Gallic acid 등으로 유발된 인간 간 조직세포 내 hydrogen peroxide 생성억제에 대한 유산균발효애엽 추출물의 영향 (Effect of Artemisiae Argi Folium Fermented with Lactobacillus Pentosus on Hydrogen Peroxide Production of Human Hepatocyte Treated with Toxicants)

  • The purpose of this study is to investigate the effect of water extract from Artemisiae Argi Folium Fermented with Lactobacillus pentosus (AFL) on hydrogen peroxide production within human hepatocyte HepG2 cells treated with gallic acid, EtOH, nicotine, acetaminophen, and acetaldehyde. AFL (0~400 ug/mL) was treated with gallic acid, EtOH, nicotine, acetaminophen, and acetaldehyde. And the intracellular productions of hydrogen peroxide were measured by dihydrorhodamine 123 (DHR) assay. AFL showed the restoration of the intracellular productions of hydrogen peroxide which were reduced by gallic acid, EtOH, nicotine, acetaminophen, and acetaldehyde in HepG2 Cells. AFL could be supposed to have the hepatoprotective effect related with hepatocytologic signaling activity against gallic acid, EtOH, nicotine, acetaminophen, and acetaldehyde.

황금 에탄올 및 열수 추출물의 피부 개선 연구 (A Study on the Skin Improvement of Ethanol and Hot Water Extracts from Scutellaria baicalensis)

  • 본 연구에서는 황금 70% 에탄올 및 열수 추출물의 항산화, 항염, 미백, 보습, 세포 재생 및 자외선 및 블루라이트에 대한 세포 보호 효능을 확인하였다. 항산화 실험 결과, 70% 에탄올 및 열수 추출물 모두 DPPH 라디칼 소거 활성을 나타내었으며, 항염 실험 결과, 70% 에탄올 및 열수 추출물 모두 NO, 전염증성 cytokine(IL-6) 및 PGE2의 생성을 억제하는 것을 확인하였다. 미백 실험 결과, 70% 에탄올 추출물과 열수 추출물 모두 멜라닌 생성을 저해하였으며, 보습 실험 결과, 70% 에탄올 및 열수 추출물 모두 보습 인자인 HA의 생성량이 증가시키는 것을 확인하였다. UVB 및 블루라이트에 대한 세포 보호 효과를 분석한 결과, 70% 에탄올 추출물과 열수 추출물 모두 UVB에 대한 세포 보호 효능을 나타내었으며, 70% 에탄올 추출물은 블루라이트에 대한 세포 보호 효능 또한 나타내었다. 또한, 세포 이동 및 증식 효능을 분석한 결과, 70% 에탄올 추출물에서 세포 성장이 촉진되는 것을 확인하였다. 이러한 결과를 바탕으로 황금 70% 에탄올 추출물, 열수 추출물은 항산화, 항염, 미백, 피부재생, 자외선 및 블루라이트에 대한 세포 보호 효과가 있는 화장품 관련 천연소재로써의 활용가능성이 있는 것으로 사료된다.

Favorable Hepatoprotective Effects of Gongjin-dan on the Acute Ethanol-induced Liver Damaged C57BL/6 Mice

  • To observe the potential hepatoprotective effects of Gongjin-dan on the acute ethanol (EtOH)-induced liver damages in C57BL/6 mice with its possible action mechanisms. EtOH-mediated acute hepatic damages were induced by oral administration of EtOH total 3 doses. The changes on the body weight, liver weight, albumin, TG, AST, ALP, ALT, hepatic TG contents, hepatic antioxidant defense system, TNF-α, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes - SREBP-1c, SCD1, ACC1, FAS, PPARγ and DGAT2 or genes involved in fatty acid oxidation - PPARα, ACO and CPT1 were observed with final liver histopathological inspections after 15 days of continuous administration of silymarin 200 mg/kg, Gongjin-dan (GJD) 400, 200 and 100 mg/kg. The results were compared with silymarin 200 mg/kg treated mice. Marked decreases of body and liver weights, increases of serum AST, ALT, Albumin and TG levels, hepatic TG contents, TNF-α level, CYP 2E1 activity and mRNA expressions of hepatic lipogenic genes or decreases mRNA expressions of genes involved in fatty acid oxidation were observed with histopathological changes related hepatosteatosis increases of immunolabelled hepatocytes, as the results of a binge drinking of EtOH in the present study. Also destroys of hepatic antioxidant defense systems were demonstrated in EtOH control mice as compared with intact vehicle control mice, respectively. The results suggest that oral administration of 400, 200 and 100 mg/kg of GJD favorably protected the liver damages from acute mouse EtOH intoxications.

Supplementation of Laurus nobilis Attenuate Ethanol-induced Psychomotor Alterations in Rats

  • Laurus nobilis (L. nobilis) is traditionally used as an herbal medicine to treat various diseases. Ethanol (EtOH) consumption entails physiological, mental and psychomotor alterations. The aim of the present study was to assess the effects of L. nobilis in attenuating the EtOH-induced psychomotor alterations. L. nobilis was administered to SD rats, 30 minutes before EtOH administration (4 g/kg), at doses of 25, 50 and 100 mg/kg. Evaluations of psychomotor activity in the open-field, accelerating rota-rod, wire, and swimming ability were done at 1, 2, 4 and 8 hours after EtOH administration. In addition, blood ethanol and acetaldehyde levels were also measured. Pre-treatment of L. nobilis significantly improved EtOH-induced psychomotor alterations and decreased blood ethanol and acetaldehyde levels. These findings suggest that L. nobilis might be an effective substance to attenuate the harmful effects of EtOH, particularly psychomotor alterations, and can potentially be considered as a functional food.

오미자(Schisandra chinensis) 착즙박 추출물의 항산화 활성 (Antioxidation activity of residue after omija (Schisandra chinensis) juice extract)

  • 오미자를 착즙하고 남은 착즙박을 이용하여 추출조건에 따른 이화학적 변화를 알아보았다. 즉, 착즙박을 열수, 50% 에탄올, 70% 에탄올 및 95% 에탄올을 추출 용매로 사용하였으며, 추출시간은 1시간에서 10시간까지 1시간 간격으로 $60^{\circ}C$에서 정치 추출하여 여과한 후, 감압 농축하여 제조된 용매별 농축액을 분석시료로 이용하였다. 오미자에 함유된 유효성분 중 하나인 schizandrin 함량은 추출 용매에 따라서 열수: 0.52 g/100 g, 50% 에탄올: 1.79 g/100 g, 70% 에탄올: 2.01 g/100 g 및 95% 에탄올은 2.37 g/100을 나타내었으며, 열수를 이용하여 추출한 경우보다 에탄올 농도가 증가할수록 schizandrin 함량이 증가하는 경향을 나타내었다. 총 폴리페놀 함량은 50% 에탄올 용매 추출물에서 16.70 mg/mL로 가장 높게 나타났으며, 2,2-diphenyl-1-picrylhydrazyl 전자공여소거능에 대한 효과는 분석 시료의 200 mg/mL 농도에서 측정하였을 때, 50% 에탄올 용매 추출물에서 86.16%로 가장 높게 나타내었다. 또한, 안토시아닌 함량은 모든 용매 추출물이 추출 시간이 증가할수록 감소하는 경향을 보였다.

추출조건에 따른 늙은 호박 추출물의 생리활성 (Physiological Activities of Cucurbita moschata Duch. Extracts with Different Extraction Conditions)

  • 늙은 호박의 부위를 나누고 추출용매를 증류수, 50% EtOH, 100% EtOH로 달리하여 건물 중량의 25배에 해당되는 부피(w/v)일 때, 추출물의 생리활성 및 항산화 효과를 측정하였다. 추출물의 수율을 측정한 결과 추출용매를 증류수로 사용한 추출물들이 가장 높았으며, 특히 섬유질 추출물이 14.62%로 가장 높게 나타났다. 전자공여 작용의 경우 50% EtOH로 추출한 섬유질 추출물이 52.37%의 높은 전자 공여능을 보였다. SOD 유사활성을 측정한 결과에서는 100% EtOH에 의해 추출된 과육 추출물이 42.48%로 다른 추출물보다 높은 유사활성을 나타냈다. 총 폴리페놀 함량의 경우 50% EtOH로 추출한 섬유질 추출물이 121.49 mg%로 가장 높게 나타났다. 아질산염 소거능 측정 결과에서 모든 추출물이 pH 1.2에서 소거능이 가장 높게 측정되었다. Tyrosinase 활성 저해능의 경우 모든 과육 추출물이 11.36~19.14%로 가장 높게 나타났으나 비교물질인 L-ascorbic acid에 비해 매우 낮은 저해능을 보였다. ACE 저해활성에서도 전자공여능과 페놀함량 측정결과와 같이 50% EtOH로 추출한 섬유질 추출물이 가장 높게 나타났다.

알콜로 유도된 흰쥐의 근위축에서 갈근(葛根)의 보호 작용과 그 기전에 대한 고찰 (The Protective Effects of Pueraria Radix against Chronic Alcohol-induced Muscle Atrophy in Rats)

  • Objectives Ethanol is a potent inhibitor of muscle protein synthesis. Muscle mass is regulated by the balance between rates of protein synthesis and protein breakdown. Both acute and chronic alcohol consumption inhibits synthesis to a greater extent than degradation. Protein synthesis is more intensely decreased in type II fibers than in type I fibers. Apoptosis has been shown to occur frequently in a variety of tissues in response to chronic alcohol feeding. Increased muscle fiber apoptosis has been shown in alcoholics with myopathy. Pueraria radix has been used for many disorders such as fevers, gastrointestinal disorders, muscle aches, allergies, respiratory problems, skin problems, high blood pressure, migraine headaches, lowering cholesterol and treating chronic alcoholism. We therefore tested the hypothesis that oral treatment with Pueraria radix could reduce the ethanol-induced muscle atrophy. Methods Young male Sprague-Dawley rats were orally given 25% ethanol (5 ml/kg, body weight) daily with Ethanol for 4 weeks. Normal group was similarly administrated with saline. The Rats of Pueraria radix treated group (EtOH+PR) were orally administrated Pueraria radix water extract, and rats of EtOH group were given with the vehicle only. After 4 week, the morphology of gastrocnemius and plantaris muscles were assessed by hematoxylin and eosin staining. The immunoreactivities of pre-apoptotic BAX and anti-apoptotic Bcl-2 proteins were also measured. Results The muscles from rats of EtOH group represented a significant reduction in average cross section area compared to Normal group. EtOH+PR group had increased fiber compared to the EtOH group. Moreover, to investigate the ethanol-induced muscular apoptosis, the immunohistochemical analysis of Bax and Bcl-2 was carried out. The treatment with Pueraria radix (EtOH+PR) significantly decreased BAX expression and increased Bcl-2 expression 4 weeks after ethanol administration when compared with Normal group. Conclusions These results suggest that Pueraria radix water extract has protective effects on chronic alcohol induced myopathy.

흰쥐의 알코올 유발성 간손상에 실비음(實脾飮)이 미치는 보호 효과 (The Protective Effects of Silbi-um Extract on the Alcoholic Liver Injury in Rats)

  • Objectives: The objective of this study is to investigate the effects of Silbi-um (SBU) extract on the alcoholic fatty liver induced by EtOH administration for 8 weeks. Methods: Male Sprague Dawley rats were used. All animals were randomly divided into 3 groups; Normal, EtOH and EtOH+SBU. The rats of EtOH group were daily treated with ethanol of 25% (v/v) for 8 weeks (n=10). EtOH+SBU group was orally treated with SBU water extract after ethanol administration (n=10). The rats of Normal group were treated with saline (n=10). After 8 weeks, the mean body weight, liver weight, and liver-body weight ratio were calculated. The serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) of all groups were measured. The morphological alterations were observed using hematoxylin and eosin (H&E) and Oil Red O staining. Moreover, the alteration of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) levels were analyzed immunohistochemistrically. Results: The histological data showed that liver sections from EtOH group displayed severe steatosis. SBU extract significantly inhibited the progression of the alcoholic liver injury. The increased serum level of ALT and AST induced by ethanol administration were decreased by SBU extract. Furthermore, SBU extract significantly decreased the liver concentrations of $TNF-{\alpha}$. Conclusions: SBU water extract attenuated the alcohol induced fatty liver by improving hepatic lipid metabolism via suppression of $TNF-{\alpha}$ protein. SBU could be effective in protecting the liver from alcoholic fatty liver.

The antioxidant and cytotoxic activities of Sonchus oleraceus L. extracts

  • This study investigated in vitro antioxidant activity of Sonchus oleraceus L. by extraction solvent, which were examined by reducing power, hydroxyl radical-scavenging activity(HRSA) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays. 70% MeOH extract had the greatest reducing power while EtOH extract had the greatest HRSA. The antioxidant activity of S. oleraceus extracts was concentration dependent and its $IC_{50}$ values ranged from 47.1 to $210.5\;{\mu}g/ml$ and $IC_{50}$ of 70% MeOH, boiling water and 70% EtOH extracts were 47.1, 52.7 and $56.5\;{\mu}g/ml$, respectively. 70% MeOH extract of S. oleraceus contained the greatest amount of both phenolic and flavonoid contents. The extracts tested had greater nitrite scavenging effects at lower pH conditions. The cytotoxic activity showed that EtOH extract had the best activity against the growth of stomach cancer cell. These results suggest that S. oleraceus extract could be used as a potential source of natural antioxidants.
