Phylogenetic Diversity and Antifungal Activity of Endophytic Fungi Associated with Tephrosia purpurea

  • Sixty-one endophytic fungus strains with different colony morphologies were isolated from the leaves, stems and roots of Tephrosia purpurea with colonization rates of 66.95%, 37.50%, and 26.92%, respectively. Based on internal transcribed spacer sequence analysis, 61 isolates were classified into 16 genera belonging to 3 classes under the phylum Ascomycota. Of the 61 isolates, 6 (9.84%) exhibited antifungal activity against one or more indicator plant pathogenic fungi according to the dual culture test. Isolate TPL25 had the broadest antifungal spectrum of activity, and isolate TPL35 was active against 5 plant pathogenic fungi. Furthermore, culture filtrates of TPL25 and TPL35 exhibited greater than 80% growth inhibition against Sclerotinia sclerotiorum. We conclude that the endophytic fungal strains TPL25 and TPL35 are promising sources of bioactive compounds.

새우난초와 은대난초 뿌리에서 분리한 내생균의 다양성 (Diversity of Endophytic Fungi Associated with Roots of Calanthe discolor and Cephalanthera longibracteata in Korea)

  • 본 연구에서는 충남, 제주, 경남, 충북 등에서 새우난초(Calanthe discolor)와 은대난초(Cephalanthera longibracteata)의 뿌리를 채취하여 표면살균한 후 내생균을 순수분리 하였다. 분리된 내생균의 형태학 특징을 관찰하고 internaltranscribed spacer (ITS) rDNA영역의 염기서열을 ITS1F와 ITS4프라이머를 이용하여 분석하였다. 서로 다른 지역에서 채집한 새우난초와 은대난초이지만, 공통적으로 Leptodontidium orchidcola가 높은 상대수도와 빈도를 나타냈으며, 내생균류의 군집차이를 분석할 결과, 지역적 차이보다는 숙주에 의한 차이가 더 큰 영향을 미치는 것으로 나타났다.

  • 경북영주의 산림약용자원연구소에서 재배하고 있는 약용작물 3종(토천궁, 참당귀, 일천궁)의 뿌리에서 내생균을 분리하였다. 분리된 균주는 형태적 특성과 internal transcribed spacer, large subunit rDNA, beta-tubulin 영역의 분자적 분석을 토대로 동정하였다. 연구과정에서 3종의 국내 미기록종 내생균 균주를 확인하였고, 확인된 종은Dactylonectria pauciseptata, Rhizopycnis vagum, Sistotrema sernanderi이다. 미기록종 내생균 균주의 형태적 특성 확인 및 계통 분석 결과에 대해 기술하였다.

고랭이류에서 분리한 내생균의 생물다양성 및 지리적 양상 (Geographical Patterns and Biodiversity of Endophytic Fungi Isolated from Scirpus L. s.l. in Korea)

  • 고랭이류의 숙주식물인 Bolboschoenus planiculmis와 Schoenoplectustriqueter에서 내생균을 조사하였다. 총 25개체의 숙주식물에서 85개의 내생균을 분리하였고, ITS 지역을 사용하여 동정한 결과 19개의 분류군을 확인하였다. 그 중 92%는 Dothideomycetes에 속하였고, 4.8%는 Sordariomyetes, 1.6%는 Agaricomycetes, 0.8%는 Eurotiomycetes에 속하였다. B. planiculmis에서 가장 많이 분리된 내생균은 Cladosporium perangustum이고 S. triqueter에서는 Macrospora scirpicola였다. 염생식물에서 내생균의 연구는 매우 드물며, 이전에 목본식물들을 대상으로 수행된 선행연구들과는 다른 내생균의 군집 구조를 확인하였다. 향후 본 연구결과는 내생균 의 생물다양성 연구에 매우 유용할 것으로 생각된다.

Characterization of Pigment Production by Endophytic Rhodotorula mucilaginosa MGI from Tagetes erecta

  • Due to the hazardous effects of synthetic pigments, natural pigments are gaining popularity. Among natural sources microorganisms have become a major source of numerous industrially essential items and their use for getting various natural products have expanded dramatically in recent years. In the present study, 9 endophytic fungal strains were isolated from Tagetes erecta. On screening, yeast strain MGI was selected for further study which was identified as Rhodotorula mucilaginosa MGI. The pigment was intracellular, and the color of the crude extract was orange. The extract was subjected to characterization by UV-visible spectrophotometer and was purified by column phase chromatography, after purification two pigmented fractions were obtained. These fractions on characterization by thin layer chromatography (TLC) and Fourier transform infrared (FTIR) spectrophotometer affirms that they belong to carotenoid group of pigments. Orange (F1) and yellow (F2) fractions were anticipated as astaxanthin and beta carotene respectively. Moreover, the bioactive potential of pigmented fractions was investigated which manifested that F1 exhibited a maximum antioxidant activity of about 77% and F2 showed the highest zone of inhibition of 38 mm against Staphylococcus aureus. Thus, this study reflects that an endophytic yeast strain has the potential for the production of bioactive microbial pigments.

태안반도에 자생하는 해안식물 뿌리에서 분리한 내생진균의 다양성 분석 (Diversity Analysis of Endophytic Fungi Isolated from the Roots of Coastal Plants in Taean Peninsula)

  • 태안반도에 자생하는 해안식물인 해국(Aster sphathulifolius Maxim.), 갯개미취(Aster tripolium L.), 갈대(Phragmites australis), 갯꾸러미풀(Puccinellia nipponica Ohwi) 의 뿌리로부터 총 42주의 내생진균이 분리되었다. 그리고 분리된 내생진균의 ITS지역의 염기서열을 이용하여 동정한 결과 모두 자낭균문에 속하며, 8종류의 목(Botryosphaeriales, Capnodiales, Diaporthales, Dothideales, Eurotiales, Helotiales, Hypocreales, and Pleosporales)과 13종류의 속(Alternaria, Aureobasidium, Cadophora, Cladosporium, Davidiella, Diaporthe, Fusarium, Gibberella, Macrophomina, Metarhizium, Neosartorya, Penicillium, and Phoma)으로 분류되었으며, Eurotiales목의 Penicillium속의 분포비율이 가장 높게 존재하였다. Shannon 다양성지수를 비교한 결과 4종의 해안식물 중 갯개미취에서 분리한 내생진균의 다양성이 가장 높게 확인되었다.

  • Endophytic fungi have currently been acknowledged as the most promising source of bioactive compounds for drug discovery, and considerable progress has been made in exploring their diversity, species richness, and bioprospecting. Fungal endophytes from unique environmental settings offer a pool of potentially useful medicinal entities. Owing to the constant stresses imposed on macroalgae by marine environments, it is believed that algae and their associated endophytic symbionts represent a good source of structurally diverse bioactive secondary metabolites. Despite the proven significance of active metabolites of algal endophytes, little have been exploited. This review highlights the latest discoveries in algicolous endophytic research, with particular focus on the bioactive metabolites from algal endophytes. Compounds are classified according to their reported biological activities, like anticancer, antibacterial, antifungal, and antioxidant properties. Present experimental evidence suggests that a majority of the bioactive metabolites were reported from Phaeophyceae followed by Rhodophyceae and Chlorophyceae. An intensive search for newer and more effective bioactive metabolites has generated a treasure trove of publications, and this review partially covers the literature published up to 2016.

Biocontrol Potential of Fungal Endophytes against Fusarium oxysporum f. sp. cucumerinum Causing Wilt in Cucumber

  • Endophytic fungi have received much attention as plant growth promoters as well as biological control agents against many plant pathogens. In this study, 30 endophytic fungal species, isolated from various plants in China, were evaluated using in vitro dual culture assay against Fusarium oxysporum f. sp. cucumerinum, causing wilt in cucumber. The results of the present study clearly showed that all the 30 endophytic fungal isolates were highly capable of inhibiting the mycelial colony growth of Fusarium oxysporum f. sp. cucumerinum with inhibition % over 66% as compared to control treatments. Among all of them, 5 isolates were highly effective such as, Penicillium sp., Guignardia mangiferae, Hypocrea sp., Neurospora sp., Eupenicillium javanicum, and Lasiodiplodia theobromae, respectively. The Penicillium sp. and Hypocrea sp. were highly effective as compared to other isolates. From in vitro results 10 best isolates were selected for greenhouse studies. The results of the greenhouse studies showed that among all of them 3 endophytic fungal isolates successfully suppressed wilt severity when co-inoculation with pathogen Fusarium. oxysporum f. sp. cucumerinum. The endophytic fungi also enhanced plant growth parameters of the host plants, the antagonistic fungal isolates increased over all plant height, aerial fresh, and dry weight as compared to control.

고창갯벌의 염생식물 뿌리로부터 분리된 내생진균의 다양성 (Diversity of Endophytic Fungi from the Roots of Halophytes Growing in Go-chang Salt Marsh)

  • 염생식물인 칠면초, 갈대, 갯질경, 해홍나물을 고창갯벌로부터 채집하였으며, 39점의 내생진균을 염 습지에 자생하고 있는 염생식물의 뿌리로부터 분리하였다. 내생진균들은 ITS1, 5.8s와 ITS2를 포함하는 ITS-rDNA 영역에 의해 분석되었다. 다양한 내생진균은 Eurotiales (36%), Pleosporales (26%), Hypocreales (18%), Incertae sedis (8%), Glomerellales (5%), Sordariales (2%), Xylariales (2%), Capnodiales (3%)등 8종류 목에 속하는 것으로 확인되었다. 39점의 내생진균은 속(genus)단계에서 Alternaria, Aspergillus, Chaetomium, Cladosporium, Colletotrichum, Coniothyrium, Dothideomycete, Fusarium, Gibberella, Macrophoma, Penicillium, Pestalotiopsis, Phaeosphaeria, Phoma, Pleosporales, Pseudozyma, Talaromyces, 그리고 Termitomyces에 포함되는 것으로 분석되었다. 그리고 내생진균은 Eurotiales목의 Penicillium속이 가장 많이 분포하고 있는 것으로 분석되었다.

Screening for Antifungal Endophytic Fungi Against Six Plant Pathogenic Fungi

  • A total of 187 endophytic fungi were isolated from 11 plant species, which were collected from 11 locations in Korea. Their antifungal activities were screened in vivo by antifungal bioassays after they were cultured in potato dextrose broth and rice solid media. Antifungal activity against plant pathogenic fungi such as Magnaporthe grisea(rice blast), Corticium sasaki(rice sheath blight), Botrytis cinerea(tomato gray mold), Phytophthora infestans(tomato late blight), Puccinia recondita(wheat leaf rust), and Blumeria graminis f. sp. hordei(barley powdery mildew) was determined in vivo by observing the inhibition of plant disease development. Twenty(11.7%) endophytic fungi fermentation broths were able to control, by more than 90%, at least one of the six plant diseases tested. Among 187 liquid broths, the F0010 strain isolated from Abies holophylla had the most potent disease control activity; it showed control values of more than 90% against five plant diseases, except for tomato late blight. On the other hand, fourteen(7.5%) solid culture extracts exhibited potent disease control values of more than 90% against one of six plant diseases. The screening results of this study strongly suggested that metabolites of plant endophytic fungi could be good potential sources for screening programs of bioactive natural products.
