택사의 항균 및 항진균 작용에 관한 연구 (Antimicrobial and Antifungal Studies on Alismae Rhizoma)

  • We previously reported the morphological, physiochemical characteristics of Alismae Rhizoma. In this study, we examined the antimicrobial and antifungal activities of 85% ethanol and water extracts of Alismae Rhizoma using disc method. Ethanol extracts of Korean Alismae Rhizoma showed antimicrobial activity on B. subtilis and S. aureus on the concentration dependent manner, whereas, Chinese Alismae Rhizoma revealed antimicrobioal activity on E. coli and N. gonorrhoeae. The water solution(500 mg/ml) of each lyophilized powder of aqueous extracts didn't show any antimicrobial activity on B. subtilis, S. aureus and E. coli, on the contrary, they stimulated cell growth slightly. Ethanol extracts of Korean samples showed antifungal activity on A. niger, but not on C. albicans. Neither A. niger nor C. albicans were affected by Chineses samples.

Ethanol but not Aqueous Extracts of Tubers of Sauromatum Giganteum(Engl.) Cusimano and Hett Inhibit Cancer Cell Proliferation

  • Background: Both alcohol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, the dried root tuber of which is named Baifuzi in Chinese, have been used for folklore treatment of cancer in Northeast of China. However, little is known about which is most suitable to the cancer therapy. Materials and Methods: Serum pharmacology and MTT assays were adopted to detect the effects of ethanol and aqueous extracts of Sauromatum giganteum(Engl.) Cusimano and Hett, prepared by heat reflux methods, on proliferation of different cancer cells. Results: Cancer cells treated with medium supplemented with 10%, 20%, 40% serum(v/v) containing ethanol extract had a decline in viability, with inhibition rates of 7.69%, 21.8%, 41.9% in MCF-7 cells, 42.8%, 48.1%, 51.8% in SGC-7901 cells, 44.1%, 49.2%, 53.7% in SMMC-7721 cells, 6.8%, 15.2%, 39.8% in HepG2 cells, 7.57%, 16.3%, 36.2% in HeLa cells, 6.24%, 12.5%, 27.4% in A549 cells, and 7.20%, 17.5%, 31.3% in MDA-MB-231 cells, respectively. Viability in the aqueous extract groups was no different with that of controls. Conclusions: An ethanol extract of Sauromatum giganteum(Engl.) Cusimano and Hett inhibited the proliferation of SMMC-7721, SGC-7901 and MCF-7 cells, which supports the use of alcoholic but not aqueous extracts for control of sensive cancers, which might include hepatocarcinoma, gastric cancer and breast cancer.

볶음처리로 생성된 인삼박 추출액의 분리와 특성 (Separation and Characteristics of the Extract Solution from Ginseng Marc by Roasting Process)

  • Roasting ginseng marc was roasted at different temperatures (140, 170, 200, 23$0^{\circ}C$) and for different periods (10, 20, 30 min) produced aqueous soluble brown pigments, gel filtration of which over Sephadex G-50 yielded 3 fractions A, B, C. The treatment at higher temperature and for longer time lead to increase of peak A and decrease of peak C. The contents of the brown pigments and the degree of brown color increased about 4 times and over 6 times, respectively, by roasting at 23$0^{\circ}C$ and for 30 min as compared to the control. 5-Hydroxymethyl furfural in aqueous and 50% ethanol extracts of treated samples at 23$0^{\circ}C$ and for 30 min was increased to 3.6 times and 8 times, respectively, and carbonyl compound in both aqueous and 50% ethanol extracts was increased 3 times. Also pyroxene-like substance was increased apparently in both aqueous and 50% ethanol extracts of treated samples.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 마치현의 억제 작용 (The Inhibitory Effects of Portulaca oleracea L. on HCl-ethanol Induced Gastritis in Rats)

  • Objectives : The objective of this study was to examine the effects of P. oleracea into the HCl-ethanol induced gastritis in rats, and to isolate and determine the chemical compounds from P. oleracea. Methods : The rats were orally administered with crude extract or fractions or isolated compounds of P. oleracea 30 mins before the induction of gastric lesion by oral administration of HCl-ethanol. The gastric lesional area was measured using pixel counting software. Then the chemical compounds from P. oleracea was isolated and determined by LC-MS and NMR. Results : The inhibition effect of oral administration of crude extract of P. oleracea at a dose of 500 mg/kg in HCl-ethanol induced gastritis was similar to cimetidine. Then, aqueous fraction at a dose of 240 mg/kg exhibited the effects similar to cimetidine. Then, the aqueous fraction was further separated by MPLC and yielded four sub fractions. Among those sub fractions, agent II at a dose of 40 mg/kg possessed the strongest effect in the HCl-ethanol induced gastritis. The water fraction yielded-Uridine, Adenosine, Guanosine, which were characterized by Mass, 1H-NMR, 13C-NMR. Conclusions : This study suggest that a P. oleracea and its compounds showed potent efficacy on the development of HCl-ethanol induced gastritis. Thus, P. olaracea can be a potential natural resource for the management of gastritis although the mechanism of action involved in the treatment remains to be explored.

Thioacetamide 유발 흰 쥐 간독성에 대한 인삼 사포닌 및 에타놀 추출물의 효과 (Effects of Saponin and Ethanol Extract of Panax Ginseng against Thioacetamide - Induced Hepatotoxicity in Rats)

  • Panax ginseng has been used for various diseases including hepatic disorders. The aim of the present study was to investigate the hepatoprotective effects of ethanol extract and saponin of Panax ginseng in thioacetamide-intoxicated rats and to compare with silymarin, a known hepatoprotective agent. Male Sprague-Dawley rats were given single intragastric administration of thioacetamide. Aqueous solutions of ethanol extract and saponin of Panax ginseng with or without silymarin were administered intragastrically daily for six days from four days before until one day after thioacetamide administration. At the end of the treatment, the rats were fasted overnight and sacrificed. As a result, thioacetamide caused significant increase in serum levels of AST, ALT, 5'-nucleotidase and bilirubin. Thioacetamide increased $Ca^++$ content but decreased protein content in liver tissue. These thioacetamide-induced biochemical changes were prevented both by ethanol extract of ginseng and silymarin, but not by ginseng saponin. Silymarin did not potentlate the effect of either ethanol extract or saponin of ginseng on these parameters. Thioacetamide-induced confluent necrosis was not protected by the test drugs. In conclusion, ethanol extract of ginseng protects the liver possibly by stabilizing the cell membrane and by inhibiting thioacetamide-induced $Ca^++$ increase in the hepatocytes, which was comparable to that of silymarin.

톳(Hizikia fusiformis) 에탄올 추출물 및 분획물의 미백활성 (Whitening Effect of Hizikia fusiformis Ethanol Extract and Its Fractions)

  • 멜라닌 생합성은 tyrosinase의 효소적 산화반응에 의해 일어난다. 톳(Hizikia fusiformis)의 미백효과를 검증하기 위하여 메탄올, 헥산, 부탄올 및 물을 용매로 이용한 분획과정을 통해 톳 에탄올 추출물로부터 분획물을 제조하였다. 에탄올 추출물과 분획물들을 대상으로 B16F10 흑색종 세포에서 tyrosinase 활성 저해효과 및 멜라닌 생합성 억제효과를 평가하였다. 생쥐의 에탄올 추출물 및 수층 분획물은 미백효과를 나타냈지만, 세포독성은 나타내지 않았다. 에탄올 추출물은 시료들 중에서 가장 높은 미백활성을 보였다. 에탄올 추출물은 $100{\mu}g/ml$의 농도에서 arbutin $10{\mu}g/ml$의 활성보다 높은 저해활성을 보였으나, kojic acid $10{\mu}g/ml$의 활성보다는 낮은 저해활성을 보였다. 또한, 메탄올, 헥산, 부탄올 및 수층 분획물은 $100{\mu}g/ml$의 농도에서 arbutin $10{\mu}g/ml$의 활성과 유사한 저해활성을 보였다. 항산화 활성은 L-ascorbic acid를 양성 대조군으로 하여 수치를 비교하여 나타냈다. 에탄올 추출물 및 수층 분획물의 DPPH 라디칼 소거활성은 다른 시료들보다 비교적 높게 나타났다. 또한, 에탄올 추출물및 수층 분획물은 $500{\mu}g/ml$의 농도에서 LPS에 의해 유도된 iNOS 발현을 각각 82와 80% 감소시켰다. 이러한 결과들로 톳의 에탄올 추출물과 수층 분획물은 미백효과 및 피부 보호효과를 가지는 화장품 소재로 이용 가능할 것으로 기대된다.

에탄올 유발 위점막 손상에 대한 황련 물 추출물의 방어효과 (Protective Effects of Coptidis Rhizoma on Ethanol-induced Gastric Ulcer in Mice)

  • The exposure of gastric mucosa to ethanol produces acute ulcers mediated by inflammatory processes, hemorrhagic erosions and increase of reactive oxygen species. The purpose of this study was to assess the effects of Coptidis Rhizoma(CR) aqueous extracts on hydrochloride (HCl)/ethanol induced gastric ulcer in mice as compared with rebamipide (30 mg/kg) and ranitidine (100 mg/kg). Stomach ulcers were induced by oral ingestion of HCl/ethanol. CR extracts (125, 250 and 500 mg/kg) were orally administered, once a day for 7 continuous days, and 1 hr after last 7th treatment of CR extracts stomach ulcers were induced. Effects of CR extracts on HCl/ethanol-induced gastric ulcer were evaluated based on gross and microscopic observations with anti-oxidant activities. All three different dosages of CR extract significantly decreased HCl/ethanol-induced gastric ulcer compared with the HCl/ethanol control mice. CR extracts also strengthened the antioxidative defense systems - decreased the level of lipid peroxidation but increased the level of catalase, superoxide dismutase and nitrate/nitrite compared with the HCl/ethanol control. The effects of CR extract 500 mg/kg were similar to that of 30 mg/kg rebamipide, and CR extract 250 mg/kg showed similar anti-ulcer effects as compared with ranitidine 100 mg/kg. These results suggest that the gastroprotective effects of CR extracts on mice ulcer models can be attributed to its ameliorating effect on oxidative damages.

In vitro antimicrobial activity of aqueous and ethanol extracts of Euphorbia hirta

  • Euphorbia hirta, locally called 'ara tanah' or 'susun nabi' in Malaysia is a small annual herb common to the tropical countries and belongs to the same family as the tic and tapioca. E. hirta has had a long history of usage in the treatment of various ailments. In this current study, in vitro sensitivity test of crude aqueous and ethanol extracts of leaves and barks of E. hirta was carried out against bacteria (Escherichia coli, Salmonella enteritidis, Staphylocccus aureus and Bacillus subtilis) and fungi (Microsporum canis, Aspergillus fumigatus, Candida albicans and Candida tropicalis) using the discs diffusion method. The extract-impregnated discs (20, 40 and $80\;{\mu}g/{\mu}l$), the E. hirta extracts inhibited the growth of all the bacteria tested. The growth of C. albicans was inhibited in a concentration dependent manner by the aqueous leaves and barks extracts. C. tropicalis was found to be sensitive to the aqueous leaves extracts. The results were compared to antibacterial drugs of chloramphenicol, ampicilin, penicillin G, and enrofloxacine; and to antifungal drug of ketoconazole, itraconazole and miconazole. In this current study, it can be concluded that this plant has antimicrobial activity that is as potent as the standard antimicrobial drugs against certain microorganisms.

Antimalarial Activity and Phytochemical Profile of Ethanolic and Aqueous Extracts of Bidara Laut (Strychnos ligustrina Blum) Wood

  • This study aimed to determine the antimalarial effect of the Strychnos ligustrina (SLW) wood extracts and to analyze its phytochemicals. The SLW powder samples were macerated with 100% ethanol (E100), 75% ethanol (E75), 50% ethanol (E50), 25% ethanol (E25), and aqueous (A100). The extracts were analyzed by LCMS/MS, and its in-vitro antimalarial activity was tested with Plasmodium falciparum. The results showed that the extract yields of E100, E75, E50, E25, and A100 were 4.3, 5.2, 5.3, 4.7, and 3.6%, respectively. The antimalarial activities of the A100, E25, E50, and E75 extracts were classified as active with $IC_{50}$ values of 38.6, 42.6, 42.9, and $43.7{\mu}g/mL$, respectively. But, the antimalarial activity of the E100 extract was classified as slightly active with $IC_{50}$ values of $87.4{\mu}g/mL$. The dominant compounds contained in the extracts of A100, E25, E50, E75, and E100 was the alkaloid compound, namely brucine with relative concentrations of 24.96, 24.55, 21.33, 11.79, and 11.62%, respectively.

Origanum vulgare and Thymus vulgaris Extract Usability to Improve Silage Hygienic Quality and Reduce Mycotoxin Concentrations

  • Silage is one of the main feed components of ruminants around the world and can make up about 50-80% of the rations of dairy cows during the winter. The aim of this study was to evaluate the usability of oregano (Origanum vulgare L.) and thyme (Thymus vulgaris L.) aqueous and ethanol extracts to improve the hygienic quality of perennial ryegrass, red clover and blue alfalfa silage samples and estimate their effect on mycotoxins concentrations. Under laboratory conditions, 63 silage samples (21 perennial ryegrass, 21 blue alfalfa, 21 red clover) were fermented with inserted aqueous and ethanol extracts of oregano and thyme and two commercial inoculants with mesophilic lactic acid bacteria. After 96 days of fermentation, in silage samples were established fermentation parameters, microbiological status and mycotoxins concentrations. It was determined that the best results for achieving hygienic quality of perennial ryegrass and red clover silage samples was by insertion of aqueous and ethanol extracts of oregano and thyme. In blue alfalfa samples, the best results of silage hygienic indicators were determined by inserting aqueous and ethanol extracts of oregano. Aflatoxin B1 (AFB1), deoxynivalenol (DON), zearalenone (ZEA) and T-2 toxin concentrations in perennial ryegrass, red clover and blue alfalfa silage samples were best reduced by inserting aqueous and ethanol extracts of oregano and thyme. The present study shows that these extracts can be used to improve silage hygienic quality, reduce mycotoxins concentrations and thus ensure the wellness of cattle.
