In vitro 검출시스템을 이용한 한약재 추출물로부터의 에스트로겐 활성의 검증 (Verification of Estrogenic Activities in Ethanol Extracts of Oriental Herbal Medicines using In vitro Detection System)

  • In order to evaluate the direct effect of estrogenic compounds in oriental herbal medicines, the estrogenic activity was measured using an in vitro detection system. For this system, human breast cancer cell line MCF7 was transfected using an estrogen responsive CAT reporter plasmid. Estrogenic activities of Platycodi radix, Astragali radix and Glycyrrhizae radix were evaluated using this system. Estrogenic activity of a 500 ㎍/ml ethanol extract of Platycodi radix was as same as that of a 10/sup -8/ M standard solution (17β-estradiol) and activity of a 50 ㎍/ml ethanol extract was between those of a 10/sup -8/ M and a 10/sup -9/ M standard solutions. In addition, estrogenic activity of a 50 ㎍/ml ethanol extract of Platycodi radix was as same as that of a 10/sup -10/ M standard solution. The same activity patterns were observed in the system which was treated by Astragali radix or Glycyrrhizae radix extracts. The most effective activity was observed in a system which was treated by Platycodi radix extract, but the least activity was observed by Glycyrrhizae radix extract. In this result, it was confirmed that Platycodi radix, Astragali radix and Glycyrrhizae radix extracts possess estrogenic compounds.

  • Stable MCF-7-ERE cells, in which pERE-Luc reporter gene has been stably integrated into the genome of the MCF-7 cells, were used to detect the estrogenic activity of various dietary flavonoids in either pure chemical or mixtures. Estradiol (E2) induced luciferase activity in dose dependent manner and this activity was inhibited by tamoxifen (Tam) concomitant treatment. A large series of flavonoids showed estrogenic activities, corresponding to EC5O values between 0.2 and 9 microM and their mixtures didn't show additive or synergistic effects. And we could find some structure and activity relationship. First, 4-methoxylation and catechol structure decreased estrogenic activities. Second, hydroxylation of 3 position reduced estrogenic effect. Third glycosides of flavonoids showed weak estrogenic activity or no activity. Interestingly, when tested at high concentrations, genistein, kaempferol, biochanin A and chrysin elicited luciferase induction higher than that of the maximum induction by estradiol. And these effects of genistein and kaempferol could not be fully inhibited with tamoxifen

감마선 처리에 의한 에스트로겐 활성 저감 연구 (Reduction of Estrogenic Activity by Gamma-ray Treatment)

  • In this study, degradation of estrone (E1) and $17{\alpha}$-ethynylestradiol (EE2) by gamma-irradiation and subsequent reduction of estrogenic activity as a function of absorbed dose were conducted using the yeast two-hybrid assay. Relative potency of E1 and EE2 compared to estrogenic activity of $17{\beta}$-estradiol (E2) was found to be 0.0144 and 0.1605, respectively. More than 90% of E1 and EE2 (both $5.0{\times}10^{-6}M$) was removed at an absorbed dose of 5 kGy, but more than 40% of estrogenic activity still remained. The addition of $TiO_2$ catalyst appeared to improve the removal efficiency of E1 and decrease estrogenic activity while there was no significant effect for EE2. Additionally, the calculated estrogenic activity of E1 and EE2 based on a regression model was well correlated with the observed activity.

염소산화에 의한 에스트로겐성 화학물질의 활성저감 (Decrease of Activity of Estrogenic Chemicals by Chlorination)

  • The effects of chlorination on the elimination of three estrogenic chemicals such as $17{\beta}$-estradiol (E2), nonylphenol (NP) and bis-phenol A (BPA) were investigated using yeast two-hybrid assay (YTA), estrogen receptor competition assay (ER-CA), and high-performance liquid chromatography/mass spectrometer (LC/MS). Results of YTA, ECA and the analysis of LC/MS indicated that the estrogenic activity of above mentioned three endocrine disruptors were significantly reduced as the result of chlorination. The decrease in estrogenic activity paralleled with decrease in estrogenic chemicals under the influence of free chlorine. One common characteristic of estrogenic chemicals is the presence of a phenolic ring. Considering that a phenolic ring is likely to undergo some sort of transformation in aqueous chlorination solution, the above mentioned results may be applied to the rest of the other estrogenic chemicals in natural waters.

E-Screen assay법을 이용한 농약화학물질의 에스트로겐 활성 연구 (Study on Estrogenic Activities of Pesticide Chemicals using E-screen Assay)

  • In this study, sixty chemicals including 47 pesticides were screened for estrogenic activity using E-screen assay. MCF-7 cell, used in E-screen assay, is known to be proliferated by addition of estrogenic chemicals. Eight of the measured pesticides showed estrogenic activity at the concentration range of 100-10,000nM. Their relative proliferative effect (RPE) and the relative proliferative potency (RPP) were 20-65% and 0.01-1.0%, respectively, when compared with 1.0nM of $\beta-Estradiol-17-acetate(E_{2}).$ DDVP and diazinone showed most strong estrogenic potency(RPP; 1.0%) and effect(RPE; 65%) of the eight pesticides. These results are in agreement with estrogenic activity of bisphenol A is known as a positive endocrine disrupter. Also, in this study, paraquat, DDVP, 4-chloro-3-methylphenol, 2,4,6-trichlorophenol and diazinon of the measured pesticides are estimated to estrogenic chemical.

Antifertility activity of hydro alcoholic extract of Moringa concanensis Nimmo: An ethnomedicines used by tribals of Nilgiris region in Tamilnadu

  • In the present study, the hydro alcoholic extract of Moringa concanensis and their different fractions were evaluated for it's anti implantation, abortifacient, estrogenic and antiestrogenic activity. Hydro alcoholic extract of Moringa concanensis has showed potent antiimplantation and abortifacient activity at 200 mg/kg and 400 mg/kg respectively and marked estrogenic activity when administered individually and anti estrogenic activity was observed when administered along with ethinyl estradiol (1 ${\mu}g/rat/day$) as well as their different fractions of Moringa concanensis showed significant antiimplantation and abortifacient activity at 100 mg/kg. Moreover, all tested fractions showed significant anti estrogenic activity when administered simultaneously with ethinyl estradiol.

  • 천연물에 포함되어 있는 에스트로겐 효과를 가지는 성분의 생체 내에서의 직접적인 효과를 검정하기 위하여, 에스트로겐 수용체를 발현하는 것으로 알려진 인체 유방암 세포주 MCF7에 에스트로겐에 반응성을 나타내도록 고안된 CAT 리포터 플라스미드를 도입한 in vitro 검출시스템을 사용하여 에스트로겐 활성을 측정하였다. 이미 여러 분야에서 그 기능성의 연구가 활발히 진행되고 있는 광합성 조류인 스피루리나(spirulina)와 파래 등의 해양식물을 대상으로 에스트로겐 반응 리포터 시스템을 이용하여 에스트로겐 활성을 측정하였다. 그 결과, 스피루리나 에탄올 추출물의 CAT 활성은 $500\mug/ml\; 와\; 50 \mug/ml$의 농도에서 표준물질인 $17\beta-estradiol$의 농도 $10^{-8}$ M과 비슷한 정도의 에스트로겐 활성 효과를 나타내었고, $5\mug/ml$의 농도에서는 표준물질인 $17\beta-estradiol$의 농도 $10^{-10}$ M과 비슷한 정도의 에스트로겐 활성 효과를 나타내었다. 하지만 파래 추출물의 경우에는 에탄올을 처리한 대조군과 비교하여 유의한 CAT활성 변화를 나타내지 않았다 한편, 불가사리와 새우 등의 해양동물을 대상으로 한 실험에서는 에탄올을 처리한 대조군과 비교하여 유의한 CAT 활성 변화를 나타내지 않았다. 이 연구 결과로 광합성 조류인 스피루리나에 에스트로겐 활성을 효과적으로 나타내는 생리활성성분이 포함되어 있을 수 있다는 가능성을 확인하였다.

Evaluation of the Estrogenic Activity of Isoflavones from the Rhizome of Belamcanda chinensis

  • Four compounds were isolated from the rhizome of Belamcanda chinensis (Iridaceae) by repeated column chromatography and were elucidated as iristectorigenin A, irigenin, tectorigenin and tectoridin by spectral analysis. The estrogenic activity of these isoflavones was tested using Ishikawa cells. Irigenin, tectorigenin and tectoridin were highly estrogenic ($EC_{50}\;=\;0.75$, 0.42 and $0.81\;{\mu}g/ml$, respectively), while iristectorigenin A exhibited weak estrogenic activity ($EC_{50}{\geq}4\;{\mu}g/ml$).

랫드 자궁비대반응시험(Uterotrophic assay)을 이용한 phthalate esters의 에스트로겐성 작용 연구 (No Estrogenic Activity of Phthalate Esters in Ovariectomized Rat Uterotrophic Assay)

  • The rodent uterotrophic assay is currently recommended as one of the primary in vivo assays far endocrine disrupting chemicals by the Organization for Economic Cooperation and Development (OECD) and Endocrine Disruptor Screening and Testing Advisory Committee (US EPA EDSTAC). Generally, this assay relies on the rapid increase in uterus and vagina weights when exposed to estrogenic compounds. Phthalate esters have been used extensively as a plasticizer in the manufacture of plastic products such as PVC films and medical devices. Recently, phthalate esters have been shown to induce endocrine system mediated responses. However, a flew studies have been conducted for the screening of their estrogenic activity. In this study the estrogenic activity of seven phthalate esters, butyl benzyl phthalate (BBP), di(2-ethylhexyl) phthalate (DEHP), di-n-butylphthalate (DBP), diethylphthalate (DEP), di-n-pentylphthalate (DPF), di-n-propylphthalate (DPrP) and dicyclohexylphthalate (DCHP), was examined in uterotrophic assay. Phthalate esters dissolved in corn oil were administered to ovariectomized (OVX) female Sprague-Dawley rats by sub-cutaneous injection for three consecutive days. fiats were sacrificed 24h after final treatment, and then uterus and vagina weights were deter mined. All phthalate esters tested in this assay did not change talc uterus and vagina weights at dosage levels up to 200 mg/kg/day treatment. These results demonstrated that phthalate esters did not exhibit estrogenic activity in vivo uterotrophic assay.

유방암 세포주를 이용한 국내 자생 콩과식물의 에스트로겐 활성검색 (Estrogenic Activity of Leguminosae Species in Korea using MCF-7 Cells)

  • Leguminosae plants are known for its phytoestrogen constituents which play a major role in the prevention of osteoporosis, cancer and heart disease. In this study, the estrogenic activity of 158 samples from 58 species, 3 subspecies and 10 varieties of Leguminosae plants growing in Korea was evaluated. An estrogen, 17β-estradiol was used as a reference compound, and the potency of each sample was expressed in relative efficacy (%) compared to that of the reference by a reporter gene assay using MCF-7 cells. As results, the estrogenic activity of methanolic extracts of Phaseolus vulgaris var. humilis, Sophora flavescens, Lespedeza × robusta, Indigofera pseudotinctoria, Maackia amurensis, Glycine soja, Wisteria floribunda, Robinia pseudoacacia, Astragalus sinicus, Pueraria lobata, Lespedeza maximowiczii var. tomentella, Trifolium repens and Crotalaria sessiliflora showed similar to or higher at 100 ㎍/ml than the positive control at 10 nM. These findings can be a potential evidence for developing estrogen alternatives resolving various types of menopause symptoms with information on proper harvest season and usage plant part. To the best of our knowledge, the estrogenic activity of Lespedeza × robusta, Indigofera pseudotinctoria, Wisteria floribunda, Robinia pseudoacacia and Lespedeza maximowiczii var. tomentella is reported for the first time in this study.
