Human cell regulatory mechanism is one of suspicious problems among biologists. Here we tried to uncover the human breast cancer cell regulatory mechanism from gene expression data (Marc J. Van de vijver, et. al., 2002) using a module network algorithm which is suggested by Segal, et. al.(2003) Finally, we derived a module network which consists of 50 modules and 10 tree depths. Moreover, to validate this candidate network, we applied a GO enrichment test and known transcription factor-target relationships from Transfac(R) (V. Matys, et. al, 2006) and HPRD database (Peri, S. et al., 2003).

We investigated the depth profiles of heavy metals in the surface sediments at Heedong reservoir in Pusan. Sampling was done at the intervals of 50 m of drift along the water channel into the reservoir. All samples were analyzed with an ICP-AES. We determined the content of Zn, Pb, Cd, Mn, Cu, Cr, and Fe. The overall mean content of these heavy metals were observed to $(2.9\pm1.2){\times}10^{-3},\;(1.3\pm0.7){\times}10^{-3},$$(1.9\pm2.1){\times}10^{-4},$$(2.3\pm1.1){\times}10^{-2},\;(1.6\pm1.0){\times}10^{-3},\;and\;(4.5\pm2.6){\times}10^{-4}$ ppm/ppmFe, respectively excluding iron data. Mean contents of Cu show an increasing trend toward the surface of sediments, while those of Cd show a decreasing trend, and those of Pb and Cr are relatively stable. Comparing with the contents of heavy metals in soils at two sites of Kumjeong mountain, enrichment factors of heavy metals in the surface sediments were determined. Among heavy metals we investigated, copper showed the largest value of enrichment factor. Considering the maximum content of heavy metals in the surface sediment, the values of enrichment factors of Cu, Cd and Cr were significant, which were 22, 8.1 and 4.0, respectively. In leaching experiment, it appeared that Pb, Cd, Cr, and Fe in sediments were hardly leached out into water, We also examined the effect of pH on the content of heavy metals.

Objectives : Because predicting the potential efficacy and mechanisms of Korean medicines is challenging due to their high complexity, employing an approach based on network pharmacology could be effective. In this study, network pharmacological analysis was utilized to anticipate the effects of YunPye-Hwan (YPH) in treating Chronic obstructive pulmonary disease (COPD). Methods : Compounds and their related target genes of YPH were gathered from the TCMSP and PubChem databases. These target genes of YPH were subsequently compared with gene sets associated with COPD to assess correlation. Next, core genes were identified through a two-step screening process, and finally, functional enrichment analysis of these core genes was conducted using both Gene Ontology (GO) enrichment analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) Pathways. Results : A total of 15 compounds and 437 target genes were gathered, resulting in a network comprising 473 nodes and 14,137 edges. Among them, 276 genes overlapped with gene sets associated with COPD, indicating a significant correlation between YPH and COPD. Functional enrichment analysis of the 18 core genes revealed biological processes and pathways such as "miRNA Transcription," "Nucleic Acid-Templated Transcription," "DNA-binding Transcription Factor Activity," "MAPK signaling pathway," and "TNF signaling pathway" were implicated. Conclusion : YPH exhibited significant relevance to COPD by modulating cell proliferation, differentiation, inflammation, and cell death pathways. This study could serve as a foundational framework for further research investigating the potential use of YPH in the treatment of COPD.

In 2010 and 2015, total 77 surface sediment samples were collected to assess the anthropogenic effects of trace metals in surface sediments of Garolim Bay, and the physical characteristics (particle size and specific surface area) and geochemical components (major (Al, Ca, Fe, K, Ba) and trace metals (Mn, Cs, Cr, Co, Ni, Cu, Zn, Pb), organic carbon and calcium carbonate) were analyzed. Mean grain size of Garolim Bay surface sediments ranged from 0.51-5.58 Ø (mean 3.98 Ø) and increased from the inlet of bay to the inner bay, and from the waterway to the land. Most of the metal concentrations except for some elements showed the similar distribution to those of mean grain size and specific surface area. As the particle size decreased and the specific surface area increased, the metal concentration increased. In order to estimate the factors controlling the concentration of trace metals, factor analysis was performed, and three factors were extracted (92.7% of the total variation). Factor 1 accounted for 71.3% of the total variation, which was a grain size factor. Factor 2 accounted for 14.2% of the total variation, Factor 3 accounted for 7.2% of the total variance. Enrichment factor was calculated using the particle size corrected background concentration. Metals with a enrichment factor of 1.5 or higher and the number of samples were 4 for Cr (St. 1, 16, 27, 39) and 1 for Pb (St. 39), but there were little differences in the concentrations of 1M HCl leached metals for these metals. The percentage of 1M HCl leached fraction to total metal concentration decreased in the order of Pb~Co>Cu>Zn~Mn>Ni>Cr. Comparing this value with contaminated and clean sediments in other coastal areas, the percentages for each metal were similar regardless of the trace metal levels in all regions. This fact might be resulted from the reaction between the 1M HCl solution and the different sediment constituents, indicating that there is a limit to apply this percentage of leached metal to the estimation of the contamination extent.

Objectives: We analyzed gene-expression profiles after 14 day odontogenic induction of human dental pulp cells (DPCs) using a DNA microarray and sought candidate genes possibly associated with mineralization. Materials and Methods: Induced human dental pulp cells were obtained by culturing DPCs in odontogenic induction medium (OM) for 14 day. Cells exposed to normal culture medium were used as controls. Total RNA was extracted from cells and analyzed by microarray analysis and the key results were confirmed selectively by reverse-transcriptase polymerase chain reaction (RT-PCR). We also performed a gene set enrichment analysis (GSEA) of the microarray data. Results: Six hundred and five genes among the 47,320 probes on the BeadChip differed by a factor of more than two-fold in the induced cells. Of these, 217 genes were upregulated, and 388 were down-regulated. GSEA revealed that in the induced cells, genes implicated in Apoptosis and Signaling by wingless MMTV integration (Wnt) were significantly upregulated. Conclusions: Genes implicated in Apoptosis and Signaling by Wnt are highly connected to the differentiation of dental pulp cells into odontoblast.

The determination of the boron isotopic ratio in solutions was achieved by means of a solid state track detector by using an alpha track. The neutron flux was optimized by using a Cd-foil to find the optimum conditions for counting the number of alpha tracks on the selected solid detector caused by the (n, $\alpha$) nuclear reaction of boron. The home-made multi-dot detector plate was utilized in this study to increase the reproducibility of the measurement by uniformly drying the boron solution within the marked circle area on the detector plate. The experimental results of this study verified that the $^{11}B/^{10}B $ isotopic ratio can be measured by observing the number of alpha tracks for different concentrated standard solutions with various isotopic compositions. This technique was applied to the determination of $^{10}B$ enrichment factor in a biological sample for a boron neutron capture therapy.
Airborne particles collected from a heavily industrialized site were analyzed by chemical speciation of seven trace metals: Pb, Cd, Cr, Cu, Ni, Zn, and As. The average concentrations were as follows: $Zn,\;502.0{\pm}230.7;\;Pb,\;176.5{\pm}310.9;\;Cu,\;111.9{\pm}82.7;\;As,\;38.0{\pm}31.0;\;Cr,\;21.5{\pm}24.4;\;Cd,\;20.8{\pm}17.4;\;and\;Ni,\;11.4{\pm}8.4\;ng/m^3$. The median enrichment factor (EF) values of Cd (7,280), As (1,030), Cu (215), Zn (214), and Pb (143), with respect to iron, were much larger than 100. We observed that Cd was found in the soluble and exchange- able form (56.9%), and that Pb and Cr were found in carbonates, oxides and the reducible form (69.8% and 61.1%, respectively). These two forms, which are the most easily absorbed into human body tissue, predominated in most of the trace metals investigated in this study.
We have previously reported that NS-398, a cyclooxygenase-2 (COX-2)-selective inhibitor, inhibited replicative cellular senescence in human dermal fibroblasts and skin aging in hairless mice. In contrast, celecoxib, another COX-2-selective inhibitor, and aspirin, a non-selective COX inhibitor, accelerated the senescence and aging. To figure out causal factors for the senescence-modulating effect of the inhibitors, we here performed cDNA microarray experiment and subsequent Gene Set Enrichment Analysis. The data showed that several senescence-related gene sets were regulated by the inhibitor treatment. NS-398 up-regulated gene sets involved in the tumor necrosis factor ${\beta}$ receptor pathway and the fructose and mannose metabolism, whereas it down-regulated a gene set involved in protein secretion. Celecoxib up-regulated gene sets involved in G2M checkpoint and E2F targets. Aspirin up-regulated the gene set involved in protein secretion, and down-regulated gene sets involved in RNA transcription. These results suggest that COX inhibitors modulate cellular senescence by different mechanisms and will provide useful information to understand senescence-modulating mechanisms of COX inhibitors.
