Aqueous extract of Laurus nobilis leaf accelerates the alcohol metabolism and prevents liver damage in single-ethanol binge rats

  • BACKGROUND/OBJECTIVES: Excessive alcohol consumption has harmful health effects, including alcohol hangovers and alcohol-related liver disease. Therefore, methods to accelerate the alcohol metabolism are needed. Laurus nobilis is a spice, flavoring agent, and traditional herbal medicine against various diseases. This study examined whether the standardized aqueous extract of L. nobilis leaves (LN) accelerates the alcohol metabolism and protects against liver damage in single-ethanol binge Sprague-Dawley (SD) rats. MATERIALS/METHODS: LN was administered orally to SD rats 1 h before ethanol administration (3 g/kg body weight [BW]) at 100 and 300 mg/kg BW. Blood samples were collected 0.5, 1, 2, and 4 h after ethanol administration. The livers were excised 1 h after ethanol administration to determine the hepatic enzyme activity. The alcohol dehydrogenase (ADH), aldehyde dehydrogenase (ALDH), superoxide dismutase (SOD), and glutathione peroxidase (GPx) activities in the liver tissue were measured. RESULTS: LN decreased the serum ethanol and acetaldehyde levels in ethanol-administered rats. LN increased the hepatic ADH and ALDH activities but decreased the alanine aminotransferase, aspartate aminotransferase, and gamma-glutamyl transferase activities in the ethanol-administered rats. In addition, LN inhibited lipid peroxidation and increased the activities of SOD and GPx. CONCLUSIONS: LN modulates the mediators of various etiological effects of excessive alcohol consumption and enhances the alcohol metabolism and antioxidant activity, making it a potential candidate for hangover treatments.

어성초 추출물을 이용한 면과 견직물의 항미생물성 염색 (Antimicrobial Dyeing of Cotton and Silk Fabrics Using Houttuynia cordata Extract)

  • Cotton and silk fabrics were dyed with Houttuynia cordata extract using aqueous ethanol solution and the dyeing and post-treatment conditions were optimized to impart antimicrobial activity to the fabrics. The dried Houttuynia cordata can be extracted at $80^{\circ}C$ for 3 hours using an aqueous ethanol solution containing 70%(w/w) ethanol. For the highest color yields. Both cotton and silk fabrics can be dyed at $100^{\circ}C$ for 60min with 10g/L of NaCl under pH 4. Silk fabrics can be dyed to higher K/S than cotton fabrics. The color fastness properties of the dyed fabrics were good when either citric acid crosslinking or aluminum alum mordanting was carried out as a post treatment. The dyed silk and cotton fabrics with the post treatments showed excellent antimicrobial activity against both Staphylococcus aureus and Klebsiella pneumoniae.

  • The ethanol extracts from Angelica gigas Nakai and Angelica acutiloba Kitagawa were fractionated to diethyl ether and aqueous partitions. Both partitions had strong antimutagenic effect on the MNNG (N-methyl-N-nitro-N-nitrosoguanidine) by Ames mutagenicity test. Diethyl ether fractions exhibited the greatest antimutagenic effect suppressing the mutagenicity of MNNG with inhibition of 78-80%. The ethanol extracts from both species showed the inhibitory effect on the growth of several human cancer cell lines. Especially, the diethyl ether fraction from ethanol extracts was most effective on human hepatocellular carcinoma cells, inhibiting 90-95% of cell growth. However, the aqueous fractions had least inhibition activity on many cancer cells. There was little cytotoxicity on human normal liver cell by ethanol extracts. Diethyl ether fraction from Angelica gigas Nakai ethanol extract had cytotoxicity less than 20% on human normal liver cells, compared with that from Angelica acutiloba Kitagawa ethanol exract. The adding of 0.5 (g/l) of diethyl ether fractions of Angelica gigas Nakai or Angelica acutiloba Kitagawa increased immune activity by enhacing human B and T cells up to three to four times. It was proven that diethyl ether fraction (0.7 g/1) from Angelica gigas Nakai could control blood pressure by suppressing angiotensin converting enzyme activity up to 98%. From TLC, it was appeared that both of diethyl ether partitions had umbelliferon, known to one of active substances from Angelica gigas Nakai and Angelica acutiloba Kitagawa.

생열귀나무 잎 추출물과 분획물의 항산화 및 NO생성 억제 활성 (Anti-oxidant and Inhibitory Activity on NO Production of Extract and its Fractions from Rosa davurica Pall. Leaves)

  • In the present study, we investigated biological activities of Rosa davurica Pall. leaves in order to evaluate the possibility as a natural biomaterial. The 80% ethanol extract and its subsequent fractions of Rosa davurica Pall. leaves were prepared using several solvents with different polarities. The extraction yield was 33.4, 36.6, 25.2, 18.7, 5.8 and 5.8% in ethanol extract, aqueous, butanol, ethyl acetate, n-hexane, chloroform fractions, respectively. The ethyl acetate fraction (661.38 mg/g), butanol fraction (396.68 mg/g) and 80% ethanolic extract (239.54 mg/g) has higher total polyphenol contents than other fractions. The antioxidant activity was detected in ethanolic extract, ethyl acetate and butanol fractions. The ethyl acetate fraction showed the highest levels of DPPH radical scavenging activity ($IC_{50}$, $4.77{\mu}g/m{\ell}$). Moreover, the ethyl acetate fraction significantly inhibited production of NO in LPS-stimulated macrophage RAW 264.7 cells without cytotoxicity. These results indicate that 80% ethanol extract and its fractions of Rosa davurica Pall. leaf, especially ethyl acetate fraction, have the properties of anti-oxidant and anti-inflammation, suggesting leaf of Rosa davurica Pall. may be a candidate for natural and functional materials.

  • This study examined the antimicrobial activity of Myagropsis yendoi (MY). The antimicrobial activity of ethanol and aqueous MY extracts were measured using a paper disc assay. The MY ethanol extract had the best antimicrobial activity. It inhibited the growth of Candida tropicalis and Gram-positive bacteria such as Bacillus subtilis, Listeria innocua, L. monocytogenes, and Staphylococcus aureus at 4 mg/mL. The minimum inhibitory concentration (MIC) of the MY ethanol extract ranged from 0.1 to 0.5%. The MY ethanol extract inhibited the growth of B. subtilis and L. innocuaat concentrations of 0.15% and 0.1%, respectively. The antimicrobial activity of the MY ethanol extract was not affected by heating at $121^{\circ}C$ for 15 min or pH 2.10. Therefore, the main substances responsible for the antimicrobial activity of the MY ethanol extract are believed to be stable with changing heat and pH.

에탄올의 약동학에 미치는 홍삼액기스의 효과 (Effects of Red-Ginseng Extract on Pharmacokinetics of Ethanol)

  • 홍삼액기스의 투여가 에탄올의 곡선하면적(AUC)과 제거속도상수(elimination rate constant)에 미치는 효과에 대하여 웅성 백서 14마리와 건강한 남성 자원자 10명을 대상으로 연구하였다. 백서에게 홍삼액기스 200 mg/kg를 투여하거나 같은 분량의 물을 투여한 후 즉시 $50\%$ 에탄올, 3.2g/kg의 용량을 구강 투여하였다. 에탄올의 곡선하면적(AUC)과 제거속도상수는 홍삼액기스를 투여할 경우 $29.2{\pm}6.2\;g{\cdot}min.{\cdot}dl^-$와, $0.51{\pm}0.06\;mg{\cdot}dl^-{\cdot}min.^-$를 나타내었으며, 홍삼액기스를 투여하지 않을 경우는 각각 $28.0{\pm}5.6\;g{\cdot}min.{\cdot}dl^-$$0.5{\pm}0.1\;mg{\cdot}dl^-{\cdot}min.^-$를 나타내었다. 이들 차이는 통계적으로 유의하지 않았다. 또한 인체에서는 일인당 홍삼액기스 3그램을 100 ml의 물에 희석하거나 또는 동일분량의 물을 투여하고 곧 이어서 소주 2.4 m/kg의 용량을 마시게 하였다. 자원자의 에탄올에 대한 곡선하면적과 제거속도상수는 홍삼액기스의 투여군이 $10.6{\pm}2.0\;g{\cdot}min.{\cdot}dl^{-}$$0.21{\pm}0.05\;mg{\cdot}dl^-{\cdot}min.^-$를,대조군에서는 $11.0{\pm}2.2\;g{\cdot}min.{\cdot}dl^{-}$$0.22{\pm}0.04\;mg{\cdot}dl^-{\cdot}min.^-$를 나타내었으며 역시 두 군간의 차이는 없었다. (평균${\pm}$표준편차) 이런 결과들로 미루어 홍삼액기스의 투여로 에탄올의 약동학에 의미있는 변화를 초래하기란 어려울 것으로 생각된다.

남천(Nandina domestica) 열매 분획 추출물의 항산화 효과 (Antioxidative Effects of Solvent Fractions from Nandina domestica Fruits)

  • 남천(Nandina domestica) 열매를 물과 에탄올 추출 후 극성이 상이한 유기용매를 순차적으로 분획하여 각 추출물의 생리활성을 측정하였다. 폴리페놀은 EE(ethyl acetate fraction of ethanol extract)에서 922.22 mg/g이었으며, 플라보노이드는 EB(butanol fraction of ethanol extract)에서 282.49 mg/g을 함유하였다. WA(aqueous of water extract)와 EH(n-hexane fraction of ethanol extract)는 0.1 mg/mL에서 93% 이상의 전자공여활성을 나타내었으며, 분획물의 농도 증가에 따른 전자공여능의 유의적 차이는 없었다. SOD 유사활성은 1.0 mg/mL의 EA(aqueous of ethanol extract)에서 56.36%로 가장 우수하였으며, EH와 EC(chloroform fraction of ethanol extract)에서도 50% 이상의 활성을 나타내었다. 아질산염 소거는 pH 1.2의 EC에서 82.03%로 가장 높았다. Xanthine oxidase 저해율은 0.5 mg/mL에서 WE를 제외한 모든 분획물에서 90% 이상의 저해율을 나타내었으나, WE는 46.75%로 가장 우수한 tyrosinase 저해효과를 나타내었다. 이상의 결과 남천 열매 분획물은 다량의 폴리페놀과 플라보노이드를 함유하고 생리활성 효과가 우수하여 천연 항산화 제재 및 이를 이용한 의약품 개발 가능성을 지닌 약용 식물자원인 것으로 판단된다.

수삼추출물 및 Glucose 또는 Arginine첨가 추출물의 특성과 항산화작용에 대하여 (The Characteristics and Antioxidant Activity of Non-enzymatic Browsing Products from Fresh Ginseng Bxtracts and Those with Arginine or Glucose)

  • Color is one of the most important quality factors of red ginseng (Hong-sam) which is processed from fresh ginseng (Panax ginseng C. A. Meyer). Therefore, a study of characteristics of browning mixtures of aqueous fresh ginseng extracts, factors which accelerate the browning of the aqueous extracts, and the antioxidant activity of the browning mixtures may contribute to the improvement of the color and other quality of red ginseng and other ginseng products such as ginseng extracts. In the present study, factors which affect the Maillard-type browning reaction of aqueous extracts of fresh ginseng roots were investigated firstly by adding various concentrations (0.001-0.5M) of arginine or glucose solutions, by varying the browning reaction temperatures and durations. Secondly, some characteristics such as brown color intensity, amounts of water-soluble and ether-soluble extracts, amounts of non-dialyzable materials, pH, viscosity, and reactivity with 2,2'- diphenyl -1 - picrylhydrazyl and antioxidant activity of the browning mixtures of the aqueous fresh ginseng extracts with small amounts of 0.1 M arginine, 0.1 M glucose, and distilled water at various browning temperatures and reaction time were studied. The results of the present study are as follows. 1. Color intensity (absorbance at 470 nm) of the browning mixtures was increased by adding various concentrations of arginine solution to the fresh ginseng extract, but the addition of the same amount of glucose solution did not increase the color intensity. 2 The amounts of water- or ether-soluble extracts, amounts of non-dialyzable materials were slightly greater in case of the browning mixtures of the fresh ginseng extract with 0.1M arginine solution than in case of the browning mixtures of the fresh ginseng extract with the same amount of 0.1 M glucose solution. In the process of the browning reaction, the pH of the browning mixtures of the fresh ginseng extract with 0.1 M arginine solution decreased slightly, while that of the browning mixtures with 0. 1 M glucose solution was almost constant. 3. The color intensity (absorbance at 470 nm) of the browning mixtures of the fresh ginseng extract with 0.1 M arginine or 0.1 M glucose solutions did not correlate well with the reducing power or the antioxidant power of the browning mixtures. The antioxidant activity of 90% ethanol extracts from the earlier stages of the browning mixtures of the fresh ginseng extract with the arginine solution was almost comparable to that of the 90% ethanol extracts from the later stages of the corresponding browning mixtures. The browning mixtures of only the fresh ginseng extract or of the fresh ginseng extract with the glucose solution showed considerable antioxidant activity, although both showed less brown color intensity than the fresh ginseng extract with he arginine solution.

  • 관중의 추출분획물별과 Bacillus subtilis에 강한 항균활성을 보였던 가자, 정향, 단삼, 관중의 추출 용매별 (ethanol과 물)로 식품 부패성 미생물인 Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Pseudomonase fluorescence에 대한 항균활성을 조사하였던 바 다음과 같은 결론을 얻었다. 1. 가자는 물추출물이, 정향, 단삼 및 관중은 ethanol 추출물이 각각 강한 항균활성을 나타내었으며, 그 중 관중의 ethanol 추출물이 가장 강한 활성을 보였고 그 활성정 도는 인공합성보존류인 안식향산나트륨 (sodium benzoate)보다 실험한 모든 미생물에 대하여 강했다. 2. 관중의 ethanol 추출물의 각분획물에 대한 항균활성은 CHCI$_3$분획물>EtOAc 분획물>BuOH 분획물>물 분획물 순이었다. 3. 관중의 각분획물은 농도 의존적으로 미생물의 성장 억제효과가 있었으며, 그 억제효과에 대한 기작은 살균작용에 의한 것이라기보다는 정균작용에 의한 것으로 사료된다.

Antiarthritic action of polar fraction from Sida rhombifolia aerial parts

  • Aerial parts of Sida rhombifolia Linn. (Malvaceae) were extracted successively using various solvents and screened for various parameters of antiarthritic activity such as adjuvant-induced arthritis, motor performance, and histopathological study. The ethanol and the aqueous extracts showed potent activity; further these extracts were fractionated by using column chromatography. The fraction ET1 isolated from ethanol extract showed the most potent antiarthritic activity.
