  • Attempts were made to investigate the sterol components of Xerocomus badius Fr. Kuhner ex Gilbert which grows wildly in Korea. Its carpophores were collected in Gyeong-Gi Province and extracted with chloroform and methanol. Two compounds were isolated from the extract and one of these compounds was identified as ergosterol by TLC, GLC and chemical tests.

  • Attempts were made to investigate the sterol components of Coriolus versicolor (Fr.) Quel, which is one of medicinal fungi and which grows wildly in Korea.Its carpophores were collected in the Gyeong Gi Province and extracted with chloroform and methanol. Four Compounds were isolated and one of these compounds was identified as ergosterol by T.L.C., G.L.C. and chemical tests. ${\beta}-Sitosterol$ appears to exist also in the carpophore.

  • Ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were screened for their ability to inhibit nitric oxide (NO) production in LPS-activated RAW 264.7 cells. Compounds 1, 2, and 3 reduced NO production in the assay with $IC_50$ values of 29.7 ${\mu}M$ (1), 15.1 ${\mu}M$ (2), and 18.4 ${\mu}M$ (3) respectively. They also suppressed the expression of protein and m-RNA of iNOS and COX-2 in a dose dependent manner by western blot analysis and RT-PCR experiment in LPS-activated microglial cells.

Antifungal Activity of an Endophytic Fungus Aspergillus versicolor DYSJ3 from Aphanamixis grandifolia Blume against Colletotrichum musae

  • An endophytic fungus strain DYSJ3 was isolated from a stem of Aphanamixis grandifolia Blume, which was identified as Aspergillus versicolor based on the morphological characteristics, internal transcribed spacer (ITS) and calmodulin gene sequences analyses. A. versicolor DYSJ3 exhibited strong antagonistic activity against Colletotrichum musae, C. gloeosporioides and Fusarium oxysporum f. sp. cubense with the inhibition rates of 61.9, 51.2 and 55.3% respectively. The antifungal metabolites mainly existed in the mycelium of A. versicolor DYSJ3, and its mycelial crude extract (CE) had broad-spectrum antifungal activities against plant pathogenic fungi. The CE had a good thermal stability, and the inhibition rate of 100 mg/mL CE against C. musae was above 70.0% after disposing at 120 ℃ for 1 h. Five secondary metabolites were isolated from the CE and identified as averufanin, ergosterol peroxide, versicolorin B, averythrin and sterigmatocystin. Activity evaluation showed versicolorin B exhibited inhibitory effects on the mycelial growth and conidial germination of C. musae, and sterigmatocystin had a weak inhibitory effect on the mycelial growth of C. musae.

  • The EtOAc and CH$_2$Cl$_2$ soluble fractions from the fruit body of Ganoderma applanatum showed strong aldose reductase inhibitory activity. Nine compounds were isolated from both fractions. They were identified by spectral data as D-mannitol (1), 2-methoxyfatty acid (2), cerebrosides [(2S,3R,4E,8E)-1-O-${\beta}$-D-glucopyranosyl-3-hydroxy-2-[(R)-2'-hydroxypalmitoyl]amino-9-methyl-4,8-octadecadiene] (3), daucosterol (4), 2,5-dihydroxybenzoic acid (5), protocatechualdehyde (6), 5-dihydroergosterol (7), ergosterol peroxide (8), and cerevisterol (9). Comparison of Liquid and Solid-State Fermentation Processes for the Production of Enzymes and Beta-Glucan from Hulled Barley

  • Solid-state fermentation using hulled barley was carried out to produce enzymes and β-glucan. The one-factor-at-a-time experiments were carried out to determine the optimal composition of the basal medium. The modified synthetic medium composition in liquid-state fermentation was determined to be 70 g/l hulled barley, 0 g/l rice bran, 5 g/l soytone, and 6 g/l ascorbic acid. Optimal pretreatment conditions of hulled barley by solid-state fermentation were evaluated in terms of maximum production of fungal biomass, amylase, protease, and β-glucan, which were 1.26 mg/g, 31310.34 U/g, 2614.95 U/g, and 14.6% (w/w), respectively, at 60 min of pretreatment condition. Thus, the solid-state fermentation process was found to enhance the overall fermentation yields of hulled barley to produce high amounts of enzymes and β-glucan.

Biological activity in hot water extract from fermented Coix lacryma-jobi L. var. mayuen Stapf. by Hericium erinaceum (Bull. : Fr.) (노루궁뎅이균사발효 율무 열수추출물의 유용성분 및 생리활성)

  • This study was conducted to determine the antioxidant, nitrite scavenging, melanin tyrosinase inhibitory, and melanogenesis inhibitory activities of fermented Coix lacryma-jobi L. var. mayuen Stapf. by Hericium erinaceum (Bull.: Fr.) mycelial hot water extract (FCLHE). Additionally, we analyzed β-glucan and ergosterol contents in FCLHE and C. lacryma-jobi hot water extract (CLHE). The ergosterol and β-glucan contents in FCLHE were 740.2 mg% and 245.3 mg%, respectively, whereas these components were not detected in CLHE. FCLHE showed higher cell viability than CLHE. When B16F10 cells were treated with 500 ㎍/mL each of CLHE and FCLHE, the FCLHE treated cells produced 8.9 uM nitric oxide (NO), which was lower than that produced by CLHE treated cells (10.6 uM). The FCLHE treated cells showed significantly greater tyrosinase inhibition and melanin production at all tested concentrations than when compared to the CLHE treated group. Antioxidant parameters such as DPPH and ABTS radical scavenging activities were higher in FCLHE than in CLHE. These results suggest that FCLHE can be used as a raw material for functional foods, for food additives, and in the cosmetic industry.

Quality characteristics of spread jam using low-salt Lentinula edodes soybean paste (저염표고된장을 활용한 스프레드잼의 품질특성)

  • In this study, a low-salt Lentinula edodes soybean paste (LSLESP) with improved palatability and storability relative to traditional Korean soybean paste was developed by utilizing low quality log cultivation of L. edodes. We also conducted quality characteristic analyses of spread jam made from low-salt L. edodes soybean paste (SJLLESP). The lowest salinity content and the highest L (brightness) value was found in LSLESP. According to proximate compositions analysis, crude protein, crude fat, and ash content were higher in commercial soybean paste than in LSLESP. Glucose, fructose, and maltose were the only free sugars detected in commercial soybean paste, whereas arabinose and fucose were additionally detected in LSLESP. Only two types of organic acid, were detected in common in commercially available soybean paste and LSLESP. Four types of organic acid, were detected in SJLLESP due to its added ingredients. The major free amino acids in commercially available soybean paste and LSLESP are histidine, glutamic acid, and arginine. Total amino acid, content was higher in LSLESP (54.81 mg%) than in commercial soybean paste (49.26 mg%). Total free amino acid, content in SJLLESP was 43.01 mg%. Ergosterol and β-glucan contents were highest in SJLLESP. The elevated ergosterol and β-glucan content in low-salt LSLESP relative to commercial soybean paste was significant. In conclusion, LSLESP and SJLLESP contain useful components from L. edodes, and offer the advantage of low salinity. LSLESP and SJLLESP could thus contribute to the development of health foods using L. edodes.

Fungicide Spray Program to Reduce Application in Anthracnose of Strawberry (살균제 살포횟수 감소를 위한 딸기 탄저병 방제프로그램)

  • The effect of various fungicides on anthracnose of strawberry, caused by Colletotrichum gloeosporioides, was determined in vitro, and in greenhouse and field trials. The $EC_{50}$ values of benomyl were clearly different between two sensitive and resistant groups of isolates. Iminoctadine tris had lower $EC_{50}$ values than mancozeb and propineb as protective fungicides and the response of mancozeb, propineb and azoxystrobin was variable depending on the isolate. In the greenhouse, pre- and post- inoculation fungicide applications significantly reduced disease compared to the non-treated control. Propineb, mancozeb and azoxystrobin were effective in controlling the disease when applied prior to inoculation. Metconazole and prochloraz-Mn treatments as ergosterol biosynthesis-inhibiting fungicides had the lowest incidence of anthracnose. In the nursery field trials in 2009 and 2010, the reduced fungicide spray program provided similar levels of disease control compared to the calendar-based applications with captan. A reduced spray program based on efficacious fungicides such as prochloraz-Mn will be useful for strawberry growers and provide more options for controlling anthracnose in Korea.

Effects of fruit body characteristics of Lentinula edodes according to irradiation intensity of the green LED with sawdust substrate cultivation (표고 톱밥배지 재배시 녹색LED 광량이 자실체 생육에 미치는 영향)

  • In previous studies, we selected the green LED(light emitting diodes) for suitable wavelength of the light by higher commercial yields and lower ratio of the abnormal fruit body in Lentinula edodes. In this study, we aimed to select efficient irradiation intensity of the green LED. Stronger irradiation intensity of the green LED resulted in more polyphenol content of fruit body. And Polyphenol content of fluorescent lamp was similar to that of the green LED $20umol{\cdot}m^{-2}{\cdot}s^{-1}$.. Ergosterol content of the green LED $5umol{\cdot}m^{-2}{\cdot}s^{-1}$ was showed higher 2.1 times than that of the fluorescent lamp. In four level of irradiation intensity, 5, 10, 15, $20umol{\cdot}m^{-2}{\cdot}s^{-1}$ there was no big difference in characteristics of the fruit body. However the yield of fruit body in the green LED $5umol{\cdot}m^{-2}{\cdot}s^{-1}$ is higher than the others. In addition, The amount of electricity used of the green LED $5umol{\cdot}m^{-2}{\cdot}s^{-1}$ was reduced 15.9% than that of the fluorescent lamp. In conclusion, we selected $5umol{\cdot}m^{-2}{\cdot}s^{-1}$ for suitable irradiation intensity in Lentinula edodes with sawdust substrate cultivation.
