mitochondrial membrane potential analysis, measurement of intracellular reactive oxygen species levels were conducted to investigate antioxidant activity of Jinsimyusaeng-hwan. Results : In comparison of DPPH free radical and ABTS cationic radical scavenging activity, it increased as the concentration of water extracts of Jinsimyusaeng-hwan(WEJ) and 70% ethanol extracts of Jinsimyusaeng-hwan (EEJ) increased. In the results of comparing the total phenol content and reducing power using the FRAP method, extracts with high total phenol content also showed high reducing power. In comparison of the protective effect against H2O2-induced oxidative stress in C2C12 myoblasts, WEJ had no significant effect, but the EEJ inhibited H2O2-mediated cytotoxicity in a concentration-dependent manner. The cytotoxic protective effect of EEJ against oxidative stress in C2C12 myoblasts was correlated with their inhibitory effects on H2O2-induced apoptosis and cell cycle arrest. In H2O2-treated C2C12 myoblasts, the apoptosis inhibitory effects of EEJ were associated with suppression of mitochondrial dysfunction and DNA damage. The protective effect of EEJ against H2O2-induced oxidative stress in C2C12 myoblasts were directly related to the inhibition of ROS generation. Conclusion : Jinsimyusaeng-hwan extracts have cytotoxic protective effects against oxidative stress, and it was better in 70% ethanol extract than in water extract.

The Effect of Powdered Herb of Aster scaber Thunb. on Antioxidant System in Ethanol-Treated Rats (참취 분말이 에탄올을 투여한 흰쥐의 항산화계에 미치는 효과)

  • The present study was conducted to investigate effect of powdered herb of Aster scaber Thunb. (chamchwi) on antioxidant system in ethanol-administrated rats. Four week-old Sprague Dawley male rats which had initial body weights of 97.10$\pm$4.50 g were randomly divided into three groups: control (ethanol treated, vitamin E-deficient group); 5% chamchwi (ethanol-treated, 5% chamchwi powder-supplemented group): 10% chamchwi (ethanol-treated, 10% chamchwi powder-suplemented group). Three groups of rats were suplemented with three experimental diets for 4 weeks and orally administrated 10% ethanol (v/v) daily via drinking water in the last experimental week. Contents of TBARS (thiobarbituric acid reactive substance). glutathione in liver and kidney and serum albumin were determined. The activities of superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) in liver and kidney were also analyzed. Relative weight of liver and spleen to body in chamchwi groups was lower than that in control group (p<0.05). The most remarkable result was that liver TBARS contents in chamchwi groups (5% chamchwi group, 46 $\mu\textrm{g}$ in MDA value; 10% chamchwi group, 35 $\mu\textrm{g}$) were significantly lower (p<0.05) than that in control group (66 $\mu\textrm{g}$). The supplement of chamchwi powder lowered the activity of manganese- superoxide dismutase (Mn-SOD), catalase in liver and GSH-px in kidney. The levels of glutathione in liver and kidney and serum albumin were not significantly different in all experimental groups (p<0.05). These results indicate that powdered herb of Aster scaber decreases lipid peroxidation and acitvity of Mn-SOD increased by alcohol-induced oxidative stress in liver of rats.

Ganoderma Lucidum Pharmacopuncture for Teating Ethanol-induced Chronic Gastric Ulcers in Rats

  • Objectives: The stomach is a sensitive digestive organ that is susceptible to exogenous pathogens from the diet. In response to such pathogens, the stomach induces oxidative stress, which might be related to the development of both gastric organic disorders such as gastritis, gastric ulcers, and gastric cancer, and functional disorders such as functional dyspepsia. This study was accomplished to investigate the effect of Ganoderma lucidum pharmacopuncture (GLP) on chronic gastric ulcers in rats. Methods: The rats were divided into 4 groups of 8 animals each: the normal, the control, the normal saline (NP) and the GLP groups. In this study, the modified ethanol gastritis model was used. The rats were administrated 56% ethanol orally every other day. The dose of ethanol was 8 g/kg body weight. The normal group received the same amount of normal saline instead of ethanol. The NP and the GLP groups were treated with injection of saline and GLP respectively. The control group received no treatment. Two local acupoints CV12 (中脘) and ST36 (足三里) were used. All laboratory rats underwent treatment for 15 days. On last day, the rats were sacrificed and their stomachs were immediately excised. Results: Ulcers of the gastric mucosa appeared as elongated bands of hemorrhagic lesions parallel to the long axis of the stomach. In the NP and GLP groups, the injuries to the gastric mucosal injuries were not as severe as they were in the control group. Wound healings of the chronic gastric ulcers was promoted by using GLP and significant alterations of the indices in the gastric mucosa were observed. Such protection was demonstrated by gross appearance, histology and immunehistochemistry staining for Bcl-2-associated X (BAX), B-cell lymphoma 2 (Bcl-2) and Transforming growth factor-beta 1 (TGF-${\beta}1$). Conclusion: These results suggest that GLP at CV12 and ST36 can provide significant protection to the gastric mucosa against an ethanol induced chronic gastric ulcer.

Antigastritic and Antiulcerative Activities of Water Extracts Derived from Scutellaria baicalensis

  • Gastritis and gastric ulcer were known to be induced by gastic acid, stress, ethanol, Helicobacter pylori and free radical, etc. This study was performed for the development of a new drug or nutraceutical from medicinal plants or natural products with anti-gastritis, anti-ulcerative and gastroprotective activities. The water extract of Scutellaria baicalensis was exhibited potent inhibition in HCl ethanol-induced gastric lesion, acetic acid-induced and Shay ulcers, indicating the effects on gastric lesion and ulcer in rats. The water extract of Scutellaria baicalensis significantly inhibited HCl ethanol-induced gastric lesions at the oral dose of 300, 500 mg/kg. In pylorus ligated rats, the treatments of the water extract from Scutellaria baicalensis showed decrease in the volume of gastric secretion and acid output and increase pH at oral dose of 300, 500 mg/kg. And significantly reduced acetic acid-induced ulcer at the oral dose of 500 mg/kg for 12 days. In this study, we have found that the water extract from Scutellaria baicalensis had significant improvement in acute gastritis and ulcer at the dose of 300, 500 mg/kg and in chronic gastritis and ulcer at the dose of 500 mg/kg. Also we evaluated the antibacterial activity against Helicobacter pylori treated with Scutellaria baicalensis. Scutellaria baicalensis had a equivalent antibacterial activity with ampicilin against H. pylori at the dose of $100\;{\mu}/ml$. In histological examination, the water extract of Scutellaria baicalensis drastically restored gastric damages induced by HCl ethanol solution, pylorus- ligature and acetic acid. Therefore, we may use the water extract from Scutellaria baicalensis as antigastritic and antiulcerative agent for the purpose of the improvement or treatment of gastritis and gastric ulcer.

Antioxidant, Anti-Wrinkle Activity and Whitening Effect of Fermented Mixture Extracts of Angelica gigas, Paeonia Lactiflora, Rehmannia chinensis and Cnidium officinale (당귀, 작약, 지황, 천궁 혼합 발효물의 항산화, 항주름 및 미백 효과)

  • Background: In this study, examined the effects of an extract of a mixture of Angelica gigas, Cnidium officinale, Paeonia lactiflora, and Rehmannia glutinosa fermented by Leuconostoc mesenteroides, with enhanced value and functionality. In oriental medicine, a mixture of these herbs is called Samultang. Methods and Results: In this study, we evaluated the effects of a fermented extract of Samultang on oxidative stress, procollagen type I expression, and melanin production. Samultang was extracted with 70% ethanol, followed by inoculation with Leuconostoc mesenteroides to obtain the fermented extract. The evaluation of viability of B16F10 cells and human foreskin fibroblast (HHF) revealed that both ethanol and fermented extracts of Samultang were non-toxic. The results of 1,1-diphenyl-2-picrylhydrazyl (DPPH) test showed that the fermented extract of Samultang ($SC_{50}value=100{\mu}g/m{\ell}$) was a more effective DPPH free radical scavenger than its ethanol extract. In addition, procollagen type I expression was higher in cells treated with the fermented extract of Samultang than in cells treated with ethanol. In the non-toxic concentration range, the fermented extract of Samultang showed strong inhibitory effect on melanin production in ${\alpha}-melanocyte$ stimulatin hormone-stimulated B16F10 cells ($IC_{50}=37.9{\mu}g/m{\ell}$). Conclusions: These results suggest that the fermented extract of Samultang has considerable protential as a cosmetic ingredient owing to its antioxidant, anti-wrinkle, and whitening effects.

Antioxidant Activity, Macamide B Content and Muscle Cell Protection of Maca (Lepidium meyenii) Extracted Using Ultrasonification-Assisted Extraction

  • This study aims to evaluate the efficacy of the Ultrasonication-Assisted (UA) extraction on the functionality of the herbaceous biennial plant maca (Lepidium meyenii). The specific objectives include comparison of the antioxidant activities among various maca extracts, determination of the macamide B content of the extracts, and in vitro evaluation of maca on cell viability and creatine kinase (CK) activity. The antioxidant activities of the water, ethanol, and UA extracts were compared by determining the total phenolic and flavonoid contents, the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities, and the ferric reducing antioxidant power (FRAP) of the extracts. The macamide B content of maca extracts were analyzed by HPLC. The effects of the extracts on muscle cell viability and creatine kinase activity were also determined using C2C12 myoblasts. UA extraction significantly increased the total phenolic content (2.90 GAE ㎍/mg, p < 0.05), without affecting the flavonoid content. DPPH radical scavenging activity did not exhibit any statistical difference among the extracts. The ethanol and UA extracts exhibited significantly higher FRAP than the water extract (p < 0.05). The macamide B content of ethanol and UA extracts were 0.087 and 0.083 ㎍/mg, respectively. The water and UA extracts exhibited higher C2C12 muscle cell viability than the ethanol extract, and both extracts resulted in a significantly lower CK level than the H2O2-treated control group. This research suggests that the maca extract can protect muscle cells and serve as an antifatigue agent under oxidative stress conditions.

Cell Viability and Hair Growth Effect on 3T3-L1 Cells of Ethanol Extract from Calendula officinalis L. Flower, Phellinus linteus Fruit Body and Houttuynia cordata Thunb. Whole Plant (금잔화, 상황, 어성초 에탄올 추출물의 세포독성 평가 및 3T3-L1 세포에 대한 육모 효과)

  • Background: Hair loss related syndromes are increasing due to environmental pollution and stress. Hair care products are mainly prepared by mixing chemicals and natural extracts, such as those obtained from medicinal plants. The purpose of this study was to investigate the effects of 70% ethanol extracts from the flowers of Calendula officinalis, fruit body of Phellinus linteus, and the whole plant of Houttuynia cordata on the growth of CCD-986 cells, hair follicle dermal papilla cells (HFDPC), and 3T3-L1 cells. Methods and Results: All sample extracts at all concentrations, except for that from P. linteus fruit body at $500{\mu}g/m{\ell}$, were cytotoxic to CCD-986 cells. However, none of the sample extracts were cytotoxic to HFDPC. The lipid differentiation of 3T3-L1 cells regulates hair regeneration via secretion of platelet derived growth factor. The 70% ethanol extract of H. cordata whole plant promoted hair growth. Adipogenesis rate significantly increased in a treatment concentration-dependent manner. Conclusions: These results suggest that 70% ethanol extracts of C. officinalis flower, P. linteus fruit body and H. cordata could be used for the development of hair care products.
