유통 한약재 중 내분비계 장애물질로서의 잔류농약에 관한 연구(I) (Monitoring Research for Residual Pesticides as Endocrine Disruptors in Natural Medicines (I))

  • Research on pesticide residues as endocrine disruptors in natural medicines was initiated by Korea Food & Drug Administration this year. We determined the presence and levels of certain pesticides in selected natural medicines. The natural medicines collected this year are Glycyrrhiza Root, Cinnamon Bark, Pueraria Root, Polygonatum Rhizome, Jujube, Schizandra Fruit, Lycium Fruit, Liriope Tuber, Eucommia Bark, Peony Root, Korean Angelica, Dioscorea Rhizome, Cnidium Rhizome, Cassia Seed, Platycodon Root, Comus Fruit, Mentha Herb, Epimedium Herb, Bupleurum Root, and Ginger, which have no data for pesticide residues and 192 samples of them were circulated in Korea, 28 samples were circulated in China. In order to analyze many pesticides in large number of samples we used simultaneous multi-residue analysis of pesticides by GC-ECD, which was followed by GC-MSD analysis to confirm the identity of the detected pesticide in each sample.

Icariin 함량에 따른 삼지구엽초 추출물의 항돌연변이 및 면역활성 (Antimutagenicity and Immuno Activity of Extracts from Epimedium koreanum Nakai Containing Different Icariin Quantity)

  • 예로부터 삼지구엽초는(Epimedium koreanum Nakai)는 한방에서 강장, 강정, 이뇨 등의 약재로 이용되어 왔다. 본 연구에서는, 삼지구엽초의 주성분인 icariin의 함량에 따른 항돌연변이 및 면역활성을 탐색하여 향후 기능성 식품소재로서 활용하고자 하였다. Ames test 결과, icariin 함량에 따른 삼지구엽초 추출물은 TA98 및 TA100 균주 모두에서 집락수가 음성대조군에 비하여 농도 변화에 따른 집락수의 큰 변화를 나타내지 않으므로 돌연변이원성이 없는 것으로 나타났다. 간접변이균인 B(a)P 항돌연변이 실험에서는 열수 및 에탄올 추출물이 각각 80% 그리고 90% 이상을 그리고, 직접변이균인 4NQO 항돌연변이균에서는 70% 이상으로 icariin 함량이 많은 순인 KE9412, KE9408, KE9405 순으로 항돌연변이 억제효과를 각각 나타내었다. 인간면역세포인 B와 T 세포를 이용한 면역활성 실험결과, 열수 추출물에서는 icariin 함량이 적은 KE9405가 1.8배로 가장 높은 활성을 나타냈으나, 에탄올 추출물에서는 열수 추출물과는 반대로 icariin 함량이 많은 KE9412가 1.74배로 가장 높은 활성을 나타내었다. 이러한 결과로 인하여, 삼지구엽초는 항돌연변이능과 면역활성 효능을 가지고 있으므로, 건강기능성 식품 소재로의 효용 가치가 높을 것으로 판단된다.

  • Effects of medicinal herb extract on nonspecific immune responses, hematology and disease resistance against Edwardsiella tarda in olive flounder, Paralichthys olivaceus were evaluated. Wormwood, Artemisia asiatica NAKAI and barrenwort, Epimedium koreanum NAKAI were mixed at a ratio of 7 : 3 (w/w) for 2-herbs extract and wormwood, barrenwort, Korean forsythia, Forsythia koreana NAKAI, chrysanthemum, Chrysanthemum zawadskii var. latilobum KITAMURA, peppermint, Mentha arvensis L. var, piperascens MALINV., great burnet, Snaguisorba afficinalis L., Lizard tail. Saururus chinensis BAILL., mulberry, Morus alba L., and star anise, Illicium varum HOOK, f, at the same weight for 9-herbs extract. Two-herbs of 9-herbs extract were prepared by heating after adding 10㎖ of distilled water per g of the herb mixtures. Fish (10.3$\pm$2.5g) were fed the experimental diets supplemented with the 2-herbs or 9-herbs extract at the different concentrations of 0%, 0.1%, 0.5% and 1% per kg diet for 12 weeks. Lysozyme and bactericidal activities of serum, and hematological characteristics were examined during experimental period. After feeding test period, all experimental groups were challenged with E. tarda. Lysozyme activity from the fish fed the diet supplemented with 0.1% or 0.5% of 2-herbs extract was significantly higher than the control. But there was no difference both in bactericidal activity and hematology among each group. Sixty seven % of relative percent survival values (RPS) in the group fed the diet supplemented with 0.1% of 2-herbs was higher than the other group and the control. These results suggest that supplenmentation of 0.1% of 2-herbs extract to a commercial diet may enhance disease resistance in olive flounder. Although both 0.1% and 0.5% 9-herbs extract did not improve non-specific immune reponses, they could enhance disease resistance of 53% RPS, respectively.
