Climate change is one of the biggest dangers facing all living creatures in the earth. It has been understood that emissions of greenhouse gases from human activity is the cause of climate change. Cars are responsible for around 12% of total EU emissions of CO2, the main greenhouse gas. The United Nations Framework Convention on Climate Change (UNFCCC or FCCC) is an international environmental treaty adopted at the United Nations Conference on Environment and Development (UNCED) on 9 May, 1992, which entered into force on 21 March 1994. The European Commission first adopted a Community Strategy to reduce CO2 emissions from cars in 1995. On 19 December 2007, the European Commission proposed "Proposal for Setting emission performance standards for new passenger cars to reduce CO2 emissions", which was adopted on 23 April 2009 as "Regulation (EC) No 443/2009". Prior to submitting the Proposal, the European Commission performed impact assessment and prepared impact assessment report which was reviewed by the Impact Assessment Board. The objective of this Regulation is to set emission performance standards for new passenger cars registered in the Community, which forms part of the Community's integrated approach to reducing CO2 emissions from light-duty vehicles while ensuring the proper functioning of the internal market. In the event that a manufacturer fails to meet its target, it will be required to pay an excess emissions premium in respect of each calendar year from 2012 onwards. On 11 March 2014, Regulation (EC) No 333/2014 amending Regulation (EC) No 443/2009 was adopted. Regulation (EC) No 333/2014 amends Regulation (EC) No 443/2009 to implement the modalities of meeting the 95g CO2/km target for new passenger cars to be reached in 2020. As industry benefits from indications of the regulatory regime that would apply beyond 2020, the Regulation includes a further review to take place by, at the latest, 31 December 2014.
본 연구는 고온고습 시험을 통하여 Cell 레벨에서의 표면관찰 및 효율저하를 분석하였다. 고온고습 시험조건은 KS C IEC-61215에서 제시한 PV 모듈하의 조건을 이용하여 온도 $85^{\circ}C$, 습도 85%, 1000hr 동안 수행하였다. EL(Electroluminescence)촬영을 통하여 Cell 표면의 이상 유 무를 분석한 결과, 시간이 경과함에 따라, 부분적으로 표면이 손상되어 변색되는 것을 확인하였다. 고온고습 시험 전 단결정 Cell 및 다결정 Cell의 효율은 각각 17.7%, 15.5%였으며, 1000hr 수행 후 15.6%, 14.0%로 각각 11.9%와 9.3%의 감소율을 보였다. 또한, 경년 시 나타나는 전기적 특성을 분석하기 위하여 FF(Fill Factor)값을 분석한 결과, 고온고습 시험 후 단결정 Cell은 78.7%에서 75.0%로 4.7%, 다결정 Cell은 78.1%에서 76.7%로 1.8%의 감소율을 보였다. 태양전지 실리콘의 원자배열 및 순도에 따라 효율 변화에 영향을 받아 단결정 Cell이 다결정 Cell보다 효율저하가 크게 나타났다고 판단된다. 또한, FF감소율보다 효율 감소율이 크게 저하된 것을 확인할 수 있었으며, 이는 Cell의 외부환경적 요인에 의한 표면 손상이 p-n접합층 접촉저항과 경년 시 나타나는 FF 감소율보다 크게 영향을 준 것으로 판단된다.
In 2015, a candidate for the second national reference standard (NRS) of Gloydius snake venom was produced to replace the first NRS of Gloydius snake venom. In the present study, the potencies of the candidate were determined by a collaborative study, and the qualification of the candidate was estimated. The potencies of the candidate were determined by measuring the murine lethal titers and lapine hemorrhagic titers of venom against the regional working reference standard (RWRS) for antivenom using the methods described in the previous report for the first NRS of Gloydius snake venom. Three Korean facilities contributed data from a total of 30 independent assays. Subsequently, two foreign national control research laboratories contributed to this collaborative study. The results were calculated using the Reed-Muench method for lethality and determined using a mixed-effects model for hemorrhage. The general common potencies of the lethal and hemorrhagic titers were obtained from the results of the 30 tests performed at three Korean facilities. The results are expressed in micrograms for 1 test dose (TD) with a 95% confidence interval as follows: a lethal titer of $90.13{\mu}g/TD$ (95% confidence interval = $87.39{\sim}92.86{\mu}g$) and a hemorrhagic titer of $10.80{\mu}g/TD$ (95% confidence interval = $10.46{\sim}11.14{\mu}g$). In addition, the candidate preparation showed good quality evaluation according to the results of the quality estimation of the candidate and is judged to be suitable to serve as the Korean NRS for snake venom. In conclusion, the second NRS of Gloydius snake venom was established in this study and will be used for national quality control, including a national lot release test of Korean antivenom products.
The prevalence of Toxoplasma gondii infection in birds has epidemiological significance because birds are indeed considered as a good indicator of environmental contamination by T. gondii oocysts. In this study, the prevalence of T. gondii in 313 house sparrows in Lanzhou, northwestern China was assayed by the modified agglutination test (MAT). Antibodies to T. gondii were positive in 39 (12.46%) of 313 samples (MAT titer ${\geq}$ 1:5). Tissues of heart, brain, and lung from the 39 seropositive house sparrows were tested for T. gondii DNA, 11 of which were found to be positive for the T. gondii B1 gene by PCR amplification. These positive DNA samples were typed at 9 genetic markers, including 8 nuclear loci, i.e., SAG1, 5'- and 3'-SAG2, alternative SAG2, SAG3, GRA6, L358, PK1, c22-8 and an apicoplast locus Apico. Of them, 4 isolates were genotyped with complete data for all loci, and 2 genotypes (Type II variants; ToxoDB #3 and a new genotype) were identified. These results showed that there is a potential risk for human infection with T. gondii in this region. To our knowledge, this is the first report of T. gondii seroprevalence in house sparrows in China.
유기용매 내성 세균의 첫 분리 보고 이후 다수의 유기용매 내성 세균들이 토양 폐수 심지어 심해 등 모든 환경에서 분리 보고되고 있다. 대부분의 유기용매 내성 세균은 그람음성세균으로 이는 그람음성 세균이 그람양성 세균 보다 유전적으로 더 내성을 보이기 때문이다. 유기용매 내성 기전은 유기용매 내성 그람음성 세균을 주로 사용하여 집중적으로 구명되어왔다. 유기용매 내성 그람양성 세균의 유기용매 내성 기전은 비교적 최근 연구에서 발견되고 있다. 유기용매는 용매에 따라 다른 독성을 보이며 유기용매 내성세균의 유기용매 내성 수준은 종과 균주에 의존적으로 매우 변화가 심하다. 그러므로 유기용매 내성세균은 다양한 변인과 다유전자에 의한 적응 전략에 의하여 용매독성과 싸우며 용매 스트레스에 적응할 수 있다. 그들은 세포형태 및 세포 행동에서의 변화, 세포표층의 수식, 세포막 적응, 용매 배출 펌프, 샤페론 그리고 항산화 반응 등의 기전을 통하여 유기용매의 과량의 농도에서도 생존할 수 있다. 본 총설에서는 대표적인 유기용매 내성 세균, 유기용매 내성 세균에서의 유기용매에의 적응 및 내성 전략들 나아가 그들의 산업적 및 환경공학적인 잠재적인 영향에 대하여 개관하고자 한다.
주어진 작물모형이 기후요소 중 어떤 것에 얼마만큼 반응하는지 상대적인 민감도를 모르는 상태에서 계산결과만으로 미래 전망을 내리는 것은 적절치 않을 수 있다. 본 연구에서는 미래기후전망에 근거한 국내 보리 품종의 민감도 변화 실험을 통해 기온, 강수량, 이산화탄소 농도의 변화가 작물의 생산량에 미치는 상대적인 영향을 분석하였다. 주요 보리 품종(올보리-수원, 알보리-밀양, 새쌀보리-익산, 삼도보리-진주)에 맞게 품종모수가 조정된 CERES-Barley를 RCP 8.5 시나리오의 2071-2100 기후변동범위(1981-2010 기준 기온 $-1^{\circ}C{\sim}+8^{\circ}C$, 강수량 -50% ~ +50%, $CO_2$ 농도 330ppm ~ 900ppm)에 근거한 99개 처리조합에 의해 구동하였다. 이들 처리조합에 의해 생성된 11,880개 종실수량 계산결과를 토대로 각 기후요소의 변화에 따른 종실수량의 민감도를 분석한 결과 이산화탄소농도 변화에 가장 민감한 반응을 보인 반면 강수량 변동에 대한 반응이 가장 무디었고, 온도변화에 대해서는 기준온도대비 상승, 하강 모두 감소하는 경향이었지만 품종 별로 차이가 뚜렷하였다. CERES-Barley는 우리나라에서 농업부문 기후변화 영향평가에 널리 사용되므로 실험설계 시 이러한 모형의 민감도를 감안하여야 현실성 있는 생육모의가 가능하다.
The regulation of neurotoxicants has usually been based upon setting reference doses by dividing a no observed adverse effect level (NOAEL) by uncertainty factors that theoretically account for interspecies and intraspecies extraploation of experimental results in animals to humans. Recently, we have proposed a four-step alternative procedure which provides quantitative estimates of risk as a function of dose. The first step is to establish a mathematical relationship between a biological effect or biomarker and the dose of chemical administered. The second step is to determine the distribution (variability) of individual measurements of biological effects or their biomarkers about the dose response curve. The third step is to define an adverse or abnormal level of a biological effect or biomarker in an untreated population. The fourth and final step is to combine the information from the first three steps to estimate the risk (proportion of individuals exceeding on adverse or abnormal level of a biological effect or biomarker) as a function of dose. The primary purpose of this report is to enhance the certainty of the first step of this procedure by improving our understanding of the relationship between a biomarker and dose of administered chemical. Several factors which need to be considered include: 1) the pharmacokinetics of the parent chemical, 2) the target tissue concentrations of the parent chemical or its bioactivated proximate toxicant, 3) the uptake kinetics of the parent chemical or metabolite into the target cell(s) and/or membrane interactions, and 4) the interaction of the chemical or metabolite with presumed receptor site(s). Because these theoretical factors each contain a saturable step due to definitive amounts of required enzyme, reuptake or receptor site(s), a nonlinear, saturable dose-response curve would be predicted. In order to exemplify this process, effects of the neurotoxicant, methlenedioxymethamphetamine (MDMA), were reviewed and analyzed. Our results and those of others indicate that: 1) peak concentrations of MDMA and metabolites are ochieved in rat brain by 30 min and are negligible by 24 hr, 2) a metabolite of MDMA is probably responsible for its neurotoxic effects, and 3) pretreatment with monoamine uptake blockers prevents MDMA neurotoxicity. When data generated from rats administerde MDMA were plotted as bilolgical effect (decreases in hippocampal serotonin concentrations) versus dose, a saturation curve best described the observed relationship. These results support the hypothesis that at least one saturable step is involved in MDMA neurotoxicity. We conclude that the mathematical relationship between biological effect and dose of MDMA, the first step of our quantitative neurotoxicity risk assessment procedure, should reflect this biological model information generated from the whole of the dose-response curve.
Aflatoxin B1 (AFB1) is produced by Aspergillus flavus growing in feedstuffs. Early detection of maize contamination by aflatoxigenic fungi is advantageous since aflatoxins exert adverse health effects. In this study, we report the development of an optimized conventional PCR for AFB1 detection and a rapid, sensitive and simple screening Real-time PCR (qPCR) with SYBR Green and two pairs of primers targeting the aflR genes which involved aflatoxin biosynthesis. AFB1 contaminated maize samples were divided into three groups by the toxin concentration. Genomic DNA was extracted from those samples. The target genes for A. flavus were tested by conventional PCR and the PCR products were analyzed by electrophoresis. A conventional PCR was carried out as nested PCR to verify the gene amplicon sizes. PCR-RFLP patterns, obtained with Hinc II and Pvu II enzyme analysis showed the differences to distinguish aflatoxin-producing fungi. However, they are not quantitative and need a separation of the products on gel and their visualization under UV light. On the other hand, qPCR facilitates the monitoring of the reaction as it progresses. It does not require post-PCR handling, which reduces the risk of cross-contamination and handling errors. It results in a much faster throughout. We found that the optimal primer annealing temperature was $65^{\circ}C$. The optimized template and primer concentration were $1.5{\mu}L\;(50ng/{\mu}L)$ and $3{\mu}L\;(10{\mu}M/{\mu}L)$ respectively. SYBR Green qPCR of four genes demonstrated amplification curves and melting peaks for tub1, afIM, afIR, and afID genes are at $88.0^{\circ}C$, $87.5^{\circ}C$, $83.5^{\circ}C$, and $89.5^{\circ}C$ respectively. Consequently, it was found that the four primers had elevated annealing temperatures, nevertheless it is desirable since it enhances the DNA binding specificity of the dye. New qPCR protocol could be employed for the determination of aflatoxin content in feedstuff samples.
Objectives: The purpose of this study was to identify the needs and improvement measures of the risk assessment system through a survey of asbestos abatement companies. Methods: This study prepared a questionnaire that includes improvement measures for the risk assessment system(six questions) and the necessity of a risk assessment system for asbestos abatement(one question). The questionnaire was sent to 2,170 asbestos abatement companies and returned by 83 companies(return rate = 3.8%). We conducted frequency analysis, ANOVA, and Chi-squared testing at the 5% significance level. Results: This study analyzed the survey results and identified six main opinions on the risk assessment system and improvement measures. First, giving an advantage to companies with a high grade(S and A grades) in public bidding and/or qualification screening showed a similar preference(agree: 50.6%, disagree: 49.4%). Second, 57.6% of the respondents wanted to allow air showers along with water showers for low risk asbestos work. However, 23.2% of the respondents asserted that only a water shower should be allowed since there is no scientific evidence supporting the removal of asbestos by air shower. Third, in order to prevent missing the enrollment of workers, simply submitting a change report should be allowed when any worker is changed (40.0%). Fourth, 43% of the respondents answered that they did not know how to calculate the proper number of negative pressure units. The reasons given were a lack of guidelines or standards(38.9%), inconvenience (36.1%), and absence of education(25%). Fifth, the respondents who were favorable toward the necessity of a risk assessment system gave higher scores compared to unfavorable respondents on the necessity of the management of workers' work history(favorable respondent = 3.96 out of 5, unfavorable respondent = 2.68) and the necessity of professional training for workers(favorable respondent = 3.71, unfavorable respondent = 2.56). Finally, the respondents favorable toward a risk assessment system showed higher scores on the awareness of calculating the proper number of negative pressure units(4.79) compared to unfavorable respondents(3.3). Conclusions: The opinions of asbestos abatement companies identified through the survey in this study can be usefully utilized as fundamental information to improve the risk assessment system.
성장호르몬 유전자는 하나의 작은 공통 선조 유전자로부터 아주 오랜 기간동안 유전자 중복에 의해 진화되어 온 그룹들 중의 하나이다. 이들에 속하는 유전자들은 동물 종간에 구조적인 상동성과 기능적 공통성 등 유사성이 비교적 높게 나타난다. 이런 연구결과들을 근거로 하여 소 성장호르몬 유전자에서 아미노산을 암호화하는 영역으로부터 새로운 아미노산의 변이(missense mutation)를 검출하였고, 이 변이의 대립유전자 빈도는 소(cattle)의 종(species) 및 품종의 지리적 분포에 따라 일정한 경향 치를 보여 주었다. 한편 변경되어진 아미노산은 Tryptophan으로 이는 생물체에 존재하는 많은 단백질들을 구성하는 아미노산중에서도 그 출현빈도가 가장 낮은 것이다. 또한 검출된 변이는 성장호르몬이 그의 수용체와 강하게 결합하는 부위로서, 성장호르몬의 구조적 변이를 초래하여 수용체와의 결합이 비정상적으로 이루어져, 이후 성장호르몬이 표적세포로의 신호전달과 같은 역할을 제대로 수행치 못하게 되고, 이로 인하여 가축의 표현되어지는 경제형질에 영향을 미칠 것으로 추정된다. 그러므로 이러한 대립유전자를 보유하는 개체는 집단에서 제거하는 방법에 의한 개량이 가능할 것으로 사료된다.
