인삼(Panax ginseng C.A. Meyer) 배유세포내 Vicilin의 면역세포화학적 분포 (Immunocytochemical Localization of Vicilin in Endosperm Cells of Panax ginseng C.A. Meyer)

  • 인삼(Panax ginseng C.A. Meyer) 종자단백질인 vicilin을 ammonium sulfate 침전, gel permeation 및 이온 교환 크로마토그래피로 정제하였다. Vicilin은 분자량 55,000(큰 소단위) 및 44,000(작은 소단위)인 두 종의 소단위를 포함하는 당단백질이다. Vicilin에 대한 항체를 토끼에서 형성시켜 DEAE-Affi-Gel Blue affinity 크로마토그래피로 정제하였다. 이 항체와 금 입자가 결합된 2차 항체를 종자의 배유세포에 반응시켰다. 금 입자는 배유세포내의 단백질체, 전자밀도가 높은 과립 및 골지체의 elaborating 과립에 표지되었다. 이러한 결과는 조면소포체에서 합성되어 골지체로 수송된 vicilin이 골지의 소포내에서 공정과정을 거쳐 전자밀도가 높은 과립이 된 다음 단백질로 수송됨을 나타낸다.

  • This study was carried out to investigate the development of endoplasmic reticulum and the formation of Protein body in the endosperm cell during seed formation of Panax ginseng C. A. Meyer with electron microscope. In the endosperm cell of early developmental process after pollination, vesicles that contain storage materials produced in rough endoplasmic reticulum incorporated into central vacuole. The central vacuole is gradually subdivided into several small-sized vacuoles and increased in number. Amorphous proteinaceous materials of high electron density are produced in rough endoplasmic reticulum. Rough endoplasmic reticulum increase in number and surround the protein body and vesicles circularly. Spherical proteinaceous granules with limited membrane appeared from the amorphous granules at the peripheral region of the rough endoplasmic reticulum. Gradually, storage materials are accumulated within the vacuole surrounded by spherosomes. Protein bodies are formed by interfusing between vacuoles and vesicles derived from rough endoplasmic reticulum which contained the amorphous protein of high electron density.

Effect of Particle Size on the Solubility and Dispersibility of Endosperm, Bran, and Husk Powders of Rice

  • Size effects of rice product powders on physical properties including suspension stability were investigated in this study. Endosperm, bran, and husk powders of rice with different size particles were prepared using the pin crusher or the ultrafine air mill. The physical properties of the powders were examined using particle size analysis, scanning electron microscopy, and spectrophotometry. The peak of the volume-weighted particle distribution of ultrafine endosperm particles was at $5.4\;{\mu}m$ whereas those of the bran and the husk appeared at 65 and $35\;{\mu}m$, respectively. Ultrafine particles of the endosperm and the husks dispersed better than larger-sized particles. As time elapsed, the dispersibility decreased, but the ultrafine particles were precipitated at the slowest rate. Our results suggest that ultrafine particles, including future nanosized particles, provide improved solubility and dispersibility resulting in better stability in the food colloidal suspension.

보리의 전분 Isogenic line들의 취반 및 제분 특성 (Cooking and Milling Characteristics of Several Barley Starch Isogenic Lines)

  • 보리의 이용성을 검토하기 위하여, 찰성 부정형 Shrunken endosperm 유전자를 결합하여 육성한 Isogenic line을 이용하여, 취반 및 제분에 대한 제특성을 조사한 바 그 결과를 요약하면 다음과 같다. 1. 보리의 배유 전분 특성이 다른 Isogenic type 간에 정곡 천립중, 회분, 단백질 함량은 별차이가 없었으나 Amylose함량, $\beta$-glucan viscosity와 취반특성은 뚜렷한 차이를 보였다. 2. 취반특성중 흡수율과 퍼짐성은 천립중, Amylose 함량과 부의 상관이 있었고, 용출고형물량과 점도간에도 부의 상관이 있었다. 3, 흡수율과 퍼짐성은 배유 전분 특성과 입의 크기에 의해 영향을 받았으며 그 결과 찰성 Isogenic type이 높았고 부정형 전분 Isogenic type이 가장 낮았으며 종실의 크기가 작은 것이 큰 것에 비해 높았다. 4. 보리밥의 Mixogram pattern은 전분 특성에 따라 차이가 있었으며 부정형 전분이 가장 많이 으깨어졌다. 5. 제분 특성은 메성이면서 부정형인 Isogenic type이 좋았고 찰성의 Shrunken endosperm Isogenic type은 대단히 불량하였다.

  • 진흥(振興)과 IR667의 비중선(比重選)된 현미(玄米)를 배아(胚芽)와 배유(胚乳)로 분리(分離) 비중등급별(比重等級別)로 전(全) amino산조성(酸組成)을 조사(調査)한 결과는 다음과 같다. 1. 현미단백질함량(玄米蛋白質含量)이 높은 IR667은 배아(胚芽)에서 진흥(振興)보다 단백질함량(蛋白質含量)이 적었으며 질소당(當) lysine과 필수(必須) amino산함량(酸含量)도 낮았다. 2. 두품종 모두 amino산으로의 질소회수율(窒素回收率)이 중등비중(中等比重)에서 배아(胚芽)나 배유(胚乳) 어느 경우도 높아서 비중(比重)이 높은 경우는 전분(澱粉)에 의(依)한 가수분해중(加水分解中)의 파괴(破壞)에 기인(起因)하는 것으로 보였다. 3. 배아(胚芽)나 배유(胚乳) 공(共)히 IR667에서는 Glu.>Asp.의 순(順)으로 많고 진흥(振興)에서는 Glu.>Arg.의 순(順)으로 많아 품종간(品種間) 질소대사(窒素代謝)의 차(差)를 보이는 것 같다. 4. 두 품종 공(共)히 배아(胚芽)에서는 His.

Characterization of a New High-lysine Mutant in Barley (Hordeum vulgare L.)

  • A chemical, MNU-induced hulless barley mutant line designated as 'Mutant 98 (M98)' was developed from a Korean hulless waxy barley cultivar, 'Chalssalbori'. The objective of the study was to determine the genetic basis of 'M98' and the possibility of using 'M98' as breeding parent to improve lysine level. Compared to 'Chalssalbori', 'M98' had large embryo and higher lysine content in both the embryo and endosperm. Significantly different lysine content in 'M98' and the other high-lysine barley mutant stocks was observed for two years. However, the genotype by year interaction was not significant. 'M98' was higher than the other high-lysine barley mutant stocks in the percentage of lysine of total amino acid composition (0.75%). The trait of shrunken endosperm of 'M98', which was typical in the high-lysine mutants, was inherited by a single recessive gene. Based on seed morphology and lysine content of $F_1$ seeds, 'M98' had a genetically different gene from the other high-lysine mutants for shrunken endosperm. Segregation of $F_2$ for plump/shrunken endosperm did not fit the expected ratio of Mendelian inheritance except for only one cross combination (GSHO1784 (lys1)/M98). The amino acid analysis of $F_5$ and $F_6$ progenies from the cross between 'M98' and 'Chalssalbori' revealed that the attempt to increase the range of lysine content of plump lines did not go beyond the limit of the average high-lysine barley germplasm.

  • The developmental processes of the protein body are studied on endosperm cells of Panax ginseng during seed maturation periods. The spherosome, mitochondria, rough endoplasmic reticulum, and ribosome are observed and then are gradually increased in early endosperm cells. Protein body developed from vesicles produced by the rough endoplasmic reticulum and was formed at the enlarged ends of rough endoplasmic reticulum. Also, vacuole-like protein body was observed in associated with rough endoplasmic reticulum. Golgi complex is observed in associated with vacuole and its vesicles containing proteinaceous granules moved and accumulated to the vacuole. Proteinaceous granules are deposited in the spherical or oval shaped vacuole and gradually, vacuole is surrounded by the multi-membranous structure. Rough endoplasmic reticulum, ribosome, Golgi complex, and vacuole are observed in associated with protein body formation.

Changes in Metabolites and Embryo Growth during Seeds Priming in Tobacco

  • Some metabolites and embryo growth of primed tobacco (Nicotiana tabacum L. cv. ‘KFI09’) seeds were observed during priming. The seeds were primed at 15 and $25^{\circ}C$ for 1, 2, 3, 5, 10 and 15 days in a -0.8 MPa polyethylene glycol 6000 (PEG) solution. The time to 50% seed germination (T$_{50}$) was greatly reduced when the seeds were primed at $25^{\circ}C$ when compared with 15$^{\circ}C$. The $\alpha$-amylase activity and sugars and amino acid contents in the seeds primed at $25^{\circ}C$ greatly increased, while $\alpha$-amylase activity was similar, and sugar and amino acid contents increased slightly in the seeds primed at 15$^{\circ}C$. When the seeds were primed at $25^{\circ}C$, growth of the embryo which was enclosed by endosperm was detected, while the endosperm became thinner as the priming duration was extended.d.

  • This study has been carried out to investigate the ultrastructural changes, formation of storage materials in endosperm cells with electron microscope during the seed formation of Panax ginseng C.A. Meyer. In the early stage of seed formation with green seed coat, the endosperm was cellular type. Cell plate was largely composed of dictyosome vesicles in early stage of wall formation after mitosis. Central vacuole was gradually subdivided into several small-sized vacuoles. During the differentiation of plastids, some proplastid was replaced by amyloplast with starch grains and lamellar structure. A number of mitochondria with well developed cristae were distributed in cytoplasm. Rough endoplasmc reticulum, dictyosome, microbody, free ribosomes and polysomes were evenly distributed in cytoplasm. Spherical spherosomes were formed from dictyosome containing the lipid materials of even electron density. Protein bodies were formed by interfusing between vacuoles and vesicles derived from rough endoplasmic reticulum which contained the amorphous protein of high electron density.

인삼배유세포에 존재하는 단백질체의 분해양상 (The Degrading Patterns of Protein Bodies in the Ginseng Endosperm Cells)

  • Protein bodies in the endosperm cells of mature red ginseng (Panax ginseng C.A. Meyer) were distributed evenly in the cytoplasm and their size varied from 1 to $8{\mu}m$. Three types of protein bodies were detected and they are spherical or egg-shaped ones containing homogeneous matrix only, spherical ones containing globoids, and irregular shaped ones. Protein bodies degraded in two patterns, one is to start the degration of the body from the surface toward the center, while the other is that the body was broken evenly and then degraded gradually. After degradation, only the limiting membrane remained, that causes the body to be empty. The limiting membranes fused with each other to form a large vacuole. Vicilin and legumin decreased in the endosperm cells as the protein bodies degraded gradually whereas they increased in the umbiliform layers.
