Red sea cucumber Stichopus japonicus is an invertebrate animal inhabiting in coasts of Korea, China, and Japan. They are traditionally used for food and medicine and well known for their distinctive biologically and pharmacologically important compounds. We investigated the effect of amyloglucosidase (AMG) enzymatic extracts of S. japonicus (AESJ) on the proliferation and cytokine secretion of murine splenocytes stimulated with concanavalin A (Con A). AESJ enhanced the proliferation of splenocytes and the production of IL-2 (Th1 cytokine), IL-1β (Th1 promoting cytokine), and IL-4, IL-10 (Th2 type cytokines) when treated alone. However, under Con A stimulation, AESJ suppressed the proliferation of splenocytes, attenuated the secretion of IL-2, IL-4, IL-10, and enhanced IL-1β secretion. These results suggest that AESJ exhibits immunomodulatory effect by moderating the proliferation of splenocytes and the secretion of IL-2, IL-1β, IL-4, and IL-10 differently depending on the absence and presence of Con A stimulation. These data evidence the immunomodulatory potential of AESJ, which can be further developed into a functional food mediating homeostasis.
This study was carried out to examine the mutagenicity of extracts from grilled pork belly and the effect of garlic on it by using Arnes test. And in order to imitate the in vivo metabolic activation system of the mutagens, the enzymatic activation system was adopted. The results are summarlized as follows: 1. The degree of browning in pork belly extracts increased with the increasing heating intensity of the grilling. 2. When pork belly grilled at "low" heating intensity, no mutagenicity was detected. However with the samples grilled at "medium" and "high" heating intersity, mutagenicity was recognized. 3. The mutagenicity of grilled pork belly extract decreased remarkabley with the addition of S-9 mix. 4. The mutagenicity of grilled pork belly extract decreased with the addition of garlic extract.

Deodorizing activity of polyphenol cxidase (PPO) extracted from apples was investigated by measuring the changes of methyl mercaptan as an indicator of halitosis in human mouths. In the studies of apple extracts on deodorizing activity, the deodorizing activity was increased with the amount of apple extracts. In the cases of adding PPO to the low molecular fraction of apple extracts, the deodorizing activities were increased with the amount of the law molecular fraction of apple extracts and the reaction time of the extracts with PPO. Deodorizing activities of PPO is thought that o-quinone as an intermidiate produced by an oxidative reaction of PPO during enzymatic browning reactions may react with methyl mercaptan to form a non-volatile and sulfur-containing compound .
This study was conducted to improve the antioxidative activity of instant noodles containing enzymatic extracts from Ecklonia cava (EEC). EEC has relatively better antioxidative activity than extracts from other indigenous plants in Jeju Island. The EEC (2.5 mg/mL) had 82.5% for the hydroxy radical, 78.4% for the DPPH radical, and 64.9% for the superoxide anion radical scavenging activities, and 65.2% for the cell viability (100 ${\mu}g/mL$). According to the texture of the dough, the DPPH free radical scavenging of uncooked instant noodles, sensory evaluation of cooked instant noodles, and turbidity of the cooking drip, the optimal EEC concentration was 1.8% for the instant noodles. The major amino acids in the instant noodles with EEC were glutamic acid (24.2%), proline (10.2%), valine (10.0%), and isoleucine (12.3%). The zinc and iron in the instant noodles were enhanced by adding 1.5-1.8% EEC. The antioxidant activity of instant noodles with EEC was 75.4% for the hydroxy radical, 74.1% for the DPPH radical, and 51.2 % for the superoxide anion radical scavenging activities.
Glycosylation of resveratrol was carried out by using the amylosucrase of Deinococcus geothermalis, and the glycosylated products were tested for their solubility, chemical stability, and biological activities. We synthesized and identified these two major glycosylated products as resveratrol-4'-O-α-glucoside and resveratrol-3-O-α-glucoside by nuclear magnetic resonance analysis with a ratio of 5:1. The water solubilities of the two resveratrol-α-glucoside isomers (α-piceid isomers) were approximately 3.6 and 13.5 times higher than that of β-piceid and resveratrol, respectively, and they were also highly stable in buffered solutions. The antioxidant activity of the α-piceid isomers, examined by radical scavenging capability, showed it to be initially lower than that of resveratrol, but as time passed, the α-piceid isomers' activity reached a level similar to that of resveratrol. The α-piceid isomers also showed better inhibitory activity against tyrosinase and melanin synthesis in B16F10 melanoma cells than β-piceid. The cellular uptake of the α-piceid isomers, which was assessed by ultra-performance liquid chromatography (UPLC) analysis of the cell-free extracts of B16F10 melanoma cells, demonstrated that the glycosylated form of resveratrol was gradually converted to resveratrol inside the cells. These results indicate that the enzymatic glycosylation of resveratrol could be a useful method for enhancing the bioavailability of resveratrol.
An efficient production method of chlorella extract was developed by enzymatic treatment using cell lytic and proteolytic enzymes. The suitable dosage of Tunicase, a cell lytic enzyme, was found to be 1.0% (w/w). Proteolytic enzymes were screened to obtain high chlorella growth factor (CGF) index, which indicates crude CGF content and solid recovery. Among the seven tested proteases, Esperase, whose optimal dosage was 1.0% (w/w), was selected. By co-treatment using optimal dosages of Tunicase and Esperase, the highest CGF index and solid recovery were obtained. The CGF index and solid recovery of co-treatment were remarkably enhanced by 250 ($4.36{\rightarrow}15.21$) and 220% ($12.65%{\rightarrow}40.15%$), respectively, than those of the non-treated extracts.
The enzymatic properties of soil cellobiohydrolase were examined and compared with those of cellobiohydrolase-active extracts from soil in the forms of enzyme-humic complex and humicfree enzyme, and cellobiohydrolase partially pruified from Aspergillus niger. The pH optima of soil cellobiohydrolase and cellobiohydrolase-humic complex were greater by 1.5-3.0 pH units than those of cellobiohydrolase in humic-free extract and from A. niger. Soil cellobiohydrolase and cellobiohydrolase-humic complex were remarkably resistant to thermal denaturation and proteolysis. These results confirm that cellobiohydrolase in soil is atable in conditions which rapidly inactivate microbial cellobiohydrolase and that its stability is due to the immobilization of this enzyme by association with humic substances. The Michaelis-Menten constants (Km) for soil, cellobiohydrolase-humic complex, humic free extract and cellobiohydrolase from A. niger were 22.1mg/ml, 11.3mg/ml, 10.6mg/ml and 4.5 mg/ml of Avicel, respectively.

The browning characteristic and dyeability of the mushroom were examined to establish the optimum condition for browning and extraction in the process of obtaining the natural dye, brown colorant. The composition of browning extracts from mushroom was also analyzed and the dyeability were investigated. The results of this study are as follows, 1. The optimum condition for obtaining the browning colorant from mushroom was the crushed phase of sample. 2. The browning reaction by enzymes in mushroom was efficient at a temperature of 35$^{\circ}C$, a duratron period of 2 hr and pH of 8. 3. The optimum condition for extraction of browning extracts from browned mushroom was at 95$^{\circ}C$ and Ihr. 4. To analyze the content of browning extracts in the mushroom, three fractions were obtained from gel filtration using Sephadex G-25 and the fraction 1 was melanin and the fraction 3 was dihydroxyphenylanine (DOPA) and glutaminyldihydroxybenzene (GDHB) . 5. The turbidity in enzymatic browning extraction of mushroom increased depending on refrigeration storage (4$^{\circ}C$) and UV radiation. The filtration of the extracts resulted in a decrease of absorbance. 6. The dyeability of the fraction 1 was greater compared to that of the fraction 3 and that of wool greater compared to the other two fabric materiales.

Although Semisulcospira libertina is generally regarded as a supplement for the alleviation of alcohol hangover, little is known about its effects on cell metabolism. Therefore, this study was conducted to analyze the constituents of the extracts prepared using different extraction methods and to compare their biochemical properties. The amino acid contents were found to be much higher in acidic and enzymatic hydrolysates than hot water extracts from S. libertina. DPPH radical scavenging activities in acidic and enzymatic hydrolysates were higher than those of hot water extracts. Three types of S. libertina hydrolysate was added to HepG2 cells damaged by acetaminophen (AAP), after which the survival rate of HepG2 cell were measured. In addition, lactate dehydrogenase (LDH) activities in the culture media were evaluated. The survival rates of HepG2 cells were $77.0{\pm}4.3%$ and $81.5{\pm}1.3%$ at 3 h and 5h enzymatic hydrolysates, respectively. These cell survival rates were higher compared to those of the negative control group ($67.8{\pm}4.3%$) treated only with acetaminophen. Cellular toxicities induced by treatment with AAP were also significantly alleviated in response to treatment with the extracts of S. libertina. In addition, the activities of 2 key enzymes that metabolize ethanol, alcohol dehydrogenase and aldehyde dehydrogenase, were upregulated by 4.7- and 2.7-fold respectively in response to treatment with a 3 h enzymatic hydrolysate of S. libertina. Taken together, these results provide biochemical evidence of the method by which S. libertina exerts its biological functions, including the alleviation of alcohol hangover and the protection of liver cells against toxic insults.
Enzymatic hydrolysis of crushed ginger was carried out to increase the yield of ginger extract, and the quality of the extract was investigated. The first extract was obtained by pressing crushed ginger, and the second extract by pressing ginger pomace hydrolyzed with $\alpha$-amylase at $90^{\circ}C$ for 1 hr. Oleoresin was extracted from the residue of enzymatic hydrolysis with 90% ethyl alcohol. The first extract, second extract and oleoresin were mixed to obtain the final ginger extract. The yield of final extract was increased by 276% on the solid base of the fresh ginger extract. The final ginger extract contained less crude fiber, starch and free amino acids (62, 48 and 40%, respectively), but contained more free sugar (270%) compared to fresh ginger extract.
