A combined approach to evaluate activity and structure of soil microbial community in long-term heavy metals contaminated soils

  • In the present study, long-term heavy metals (HMs) contaminated soil samples from a well-known Pb/Zn smelting area in the southwest of China were collected, and physicochemical and biological characteristics of these samples were evaluated. Soil samples contained different concentrations of HMs, namely Pb, Zn, Cu, and Cd. Enzyme activity analyses combined with microcalorimetric analysis were used for soil microbial activity evaluation. Results showed that two soil samples, containing almost the highest concentrations of HMs, also shared the greatest microbial activities. Based on correlation coefficient analysis, high microbial activity in heavily HMs contaminated soil might be due to the high contents of soil organic matter and available phosphorus in these samples. High-throughput sequencing technique was used for microbial community structure analysis. High abundance of genera Sphingomonas and Thiobacillus were also observed in these two heavily contaminated soils, suggesting that bacteria belonging to these two genera might be further isolated from these contaminated soils and applied for future studies of HMs remediation. Results of present study would contribute to the evaluation of microbial communities and isolation of microbial resources to remediate HMs pollution.

Antifungal activity of Saccharomyces cerevisiae peroxisomal 3-ketoacyl-CoA thiolase

  • Peroxisomes play an important role in cellular defense systems and generate secondary messengers for cellular communication. Saccharomyces cerevisiae containing oleate-induced peroxisomes were subjected to buffer-soluble extraction and two chromatographic procedures, and a protein with antifungal activity was isolated. The results of MALDI-TOF analysis identified the isolated protein as peroxisomal 3-ketoacyl-CoA thiolase (ScFox3). Purified yeast ScFox3 exhibited thiolase activity that catalyzed the thiolytic cleavage of 3-ketoacyl-CoA to acetyl-CoA and acyl-CoA. ScFox3 protein inhibited various pathogenic fungal strains, with the exception of Aspergillus flavus. Using ScFox3-GFP and PTS2 signal-truncated ScFox3M-GFP, we showed that only ScFox3-GFP, with an intact PTS2 peroxisome signal sequence, was able to translocate into peroxisomes. Yeast ScFox3 is a natural antifungal agent found in peroxisomes.

Determination of Medium Components in the Flocculating Activity and Production of Pestan Produced by Pestalotiopsis sp. by Using the Plackett-Burman Design

  • Optimization for the production of Pest an was followed by the Plackett-Burman Design, using modified Czapek-dox medium as the starting point. At the flask level, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$, and aeration variables positively affected the Pestan production, DCW (dry cell weight), apparent viscosity, and flocculating activity response. KCI and $FeSO_4{\cdot}7H_2O$ negatively affected the Pestan production, DCW, apparent viscosity, and flocculating activity response. Aeration variable was shown to have a positive effect on only the flocculating activity response among Pestan production, DCW, and apparent viscosity responses. In comparison of the positive and negative variables media conditions, Pestan production and flocculating activity differed by about 9 and 125 times, respectively. In particular, at the jar fermentor level, the aeration variable was the most important factor of the all responses (pestan production, DCW, apparent viscosity, flocculating activity, and anionic charge density). The flocculating activity and apparent viscosity of Pestan were closely related to the molecular chain length and charge density.

  • The article proposes a method of modeling a comprehensive indicator for evaluating the effectiveness of regional management of innovation activity. This will make it possible to assess the effectiveness of personnel, financial and credit and foreign economic activity of the regions from the standpoint of an integrated approach. The modeling technique is proposed to be carried out using the tools of taxonomic analysis and the calculation of a complex indicator of the effectiveness of the innovation activity management.

Effects of acute di-n-butyl phthalate administration on oxidative stress parameters

  • Di-n-butyl phthalate (DBP) is used extensively in the plastic industry and has been known as an environmental hormone (endocrine disruptor). Present study was undertaken to examine whether DBP can induce oxidative stress in mice. In this study, oxidative stress was measured in terms of the modification of lipid peroxidation and gamma-glutamyltranspeptidase (${\gamma}-GT$) activity. The activity of ${\gamma}-GT$, the level of lipid peroxidation and serum toxicity index were measured in male ICR mice after treatment with DBP (5 g/kg, po). Administration of DBP was found to significantly increase the level of lipid peroxidation approximately 2 fold in liver. The activity of ${\gamma}-GT$ in the liver of DBP-exposed animals was also increased approximately 2.5 fold. However, DBP did not alter the parameters for hepatotoxicity and nephrotoxicity such as alanine aminotransferase (ALT), aspartate aminotransferase (AST) and creatinine. These results indicate that DBP can induce oxidative stress in mice. The ${\gamma}-GT$ activity is considered to be increased as one of the adaptive defense mechanisms to oxidative stress induced by DBP.

  • Alum은 단분자성 알루미늄이 주종이었고, PAC는 고분자성 알루미늄을 함유하고 있었다. Alum과 PAC 모두 응집제 주입에 따른 총인 제거는 향상되었다. 응집제 사용은 미생물 활성도에 영향을 미치는 것으로 나타났다. Alum에 비해 PAC가 미생물 활성도에 대한 영향을 적게 미치는 것으로 나타났다. 그리고 슬러지 반송에 따른 미생물 활성도와 개체수에 대한 영향은 미미한 것으로 나타났다.

  • The purpose of this study is to analyze the content of environmental inquiry activities and values of elementary school students written in their reports of the Energy Environment Inquiry Activity Contest which is a unit contest of the Youth Science Festival held annually in Korea and get some ideas for the valuable way of Korean environmental education. 10 teams' reports were gathered and the locality, class study area, inquiry theme, inquiry method and values were analyzed and introduced as case studies to get some insights to teach elementary school students environmental values through inquiry activities of science subject approach. Each teams set various inquiry themes in the content domain of energy resources and earth environment conservation focusing on each team's geographical characteristics and occurring environmental problems in their local areas. There were 16 sentences representing technocentrism and 12 sentences representing ecocentrism. 5 of 10 teams showed both of technocentrism and ecocentrism in their reports. But their was only scientific knowledge without any environmental values in one team's report. Inquiry activity is a good teaching-learning method to develop environmental literacy. There should be environmental values involved as far as it is the environmental inquiry activities using environmental subject matter. The result of this study proposes that environmental educators should actively intervene for inquirers to set a stage in the whole process to search for an alternative to solve environmental problems involved in their project activities. The reason is because the environmental education in the elementary school is established to operate by the infusion way into various subjects already existed in the Korean curriculum.

제7차 중학교 ‘환경’ 교육과정과 청소년 환경교육 (Middle School Environmental Education of the 7th National Curriculum and Application to Teen-agers Practice of Environmental Education)

  • The Quality of human living depends on the environmental quality of the region sustaining the life. The environmental deterioration of the modern society is due to mechanical environmentalism. For the better quality of the life, The changes of recognition and attitude on the environments are required. These changes of mind are also important in environmental education for teenagers. The 7th national curriculum, officially anounced December 1998, focuses on the change of attitude to environments and practical behavior in real life for “Environments”, the environmental education curriculum in middle school. Basic elements of the curriculum are cultivation of the pro-environmental thinking, multi-levelling of teaching materials and methods, and encouraging of student participating activity. Actually, the curriculum construction is composed of stepped-levelling of teaching and learning, reasonable contents volume, encouraging of student practice, and suggesting of evaluation standards of textbook writing. Three main subjects of environmental education for middle school consist of (1) man and environment, (2) recognition of environmental problem, and (3) protection activity for environment. Methodology of environmental education can include multi-disciplinary approaches, variable teaching methods, and continuing evaluation of student practice and participation attitude. Environmental education for teenagers relating to the 7th national curriculum focuses on recognition of the environmental problems and practice activity in daily life. The recognition includes considering relationship of human life to environment, solving environmental problems in regional context, and development of comprehensive understanding concept of the environments. For the practice education, variable teaching methods, such as field survey and application of multi-media, are needed.

영과잉 음이항 회귀모형을 적용한 농촌지역 성인 신체활동의 지역사회환경 요인 분석 (Neighborhood Environment Associated with Physical Activity among Rural Adults: Applying Zero-Inflated Negative Binominal Regression Modeling)

  • Purpose: This study was conducted to determine the neighborhood environmental factors associated with physical activity among adults living in rural communities. Methods: A cross-sectional descriptive survey was conducted with a convenience sample of 201 adults living in three Ri in Y-city, Gyeonggi-do. Data were collected from face-to-face interview by trained interviewers and were analyzed using a zero-inflated negative binominal regression model. Results: Participants reported engaged in moderate or vigorous physical activity was 76.1%; 10.5% of participants reported that they met moderate physical activity recommendations and 14.5% of participants reported that they met vigorous physical activity recommendations. Zero-inflated negative binominal regression analysis showed association of increasing days of physical activity with social cohesion (${\beta}=.130$, p=.005), social network (${\beta}=-.096$, p=.003), and safety for crime (${\beta}=-.151$, p=.036), and no days of physical activity was associated with no attainment of education and marginally associated with increasing BMI. Conclusion: Neighborhood environmental factors including social cohesion, social network, and crime for safety were significantly associated with physical activity of rural adults. Community health nurses should expand an approach for individual behavior change to incorporate rural adults' specific neighborhood environmental factors into physical activity interventions.

홍삼 투여가 TBTO의 성호르몬 활성에 미치는 영향 (Effect on Sex hormone activity of TBTO against injected red Ginseng extract)

  • The purpose of this study finds out the effect of red ginseng extract (1.0g/kg) on TBTO (10, 20, and 40mg/kg) which poisons against some organs like thyroids gland, liver, kidney, testis, ovary, sex hormone activity of rats are examined by gastric tubing for 3 weeks. The weight of each organ in treated group were increased, especially liver in female and those of testis in males were significantly increased at 10, 20 and 40mg/kg (P<0.05, P<0.01). In case of sex hormone activity of each sex, the estradiol activity of female and testosterone activity of males were significantly decreased rather than the control group (P<0.05, P<0.01) According to between the TBTO treated group and 10+ rGe group of the testosterone activity each sex were significantly increased (P<0.01).
