To develop Epicoccosorus nematosporus as a mycoherbicide of Eleocharis kuroguwai, the optimum temperature and humidity for sporulation of the pathogen were studied. Conidial production was most abundant at $28^{\circ}C$ with RH 60%, which yielded 661 mg in 9 cm Petri dish. Light intensity of 3,000 up to 7,500 lux was effective in stimulating conidial production of E. nematosporus on oatmeal agar, Light intensity affected sporulation more significantly than temperature. In the pot test, at least 12 h of dew period at $20^{\circ}C$ and $25^{\circ}C$ was required to achieve satisfactory conidial germination and appressorial formation. Few were killed at 8 h of dew period regardless of temperature. Sixteen hours of a single dew treatment immediately after inoculation killed more plants than did two or three repetitive dew treatments of 8-12 h.
Epicoccosorus nematosporus was detected in all 22 counties surveyed, but the frequency of occurrence of fingerprint stem blight disease (FSBD) on Eleocharis kuroguwai caused by the fungus varied with fields and regions. The disease occurred more frequently in mountain areas than in plain areas. E. nematosporus was detected in less than 20% of plain areas such as Cimjae and Milyang, whereas, it occurred in 40-60% of mountain areas such as Sangiu and Jangsoo. In Milyang, mean temperature in July to August 1993 ranged from 17 to $27^{\circ}C$ with 14.3 h of dew period. Meanwhile, in the mountain area such as Sangiu, Gyeongbook, temperature ranged from 17 to $24^{\circ}C$ with 16.1 h of dew period. Plant mortality was 61% in Milyang and 82% in Sangju. Underground tuber formation was highly suppressed at 16 and 33 tubers per plot in Sangju and Milyang, respectively. In 1992 and 1995, plants infected ranged from 40 to 78% in July to September. This sharply decreased to an average of 15% in 1999.
한국의 수도 재배 지대에서 문제가 되는 다년생 논잡초 올방개에 자연상태에서 기주특이적으로 기생하여 올방개 줄기를 고사시키는 병원균인 Epicoccosorus nematosporus를 우리나라에서 처음으로 분리, 동정하였으며, 이 병을 올방개 지문무늬병(Finegerprint stem blight disease of water chestnut)으로 명명하였다. 분생포자는 분생자층 위에서 형성이 되며 무격막, 단핵이며, 44.5~72 $\mu\textrm{m}$ 크기의 양끝이 뾰족한 선충 모양이다. 갈색의 병반상에서 흑색의 분생자층은 발달하여 특이한 지문무늬를 형성하는 것이 특징이다. 이 병균은 전국의 올방개가 자라는 논에서 분포하며 매년 발생한다. 온실 접종시 초기의 병징은 접종후 4~5일만에 바늘끝 모양의 회흑색의 반점으로 나타나며 시간이 경과하면서 병반은 급속도로 진전하여 접종후 10~15일 정도 후 약 10mm 내외의 갈색병반이 형성이 되어 올방개 줄기를 감싸고 그 줄기 전체는 고사하게 된다. 분생포자 현탁액(3.2$\times$10\ulcorner conidia/ml)을 온실 조건하에서 분무접종 하였을 때 20일 이내에 약 78.0~93.3% 정도의 고사율을 나타내었다. 올방개 지문무늬병균의 분생포자 현탁액(3.2$\times$10\ulcorner conidia/ml)을 온실 조건하에서 벼 등 31개 작물과 피 등 15개 잡초의 유묘에 접종하였을 때 올방개를 제외한 어떤 식물에도 병원성이 없었다.
Effects of temperature and dew period on sporulation of a mycoherbicide, Epicoccosorus nematosporus, on the lesion of its host, Eleocharis kuroguwai were determined. Conidia formation was first observed after 10 days on plants incubated for either 12 or 16 h in a dew chamber at 28$^{\circ}C$; 16 h dew period resulted in more conidia formation. As the dew period was decreased to less than 8 h, fewer conidia formed. Conidial production was most abundant at 28$^{\circ}C$ and produced as much as 3.3$\times$10$^4$conidia per lesion, while 0.1$\times$10$^3$and 2.3$\times$10$^3$conidia per lesion were produced at 16$^{\circ}C$ and 36$^{\circ}C$, respectively. Alternating temperature regimes, i.e., 30/15, 30/20, 28/20, and 28/15$^{\circ}C$ (day/night) were much better than constant temperature, i.e., 30/30, 28/28/, and 20/2$0^{\circ}C$ for sporulation. In the second sporulation, there were as much as 3.1$\times$10$^4$conidia per lesion (ca. <50% of the first sporulation). Then, sporulation dropped sharply to 6.2$\times$10$^2$conidia per lesion in the third sporulation. Results of this study suggest that temperature combined with dew period is the primary limiting factor in the use of E. nematosporus as a mycoherbicide off, kuroguwai.
The study was conducted to determine the cultural conditions and the effect of inert fillers for melanization and sporulation abilities of sodium alginate pellets, and the weeding efficacy of the formula in the field. Melanin production of E. nematosporus was affected by striking frequency. Percentage of melanized beads was increased to 80.6% at higher rpm up to 180. The melanized pellets produced more conidia with abundant mucilage than unmelanized pellets. Shaker culture of Epicoccosorus nematosporus with sodium alginate yielded a total of 55 mg per 100 pellets. Percentage of melanized pellets was highest with 81.0% and 83.3% of melanization, when wheat bran and rice polish were amended and produced the conidia with 65.4 and 68.4 mg per 100 pellets, respectively. When 1 L of conidial suspension of 6.0$\times$$10^5$ conidia per ml was applied on 30-day-old plants in a plot, 74.5% of the plants were killed within 20 days, whereas, its melanized sodium alginate pellets killed 57.8% of the plants in the same period. The number of tuber formation of Eleocharis kuroguwai in the untreated control plots was 128.5 per plot, but those of the plots treated with conidial suspension and melanized pellets were 22.1 and 39.7, respectively, at the end of the season. Results of this study showed that melanization of mycelia-mixed sodium alginate are an important sporulation factor in E. namatosporus as a mycoherbicide.
In greenhouse studies, control efficacy of water chestnut (Eleocharis Kuroguwai) by Epicoccosorus nematosporus was affected by temperature and dew condition. The appressoria were formed abundantly in the range of 20~28$^{\circ}C$. When stem segments o(30 cm long) of the water chestnut were inoculated with the conidial suspension of E. nematorporus, the mean conidial number attacted amounted to 2,545 conidia. Out of 2545 conidia attacted to the stem pieces, 1,733 (68%) conidia formed appressoria. When these stem pieces were treated for 24 hr at 28$^{\circ}C$ under dew condition, 183,1 (7.2%) lesions were formed 10 days after incubation. The time necessary for the death of the plants was about 24 days. Appressoria were formed at 15~35$^{\circ}C$, but decreased rapidly in their numbers at the temperature lower than 1$0^{\circ}C$ or at 35$^{\circ}C$. The appressoria formation seemed to be depended on the dew duration, which was effective to the lesion formation and plant mortality. Under dew duration of 16~24 hr with temperature range of $25^{\circ}C$ to 3$0^{\circ}C$, the weed control was increased up to 93.9%. There were no differences between the first and second or third dew treatments. A delay of 2 or 3 days in dew treatment did not increase the mortality of plants. For the use of E. nematosporus as a mycoherbicide of water chestnut, a conidial suspension should be applied when dew conditions are kept for 12 hr after inoculation.
To find optimum application methods of Epicoccosorus nematosporus for control of water chestnut, five different concentration of conidial suspensions ($10^[3}$ conidia/ml to $10^[7}$ conidia/ml) fo the fungus were applied 1 to 4 times on 10~40 days old seedlings of water chestnut in greenhouse. Inoculum levels equal to or greater than $10^[5}$ conidia/ml killed significantly more shoots (82.6%~92.1%) and suppressed significantly underground tuber formation compared to inoculum concentration less than $10^[4}$ conidia/ml. When the conidial suspension of ($6.3 {\times} 10^{5}$conidia/ml of E. nematosporus was sprayed 2 times in 7 days interval, percentage of the killed plants was up to 98.7%. Numbers of reshoots and tubers were alsosuppressed significantly compared to one time application. The percents of killed shoots were similar between 10-day-old and 20-day-old seedlings, and significantly higher than those treated on 30- or 40-day-old seedlings. The fungus treated on 20-day-old seedlings was the most effective because there was high number of reshoots from 10-day-old seedlings. Therefore, optimal application conditions for E. nematosporus is 2~3 times of application in 7 days interval with $10^[5}$ conidia/ml on 20-day-old seedling age.
This study was conducted to determine the efficacy of Epicoccosorus nematosporus for the control of Eleocharis kuroguwai and to evaluate the meteorological factors which affect weeding efficacy in field conditions for three years (1996-1998). The best time to control E. kuroguwai with E. nematosporus as a biological control agent in the field was in July, when temperature ranged from 20.4 to $23.4^{\circ}C$; the surface wetness duration was 12.6-16.1 hours, and application time of 6:00 p.m. and 8:00 p.m.; and weeding efficacy was 81-90%. On 10 June 1996 in Milyang area, where the field experiments were performed, mean temperature was $16.5^{\circ}C$ with 11.3 hours of dew duration. Meanwhile, on 20 Aug. 1996 the temperature was $21.3^{\circ}C$ with 15.4 hours of dew duration. During these periods, the weeding efficacy was recorded at 61.8 and 60.8%, respectively. Time required for complete plant death was 25.8 and 25.6 days at application times 10 June and 20 Aug., respectively. At the time of application on 7,18, and 27 July 1996, mean temperature was 20.4-$23.4^{\circ}C$ with 12.6-16.5 hours of dew duration. The weeding efficacies of these periods were very hi일 with 81.4-90.8%. Three years of field observations from 1996 to 1998 showed that infection in the field can occur at any time through the summer season, although total infection rates vary between months and between years. In 1996, plant infection rapidly increased from 56% on 30 June, 82.4% on 15 July, 94.6% on 15 August, and 92.8% on 15 September under favorable meteorological conditions such as minimum temperature of $17.6^{\circ}C$ and maximum temperature of $27.1^{\circ}C$, with 86% relative humidity and 977.5 mm of rainfall during E. kuroguwai growing season. However, in 1997, the disease incidence was very low because of unfavorable weather conditions brought about by the hot temperature and the low amount of rainfall at 321.5 mm. Disease progress was slow from 24.4% on 30 June to 49.2% at the end of the growing season.
The paper was reviewed the research results on weed dynamics and effective control methods in direct-seeded rice crop. Direct seeding method resulted in drastic increment of weed growth compared to transplanting method and also changed in troublesome weed flora. Two to three fold more weeds were harvested at the direct seeded rice and weed flora of dominant species shifted toward $C_4$type grass weeds. Some of the important troublesome weeds in direct seeded rice were Echinochloa crus-galle, Oryza saliva ssp spontanea, Leptochloa chinensis. Setaria viridus. Digitaria adsendens, Sesbania exaltata, Aeschynomene indica, Algae, etc. Yield loss due to weed competiton was about 40-60% for water-seeded and about 70-100% for dry-seeded rice while these for transplanted rice were about 25-35% for mechanical transplanting and about 10-20% for manual transplanting, respectively. Integrated weed management concept was neede to approach weed control effectively. Several cultural technologies were very effective to suppress the weed growth. These were tillage operation, water management, seeding date and seeding rate. Crop residues of barley, rice, wheat, oat and italian ryegrass were also effectivly suppressed the paddy weeds particularly to Potamogeton distiuctus, a perennial broadleaf weed. A pathogen of Epicoccosorus nematosporus identified from Eleocharis kuroguwai was an excellent potential bioagent to control the most troublesome perennial sedge weed of E. Kuroguwai without arising any detrimental effect. The herbicidal efficacy of this pathogen was as high as bentazon herbicide. Plant growth regulator of paclobutrazol (pp-333) was another possible alternative to reduce the herbicide use. In current, herbicide exhibited the most conspicuous results to control weeds in direct-seeded rice even though the application technologies were not fully established. Recommendations for herbicide application were suggested for in both water-and dry-seeded rice in USA, Japan and Korea, respectively. To make better and comprehensive recommendations further studies on weed ecology and herbicide development were emphasized.
