Effects of taurine on plasma and liver lipids, erythrocyte ouabain sensitive Na efflux and platelet aggregation in Sprague Dawley rats

  • The effects of taurine on plasma and liver cholesterol, erythrocyte ouabain sensitive Na efflux and platelet aggregation were examined in Sprague Dawley rats fed control or 0.5% cholesterol with 0.2% cholate diet. Plasma and liver levels of total cholesterol were increased significantly (p<0.05) in rats fed cholesterol diet compared to the control, and taurine significantly decreased the elevated plasma level of cholesterol in rats fed cholesterol diet (p<0.05). HDL-cholesterol was decreased in groups fed the cholesterol diet regardless of taurine supplementation and the difference between groups with and without cholesterol was significant (p<0.01). Plasma triglyceride was decreased and liver triglyceride was increased both significantly (p<0.05) in rats fed cholesterol compared to the control. Plasma and liver triglyceride in rats fed taurine was decreased significantly compared to the control (p<0.05). Intracellular Na tended to be lower in rats fed cholesterol or taurine and higher in rats fed cholesterol plus taurine compared to the control. Na efflux through Na-K ATPase and the passive leak of Na was somewhat reduced in rats fed cholesterol or taurine and was augmented in rats fed cholesterol plus taurine compared to the control, which showed a similar trend to the intracellular Na. Taurine supplementation caused a suppression of Na efflux in groups fed control diet and restored the suppressed Na efflux in groups fed cholesterol. Platelet aggregation was significantly decreased in the group fed taurine compared to the control (p<0.05) and the group fed cholesterol plus taurine was also a little lower in aggregation than the group fed cholesterol. Microscopic examination showed that taurine prevented fatty liver in rats fed cholesterol diet. Taurine known for stimulating Na-K ATPase in some cell types rather decreased erythrocyte ouabain sensitive Na-K ATPase in the present study. Taurine had hypolipidemic and hypocholesterolemic effects and inhibited platelet aggregation which may be favorable for prevention of cardiovascular diseases.

Live Blood Analysis on Interior Vascular Laser Irradiation Therapy and Exterior Vascular Laser Irradiation Therapy (혈관레이저 치료와 관련한 생혈액 검사의 진단 의미 고찰)

  • Objectives : The purpose of this study was to investigate the usability of live blood analysis on interior and exterior vascular laser irradiation therapy. Methods : We had analyzed the changing forms of the live blood sample with microscope before and after vascular laser irradiation therapy of blood. The live blood analysis was operated on Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals, red crystals, protoplasts. First, we analyzed all patients on each item, then did same thing classified two groups, Interior and exterior. Results : Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals, red crystals, protoplasts were decreased significantly, after interior and exterior aggregation, uric acid crystals. Interior vascular laser irradiation therapy was more effective than interior on Rouleau of red cell, erythrocyte aggregation, thrombocyte aggregation, uric acid crystals. Interior vascular laser irradiation therapy was more effective than exterior on red crystals, protoplasts. Conclusions : This study suggests that live blood analysis has the usability on vascular laser irradiation therapy. Then according to interior and exterior vascular laser irradiation therapy, the result has some different on each item. So it is better that choose the method, interior or exterior, for more effective therapy.

Transient microfluidic approach to the investigation of erythrocyte aggregation: comparison and validation of the method

  • A method based on transient shear flow dynamics of red cell aggregates was developed to investigate reversible re-aggregation processes with decreasing shear flow. In the microchannel-flow aggregometry, the aggregated red blood cells that are subjected to continuously decreasing shear stress in microchannel flow were measured with the use of a laser-scattering technique. Both the laser-backscattered intensity and pressure were simultaneously measured with respect to time, resulting in shear stress ranging from $0{\sim}35\;Pa$ for a time period of less than 30 seconds. The time dependent recording of the backscattered light intensity (syllectogram) yielded an upward convex curve with a peak point, which reflected the transition threshold of aggregation in the RBC suspensions. Critical-time and critical-shear stress corresponding to the peak point were examined by varying the initial pressure-differential and the micro channel depth, and these results showed good potential for being used as new aggregation indices. In the present study, these newly proposed indices were also validated by differentiating the effect of fibrinogen on RBC aggregation and then these indices were compared to the conventional indices that were measured by a rotational aggregometer.

A Clinical Study of the Live Blood Condition of Women′s Shoulder Measurement (여성 견비통 환자의 생혈액 형태에 대한 임상적인 연구)

  • Objectives : To study the Live Blood Condition of chronic shoulder measurement. Methods : Twenty-one patients were studied. The observation items of the Live Blood Condition are the form's abnormality and cohesion of red blood cells, the abnormality of hemoglobin and the toxicity in plasma. Results : 1. The form's abnormality and cohesion of red blood cells observed were erythrocyte aggregation (8 persons), Rouleau (3 persons), target cells (7 persons), ovalocytes (3 persons), poikilocytes (2 persons). Double conditions observed were erythrocyte aggregation & target cells (3 persons), erythrocyte aggregation & poikilocytes (1 person), target cells & ovalocytes (l person). 2. The abnormality of hemoglobin and the toxicity in plasma observed were cholesterol crystals (5 persons), atherosclerotic plaque (3 persons), chylous (6 persons). 3. The form normality of red blood was generally observed in one woman. The normality of hemoglobin and the non-toxicity in plasma were generally observed in seven women. Conclusions : According to this study of the Live Blood Condition of women's shoulder measurement, these results suggest that the twenty-one patients evidence the conditions of extravasated blood and phlegm.

Effect of Blood Donation on the Donor's Hemorheological Properties (헌혈이 헌혈자의 혈유변학적 지표에 미치는 영향)

  • The circulatory system is closely related to the inter-relationship between the anatomy of the heart and blood vessels, and the fluid dynamic properties of blood. The physical properties of blood, which affect blood flow, are called hemorheologic factors. Hemorheologic factors, such as blood viscosity and erythrocyte aggregation, are influenced mainly by hematocrit. A higher hematocrit level results in an increase in blood viscosity, erythrocyte aggregation, which impedes the circulation itself, and tissue oxygenation. An excess of serum ferritin causes injury to vascular endothelial cells and erythrocytes via oxygen free radicals. In addition, an excess of blood can aggravatee the adverse effects of the hemorheologic parameters and induce atherogenesis, microcirculatory disturbances, and major cardiovascular events. A preventive and therapeutic approach with a phlebotomy or blood donation has been stimulated by the knowledge that blood loss, such as regular donations, is associated with significant decreases in key hemorheologic variables, including blood viscosity, erythrocyte aggregation, hematocrit, and fibrinogen. Major cardiovascular events have been improved in regular blood donors by improving blood flow and microcirculation by decreasing the level of oxidative stress, improving the hemorheologic parameters, and reducing the serum ferritin level. Confirmation of the positive preventive and therapeutic effects of blood donations on cardiovascular disease by a well-designed and well-controlled Cohort study may be good news to patients with cardiovascular disease or at risk of these diseases, as well as patients who require a transfusion.

The Effect of Woohwangcheongsim-won on Circulatory Disturbance in Diabetes (우황청심원이 당뇨병 Rat의 혈액순환장애에 미치는 영향)

  • Object: Death rate due to hypertension, atherosclerosis, ischemic heart disease and cerebral infarction induced by Westernized diet and increased average life span is on the rise. Decrease in blood circulation, activation of thrombus generation and intravascular lipid accumulation, cited as the principal causes of the above mentioned diseases in recent studies, result in circulatory disturbance and blood vessel obstruction leading to ischemic cell death of heart, brain and peripheral vessels. Method: We investigated the biochemical changes in microvascular permeability, aggregation of platelet and the intravascular lipid accumulation in induced-diabetic rat using Streptozotocin. We also studied the effects of Woohwangcheongsirn-won after oral administration on blood circulation, platelet function and lipid metabolism. The results are as follows: I. Woohwangcheongsim-won increased blood circulation in microvessels. 2. Woohwangcheongsim-won increased the reduced erythrocyte deformability in diabetes. 3. Woohwangcheongsim-won induced the reduction of contents of 2, 3-DPG, but failed to affect the reduced contents of ATP in erythrocyte in diabetes. 4. Woohwangcheongsim-won reduced the activity of Ca/sup 2+/-ATPase in the membrane of erythrocyte. 5. Woohwangcheongsim-won reduced the platelet aggregation evoked by platelet agglutinin factor. 6. Woohwangcheongsim-won reduced the production of platelet-derived granules. 7. Woohwangcheongsim-won reduced the production of metabolites of arachidonic acid in diabetes, and also reduced the production of increased thromboxane B2. 8. Woohwangcheongsim-won reduced the synthesis of oxidized LDL-cholesterol. In conclusion, Woohwangcheongsim-won enhanced blood circulation in microvesseles, erythrocyte deformability and inhibited the increased platelet aggregation and the synthesis of oxidized LDL-cholesterol in diabetes. Therefore Woohwangcheongsim-won is believed to positively affect blood circulation (J Korean Oriental Med 2002;23(2):164-179)

Free Radical Scavenging of Flavonoids and Their Effects on Erythrocyte Na Leak, Platelet Aggregation and TBARS Production

  • We compared the radical scavenging activity of flavonoids and their antioxidant effects on erythrocyte Na leak, platelet aggregation and TBARS (thiobarbituric acid reactive substance) production, using Sprague Dawley rats. The concentrations of flavonoids needed for scavenging radicals by 50% ($SC_{50}$) in 0.1mM DPPH (2,2 Diphenyl 1-picryl hydrazyl) were: Quercetin, 7.4/$\mu$M; Catechin, 10.6$\mu$M; Morin, 22$\mu$M; Hesperidin, 400uM; and Naringin, 3.95mM. Morin completed its antioxidant activity in 2 minutes, while catechin, hesperidin and naringin had slow but long lasting antioxidant activity. Whole blood platelet aggregation, when incubated with quercetin or catechin, was significantly decreased (P<0.05) compared with the control. Sodium leak in intact erythrocytes was significantly lower when incubated with quercetin, compared with other flavonoids (P<0.05). Morin, hesperidin and naringin somewhat increased Na leak in intact erythrocytes. Sodium leak in erythrocytes treated with phenazine methosulfate (PMS) was increased overall, but was not affected by flavonoids. Intracelluar Na and K were not affected by treatment with PMS. TBARS production in platelet rich plasma (PRP) was significantly lower (P<0.05) than the control when incubated with quercetin or hesperidin. PMS treatment caused an increase in TBARS production regardless of flavonoids. In the present study antioxidant effects of flavonoids were not well correlated with their radical scavenging activities, although quercetin, which showed the strongest radical scavenging activity, had the greatest antioxidant effect.

Ultrasound Backscattering from Erythrocyte Aggregation of Human, Horse and Rat Blood under Rotational Flow in a Cylindrical Chamber

  • Human, horse and rat bloods in a cylindrical chamber where flow was controlled by a stirring magnet were used for studying red blood cell aggregation. Ultrasound backscattered powers from blood were obtained from the backscattered signals measured by a 5 MHz focused transducer in a pulse-echo setup. The experimental results showed the differences in red blood cell (RBC) aggregation tendency among the three mammalian species with an order of horse > human > rat. The ultrasound backscattered power decreased with stirring speed in human and horse blood, but no variations were observed in rat blood. Sudden flow stoppage led to the slow increase of the backscattered power for human and horse blood. There was no self-aggregation tendency in rat blood. The enveloped echo images showed the spatial and temporal variations of RBC aggregations in the cylindrical chamber. These observations from the different mammalian species may give a better understanding of the mechanism of RBC aggregation.
