For mass production of poly(3-hydroxybutyrate) (PHB), high cell density cultures of Ralstonia eutropha were carried out in 2.5-1 and 60-1 fermentors by two fed-batch culture techniques of nitrogen and phosphate limitation. When the nitrogen limitation technique was employed using both an on-line glucose monitoring and control system, a high concentration level of PHB (121g/l) was obtained in the small-scale fermentor of 2.5 1. However, the PHB concentration obtained in a large-scale fermentor of 60 1 only turned out to be 60g/l. In contrast, when another fed-batch culture technique of the phosphate-limitation employing dissolved oxygen (DO) stat glucose feeding was used, a large amount of PHB was successfully produced in both 60-1 and 2.5-1 fermentors. In a 2.5-1 fermentor, concentrations of PHB and cells obtained in 58 h were 175 and 210 g/l, respectively, which corresponded to the PHB productivity level of 3.02 g/l/h. In a 60-1 fermentor, a final cell concentration of 221 g/l and a PHB concentration of 180 g/l with PHB productivity level of 3.75 g/l/h were obtained in 48h. PHB content and yield from glucose were 81% and 0.38g PHB/g glucose, respectively. These data suggest that the phosphate limitation technique is more effective compared to nitrogen limitation in the mass production of PHB by R. eutropha of a large scale.
A cell-based in vitro exposure system was developed to determine whether oxidative stress plays a role in the cytotoxic effects of volatile organic compounds (VOCs) such as benzene, toluene, xylene, and chlorobenzene, using human epithelial HeLa cells. Thin films based on cysteine-terminated synthetic oligopeptides were fabricated for immobilization of the HeLa cells on a gold (Au) substrate. In addition, an immobilized cell-based sensor was applied to the electrochemical detection of the VOCs. Layer formation and immobilization of the cells were investigated with surface plasmon resonance (SPR), cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). The adhered living cells were exposed to VOCs; this caused a change in the SPR angle and the VOC-specific electrochemical signal. In addition, VOC toxicity was found to correlate with the degree of nitric oxide (NO) generation and EIS. The primary reason for the marked increase in impedance was the change of aqueous electrolyte composition as a result of cell responses. The p53 and NF-${\kappa}B $ downregulation were closely related to the magnitude of growth inhibition associated with increasing concentrations of each VOC. Therefore, the proposed cell immobilization method, using a self-assembly technique and VOC-specific electrochemical signals, can be applied to construct a cell microarray for onsite VOC monitoring.
Antibiotic pollution is one of the factors contributing to the spread of antibiotic-resistant bacteria in the environment. Advanced oxidation and irradiation processes have been introduced to eliminate antibiotics from water and wastewater. However, few studies have reported the toxic effects of residual antibiotics and their byproducts induced by a treatment system. In this study, we compared the efficacies of chemical (high-performance liquid chromatography (HPLC)) and biological (antimicrobial susceptibility test) assays for measuring the concentrations of residual antibiotics after gamma irradiation for degrading amoxicillin, cephradine, lincomycin, and tetracycline. The concentrations of residual antibiotics estimated using the two assay methods were almost identical, except cephradine. In the case of cephradine, inhibited bacterial growth was observed that was equivalent to twice the concentration measured by HPLC in the samples subjected to gamma irradiation. The observed inhibition of bacterial growth suggested the generation of potentially toxic intermediates following antibiotic degradation. These results indicate that biological and chemical assays should be used in concert for monitoring antibiotic contamination and the toxic derivatives of antibiotic degradation. The results demonstrate that these four antibiotics can be decomposed by 2.0 kGy gamma-irradiation without toxic effects of their byproducts.
There are fifteen non-point pollution management areas in Korea and three of them (Doam lake, Daegi district and Golji-cheon) are located in the upstream of the Namhan river watershed. Many efforts to reduce non-point sources (NPS) pollution have been conducted, however, water quality pollution in the watershed is still serious. To solve these problems, it is a priority to grasp water quality using statistical techniques. In this study, a trend analysis was conducted to evaluate the effect of NPS management in the watershed. The long-term trends from 1996 to 2018 of water quality properties were analyzed using data collected from the water environment information system. Seventeen monitoring stations were selected along the main stream in Namhan river basin. Monthly water quality properties (BOD, COD, TN, TP, TN/TP ratio, Conductivity, SS and Chlorophyll-a) were collected and analyzed by Mann-Kendall test and LOWESS. The results showed that the Conductivity tended to increase in all regions and was the highest level in Jijangcheon. Organic pollution such as BOD and COD tended to increase in the Jungseon area. SS did not show a large tendency, but it showed high concentration in the Doam watershed. In all regions, 40% of water quality properties showed a tendency to 'UP', 15% of water quality properties tended to 'DOWN', and 46% indicated no tendency. In order to determine the cause of this, additional research and measures for improvement are necessary. This study will be used for the establishment of water quality policy in the future.
본 연구에서는 Chlorophyll-a (Chl-a)값을 빈도분석인 히스토그램(histogram)을 통해 녹조관리가 필요한 지점을 분류 하고자 하였다. 누적도수분포곡선(ogive curve)에 따라 지점별 녹조발생정도를 분석하였고, 히스토그램의 형태에 따라 관리지점을 3단계로 분류하였다. 4대강 중 한강에서 저농도의 Chl-a가 가장 많이 나타났고, 금강과 영산강 지점에서 높은 농도의 Chl-a가 빈번하게 나타났다. 히스토그램의 패턴에 따라 녹조의 관리 정도를 분류 할 수 있었는데, 한강의 경우 집중관리가 필요한 지점이 나타나지 않았고, 금강과 영산강은 대부분의 지점에서 녹조관리가 필요한 것으로 나타났다. 마지막으로 낙동강은 중류 및 하류 지점부터 녹조관리가 필요한 지점으로 분류되었다. 본 연구결과를 통해 빈도분석이 녹조발생의 우심지역 판단의 보조지표 활용 가능성 여부를 확인하였다.
스마트팜은 원예작물의 생산성과 품질을 제고하기 위한 생력화 방법으로 최근 많은 주목을 받고 있다. 하지만 국내의 스마트팜은 단순한 환경 모니터링과 환경제어만 가능한 초기단계에 머물고 있으며, 작물 생육에 최적화된 환경을 모의하는 의사결정도구의 개발은 미흡한 상태이다. 본 연구에서는 의사결정도구로써의 작물생육모형의 활용가능성을 확인하기 위해 국내에서 개발된 GreenTom모형의 품종모수를 추정하고 모형의 모의 성능을 검증하였다. 적엽은 시설토마토 재배에서 흔히 행해지는 농작업이지만 기존 모형은 이를 모의하지 않아 지상부 생육 모의에 문제를 나타냈다. 이를 해결하기 위해 적엽 알고리즘을 개발하여 기존 모형에 추가하고 모의 성능을 검증한 결과, 개선된 모형은 시설재배 토마토의 발달과 생육을 비교적 잘 모의하여 본 모형이 의사결정도구로 활용될 수 있음을 확인하였다.
A novel three-phase hollow fiber liquid phase microextraction was developed for the determination of malachite green (MG) in environmental waters, which selected [BMIM][$PF_6$] mixed with 1% trioctylphosphine oxide (TOPO) as supported phase. Several parameters (accepter phase pH, sample pH, supported phase membrane, volume of accepter phase, salinity, extraction time) that could affect extraction performance were investigated. Under the optimal extraction conditions, the established approach showed excellent characters as: high enrichment factor (212), wide linear range ($0.20-100{\mu}gL^{-1}$), low detection limit ($0.01{\mu}gL^{-1}$), good reproducibility (RSD, 8.9%, n=5) and satisfactory recovery (84.0-106.2%). The method was applied to detect MG at Yangtze River and pond waters in Zhenjiang, Jiangsu province, and 4 sites among 15 sampling sites were found MG with the concentration of $1.73-11.06{\mu}gL^{-1}$, which confirmed that the proposed environmentally friendly method was simple and effective for monitoring MG in aquatic system.
This study aims to establish baseline data for sustainable monitoring by applying the green city index (GCI), which is set up to evaluate the city level, to the city of Gangneung-si, which was designated as a pilot city for the Low-carbon Green Growth City project by the Ministry of Land, Infrastructure, and Transportation. The GCI was applied in the framework of European systems, while considering the social and economic status of Korea. Indicators from 7 areas-$CO_2$, energy, building, transportation, water, waste, and quality of atmosphere were analyzed, except for qualitative indicators. Results indicate that total $CO_2$ emissions were 30.8 tons per capita, or 2.2 tons per one million units of real GDP. The total final energy consumption was 0.231 TOE/capita, or 0.317 TOE per one million units of real GDP. The percentage of total energy derived from renewable resources was 0.41% and energy consumption by the building was $433.5Mwh/1,000m^2$. The total percentage of the working population travelling to work daily by public transportation (limited to bus) was 19%. Further, the total annual water consumption was $99m^3/capita$, and the water lost in the water distribution system was $0.057m^3/capita/day$. The total annual waste collected was 0.0077 ton per capita, The annual mean emission were 0.014 ppm/day for $NO_2$, 0.005 ppm/day for $SO_2$, and 0.019 ppm/day for $O_3$. The annual mean for PM10 emissions was $39{\mu}g/m^3/day$.
The Canton Tower is a high-rise slender structure with a height of 610 m. A structural health monitoring system has been instrumented on the structure, by which data is continuously monitored. This paper presents an investigation on the identified modal properties of the Canton Tower using ambient vibration data collected during a whole day (24 hours). A recently developed Fast Bayesian FFT method is utilized for operational modal analysis on the basis of the measured acceleration data. The approach views modal identification as an inference problem where probability is used as a measure for the relative plausibility of outcomes given a model of the structure and measured data. Focusing on the first several modes, the modal properties of this supertall slender structure are identified on non-overlapping time windows during the whole day under normal wind speed. With the identified modal parameters and the associated posterior uncertainty, the distribution of the modal parameters in the future is predicted and assessed. By defining the modal root-mean-square value in terms of the power spectral density of modal force identified, the identified natural frequencies and damping ratios versus the vibration amplitude are investigated with the associated posterior uncertainty considered. Meanwhile, the correlations between modal parameters and temperature, modal parameters and wind speed are studied. For comparison purpose, the frequency domain decomposition (FDD) method is also utilized to identify the modal parameters. The identified results obtained by the Bayesian method, the FDD method and a finite element model are compared and discussed.
효과적인 가변제한속도(Variable Speed Limit: VSL)는 교통사고를 예방하고 교통정체를 완화시키는데 기여하는 중요한 교통류 제어방안이다. 본 연구에서는 교통사고 예방을 목적으로 가변제한속도를 적용하는 방법론을 제시하였다. 가변제한속도를 적용하는 조건을 두 가지로 분류하였는데, 사고위험도 예측모형에 따른 제한속도 변화와 도로환경조건의 변화에 따른 제한속도 조정 방안을 제안하였다. 예측된 사고위험도에 따른 가변제한속도의 적용은 검지기에서 추출되는 교통량, 속도, 점유율자료와 교통사고자료로 이항로지스틱회귀분석 기법을 사용하여 구축된 사고위험도 예측모형을 사용하였다. 한편, 도로환경조건에 따른 가변제한속도 적용에서는 안개, 폭우, 폭설로 인한 시거제약 상황과 결빙, 비, 눈으로 인한 노면마찰력감소상황 시 최소정지거리 계산을 통해 각 상황에 적용되어야 하는 제한속도를 제시하였다. 이를 바탕으로 통합도로환경조건 모니터링 시스템 하에서 사고위험도와 도로환경조건을 관측하고 제한속도를 변화시키는 가변제한속도 시스템 알고리즘을 제시하였으며, 제안된 알고리즘의 현장 적용 시 고려되어야 할 기술적 이슈를 논의하였다.
