Experimental Study on the Immunoregulative Action of Kamiboatang (가미보아탕(加味補兒湯)의 면역조절작용(免疫調節作用)에 대(對)한 실험적(實驗的) 연구(硏究))

  • The purpose of this study was to investigate the effects of Kami boatang(KBT) on the immune cells in Balb/c mice. KBT (500mg/kg) was administerd p.o. once a day for 7 days. KBT enhanced the proliferation of thymocytes and splenocytes. KBT enhanced the subpopulation of helper T(Th) cells, but did not affect the subpopulation of Thyl/B220 cells and Th/Tc cells in splenocytes. KBT enhanced the production of ${\gamma}$-interferon and interleukin-2 in thymocytes, but decreased the production of interleukin-4. KBT enhanced the production of ${\gamma}$-interferon, interleukin-2 and interleukin-4 in splenocytes and serum. KBT suppressed the production of nitric oxide, and enhanced the phagocytic activity in peritoneal macrophages. These results suggest that KBT has a potent activity on the immune response via the increase of the production of cytokines and phagocytic activity in vivo.

  • New post plasma nitriding can achieve a high uniformity that have been difficult in DC nitriding and have a high productivity comparable to gas nitriding. However, it has not a enough high nitriding potential for a rapid nitriding, because surface activation or ion etching in the general plasma nitriding cannot be expected. Thus, in this study, the effects of pre-treatments with oxidation and reduction gas have been investigated to improve the nitriding kinetics of post plasma nitriding. An effective pre-treatment consisting of oxidation and reduction resulted in the increase of surface energy of STD 11. This induced the surface hardness and the effective nitriding depth of STD 11. It is thought that the increase of the surface energy and the surface area with pre-treatment promote the nucleation of nitriding layer.

  • This paper presents the ability of plasma electrolytic polishing technology to polish surface of stainless steel materials. The results show that the surface of its can be polished clearly using potentiostatic regimes in various concentration of $(NH_4)_2SO_4$ solution that had been warmed to a certain initial temperature. The equipment and deposition produces for polishing process are described and the effect of processing parameters on the characterizations polishedsamples has been has been investigated.

  • The purpose of this research was to investigate the effects of Seungyangikkitang (SIT) on the immune cells in BALB/c mice. SIT (500mg/kg) was administerd p.o. once a day for 7 days. SIT enhanced the proliferation of thymocytes, but decreased the proliferation of splenocytes. SIT enhanced the subpopulation of cytotoxic T cells in thymocytes and helper T cells in splenocytes, but did not affect the subpopulation of B220/Thy1 cells. SIT enhanced the production of γ-interferon and interleukin-2 in thymocytes, splenocytes and serum, but did not affect the production of interleukin-4. SIT suppressed the production of nitric oxide, but enhanced the lucigenin chemiluminescence and the engulfment of FITC-conjugated E. coli particles in peritoneal macrophages. These results suggest that SIT has a potent activity on the specific immunity via the cytokine secretion of Th1 cells and the non-specific immunity via the phagocytic activity of macrophages in vivo.

  • Flavonoid production by suspended cells of Scutellaria baicalensis Georgi was studied and the medium was optimized for cell growth and baicalin production. In SH medium the flavonoid production was not closely associated with the cell growth. A modified SH medium, FPM, was therefore designed for enhanced baicalin production. In FPM, both cell growth and baicalin production were increased by 1.5 times and 1.67 times than in the original SH medium, respectively. The increases could be attributed to the increased metabolic activities involved in the flavonoid biosynthesis as represented by enhanced activities of phenylalanine ammonia-lyase.

  • Ultraviolet B (UVB) induces multiple inflammatory and carcinogenic reactions. In skin, UVB induces to secrete several kinds of inflammatory cytokines from keratinocytes and also increases angiogenic process via the modulation of vascular endothelial growth factor (VEGF) production. Interleukin-21 (IL-21) is an inflammatory cytokine and produced by activated T cells. The biologic functions of IL-21 have not yet extensively studied. In the present study, we investigate the production of IL-21 from human keratinocyte cell line, HaCaT and its biological effect after exposure to UVB. First, we confirmed the IL-21 production and its receptor expression in HaCaT. And then, the change of IL-21 and VEGF production in HaCaT by UVB irradiation was examined. Not only IL-21 but also VEGF production was enhanced by UVB irradiation. Next, to determine relationship of enhanced production of IL-21 and VEGF, we detected VEGF production after neutralization of IL-21. VEGF production was reduced by IL-21 neutralization, which indicates that the IL-21 is involved in the VEGF production. Taken together, our results suggest that IL-21 and VEGF production is enhanced by UVB irradiation in HaCaT. In addition, it seems that IL-21 plays a role in the angiogenic process in skin via the modulation of VEGF production.

Enhanced Production of Phaeodactylum tricornutum (Marine Diatoms) Cultured on a New Medium with Swine Wastewater Fermented by Soil Bacteria

  • There have been a number of studies of methods for recycling animal wastewater to provide new bioresources. In the present work, a marine algal culture medium, designated KEP II, was prepared by adding swine waste (3% v/v) fermented by soil bacteria to a dilution of f/2 culture medium (CT). When Phaeodactylum tricornutum was grown in batch culture in KEP II, the cells lasted long at the exponential phase producing the specific growth rate and biomass; the production of total amino acids and secondary metabolites rose up to 5-fold. It also substantially enhanced the maximum quantum yield of photo system (PS) II of P. tricornutum, greatly increased the level of thylakoid membranes containing PS, and stimulated the production of pyrenoids, including enzymes for $CO_2$ fixation in chloroplasts. KEP II should improve the cost efficiency of industrial mass batch cultures and the value of microalgae for long-term preservation of fresh aquaculture feed as well as production of anticancer and antioxidant agents. Specifically, a low-cost medium for growing the diatoms of aquaculture feed will be economically advantageous.

  • Newcastle disease virus (NDV) vaccines were produced from Vero cells by using lively attenuated virus strain. The MOI of 0.1.' serum concentration of 2%. initial pH of 8.0. and infection time of 3 days were found to be optimum conditions for vaccine production. The treatment of polycation enhanced the virus production. When ascorbic acid was added as an antioxidant, NDV production was also enhanced. Utilization of $CaCl_2$ showed an inhibitory effect on the propagation of NDV. It was also found the ammonium ion concentration higher than 4mM inhibited virus production. Thus ammonium ion removal system was tried for the efficient production of NDV vaccine.
