Rice Cultivar Breeding for Rice Flour with Low Amylose by Tissue Culture (저아밀로스 특성 및 쌀가루 적합 벼 품종 육성)

  • Rice consumption in Korea is continuously decreasing; therefore, methods for reducing rice stocks are now being proposed. Increasing the consumption of rice and reduce these stocks will require that the rice processing industry be developed beyond simply using cooked rice as food. The Korean people's eating habits have also diversified recently, and the consumption of bread is increasing. Therefore, breeding a cultivar for rice flour is now necessary. Here, a line suitable for processing into rice flour was cultivated by deriving a mutant line by tissue culture of Ilpum. Mutant populations were investigated for major agricultural traits in the field, and grain characteristics and endosperm characteristics were investigated after harvest. Among the lines, line 29111 had similar agricultural characteristics to the background Ilpum, and excellent grain quality, and was selected for further analysis. The physicochemical properties that determine taste were similar or improved. However, unlike Ilpum, 29111 had a non-uniform starch structure and a characteristic powdery endosperm because large and male starch grains are evenly distributed. Therefore, 29111 was viewed as a breeding line suitable for processing and for excellent agricultural characteristics and taste. The added value can be increased by diversifying the uses of rice. The taste was improved and a method for breeding cultivars with grain characteristics for glutinous rice that would satisfy both producers and consumers was presented.

Seedborne Fungi and Fungicide Seed Treatment of Ginseng

  • The incidence of fungi and their possible contribution to low vigour were examined in a collection of ginseng (Panax quiquefolius) seed from Ontario. When examined after one winter of stratification in the field and storage at 4f for five months in the laboratory, the collection exhibited low vigour (plant stand 16.7% of seeding rate six weeks after seeding) and high incidence (94%) of discolored or soft seed. Fungi isolated (and incidence) from 1,304 endosperm halves recovered from surface-sterilized seed were, in order of abundance, Fusarium rostrum (22.2%), Chaetomium crispuum (14.3%), Funriud oxysporum (9.0%), Fusarium sdani (9.0%), iwmor sp. (8.4%), Alternaria sp. (8.1%), Zowieua lucotricha (7.8%), Cylindruarpn sp. (0.9%), Fusarium avenacmm (0.9%), and Vdudla iliata (0.4%). Most of these fungi, including known and potential pathogens of ginseng (species of Alerraria, Cylindrocarpon, Fusarium, and Trichodirma), were associated with both healthy and diseased seed. Application of Benlate (benomyl), Thiram (thiram), or UBI-2584 (tebuconazole) to seed caused slight to pronounced reduction in emergence and did not significantly affect plant stand six weeks after seeding. The study demonstrated the high level of infection by fungi, including known and potential pathogens of the cry, in an arbitrary collection of ginseng seed from commercial sources, and the phytotoxicity of the fungicides tested when applied to moist stratified seed. The lack of efficacy of the fungicides precluded determination of the contribution of seedborne fungi to low vigour of the seed.

Optimum Chilling Terms for Germination of the Dehisced Ginseng(Panax ginseng C. A. Meyer) Seed (개갑 인삼종자의 발아 적정 저온감응기간)

  • Experiments were conducted to study the optimum chilling period for breaking physiological dormancy of dehisced ginseng(Panax ginseng C. A. Meyer) seed. Embryo of ginseng seed is too small to be noticed with naked eyed on harvesting time. Embryo grew to half size of endosperm after seeds were stratified for 3 months. It takes 6 months for this embryo to reach the size enough for germination. And it grew faster indoors than outdoors. Dehisced ginseng seed with full-size embryo did not germinate at room temperature and required chilling treatment for 75days in outdoors and 90 days in cold chamber. While seed receiving sufficient chilling treatment were left to be in room temperature, the chilling effect decreased remarkably.

Changes in Mining Properties During Maturation of Wheat Kernel (소맥의 등숙에 따른 제분특성의 변화)

  • Development and maturation of wheat were studied with reference to the quality of grain and milling properties. 1000-kernel weight and test weight increased as the wheat matured and as the orginal moisture decreased. The time of maturity was estimated 40 days after heading. Moisture content of wheat grain had a correlation coefficient of -0.877** with 1000-kernel weight, of -0.761** with test weight, and of 0.915** with pearling index. The milling data suggest that even in the early stages of maturing, the endosperm represented at large proportion of the grain. However, milling score was relatively constant at about 40 days. Break-Reduction flour ratio was a great difference between wheat varieties. 1000-kernel weight had a high significant correlation of +0.603** with milling yield, of -0.958** with ash content, and of +0.956** with milling score.

  • Sophisticated modern research tools for evaluation of medicinal plants are available but microscopic methods are one of the simplest and cheapest methods to establish the identity of the source materials. Pharmacognostical investigation of the dried, powdered and anatomical sections of the fruits of Trachyspermum roxburghianum (DC) Craib was carried out to determine its macro and microscopical characteristics along with its physical constants. Externally, the fruits, yellowish or greenish brown in colour are elongated, elliptical, slightly curved, prominently ridged and longitudinal. As seen in transectional views of the fruits from Trachyspermum roxburghianum, the mericarp has concave sides called commissural surfaces and a convex outer side called the dorsal surface. The mericarp has three primary ridges alternating with two secondary ridges on the dorsal side. On the commissural side, there are two primary ridges which are lateral in position and two secondary ridges in the commissural side. The seed is attached to the pericarp by a short stalk called a raphe. Circular, four-lobed calcium oxalate crystals are fairly abundant in the endosperm. Phytochemical studies revealed the presence of phenolic compounds, triterpenoids, proteins and sugars. The pharmacognostical profile of the fruits will assist in standardization for quality, purity and sample identification.

Physicochemical Properties of Non-glutinous, Dull, and Glutinous Rice Grain in Segregating Populations of Dull/Glutinous Crosses

  • Dull grains segregated from F$_3$ and F$_4$ of the crosses between two dull mutants and a glutinous cultivar were compared with non-glutinous and glutinous segregants for their physicochemical properties. Amylose content of dull rice grain segregated from the dull/glutinous cross showed the intermediate value between glutinous and non-glutinous rice grain, whether it is controlled by the recessive or dominant gene. Alkali digestibility value (ADV) of dull rice grain was lower than that of glutinous or non-glutinous rice. A positive correlation was found between ADV and amylose content of homozygous non-glutinous or dull F$_4$ grains, but a negative relationship was observed in glutinous grains. Protein content of dull grain was significantly higher than that of glutinous or non-glutinous grain segregated from the same cross, while those of glutinous and non-glutinous grains were not different. Among gelatinization characteristics, initial pasting temperature and peak viscosity of dull grains were higher than glutinous rice, and were not different with non-glutinous grain. Hot, cool and consistency viscosities of dull grain were intermediate between glutinous and non-glutinous rices. Dull grains showed the highest breakdown viscosity and the lowest setback viscosity among the three endosperm types.

Physical and Structural Characteristics of Endosperm Starch of Four Local Barnyardgrass (Echinochloa crus-galli L.) Collections in Korea

  • Physical and structural characteristics of four local barnyardgrass flour and starch were investigated. Amylose content ranged from 6.9 to 12.5%. The BP 3563 and BP 3606 were low amylose lines, and the BP 3592 and BP 3610 showed very low amylose lines. Peak viscosities of four barnyardgrass collections BP-3563, BP-3592, BP-3606, and BP-3610 ranged 264 to 2397. All barnyard grass lines displayed an A-type X-ray diffraction pattern showing the strong diffraction peak at around $2{\theta}$ values $15^{\circ}$, $17^{\circ}$, $18^{\circ}$ and $23^{\circ}$, and a small peak at $20^{\circ}$ $2{\theta}$. Scanning electron microscopy revealed that the starch granule sizes ranged $5.5-12.5{\mu}m$ in diameter showing polygonal and rarely spherical in shape with round edges and some pores at the surface. Starch crystallinity of BP 3563, BP 3592, BP 3606, and BP 3610 was 23.9, 25.0, 24.7, and 25.5%, respectively.

  • The experiment was conducted to study on the physiology of developing embryo and endosperm degradation during germination of Zea mays seeds treated with GA 20 and B-Nine 5000 ppm. Data obtained can be summerized as follows : 1. Swelling of seminal root increased the section of GA treatment more or less, on the other hand, the section of B-9 treatment inhibited slightly. 2. According to elapsing of seeding date, epithelial cell of Scutellum expansed in size, and the space of cell increased, that degree was enlarged as follows ; the section of GA, the section of Control, and the section of B-9. 3. According to the elapsing of seeding date, the formation of vascular organization embryo became clearer little by little, the lignification of vascular B-9 treatment section rather higher than the other section. 4. The degradation of Starch Grain is composed of near part of epithelial cell of Scutellum, the shape of degradation radiate from element of a disk shape, and the speed of degradation is the section of GA, the section of Control, the section of B-9 in turn.

Micropropagation of Medicinal Woody Eleutherococcus pedunculus via Somatic Embryogenesis

  • Zygotic embryos just after harvest of seeds were immature globular to heart stage. Maturation of zygotic embryos rapidly proceed when zygotic embryos together with small excised parts of endosperm were cultured on 1/3-strength MS solid medium with 2% sucrose, and the zygotic embryos were germinated within two months. Embryogenic callus was formed from the excised segments of germinating zygotic embryos of Eleutherococcus pedunclus on Murashige and Skoog (MS) medium with $4.5{\mu}M$ 2,4-D. The embryogenic callus formation occurred at a low frequency (less than 7%) from hypocotyl segments. The embryogenic calli were maintained on the same medium as primary medium. High frequency somatic embryogenesis was obtained after the cells were transferred to medium lacking 2,4-D. Cotyledonary embryos were germinated and converted into plantlets on medium with $20{\mu}M$ $GA_3$. Embryogenic callus and somatic embryos were produced spontaneously on the surfaces of roots and/or hypocotyls of plantlets. The frequency of embryogenic callus formation was 85% in roots and 34% in hypocotyls. Therefore maintain of cell lines performed very easily. Plantlets with developed epicotyls at more than 3 cm acclimatized at high frequency (89%). While plantlets with small epicotyls (less than 1 cm) were acclimatized at low rate (32%). The soil survived plantlets produced new sprouts after over wintering in the field.

Identification of Fungus-infected Tomato Seeds Based on Full-Field Optical Coherence Tomography

  • The morphological changes of anthracnose (fungus) -infected tomato seeds have been studied to identify the infection and characterize its effect. Full-field optical coherence tomography (FF-OCT) has been utilized as a nondestructive but efficient modality for visualizing the effects of fungal infection. The cross-sectional images extracted from a stack of en face FF-OCT images showed significant changes with infection in the seed structure. First of all, the seed coat disappeared with the infection. The thickness of the seed coat of a healthy seed was measured as 28.2 ㎛, with a standard deviation of 1.2 ㎛. However, for infected seeds the gap between surface and endosperm was not appreciably observed. In addition, the measurements confirmed that the dryness of seeds did not affect the internal seed structure. The reconstructed three-dimensional (3D) image revealed that the permeability of the seed coat, which plays the vital role of protecting the seed, is also affected by the infection. These results suggest that FF-OCT has good potential for the identification of fungus-infected tomato seeds, and for many other tasks in agriculture.
