Percoll Process Can Improve Semen Quality and Fertility in Turkey Breeders

  • A percoll density gradient technique was developed for producing high quality turkey semen and improving the fertility by removing deleterious cellular components, including spermiophages, bacteria, abnormal or dead spermatozoa, and other cellular debris. The combination of three different percoll densities, 1.05, 1.07, and 1.08 showed the best resolution and was selected to prepare a discontinuous percoll density gradient to obtain healthy spermatozoa from semen smples. Bacteria, spermiophages, and abnormal or dead spermatozoa were detected from the density range from 1.05, 1.05 to 1.07, and 1.07 to 1.08, respectively. Healthy spermatozoa were collected from the density greater than 1.08. Spermatozoa obtained from percoll density gradient centrifugation showed better sperm motility than those from unprocessed pooled semen. Bacteria including Escherichia coli, Staphylococcus aureus, and Proteus spp., were predominant contaminants in turkey semen, and the numbers of cells were approximately $5{\times}10^5$ to $1{\times}10^9cfu/ml$. The overall fertility rates in hens inseminated with processed percoll density gradient were higher than those in hens with unprocessed semen especially for unhealthy sperm. In conclusion, semen quality can be improved by percoll density gradient centrifugation, which augmented the fertility of turkey breeders.

Relationship Between Drug Resistance and Citrate Utilizing Ability of Citrate-Utilizing Variants of Escherichia Coli from Cattle (Citrate이용대장균(利用大腸菌) 변이주(變異株)의 약제내성(藥劑耐性)과 Citrate이용성(利用性)과의 관계(關係))

  • Transfer experiment of drug resistance and citrate utilizing ability were performed to show the relationship between drug resistance and citrate utilizing ability of 76 Citrate-Utilizing variants of Escherichia coli(Cit $^+E$. coli) isolated from cattle. The results obtained were summarized as follows; 1. Of seventy-six Cit $^+E$. coli, 36(47.4%) strains transfered their drug resistance to the E. coli ML 1410 by mating. 2. Tetracycline(TC) resistance determinants and citrate utilizing ability were transmitted in the recipient strains selected for TC, but it can be seen the segregation of streptomycin (SM) resistance determinants and citrate utilizing character in 9 recipient strains selected for SM. 3. Of seventeen TC resistance Cit $^+E$. coli, 10 strains transfered citrate utilizing character, alone. 4. The transmission of the ability to utilize citrate on Simmons citrate agar at $37^{\circ}C$, was demonstrated 52(68.4%) out of the 76 Cit $^+E$. coli.

Effect of Experimental Electrization Stress on the Pathogenesis of Escherichia coli in Mice (실험적 감전 Stress 가 Escherichia coli 감염마우스의 기병에 미치는 영향)

  • A hypothesis that is a stress condition in animal may cause either enhancement or reduction of the host resistance against microbial infection was experimentally studied. Among of many processes for stress formation an experimental electrization in mice was devised, on the bases of blood picture analysis, and studied the effect of experimental electrization of mice on E. coll infection. The results obtained were as follows. 1. Electrization with ordinary current, A. C. 60 cps., on the path of symmetrical line of both posterior limbs at 20 to 100 volts (less than 10 mA) for 15 to 30 seconds was able to induce a stress reaction in blood pattern without showing any dangers of electrocution, electric burns and other residual signs, and no correlation between blood pattern of the reaction and an amount of current between 20 to 100 volts was observed. As the electrodes, two of 21 gauge hypodermic needles were used, when the electrization each of them were inserted into the center of toe tissue of the both legs. 2. Serum protein fractions following the experimental electrization showed a tendency of a low A/G ratio and a high value of ${\alpha}$-globulin. 3. In the studies on the effect of electrical stress on the pathogenesis of E. coli in mice, a group in which a simultaneous electrization and infection, and a group infected two hours after electrization showed 80 per cent mortality. On the other hand, infection after 20 hours electrization and control groups showed their mortality of 40 and 60 per cent respectively.

Studies on Antibacterial Substance from Lactobacillus bulgaricus (Lactobacillus bulgaricus가 생산한 항균물질(抗菌物質)에 관한 연구)

  • This study was carried out to confirm the agent responsible for the antibacterial activity in milk culture or Lactobacillus bulgaricus to extract and purify it. The following results were summarized as followings : The antibacterial agent was extracted from the cultured skim milk with methanol and acetone and was purified by Sephadex G-50 gel filteration and thin layer chromatography on silica gel. The antibacterial substance other than lactic acid was confirmed by turbidimetric technique using the neutralized culture filtrate which inhibited the growth of Bacillus subtilis. The purified agent showed inhibitory activity against Bacillus subtilis, Escherichica coli, Pseudomonas fluorescens Staphylococcus aureus, Proteus vulgaris and Shigells $dysenteria^2$. The agent obtained from thin layer chromatography was free from $H_2O_2$ or lactic acid.

Differentiation of mixed bacterial populations by modified gram stain (수정된 Gram 염색법에 의한 혼합세균 개체군의 분별 측정)

  • Attempts were made to enumerate the number of Gram positive and negative bacteria in the development of natural fermentation rapidly and simultaneously. A general Gram stain was applied to this study. The number of cells by Gram stain was proportional to the cell turbidity by spectrophotometer within a range of 0.7 absorbance at 610nm. The cells washed out during procedures were not exceeded about 8 percentage. The standard error of separate counts in the mixture of Cscherichia coli and Micrococcus luteus was $5.1\pm2.3$%. The possible range of counting was $5.5\times 10^{7}-1.0\times 10^{9}$ cells/ml. Therefore, it is believed that a general Gram stain could be applied to the separate counting of mixture of Fram positive and negative bacterial populations too. In practice, growth kinetics of hemp retting and Kimchi fermentation were presented.

  • Nine strains of xyl mutants that could not utilize xylose as a carbon source were isolated from E. coli JM109 by the treatment of NTG in order to investigate the regulation of xylose operon and to use recipient cells for the cloning of xylR gene. For the characterization of all isolated mutants, colony colors of all mutants on MacConkey-xylose and MacConkey-xylulose agar plate were observed for the utilization of xylose and xylulose, and the growth level and the activity of xylose isomerase and xylulokinase were determined in need. The isolated xylR/T mutants formed the white colony on MacConkey-xy-lose and MacConkey-xylulose agar plate. They did not detect the activity of xylose isomerase, and the activity of xylose isomerase was not restored in transformants of xylR/T mutant with pEX13 which contained xylA gene. xylR and xylT mutants were classified from xylR/T mutants depending upon the growth level in minimal medium. xylT mutants; DH13, DH121 and DH125 could grow a little in that medium, but xylR mutants; DH10, DH53, and DH60 could not grow that medium.

  • The transformation of Saccharomyces cerevisiae with YIp26, YRp7 and YEp13 was investigated. Transformation frequences of YIp5, YIp26, YRp7 and YEp13 in Escherichia coli HB101 was $5.1{\times}10^{-4}$, $1.5 {\times}10^{-3}$, $1.3{\times}10^{-3}$, $3{\times}10^{-3}$, respectively. When plasmids were used in covalently closed circular form, transformation frequency of YEp13 was $1.2{\times}10^{-4}$ in S. cerevisiae DBY747 and $3.3{\times}10^{-4}$ in S. cerevisiae MC16 and that of YRp7, YIp26 was $3{\times}10^{-6}$, below $6{\times}10^{-8}$ respectively in S. cerevisiae DBY747 by the method of Ito. Cotransformation of YIp26 and YEp13 in linear form increased the frequency of transformation with efficiences 270-fold higher than transformation of YIp26 only in S. cerevisiae DBY747. In cotransformation of YIp5+YEp13 and YIp26+YRp7 with S. cerevisiae DBY747 by Beggs' method. Expression frequency of YIp5+YEp13 and YIp26+YRp7 was $4{\times}10^{-6}$, $1.5{\times}10^{-6}$, respectively. The recombinant plasmid of cotransformant was thought that YIp26 and YEp13, YIp5 and YEp13, and YIp26 and YRp 7 were ligated in vivo in S. cerevisiae DBY747.

Distribution and Antibiotic Production Characteristics for Streptomyces (Streptomyces의 토양중(土壤中) 분포(分布) 및 항생물질생산(抗生物質生産))

  • Out of 826 isolates of Streptomyces isolated from different soils, their distribution and antibiotic productivity were investigated. Distribution of the organism in the soil was affected by the soil conditions and plants. The highest isolation frequency was occurred from Quercus forest, Robinia forest and grass field, while soils from orchards and cultivating fields showed low density of Streptomyces. More than 49% of the isolates showed antibacterial activity against Bacillus subtilis, Erwinia carotovora subsp. carotovora and Xantomonas campestris pv. oryzae and about 40% of the isolates showed antiyeasty activity against Saccharomyces cerevisiae but only a few isolates showed antibiotic activity against E. coli and Pseudomonas solanacearum. Forty isolates of the Streptomyces showed strong antifungal activity against Pyricularia oryzae. Rate of isolation of Streptomyces was the highest on starch agar among the eight media tested. Antibiotic productivity of the isolates was the highest on potato sucrose agar medium among the 5 media tested.
