The activity of ${\alpha}$-amylase and ${\alpha}$-glucosidase as anti-diabetic function

  • BACKGROUND/OBJECTIVES: We investigated total 26 ingredients of Saengshik which will be commercially produced as an anti-diabetic dietary supplement. SUBJECTS/METHODS: Thirteen vegetables, nine cereals, three legumes and one seed were extracted with aqueous ethanol for 2 h at $60^{\circ}C$, and evaluated for their inhibitory effects against ${\alpha}$-amylase and ${\alpha}$-glucosidase and for total phenolic and flavonoid contents. RESULTS: All ingredients inhibited ${\alpha}$-amylase activity except cabbage. Strong inhibitory activity of ${\alpha}$-amylase was observed in leek, black rice, angelica and barley compared with acarbose as a positive control. Stronger inhibition of ${\alpha}$-glucosidase activity was found in small water dropwort, radish leaves, sorghum and cabbage than acarbose. All Saengshik ingredients suppressed ${\alpha}$-glucosidase activity in the range of 0.3-60.5%. Most ingredients contained total phenols which were in the range of 1.2-229.4 mg gallic acid equivalent/g dried extract. But, total phenolic contents were not observed in carrot, pumpkin and radish. All ingredients contained flavonoid in the range of 11.6-380.7 mg catechin equivalent/g dried extract. CONCLUSIONS: Our results demonstrate that Saengshik containing these ingredients would be an effective dietary supplement for diabetes.

향나무 추출물의 항말라리아 효과 (In vitro Anti-malarial Activity of Juniperus Chinensis Extract)

  • This study was carried out to investigate the anti-malarial activity of Juniperus chinensis by in vitro and in vivo system using Plasmodium falciparum chloroquine-sensitive(3D7) and P. falciparum chloroquine-resistant(S20) strains. According to cytotoxicty test on NIH 3T3 cell, the ethanol extract(EtOH), ethylacetate(EtOAc) fraction and aqueous fraction possessed significant anti-malarial activity against both 3D7 and S20 strains at non-toxic concentrations(<100 /). In vitro assay, EtOAc fraction showed notable activity against 3D7 and S20 strains of P. falciparum with $IC_{50}$ values of $37{\pm}2{\mu}g/ml$ and $36{\pm}6{\mu}g/ml$. In animal test using P. falciparum infected human erythrocytes, the treatment of EtOAc fraction significantly inhibited parasitaemia in mice in a dose-dependent manner that is parasitaemia of 42%, 34% and 31% in doses of 10 mg/kg, 20 mg/kg and 40 mg/kg, respectively. The study provides data to support the medicinal importance of the J. chinensis.

Antimicrobial Activities of White, Red, and Extruded Ginsengs with Different Extraction Conditions

  • White, red, and extruded ginsengs were studied against 8 strains of food-borne pathogens and/or food spoilage microorganisms. The ginseng powders were extracted with different extractants and screened for antimicrobial activity using the disc diffusion and broth dilution techniques. The results showed that the yield of extraction was higher with increase of aqueous solution content and temperature. Preliminary screening revealed that the red ginseng extracts were most active, that has been found to be highly effective against all tested microbe except Listeria monocytogenes. Moreover, Bacillus subtilis has shown highly susceptible, which the diameters of inhibition zone values of 28 extracts were between 7 and 14 mm. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) recorded for the different crude ginseng extracts against microorganism using ranged from 6.25 to 100 mg/mL, indicated that the methanol extract of ginseng were more effective than ethanol and water extracts. The 60% methanol extract of red ginseng had the greatest effects against B. subtilis with MIC and MBC values at 6.25 mg/mL.

비방탈명산(秘方奪命散)의 항산화 ${\cdot}$ 항암 ${\cdot}$ 항균 효과 연구 (Study on the Anti-oxidative, Anti-microbial and Anti-cancer effect of Bibangtalmyungsan.)

  • Objectives : This study was carried out to evaluate anti-oxidative, anti-microbial and anti-cancer effect for clinical application of Bibangtalmyungsan (BTS) Results : 1. The oxidative effects were measured by polyphenol, DPPH radical scavenging activity. BTS water extract was showed more effective than ethanol extract and also various solvent fractions from BTS water extract showed effective in the following order : ethyl acetate fraction > butanol fraction > hexane fraction ${\fallingdotseq}$ chloroform fraction ${\fallingdotseq}$ aqueous fraction 2. The results of anti-microbial effects were as follows. 1) Antibacterial activities of BTS extracts against Gram's negative and positive bacteria were ineffective 2) Antifungal activities of the BTS extracts against Aspergillus spp. , Trichohyton mentagrophyte KTCC 1077 were not effective. 3. The result of anti-cancer effects were as followings: 1) BTS ethanol extract was more effective than water extract against Caco-2, Calu-6, but it had cytotoxic effect against NIH3T3. 2) On the Caco-2, effective only in ethyl acetate $fraction(IC_{50}:$ 35.81 ${\mu}g/ml)$. 3) On the Calu-6, the most effective in ethyl acetate $fraction(IC_{50}:$ 189.65 ${\mu}g/ml)$ and effective In butanol $fraction(IC_{50}:$ 299.74 ${\mu}g/ml)$ and hexane $fraction(IC_{50}:$ 345.13 ${\mu}g/ml)$ 4) On the SUN-601, $IC_{50}$ value was within 80 ${\mu}g/ml$ in ethyl acetate fraction and hexane fraction. 5) On the HCT-1l6, the most effective in ethyl acetate $fraction(IC_{50}:$ 82.94 ${\mu}g/ml)$ and effective in hexane $fraction(IC_{50}:$ 374.56 ${\mu}g/ml)$. 6) On the AML-2/WT, the most effective in ethyl acetate $fraction(IC_{50}:$ 41.44 ${\mu}g/ml)$ and effective In hexane $fraction(IC_{50}:$ 303.01 ${\mu}g/ml)$. 7) On the NIH3T3, effective only in ethyl acetate $fraction(IC_{50}:$ 203.42 ${\mu}g/ml)$, but it was more ineffective than other cancer cells. Conclusion : These result suggest that BTS has antioxidative, antifungal activities and cytotoxic effects against Caco-2, Calu-6, SUN-601, HCT-116, AML-2/WT and NIH3T3, especially ethyl acetate fraction from water extract has more effective in antioxidative and anticancer effects.

건조방법에 따른 께묵뿌리 추출물의 수소공여능 조사 (Changes in Hydrogen Donating Activities of the Extract from Hololeion maximowiczii Root by Drying Methods)

  • 께묵(Hololeion maximowicizii)뿌리를 냉동건조, 열풍건조 및 증숙후 열풍건조의 3가지 방법으로 건조한 후 용매추출하여 DPPH 방법에 의한 수소공여능이 변화를 조사하였다. 증숙한 후 열풍건조구가 가장 큰 수소공여능을 나타냈으며, 여러가지 용매 추출물중에서 물추출물의 색도가 가장 높았으나 수소공여능은 50% ethanol 추출물에서 가장 높게 나타났다. 냉동건조한 께묵뿌리를 $100^{\circ}C$에서 120분까지 열처리 한 위에 흡광도를 조사한 결과는 가열 20분까지는 감소하다가 증가하였으나 수소공여능은 계속 증가하였다. 50% ethanol 추출물(6.05mg/ml)은 $2{\times}10^{-4}M$${\alpha}-tocopherol$과 비슷한 수소공여능을 보였으며, 투석외액과 내액의 수공여능을 비교한 결과 외액이 내액보다 강하였다.

번데기동충하조(Cordyceps militaris) 추출물의 세포독성 및 유전독성 억제효과 (Cytotoxicity and Antigenotoxic Effects of Cordyceps militaris Extracts)

  • 각종 암세포에 대한 번데기동충하초(Cordyceps militaris) 추출물 및 분획물의 세포독성을 규명하기 위하여 암세포로 A549, MCF-7 HeLA, HT1080, Hep3B, KATOIII 및 K562를 이용하였고 그 결과, SRB assay에 의한 1 mg/mL의 에탄올 추출물 농도에서 HT1080, HeLa, Jep3B 그리고 A549는 각각 89.4%, 85.7%, 72.9%그리고 65.5%의 억제효과를 나타내었다. MCF7, HeLa 그리고 HT1080 세포의 경우는 핵산 분획물 1 mg/mL 농도에서 각각 92.9%, 90.3%그리고 97.0%로 다른 분획물보다 현전히 높은 억제효과를 나타냈다. KATOIII세포에 대한 억제효과는 1mg/mL 투여시 에탄올 추출물의 경우 61.5%를 보였고 부탄올과 물 분획물에서 각각 83.7%와 80.4%로 다소 높은 억제효과를 보였다. K562 세포에 대한 시료의 억제효과는 에탄올 추출물(1mg/mL)이 60.5%의 억제율을 보인데 반해 같은 농도에 서 에틸 아세테이트, 부탄올 및 핵산 분획물에서는 각각 88.6%, 806% 및 75.6%로 높은 억제 효과는 양성대조군의 11.2$\pm$0.8에 비하여 10, 20, 40, 80 mg/kg 의 농도에 10.5$\pm$0.8, 8.3$\pm$0.5, 7.8$\pm$0.3, 3.7$\pm$0.6로 8.7, 25.9, 30.4그리고 67.0%의 유의적인 억제효과를 나타내었다. 또한 각각의 분획물에 대한 소핵생성 억제효과는 유전손상물질(MNNG;150 mg/kg, I.P)만 투여한 양성대조군에 비해서 시료 농도를 10, 20, 40 80 mg/kg 처리하였을 경우 동이라한 시료 농도 80 mg/kg 에서 부탄올, 에틸 아세테이트, 물, 그리고 클로로포름 분획물이 각각 3.1$\pm$0.3, 3.7$\pm$0.6, 4.2$\pm$0.3, 4.8$\pm$0.3으로 72.3%, 67.0%, 63.3%그리고 57.1%의 순으로 소핵생성 억제효과를 나타내었으며, 그중 핵산 분획물이 2.8$\pm$0.5의 75.0%로 가장 높은 소핵생성 억제효과를 나타내었다.

Simplified the Screening and In Vitro Appraisal of Antioxidant, Cytotoxic, Thrombolytic, Antimicrobial and Membrane Stabilizing Activities of Lablab Purpures at a Time

  • To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.

블루베리 추출물의 항산화 활성 (Antioxidant activity of extracts from Blueberry)

  • 본 연구에서는 제주산 블루베리의 항산화활성을 알아보기 위하여 블루베리로부터 에탄올과 효소적 가수분해를 이용하여 각각의 추출물을 제조한 후 이들로부터 활성산소종의 소거활성을 측정하였다. 여기에서 효소 추출물보다 에탄올 추출물이 추출수율은 낮았으나, 폴리페놀 함량과 DPPH radical 소거활성이 모두 높게 나타났다. 특히 70% 에탄올 추출물은 효소추출물보다 폴리페놀 함량은 1000 mg 이상 높게 나타났으며, DPPH radical 소거활성은 125 ${\mu}g$/mL sample로 측정하였을 때에 크게는 53%작게는 27%높게 나타났다.

국내 천연물 항 당뇨 실험연구의 체계적 논문 고찰 - 2000년 ~ 2010년 (Systematic Reviews of Current Domestic Studies of Herbaceous Plants on Anti-diabetes - since 2000)

  • This study tried to integrate the traditional oriental medical theories and results of experimental studies of herbaceous plants on anti-diabetes. And I tried to analyze recent experimental study trend on the anti-diabetic herb. I searched anti-diabetic herb studies on 4 korean databases and 10 korean journals by keywords, 'diabetes', 'blood glucose', 'glycometabolism', 'pancreatic ${\beta}$-cell', etc. In order to see detail review, searching was performed from 2000 to 2010. And I searched 125 study cases concerning anti-diabetic herb and 72 varieties herbaceous plants used in study of anti-diabetes. and I analyzed the choice motives of each herb for anti-diabetic study, the extract methods and anti-diabetic evaluation contents. And I analyzed anti-diabetic herbs from a traditional oriental medical point of view. When the researchers chose herb for anti-diabetic experiment, just 8.8% of the choice was based on the oriental medical evidences. I found that 60.6% of the herb shown to be effective in diabetes experimentally had oriental medical theory-based Properties(性). There were studies with whole plants(16.8%), aqueous extract(45.6%), methanol extract(8.0%), ethanol extract(8.0%) and comparative studies of more than 2 types of extracts or various fractions(18.4%). The most frequent experimental diabetic models was diabetic mouse induced by streptozotocin(STZ)(87.8%). And there were db/db mouse(6.7%), ob/ob mouse(1.1%), etc. 33.6% of all studies just measured hematological indices of diabetes, and 66.4% researches analyzed details. To improve herbaceous plants study on diabetes, we oriental medical scientists have to integrate the oriental medical theories and results of experimental studies.

갈파래(Ulva lactuca) 용매별 분획의 항산화활성 (Antioxidant Activities of Ulva lactuca Extracts with Different Solvents)

  • 갈파래(Ulva lactuca)의 용매별 분획의 항산화활성 측정을 위해 갈파래를 95% ethanol로 추출하고 diethyl ether, ethyl acetate, $H_2O$ 등의 용매로 분획하였다. 각 분획을 HPLC로 분석한 결과 각 분획에서 단일 peak가 검출되었으며 용출시간이 서로 달랐다. 각 분획의 항산화 활성을 DPPH 라디칼 소거능, 지질과산화억제활성 방법으로 검증하였다. 각 분획의 DPPH 라디칼 소거활성은 농도의존성을 나타내었고 활성은 수층, ethyl acetate 층, diethyl ether층 분획의 순서였다. 지질과산화 억제활성은 diethyl ether 분획에서 가장 높게 나타났으며 같은 농도의 $\alpha-tocopherol$과 비슷하였다. 본 연구 결과는 갈파래를 이용한 천연 항산화제 생산에 응용이 가능할 것으로 생각된다.
