The effects of Haematococcus pluvialis extracellular products on microbial growth and bacteriocin production were investigated to improve bacteriocin synthesis during the growth cycle of Lactobacilli. Lactobacillus pentosus KJ-108, L. plantarum KJ-10311, and L. sakei KJ-2008 were cultured in MRS and enriched medium (ERM) with or without supplement of the extracellular products obtained from a late exponential phase culture of Haematococcus pluvialis in modified Bold's basal medium (MBBM). In both MRS and ERM, the extracellular products strongly enhanced the growth as well as the bacteriocin production of all the lactic acid bacteria tested. The enhancing effect was observed in ERM with pH adjusted at 5 and 6. In addition, some difference in growth effects with the extracellular products of H. pluvialis was observed between pH 5 and 6 in ERM, but no effect was observed in the minimal medium. The final biomass and the final concentration of bacteriocin activity were associated with the cell growth that was promoted by the extracellular products of H. pluvialis, and the enhanced cell growth of the three lactic acid bacterial strains induced the increase of the specific bacteriocin production. Therefore, bacteriocin production and activity were influenced by the addition of the extracellular products of H. pluvialis in the culture medium.
Genetic determinant for the secondary metabolism was studied in heterologous expression in Streptomyces lividans TK-24 using Streptomyces griseus ATCC 10137 as a donor strain. Chromosomal DNA of S. griseus was ligated into the high-copy number Streptomyces shuttle plasmid, pWHM3, and introduced into S. lividans TK-24. A plasmid clone with 4.3-kb BamHI DNA of S. griseus (pMJJ201) was isolated by detecting for stimulatory effect on actinorhodin production by visual inspection. The 4.3-kb BamHI DNA was cloned into pWHM3 under the control of the strong constitutive ermEp promoter in both directions (pMJJ202); ermEp promoter-mediated transcription for coding sequence reading right to left: pMJJ203; ermEp promoter-mediated transcription for coding sequence reading left to right) and reintroduced into S. lividans TK-24. The production of actinorhodin was markedly stimulated due to introduction of pMJJ202 on regeneration agar. The introduction of pMJJ202 also stimulated production of actinorhodin and undecylproidigiosin in submerged culture employing the actinorhodin production medium. Introduction of pMJJ203 resulted in a marked decrease of production of the two pigments. Nucleotide sequence analysis of the 4.3-kb region revealed three coding sequences: two coding sequences reading left to right, ORF1 and ORF2, one coding sequence reading right to left, ORF3. Therefore, it was suggested that the ORF3 product was responsible for the stimulation of antibiotic production. The C-terminal region of ORF3 product showed a local alignment with Myb-related transcriptional factors, which implicated that the ORF3 product might be a novel DNA-binding protein related to the regulation of secondary metabolism in Streptomyces.
Logistics in a supply chain network has become an important operational strategy in a competitive market. A number of internal and external risks involved in the logistics operations in a company tend to create problems in fulfilling customer orders. This research presents how ERM (enterprise risk management) can be used to identify, assess, and control logistics risks. An electronic company's logistics activities were used as an industry case to demonstrate a way to identify and assess risks surrounding global logistics function. This paper has further presented action plans to mitigate the impact of the risks that occur.
최근 IT 거버넌스 개념이 기업경영 현장에 적용되고 있다. 본 논문에서는 클라우드 컴퓨팅 환경에서 IT 거버넌스를 기반으로 기업의 정보보호 위험을 최소화할 수 있는 비즈니스 모델에 대해 연구한다. 특히, 정보보호 거버넌스 세부 주제인 위험관리를 연계하는 상호작용 모델(IT 거버넌스 프레임워크인 COBIT과 연계하여 시너지를 높일 수 있는 구조)을 제안하고자 한다. 여기서, 시너지는 효과적, 전략적이고 안전한 비즈니스 지원을 의미하며, BMIS의 4가지 요소, 6가지 역동 연결자와의 상호작용성에 대해 분석이 요구된다. 따라서 COSO ERM이나 COBIT Risk IT Framework를 기반으로 위험관리와 연계될 수 있는 상호작용 모델을 제안한다.
The aim of this study was to investigate the prevalence of antimicrobial resistance and virulence factors in staphylococci isolated from canine otitis externa. A total 295 causative microorganisms were isolated. The most common isolated species were Staphylococcus (S) pseudintermedius (94 isolates) followed by Pseudomonas aeruginosa (60 isolates), S. schleiferi (25 isolates), Escherichia coli (23 isolates) and Proteus mirabilis (20 isolates). Staphylococci isolates were showed high resistance to penicillin (78.6%), erythromycin (55.9%), tetracycline (52.4%), clindamycin (51.7%) and ciprofloxacin (42.8%). Of the 145 staphylococci isolates, 49 (33.8%) methicillin-resistant staphylococci (MRS) were observed, distributed among S. pseudintermedius (n=34), S. schleiferi (n=6), S. epidermis (n=4), S. hominis (n=2), S. aureus, S. caprae and S. saprophyticus (n=1, respectively). Forty-three (87.8%) of 49 MRS and 10 (10.4%) of 96 methicillin-susceptibility staphylococci harbored mecA gene. About 80% of MRS were multidrug-resistant with resistance to at least one antibiotic in three or more antibiotic classes. Resistance genes blaZ (93/114, 81.5%), ermB (35/81, 43.2%), ermC (3/81, 3.7%), aacA-aphD (50/54, 92.5%), tetM (69/76, 90.7%) and tetK (6/76, 7.8%) were detected among resistant isolates. Virulence factors genes lukF and lukS were found in 100%(145/145) and 43.4%(63/145), respectively. Genes encoding ermA, eta, etb and tsst were not detected. To the best of our knowledge, this is the first study which investigated for the presence of genes encoding antimicrobial resistance and staphylococcal toxins in staphylococci isolated from canine otitis externa. A continuous monitoring and surveillance program to prevent antimicrobial resistance in companion animals is demanded.

The ezrin-radixin-moesin (ERM) proteins are a family of membrane-associated proteins known to play roles in cell-shape determination as well as in signaling pathways. We have previously shown that amphetamine decreases phosphorylation levels of these proteins in the nucleus accumbens (NAcc), an important neuronal substrate mediating rewarding effects of drugs of abuse. In the present study, we further examined what molecular pathways may be involved in this process. By direct microinjection of LY294002, a PI3 kinase inhibitor, or of S9 peptide, a proposed GSK3β activator, into the NAcc core, we found that phosphorylation levels of ERM as well as of GSK3β in this site are simultaneously decreased. These results indicate that ERM proteins are under the regulation of Akt-GSK3β signaling pathway in the NAcc core. The present findings have a significant implication to a novel signal pathway possibly leading to structural plasticity in relation with drug addiction.
The SCO3388 gene from Streptomyces coelicolor is homologous to tmrB, the tunicamycin resistance gene of Bacillus subtilis. The SCO3388-inactivation strain (SY-tbl-1) was generated by replacing SCO3388 with thiostrepton resistance gene. Spores of S. coelicolor derivatives were prepared on mannitol-soy flour (MS) agar on which SY-tbl-1 displayed no significant defect in growth and development. When plated on R4 agar, spores of SYtbl-1 displayed retardation in growth and sporulation, whereas its mycelium gave rise to normal growth. Thus, SCO3388 is suggested to be involved in the dormant spore germination. Expression of SCO3388 under the ermE1 promoter restored but only partially the ability to sporulate in SY-tbl-1. Neither SY-tbl-1 nor SY-tbl-1/ermE1p-SCO3388 showed a difference in tunicamycin resistance to the wild type whereas, interestingly, the introduction of ermE1p-SCO3388 dramatically enhanced spore survival to heat and detergent treatments, suggesting that SCO3388 might play a role in the maintenance of spore cell wall integrity.
본 연구는 하천복원에 대해 하천기술자들과 생태학자들 간 이해와 사고의 간격을 확인하고 그 간격을 줄이기 위해 하천복원의 정의와 의의를 학술적, 논리적으로 재 고찰 하는 것이다. 이를 위해 우선 하천생태계의 기능, 재화와 용역에 대해 de Groot et al. (2002)의 자료에 근거하여 분석하였다. 이를 토대로 지금까지 알려진 하천복원을 포함한 유사 활동의 정의와 복원전략을 재고찰 하였다. 그 다음 생태복원모형 (ERM), 어메너티복원모형 (ARM), 준생태복원모형 (semi-ERM) 등 기존의 하천복원모형의 개념을 보완하였다. 이 연구는 그 동안 하천복원에 대해 이해와 생각을 달리한 전문가들이 서로 간 사고의 간격을 줄이는데 도움이 될 것으로 기대된다.
본 연구는 낙동강 하구 인근의 해양퇴적토에 포함된 PAHs의 농도를 분석하여 퇴적토의 생태학적 위해성을 평가하고자 하였다. 퇴적토의 주성분은 $SiO_2$, $Al_2O_3$, CaO, $Fe_2O_3$였으며, 강열감량은 2.97%에서 8.39%의 분포를 보였다. 총 PAHs의 농도는 $128.4ng\;g^{-1}$에서 $507.4ng\;g^{-1}$의 범위를 가졌으며 2고리와 4고리 형태의 PAHs로 주로 구성이 되어있었다. 8개 지역의 각각의 PAHs의 농도는 모두 ERL 이하로 생태학적 위해성은 낮은 것으로 판단되었다. 지역별 PAHs의 생태학적 위해성 평가는 M-ERM-Q를 사용하여 이루어졌고, 계산 값 모두 0.1 이하로 생태학적 위해성은 낮음으로 분류되었다. 발생원 분석을 통해 각 지역의 PAHs의 발생원은 N-1과 N-7은 풀, 나무, 석탄 등의 연소이며, N-5와 N-6는 석유연소, N-2, N-3, N-4, N-8은 석유오염인 것으로 판단되었다.
