Effects of the aqueous or 50% ethanolic extracts prepared by various extraction procedures on macrophage activation were determined by using the mouse macrophage cell line RAW264.7 cells as a indicator cell. The results demonstrated that the fractions prepared by aqueous extraction for 2 h and by microwave extraction with 50% ethanol at 60 W for 3 min had the greatest inducing abilities for NO production, and that the greatest ROS scavenging abilities were found in the fractions prepared by hot water extraction for 2 h or 3 h, by microwave extraction with 50% ethanol at 60 W for 3 min and by 0.5% HCl extraction, respectively. Phagocytotic activities against Candida albicans were found to be highest for the 50% ethanolic extracts prepared by microwave extraction for 3 min at 60 W, 80 W and 12 W, respectively. Especially, we found that a extract prepared by microwave extraction with 50% ethanol at 60 W for 3 min enables to induce effectively overall functional activation of macrophage, such as NO production, ROS scavenging and phagocytosis of C. albicans, respectively. These results demonstrated that a 50% ethanolic extraction using microwave at 60 W for 3 min would be useful for enrichment of macrophage-activating components contained in Hericium erinaceus, implying participation of protein-bound polysaccharides as a active factor.

In order to investigate the potential of a Angelica keiskei Koidzumi extract and its fractions as an active ingredient for whitening cosmetics, we prepared Angelica keiskei Koidzumi extract(70% ethanol), and the aqueous suspension was successively extracted with hexane, ethyl acetate(EA), and n-butanol fraction. We measured their inhibitory effects on mushroom tyrosinase and melanin synthesis in B16 melanoma cells and normal human kerationocytes in vitro. They did not show inhibitory activity against mushroom tyrosinase and the melanin synthesis except hexane and EA fractions. Hexane and EA fractions markedly inhibited cellular tyrosinase activity at a lower concentration(25 and 5 ${\mu}g/mL$, respectively) than arbutin(250 ${\mu}g/mL$). We also quantified the released amount of endothelin-1(ET-1), a mitogen of melanocyte, and interleukin-$1{\alpha}$(IL-$1{\alpha})$, a mediator of UVB-induced inflammation. Hexane and EA fractions did not affect IL-$1{\alpha}$ production, but they decreased UVB-induced ET-1 production in normal human keratinocytes in a dose dependant manner. As a result, hexane and EA fractions could inhibit the melanogenesis through the inhibition of UVB-induced ET-1 production in normal human keratinocytes. This result suggests that hexane and EA fractions of Angelica keiskei Koidzumi extract could be used as an active ingredient for cosmetics.

Nypa fruticans Wurmb is a species of palm, which is widely distributed in the mangrove forest of Southeast Asia. Various parts of N. fruticans has been used as a traditional medicinal plant. However, the physiological activities of N. fruticans has not yet been clarified well. Therefore, in this study, the 50% ethanol extract and its aqueous and ethyl acetate fractions of young shoots of N. fruticans were investigated for their antioxidant, cytoprotective effect, and antimicrobial activities. Every sample possessed very high free radical and various ROS scavenging capacities assessed by employing different in vitro assays such as $DPPH^{\cdot}$, $O_2^{{\cdot}-}$, ${\cdot}OH$, and $^1O_2$ scavenging activities. Based on these results, the cytoprotective effect was investigated using the oxidative hemolysis of erythrocyte. We found that the extract and fractions provide a greater protective effect compared with (+)-${\alpha}$-tocopherol. Furthermore, antimicrobial activities were confirmed against skin pathogens by broth microdilution assay. The ethyl acetate fraction had much higher antimicrobial activities than methyl paraben against Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, and Candida albicans. Taken together, our results indicated that the young shoots of N. fruticans may have the potential role as a natural active ingredient through their antioxidant, cytoprotective effect, and antimicrobial activities.

Smilax china root has been used as traditional medicinal remedy in China and Korea and reported to have various biological activities such as anti-inflammatory, antimutagenic and antimicrobial activities. In this study, the possibility of development as natural antioxidants of Smilax china root extracts was investigated. For the evaluation of antioxidant activity, aqueous- and 25% EtOH extract from Smilax china root were prepared and six different evaluation assay methods, i.e., measurement of total phenolics, radical scavenging effects on 1,1-diphenyl-2-picrylhydrazyl (DPPH), nitric oxide (NO) and nitrite $(NO_2)$, reducing power, and inhibitory effect on tyrosinase activity, were used. The total phenolics content of two extracts was high as the level of 36 mg of gallic acid equivalent per 1 g of dried sample tested. The radical scavenging activities of ethanol extract toward DPPH and NO were better than those of aqueous extract (p<0.05). The $NO_2$ scavenging activity of both extracts showed the highest value at pH 1.2 (98%). Especially, the $NO_2$ scavenging activities of EtOH extract were significantly stronger than those of aqueous one at pH 4.2 (51%) and pH 6.0 (32%), respectively. In the reducing power test, both extracts revealed higher ferric ion reducing activity than known antioxidant, vitamin C at the level of $0.05\sim0.1mg/mL$ (p<0.01). The 1 mL of aqueous and 25% EtOH extract showed effective inhibition activity on tyrosinase activity as 45% and 53%, respectively. Therefore, these results suggest that two extracts from Smilax china root may serve as useful natural antioxidants.

Purpose: Dendropanax morifera Leveille (DML) exhibits diverse biological and pharmacological activities, including anti-oxidative effect, anti-cancer activity, hepatoprotection, immunological stimulation, and bone regeneration. As part of the identification for novel functions of DML, we investigated the therapeutic effects of DML on diabetes induced by streptozotocine (STZ) treatment. Methods: First, the four extracts including the water extract of leaf (DLW), the ethanol extract of leaf (DLE), the water extract of stem (DSW), and the ethanol extract of stem (DSE) were collected from the leaf and stem of DML using a hot water and ethanol solvent. Alterations in body weight, glucose concentration, insulin level, and pancreatic islet structure were investigated in diabetic mice after treatment with extracts of DML for 2 weeks. Results: Among four extracts, the highest level of total polyphenols and total flavonoids was detected in DLW, while the lowest level of these was measured in DSE. The radical scavenging activity was also higher in DLW than in the other three extracts at the concentration of $25-100{\mu}g/mL$, although this activity was maintained at a constant level in all groups at the concentration of $500{\mu}g/mL$. Based on the results of anti-oxidant activity, DLW and DLE were selected for examination of anti-diabetic effects in a diabetes model. Body weight was gradually decreased in all STZ treated groups compared with the No treated group. However, four STZ/DML treated groups maintained a high level of body weight during 7-14 days, while the STZ/vehicle treated group showed a gradual decrease of body weight during the same period. Also, a significant decrease or increase in the concentration of glucose and insulin in the blood of the diabetes model was detected in a subset of groups, although the highest increase was detected in the STZ/DLE-200 treated group. In addition, the histological structure of pancreatic islet was significantly recovered after treatment with DLW and DLE. Conclusion: These results suggest that DLW and DLE may contribute to attenuation of clinical symptoms of diabetes as well as prevent the destruction of pancreatic ${\beta}$-cells in STZ-induced diabetes mice.

This study investigated the biological activities and chemical characteristics of Monascus-fermented Korean red ginseng (MFRG). The comparative activities of water, ethanol, and methanol extracts from MFRGE and Korean red ginseng (RG) were tested in vitro of anti-oxidative models of linoleic acid peroxidation by thiocyanate and thiobarbituric acid (TBA) methods and DPPH ($\alpha,\alpha'$-diphenyl-$\beta$-picrylhydrazyl)radical scavenging activities. In addition, measurements of their bioactive total phenolic compounds and minerals, and extract yield, were obtained. The extract yield of each solvent extracted from MFRG and RG was aqueous by 6.58% and 5.83%, ethanol by 0.62% and 0.98%, and methanol by 1.27% and 3.04%, respectively. Total phenolic compounds were higher in all solvents extracted from MFRG than those from RG. Major mineral contents (ppm) of MFRG and RG were K by 16,936 and 22,386, Ca by 2,310 and 3,693, Mg by 2,703 and 2,647, respectively. The DPPH radical scavenging activities were higher in all solvents extracted from MFRG than those from RG, however, all these extracts exhibited a relatively low level of radical-scavenging activity compared to the butylated hydroxytoluene (BHT). In antioxidative activities determined by TBA method using linoleic acid peroxidation, 70% methanol extract from MFRG and RG showed the highest antioxidative activity at a concentration of 0.1%. These results may provide the basic data to understand the biological activities of bio-active materials derived from MFRG.

Antioxidant activities such as reducing power, electron donating ability(EDA) and oxidation stability of Rumex crispus extracts with ethanol were examined, and adsorption of Pb(II) and Cd(II) with dried Rumex crispus powder in aqueous solution was experimented. Extraction yields based on the soluble solid contents ranged 1.9% to 3.0%, and were inversely proportional to solvent ratio but proportional to extraction temperature. Reducing power and EDA were also increased with extraction temperature but decreased with solvent ratio. These were corresponded to 86.4% and 91.0% of ascorbic acid reducing power and EDA, respectively, on 20 folds of solvent ratio and $75^{\circ}C$ Oxidation stability of Rumex crispus extracts on the corn seed oil was 86~91.3% of butylated hydroxytoluene. Pb(II) adsorptivity of Rumex crispus was higher than Cd(II), and adsorption rate of Pb(II) was 88% in 20 ppm concentration and 60min.

The stems of Juncus effusus were extracted with 70% aqueous ethanol and the concentrated extract was partitioned with ethyl acetate, n-butanol and $H_2O$, successively. Two compounds were isolated from the ethyl acetate fraction through the repeated silica gel and Sephadex LH-20 column chromatographies. According to the results of physico-chemical and spectroscopic data including NMR and IR, the chemical structures of the compounds were determined as dehydroeffusol (1) and effusol (2). Dehydroeffusol and effusol exhibited potent scavenging activity for DPPH and ABTS radicals with the $IC_{50}$ values as $130{\pm}3.21$ and $79{\pm}1.53{\mu}M$ in DPPH assay, and as $39{\pm}3.51$ and $24{\pm}2.73{\mu}M$ in ABTS assay, respectively. The compounds also significantly inhibited the proliferation of human cancer cell lines, AGS and A549.
ABSTRACT - To examine antihypertensive components of Ganoderma lucidum in Korea, two kinds of fruiting bodies (J and K) were used for extraction with water and the extracts were purified by ethanol precipitation and dialysis. Three fractions, i.e., the aqueous total extract(A), the ethanol supernatant(B} and the purified precipitate(C), were compared for antihypertensive activity in anesthetized spontaneously hypertensive rats (SUR). Although fractions A and B showed the activity, fraction C did not. Particularly, fraction B of sample K produced 44.3% reduction in diastolic blood pressure and 30.6% reduction in heart rate after i.v. administration of a dose of 10 mg/kg. Direct effects of this fraction B to the heart were observed in the isolated blood perfused heart preparation of the dog. It induced positive chronotropic and inotropic responses dose-dependently in the case of sample J. In the case of sample K, marked chronotropic and inotropic effects on atrial muscle but not on ventricular muscle were induced. In both samples, coronary blood flow (CBF) was dose-relatedly increased.reased.

Background: Eucommia ulmoides has long been used as an herbal medicine for the treatment of diabetes, hypertension and other diseases in many Asian countries. Methods and Results: This study aimed at evaluating the antioxidant and anti-inflammatory properties of its water (EU-DW, and REU-DW) and ethanol (EU-EtOH, and REU-EtOH) extracts, as well as those of non-roasted E. ulmoides (EU) and roasted EU (REU) cortex. The following were assessed in each extract: total phenolic and flavonoid contents, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and nitrite radical scavenging activities, reducing power, DNA damage prevention activity, and nitric oxide (NO) inhibition activity. Both EU and REU extracts showed high phenolic and flavonoid content, dose-dependent DPPH radical scavenging capacity, high reducing activity, and considerable DNA damage prevention activity. EU extracts showed remarkable ABTS free radicals scavenging capacity. REU extracts showed a higher radical scavenging capacity and played an important role in inhibiting NO production. Conclusions: The results of this study suggested that aqueous and ethanol extracts of EU and REU possess antioxidant capacities, and prevent oxidative damage to DNA, probably owing to their phenolic and flavonoid content. Therefore, EU and REU could be candidates antioxidant supplements.
