On the Accumulation of Radioactive Materials in Marine Organisms Along the Coast of Korea 2. Strontium-90 Activities in Several Edible Marine Algae

  • Continued to the previous paper, the present investigation deals with stronitium-90 activities and calcium contents among 54 samples of edible marine algae collected along the coast of Korea during September, 1973 and April, 1974. The calcium contents are variable 2.0-17.8%, and 6.38% on an average. Most of the members investigated contain 2-4% in green and red algae, and 8-10% in brown algae. Strontium-90 activities are 0.32-0.37 pCi/l in sea-water, and 0.80-28.66 pCi$\^$90/Sr/g Ca in edible seaweeds. Among the algal phyla, they are 12.49 in green, 3.34 in brown, and 9.39 pCi$\^$90/Sr/g Ca in red algae, while they are 6.25 pCi$\^$90/Sr/g Ca on an average. In a single species collected at the same season, the highest activities appear mostly from the eastern coast, and from the western and southern coasts, in turn. The green algae, Capsosiphon- Enteromorpha complex show about 3 times higher activities, 28.66 pCi$\^$90/Sr /g Ca compared with the other members showing rather higher activities, and would be an indicator plant of strontium-90 activities of the marine algae along the coast of Korea.

Effect of Seaweed Extracts on the Viability of the Crustose Coralline Lithophyllum yessoense

  • The addition of seaweed extracts was found to regulate the viability of cultures of the crustose coralline alga Lithophyllum yessoense. The viability was quantitated using a triphenyltetrazolium chloride assay, and the methanol-soluble extracts from 18 prevalent seaweed species were tested. Extracts from Codium fragile and Enteromorpha linza inhibited viability, and a Hizikia fusiform is extract slightly increased viability. The methanol extract of C. fragile, which had the strongest inhibitory activity, decreased viability to 72 or $52\%$ that of the control following addition of 0.2 or 2 mg/mL of extract to the culture, respectively. The main active compound in the C. fragile was lipid. This information is a preliminary result related to the exploration of seaweed restoration in the algal whitening area.

Mineral Contents of Edible Seaweeds Collected from Gijang and Wando in Korea (기장산과 완도산 식용해조류 중의 미네랄 함량)

  • Mineral and heavy metal contents were determined in three brown edible seaweeds (sea mustard, Undaria pinnatifida; seaweed fusiforme, Hizikia fusiforme; sea tangle, Laminaria japonica) collected from Gijang and Wando in Korea, and two green seaweeds (sea lettuce, Enteromorpha prolifera; green laver, Monostroma nitidum) collected from Gijang. Seaweeds contained high proportions of ash (13.8-24.8%) and sulfate (1.08-3.25%). In brown seaweeds, ash contents (24.5-24.8%) were higher than in green seaweeds (13.8-15.2%). The differences of mineral contents were remarkable in both Gijang and Wando and in 5 various kinds of seaweeds. Brown seaweeds contained higher amounts of both macro-minerals (657-13,947 mg/100 g; Ca, Mg, Na, K) and trace elements (0.27-12.98 mg/100 g; Fe, Al, Zn, Mn, Cr, Cu, Ni), and green seaweeds also had relatively higher amounts of both macro-minerals (233-5,279 mg/100 g) and trace elements (0.26-10.61 mg/100 g).

Hepatoprotective Effect of Extracts and Phenolic Compound from Marine Algae in Bromobenzene-treated Rats (해조류 추출물과 페놀성화합물의 in vitro 및 in vivo 간보호활성)

  • The methanol extracts of some marine algae were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzyme in vitro in bromobenzene-treated rat. The extracts of Enteromorpha compressa, Capsosiphon fulvescens, Gelidium amansii, Hizikia fusiformis, Sargassum siliquastrum and Sargassum thunbergii which decreased the formation of lipid peroxide, inhibited the activity of xanthine and aldehyde oxidases by adding of each extracts. Phloroglucinol isolated from Ecklonia stolonifera reduced bromobenzene-induced hepatic lipid peroxidation. This compound administered daily over one week before intoxication with bromobenzene did not affect the activities of aminopyrine N-demethylase, aniline hydroxylase and glu tathione S-transferase. Epoxide hydrolase activity was decreased by bromobenzene, which was restored by pretreatment of phloroglucinol, The results suggest that the bromobenzene-induced hepatic lipid peroxidation by phloroglucinol is reduced by enhancing the activity of epoxide hydrolase.

Effect of Green Laver on the Extention of Shelf-life of Muk (Starch Jelly) (파래첨가가 묵의 저장성 향상에 미치는 영향)

  • The purpose of this study was to evaluate the antimicrobial effect of green laver (Enteromorpha linza) on the muk (starch jelly) preservation. The methanol extract of green laver completely inhibited the growth of Vibrio parahaemolyticus at 500 $\mu\textrm{g}$/ml level, and inhibited the growth of Bacillus subtilis, Escherichia coli and Staphylococcus aureus at 100 $\mu\textrm{g}$/ml level. The Listeria monocytogenes was inhibited by 98.89% at the same concentration. When various amounts of green laver powder were added in muk (starch jelly), 5% of green laver powder-added muk showed very low level of total bacterial count compared with the control group. Also, green laver did not give negative sensory qualities such as color, flavor and overall quality. Five % green laver muk was selected as the most preferable sample among the green laver muks in overall quality test.

In vitro Antioxidant Activity of Cooked Rice Containing Various Seaweeds (다양한 해조류를 첨가하여 제조한 밥의 in vitro 항산화 활성)

  • An emerging interest in healthy eating has led to an increase in the consumption of rice mixed with various types of grains. Cooked rice was prepared with five different seaweeds, Hizikia fusiformis, Sargassum fulvellum, Enteromorpha compressa, Undaria pinnatifida, and Gracilaria verrucosa, and the antioxidant activity was measured. In addition, the antioxidant activities of 80% ethanol and methanol extracts of the five seaweeds were compared. Total phenolic content (TPC), total reducing power (TRP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) cation radical scavenging activities of the ethanol extracts were higher than those of the methanol extracts. The TPC of raw seaweed ethanol extracts was from 7.58 to 26.27 mg gallic acid equivalent (GAE)/g. The antioxidant activities of both extracts of Hizikia fusiformis were the highest among the five seaweeds, and the antioxidant activities of the cooked rice were lower than those of the raw seaweed extracts. The DPPH radical scavenging activities of cooked white rice, mixed grains, barley, and a mixture of white rice and barely added with Hizikia fusiformis, Sargassum fulvellum and Undaria pinnatifida were 3.17, 23.12, 31.11 and 10.66%, respectively. These results demonstrate the addition of seaweeds to cooked rice helps to improve the antioxidant activity compared to white rice alone.

Inhibitory Effects of Marine Natural Products on Melanogenesis in B16 Melanoma Cells (B16 멜라닌 세포에서 해양소재 추출물의 멜라닌 생성 저해 효과)

  • Objectives : Under normal condition melanin protects the skin from extracellular stimuli including ultraviolet (UV)-induced oxidative skin damages, but excess production and accumulation of melanin can induce hyperpigmentation causing esthetic problems. Therefore, in this study we tried to search for natural skin whitening materials from marine natural resources. Methods : Water and ethanol extracts of marine natural resources were prepared from Porphyra thalli (PT), Laminariae thallus (LT), Ostreae concha (OC), Sargassum thallus (ST), Undaria thallus (UT), Codium thalli (CT), Enteromorpha thalli (ET), Syngnathoides biaculeatus (SB), and Hippocampus coronatus (Hc). Their effects against UVB and ${\alpha}$-melanocyte stimulating hormone (${\alpha}$-MSH)-induced melanogenesis were investigated based on melanin formation in B16 mouse melanoma cells. The mRNA and protein expression of enzymes involved in the melanogenic process were further examined by reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blot analysis, respectively. Results : Water extract of Ostreae concha (OCW/E) effectively inhibited UVB and ${\alpha}$-MSH-induced melanin production in B16 melanocytes, which seemed to be mediated by inhibition of mRNA expression of tyrosinase and tyrosinase-related protein 1 (TRP-1). In another experiment, ethanol extracts from Porphyra thalli (PTE/E), Laminariae thallus (LTE/E), Sargassum thallus (STE/E), Undaria thallus (UTE/E), Codium thalli (CTE/E), Syngnathoides biaculeatus (SBE/E), and Hippocampus coronatus (HcE/E) significantly suppressed UVB and ${\alpha}$-MSH-induced melanin formation. Furthermore, ethylacetate fraction isolated form LTE/E (LTE/EEt) decreased UVB and ${\alpha}$-MSH-elevated extracellular melanin levels via inhibition of tyrosinase protein expression. Conclutions : These results suggest that marine natural resources such as Porphyra thalli, Laminariae thallus, Ostreae concha, Sargassum thallus, Undaria thallus, Codium thalli, Syngnathoides biaculeatus and Hippocampus coronatus have anti-melanogenic effects, thereby exhibiting high potentials to be utilized as one of the ingredients for the development of new whitening functional cosmetics.

Ethanol Production from Red, Brown and Green Seaweeds and Biosorption of Heavy Metals by Waste Seaweed Slurry from Ethanol Production (홍조류, 갈조류, 녹조류를 이용한 바이오에탄올 생산 및 폐 해조류 슬러리의 중금속 생물흡착)

  • The seaweeds with high carbohydrate ratio Gelidium amansii, Saccharina japonica and Enteromorpha intestinalis were used as red, brown, and green seaweeds, respectively. Thermal acid hydrolysis, enzymatic saccharification and fermentation were carried out using those seaweeds to produce ethanol. The ethanol concentrations from red, brown and green seaweed were 14.8 g/L, 11.6 g/L and 9.9 g/L, respectively. After the production of ethanol, the seaweeds were reused to absorb heavy metal. The maximum biosorption ratio was Cu(II) (89.6%), Cr(III) (82.9%), Ni(II) (66.1%). Cu(II) had the highest affinity with 3 waste seaweeds. Red seaweed was verified the most effective substrates to both process.

Effects of Dietary Enteromorpha compressa on Growth and Blood Properties in Israeli Strain of Common Carp (Cyprinus carpio) (파래첨가 사료가 이스라엘계 잉어(Cyprinus carpio)의 성장 및 혈액성상에 미치는 영향)

  • In order to study the effects of dietary E. compresa on the growth and blood properties in young Israeli strain of common carp (mean. 8 g BW.). Fish were fed the commercial diet (control) and three experimental diets, $1\%,\;5\%\;and\;10\%$ of E. compressa powder were added to the control diet. After four months of feeding trial fish were bled by cardiac puncture and some hematological parameters were analyzed as physiological indices. 1) Following supplementation, positive responses in plasma albumin and glucose levels, and decreased body weight gain, feed efficiency and hematocrit. 2) Condition factor (fatness) in creased up to $50\%$, but decreased with $10\%$ supplementation. 3) Hb, GOT and GPT did not show any remarkable difference among all diet groups.

Effects of Thermal Effluents from Wolseong Nuclear Power Plant on Macroalgal Composition and Community Structure (월성원자력발전소 온배수가 해조류 종조성 및 군집구조에 미치는 영향)

  • Marine algal flora and community structure were seasonally examined at three sites in the vicinity of the Wolseong nuclear power plant in Korea from February to November 2006. A total of 126 seaweeds including 25 green, 31 brown, 70 red algae, and 1 marine plant were identified. The greatest number of species occurred at Jeonchonri (101 species) followed by intake (88 species) and discharge (29 species) during the study period. Of 126 seaweeds, 76 annuals and 13 warm tolerance species were recorded. Dominant species based on important value (IV > 10) were Ulva pertusa and Enteromorpha linza at intake, U. pertusa and Padina arborescens at discharge, and Sargassum horneri and Corallina pilulifera at Jeonchonri shore. Annual average biomass exhibited a wide range of variations, from 40.67g m-2 in dry weight at discharge to 133.69g m-2 at Jeonchonri. Among six functional groups, dominant group was coarsely-branched form at intake and Jeonchonri, but it was different at discharge site as filamentous form. Seaweed community structures of discharge site were distinguishable in decreasing species richness, biomass, species diversity index (H’), richness index (R), and evenness index (J’). Also, the ratios of green algae, warm tolerance species, annual algae, filamentous form and dominance index (DI) remarkably increased. Such phenomenons of discharge site are usually found under environmentally stressful conditions such as high disturbance. Thus, I can conclude that the heated effluents of nuclear power plant act as environmental stress influencing seaweed community structures and it can be detected with various community indices.
