Fungal Diversity and Plant Growth Promotion of Endophytic Fungi from Six Halophytes in Suncheon Bay

  • Endophytic fungi were isolated from roots of six halophytes in Suncheon Bay. The endophytic fungi of 35 species isolated from halophytes were identified by internal transcribed spacer (ITS) containing the ITS1, 5.8s, and ITS2 regions. All fungal strains were analyzed to diversity at the genus level. Fungal culture filtrates (FCF) of endophytic fungi were treated to Waito-c rice (WR) seedling for plant growth-promoting verification. It was confirmed that fungal strain Sj-2-2 provided plant growth promotion (PGP) to WR seedling. Then, PGP of Suaeda japonica was confirmed by treating culture filtrate of Sj-2-2. As a result, it was verified that culture filtrate of Sj-2-2 had more advanced PGP than positive control when treated to S. japonica. The secondary metabolites involved in culture filtrate of Sj-2-2 were identified by HPLC and GC-MS SIM analysis. The presence of physiologically bioactive gibberellins (GAs) and other inactive GAs in culture filtrate of Sj-2-2 was detected. The molecular analysis of sequences of Sj-2-2 showed the similarity to Penicillium sp. of 99% homology. The PGP of Sj-2-2 as well as symbiosis between endophytic fungi and halophytes growing naturally in salt marsh was confirmed. Sj-2-2 was identified as a new fungal strain producing GAs by molecular analysis of sequences. Consequently, the Sj-2-2 fungal strain was named as Penicillium sp. Sj-2-2. In this study, the diversity of endophytic fungi isolated from roots of halophytes in salt marsh and the PGP of a new gibberellin-producing fungal strain were confirmed.

First Report: Diversity of Endophytic fungi Possessing Antifungal Activity Isolated from Native Kougoed (Sceletium tortuosum L.)

  • Forty-three (n = 43) endophytic fungi with different morphologic characteristics were from a medicinal plant Sceletium tortuosum, were utilized to investigate their antifungal effectiveness against pathogenic fungi. All fungal isolates exhibited antifungal activity against one or more pathogens in the dual culture test whereas only 33 fungal culture filtrates (77%) showed decent antifungal effect. Fusaria and Aspergillus were the dominate genus that displayed significant antifungal activity. Isolates GG02, GG09, ND15, and ND17 showed the broadest spectrum of antifungal activity. Furthermore, culture filtrate of Fusarium sp. DR08 exhibited a broad range of antifungal activity against all the pathogens. The results suggest endophytic fungi isolated from medicinal plant might be a source of novel bioactive molecules. To the best our knowledge, this is the first report on endophytic fungi isolated from native kougoed exhibiting antifungal activity against plant fungal pathogens.

Three Unrecorded Endophytic Fungal Species Isolated from Plants in Taean-gun, Korea

  • In the present study, we isolated endophytic fungi from different tissues of two plants, Pseudosasa japonica and Quercus serrata from Taean-gun, Korea. Morphological characteristics of the isolated fungal strains were observed. We identified the fungi based on the phylogenetic analysis using DNA sequences from the region of internal transcribed spacer, large subunit rDNA, β-tubulin, and translation elongation factor 1-α DNA. Three endophytic fungi that were not previously recorded in Korea, namely Geomyces asperulatus, Leptoxyphium fumago, and Tubakia oblongispora were identified. Here, we describe the morphological characteristics of these unrecorded fungi and present the results of the phylogenetic analysis.

제주 수생식물에서 분리한 내생균류의 다양성 (Diversity of Endophytic Fungal Strains from Jeju Aquatic Plants)

  • 내생균류는 숙주식물 내에서 생물학적 그리고 비생물학적 스트레스로부터 저항성을 갖는데 도움을 준다. 수생식물은 수생환경에 서식하며 육상식물보다 수분스트레스에 더 많이 노출되어있다. 본 연구에서는 제주에 있는 남생이못, 수장동습지, 용수저수지, 강정천 4개의 습지에서 11개의 수생 수변식물을 채집하여 연구하였다. 전처리를 통해 식물 외생균을 제거하고 뿌리에서 내생균류를 분리하였다. 남생이못에서 126균주, 수장동습지에서 22균주, 용수저수지에서 44균주, 강정천에서 32균주가 분리되어 총 224개의 내생균류를 분리하였다. 분리된 균은 ITS를 이용해 동정한 후, 다양성 분석을 하였다. 남생이못에서 분리된 내생균류는 4개 문(Phylum), 7개 강(class), 12개 목(order), 19개 과(family), 30개 속(genus)로 구분되었다. 수장동습지에서 분리된 내생균류는 4개 문, 5개 강, 6개 목, 11개 과, 11개 속으로 구분되었으며, 용수저수지에서 분리된 내생균류는 4개 문, 5개 강, 7개 목, 12개 과, 13개 속으로 구분되었다. 강정천에서 분리된 내생균류는 1개 문, 2개 강, 5개 목, 7개 과, 9개 속으로 구분되었다. 4개의 습지에서 공통으로 분리된 속은 Alternaria속, Colletotrichum속, Fusarium속이었다. 향후 내생균류의 다양한 생태학적 분포와 다양성 규명에 대한 연구에 기초자료로 이용될 것이다.

태안반도에 자생하는 염생식물의 뿌리로부터 분리한 내생 진균의 다양성 (Endophytic Fungal Diversity Isolated from the Root of Halophytes in Taean Peninsula)

  • 7종의 자생 염생식물은 천일사초, 갯질경, 퉁퉁마디, 나문재, 칠면초, 해홍나물, 그리고 지채로서 태안반도에서 채집되었다. 염생식물의 뿌리로부터 분리된 37균주의 내생진균은 ITS영역 염기서열에 대하여 분석하였다. 모든 내생진균들은 다양성 지수들을 이용하여 분석되었다. 분리된 내생진균은 Eurotiales (78%), Capnodiales (5%), Hypocreales (5%), Agaricales (3%), Corticiales (3%), Glomerellales (3%), Pleosporales (3%)으로 7개 목으로 분류되었다. 그리고 분리된 내생진균의 속을 분석한 결과 자낭균문과 담자균문에 포함되는 Alternaria, Aspergillus, Cladosporium, Paecilomyces, Penicillium, Phanerochaete, Schizophyllum, Talaromyces, Verticillium등 9개 속으로 분류되었다. 내생진균을 분석하였을 때, Penicillium이 염생식물로부터 가장 많이 분포하고 있었다. 본 연구는 태안반도에 자생하는 염생식물들에 대한 내생진균의 분포와 다양성을 분석하였다.

Molecular Diversity of Fungal Endophytes Isolated from Garcinia mangostana and Garcinia parvifolia

  • Garcinia is commonly found in Malaysia, but limited information is available regarding endophytic fungi associated with this plant. In this study, 24 endophytic fungi were successfully recovered from different parts of two Garcinia species. Characterization of endophytic fungi was performed based on the conserved internal transcribed spacer (ITS) region sequence analysis and the antimicrobial properties. Results revealed that fruits of the plant appeared to be the highest inhabitation site (38%) as compared with others. Glomerella sp., Guignardia sp., and Phomopsis sp. appeared to be the predominant endophytic fungi group in Garcinia mangostana and Garcinia parvifolia. Phylogenetic relationships of the isolated endophytic fungi were estimated from the sequences of the ITS region. On the other hand, antibacterial screening showed 11 of the isolates possessed positive response towards pathogenic and nonpathogenic bacteria. However, there was no direct association between certain antibacterial properties with the specific genus observed.

  • 송이버섯 생육지의 소나무세근으로부터 내생균이 분리되었다. 분리된 내생균을 rDNA-ITS 영역으로 동정한 결과 18종이 확인 되었으며, 이중에는 자낭균류 15종, 털곰팡이 아문 3종이 포함되어 있음이 확인 되었다. 본 연구의 내생균 중에는 Penicillium속 균주가 가장 높은 빈도로 생육하고 있음이 확인되었다.

  • We investigated the diversity of the foliar endophytes of Korean ginseng. Endophytic fungi were isolated from healthy leaves of mountain-cultivated ginseng (MCG) and field-cultivated ginseng (FCG) at 4 sites in Chungbuk Province. A total of 24 species of fungal endophytes were identified using molecular approaches. Additionally, the diversity of these endophytic fungi was compared between MCG and FCG. The major isolated endophytes were Edenia gomezpompae and Gibberella moniliformis in the MCG and FCG samples, respectively. The results suggest that ginseng endophytes have different community structures in different environments, and this understanding may prove useful in ginseng cultivation.

소나무 침엽에서 분리된 3종의 국내 미기록 내생균 (Three Unreported Endophytic Fungi Isolated from Conifer Leaves of Pinus densiflora in Korea)

  • 본 연구에서는 전북 부안군에 서식하는 소나무의 침엽에서 내생균을 분리하였다. 분리된 균주는 internal transcribed spacer 영역, rDNA의 large subunit 영역, beta-tubulin 영역의 DNA 염기서열을 분석하여 동정하였다. 연구 과정에서 3종의 국내 미기록 내생균 균주를 확인하였으며, 확인된 종은 Paracamarosporiumhawaiiense, Tubakia dryina, Zasmidiumfructigenum이다. 확인된 미기록 균주의 형태적 특성 및 계통적 분석의 결과에 대해 서술하였다.

Endophytic Fungi as a Source of Biofuel Precursors

  • Endophytic fungi, isolated from a number of different species of tropical plants, were investigated for lipid biodiesel precursor production. The extracts produced from liquid cultures of these fungi were subjected to acidcatalyzed transesterification reactions with methanol producing methyl esters and then analyzed through chromatographic (GC-FID) and spectrometric techniques (MS, NMR $^1H$). The European Standard Method, EN 14103, was used for the quantification of methyl esters extracted from the fungi of the species and genera studied. Xylariaceous fungi exhibited the highest concentrations of methyl esters (91%), and hence may be a promising source for biofuel.
