흰쥐 골격근에서 안정화 이산화염소(Stabilized $ClO_2$)의 부패억제에 관한 현미경적 연구 (Microscopic Study of Decomposition-Inhibition in Stabilized $ClO_2$ Gas in Skeletal Muscle of Rat)

  • 강한 산화력과 소독효과로 인해 광범위하게 사용되고 있는 안정화 이산화염소(S-$ClO_2$)의 근육조직에 대한 부패억제 효과를 알아보고자 본 연구를 수행하였다. 본 실험을 위해 8주령 SD계 흰쥐 골격근(skeletal muscle)을 사용하였고, 안정화 이산화염소를 처리하지 않은 대조군과, 안정화 이산화염소의 분말과 수용액을 처리한 실험군으로 분류하여 광학 및 투과전자현미경으로 관찰하였다. 광학현미경 관찰 결과, 대조군 1일부터 근섬유막이 분리되기 시작하였고 핵의 붕괴가 관찰되었다. 2일군에서는 근섬유막이 붕괴되어 근섬유의 형태만 남아 있었고, 근원섬유도 붕괴되는 양상을 나타내었다. 3일군 이후 대조군의 경우는 대부분의 근섬유막과 결합조직의 붕괴로 근섬유의 구분이 불분명하였다. 실험군의 경우, 3일군에서 근섬유 사이 결합조직의 부분적인 붕괴가 관찰되었다. 그러나 4, 5일군에서도 3일군과 유사한 양상을 나타내었다. 한편, 전자현미경적 소견은 대조군의 경우 1일군부터 근섬유막의 붕괴로 근섬유의 경계가 불분명하였고, 미토콘드리아막도 부분적인 파괴가 관찰되었다. 2일째부터 근섬유막이 모두 붕괴되었고 근섬유 사이에서 세균이 관찰되었다. 3일군 이후는 근육의 형태를 구별할 수 없었다. 그러나 실험군의 경우, 1일과 2일군은 전형적인 근육의 미세구조를 관찰할 수 있었고, 3일군부터 부분적인 미토콘드리아막의 붕괴가 관찰되었다. 이후 시간경과에 따라 결합조직이 붕괴되어 동심원상의 구조물이 일부 관찰되었다. 따라서 본 연구결과, 대조군은 1일군부터 조직의 붕괴가 시작되어 3일군 이후는 대부분 붕괴되어 형태를 구분할 수 없었으나, 실험군의 경우는 3일군부터 부분적인 붕괴가 시작되었고 이후 시간이 경과되어도 전체적인 근육의 형태가 유지되어 안정화 이산화염소가 근육에 부패억제 효과가 있는 것으로 확인되었다.

Combined Effects of High Pressure and Heat on Shear Value and Histological Characteristics of Bovine Skeletal Muscle

  • Changes in shear force value, transverse sections, myofibrils and intramuscular connective tissue of bovine skeletal muscle exposed to the combination of high-pressure up to 400 MPa and heat (30 and $60^{\circ}C$) were studied. The shear force value decreased by pressure-heat treatment up to 200 MPa at 30 and $60^{\circ}C$, and then slightly increased over 200 MPa at $30^{\circ}C$. Shear force values of treated muscles were lower than those of untreated ones. Gaps between muscle fibers in the untreated muscle were a little clear, and then they became very clear in the treated muscles up to 200 MPa at 30 and $60^{\circ}C$. However, the gaps reduced significantly over 200 MPa at $30^{\circ}C$. The remarkable rupture of I-band and loss of M-line materials progressed in the myofibrils with increasing pressure applied. However, degradation and loss of the Z-line in myofibrils observed in the muscle treated at $60^{\circ}C$ was not apparent in the muscle treated at $30^{\circ}C$. The length of the sarcomere initially contracted by pressure-heat treatment of 100 MPa at $30^{\circ}C$ seemed to have recovered with increase of the pressure up to 400 MPa. In the muscle treated at $60^{\circ}C$, the length of sarcomere gradually decreased with increase of the pressure up to 400 MPa. In the treated muscles, changes in the honeycomb-like structure of endomysium were observed and accelerated with increase of the pressure. A wavy appearance clearly observed at the inside surface of endomysium in the untreated muscles gradually decreased in the treated muscles with increase of the pressure. Tearing of the membrane was observed in the muscles treated over 150 MPa at $30^{\circ}C$, as observed in the sample pressurized at 100 MPa at $60^{\circ}C$. The roughening, disruption and fraying of the membrane were observed over 200 MPa at $60^{\circ}C$. From the results obtained, the combination of high-pressure and heat treatments seems to be effective to tenderize tough meat. The shear force value may have some relationship with deformation of intramuscular connective tissue and myofibrils.

  • 육(肉)의 가열시(加熱時) 육조직내(肉組織內)에 어떤 변화가 일어나는지를 관찰하기 위하여 우육(牛肉)을 $80^{\circ}C$로 가열(加熱)한후 scanning 및 transmission 전자현미경으로 육(肉)의 endomysial connective tissue, sarcolemma, transverse fidge, Z-disc, A 및 I-band등(等)을 관찰한바 $80^{\circ}C$에의 가열(加熱)로 endomysium, sarcolemma 등(等)이 응고됨과 아울러 심한 파괴현상을 보였으며 근원섬유 구조에 있어서도 I-band가 심한 절단현상을 보임과 아울러 A-band에 있어서는 심한 응고현상이 관찰되었으나 Z-disc는 비교적 열(熱)에 잘 견디는 것으로 관찰되었다.

《영추(靈樞)·경근(經筋)》에서 근(筋)의 해부학적 의미에 대한 연구 (Study on the Anatomical Meaning of 'Geun(筋)' in 『Yeongchu·Gyeonggeun(靈樞·經筋)』)

  • Objectives: This study was done to establish the anatomical meaning of the term 'Geun(筋)'. Methods: Through analysis of 《HwangJeNaeGyeong(黃帝內經)》, the meaning of 'Geun(筋)', 'GeunMag(筋膜)', 'Yug(肉)', and 'Gi(肌)' were established. Based on analysis, the anatomical meaning of the 'Meridian-muscle(經筋)' was studied by comparing it with anatomy. Results & Conclusions: 'Gyeong(經)' is recognized as a metaphysical expression and "Geun(筋)" means myofascia in anatomy. The concept of 'Geun(筋)' includes the epimysium and perimysium, as well as tendons and ligaments, which are extensions of these. 'Fascia', refers to the fascia of the whole body, and also appertain to 'Geun(筋)'. 'Yug(肉)' means endomysium, muscle fiber, and adipose tissue and layer. The word 'GeunMag(筋膜)' used in the 《HwangJeNaeGyeong(黃帝內經)》 means anatomically a 'tendon'. Therefore, 'Muscle' should be translated as 'GeunYug(筋肉)' in Traditional medicine. 'Meridian-muscle(經筋)' can be defined as the longitudinal muscle and fascia system, which is the basis of whole body encompassing dynamics.

강제급수(强制給水)가 우근육(牛筋肉)의 함수율(含水率)과 조직학적성상(組織學的性狀)에 미치는 영향(影響) (Effect of Forced Oral Administration of Excessive Tap water on the Water Content and Histological Changes of Some Muscles of Korean Cattle)

  • Experimental studies on the water content of muscles and the histological changes of muscles, digestive tract and kidney, influenced by forced oral administration of excessive tap water, were performed by using 10 Korean bulls. Experimental animals were divided into two groups, five heads for test group and five for control group. All of the bulls used in this experiment were weighing between 250 kg and 300 kg and 3 and 4 years of age. In each test animal, a total of 140 litters of tap water was passed in the forestomach by means of catheterization within 12 to 18 hours. And each of them was slaughtered immediately after the animals showing symptoms of respiratory distress. In control group, the animals were allowed to drink tap water normally. From test and control animals after slaughter, each 10 gm of M. biceps femoris, M. satorius, M. adductor, M. gluteous supercialis, M. iliocostalis lumborum, and M. transversus costarum were taken from definite parts in order to measure water content. In the histopathological studies tissues of rumen, reticulum, omasum, abomasum, duodenum, colon and kidney were taken as wall as the above mentioned muscles. The results obtained were as follows; 1. Increase of water content in the muscles of test group were 4.6% in M. satorius, 4.24% in M. transversus costarum, 4.14% in M. gluteous supercialis, 4.02% in M. adductor, 3.88% in M. biceps femoris, and 2.46% in M. iliocostalis lumborum respectively. The highest increase was found in M. satorius and the lowest in M. iliocostalis lumborum. 2. In test animal, average increasing value of water content in muscles was 3.9% and shown highly significant (p<0.01). 3. On the microscopical findings of carcass, marked watery edema was observed all of the subcutaneous tissues and intermuscular connective tissues. 4. Microscopically, the epimysium, perimysium and endomysium were widened, and the muscle fibers were lacerated. The cells of stratum lucidum in the epithelium of rumen and reticulum were shown marked vacuolization. In the kidney, dilatation of Bowman's spaces and proximal tubles was observed.

전기자극이 흰쥐의 정상 가자미근 형태에 미치는 영향 (Effects of Electrical Stimulation on Normal Soleus Muscle in Rat)

  • 전기자극이 흰쥐의 정상근에 어떠한 영향을 미치는지를 알아보기 위하여 좌골신경의 표적근인 가자미근을 이용하여 조직계측학적, 조직화학, 전자현미경적 관찰을 하였다. 정상적으로 활동하는 흰쥐의 골격근을 매일 전기자극한 결과 전기자극군은 자극을 시작한 후 2주까지는 근섬유의 굵기와 무게가 증가하고 그 이후로는 큰 변화가 없었다. 특히 근섬유의 종류별로는 적색섬유(red muscle fiber)의 채적밀도가 증가하였고 백색섬유(white muscle fiber)는 줄어드는 양상을 보였다. 조직화학적 검사 결과 전기자극군은 근섬유가 다소비대(hypertrophy)해지고 내근주막과 외근주막 사이가 좁아져 있었다. 정상근에서는 당원이 근섬유의 뚜렷한 구별없이 양성반응을 보였으나 실험군은 특정 근섬유에만 반응이 나타났었다. 또한 NADH-TR반응 결과 전기자극군은 적색섬유가 유의하게 증가하여 생체계측학적 결과와 일치하였고 미세구조적 관찰 결과 정상근의 적색섬유는 근섬유와 평행으로 mitochondria가 형성되어 있었고 백색섬유에서는 근절부위에서 관찰되었다. 그러나 전기자극군에서는 적색섬유는 근초부위에 mitochondria가 많이 관찰되고 백색섬유도 근절부위에 작은 mitochondria의 수가 증가되는 것을 관찰할 수 있었다.

수컷 생쥐의 골다공증과 근위축에 대한 익지인(益智仁)의 효과 (The Effects of Alpiniae Oxyphyllae Fructus on Osteoporosis and Muscle Dystrophy of Male Mice)

  • Objective: To investigate the effect of Alpiniae oxyphyllae fructus (AOF) on the alleviation of musculoskeletal disorders caused by aging, we conducted experiments on osteoporosis and muscle atrophy. Methods: The experimental group was classified into a control group, aging-elicited (AE) group and AOF group. The control group comprised 8-week-old Institute of Cancer Research (ICR) mice. The AE and AOF groups were ICR mice at 50 weeks of age. For the AE group, 10 mL of distilled water was administered once a day for 180 days without any treatment. An AOF extract (0.54 g/kg) was dissolved in distilled water and administered to the mice in the AOF group once a day for 180 days. Results: In the experiment on the alleviation of osteoporosis, the distribution of glucosaminoglycan in the bone matrix of the femoral bone was increased in the AOF group; moreover, the osteocalcin (OCN) positive reaction was increased and 8-OHdG positivity was decreased. In addition, AOF positively decreased RANKL, positively increased OPG, and positively decreased MMP-3. Muscle fiber loss in the endomysium following muscle degeneration of the quadriceps was reduced more in the AOF group compared with the AE group, and caspase-3 positive responses were also decreased. In addition, the 8-OHdG and p-lkB positivity in the AOF group decreased compared with the AE group, and the Myo-D positivity increased. Conclusion: We found that increasing bone formation alleviates osteoporosis, and that reducing bone loss alleviates muscle atrophy by reducing muscle loss and increasing muscle development.

Influence of freeze-thawed cycles on pork quality

  • Objective: This study was conducted to determine the effect of freeze-thawed cycles (Fresh meat, F-T 1 cycle and F-T 2 cycles) on the quality characteristics of porcine longissimus dorsi muscle. Methods: A total of 20 three-crossbred pigs (Duroc×[Large White×Landrace]) were randomly obtained from a commercial slaughterhouse in Thailand. Muscle samples were immediately taken from 10 to 11th of the longissimus dorsi for histochemical analysis. The muscles were cut into 2.54 cm-thick chops. A minimum of 20 chops were used for each treatment (fresh meat, freeze-thawed 1 and 2 cycles). Individually chops were packaged in polyethylene bags and frozen at -20℃ for 6 months followed by thawing in refrigerator at 4℃ for 24 h (the 1st freeze-thawed cycle). The freeze-thawed procedure was repeated for two cycles (the 2nd freeze-thawed cycle). Thawing loss, shear force value, citrate synthase activity and muscle fiber characteristics were determined on the muscles. Results: Results showed that increasing of freeze-thawed cycle increased the thawing loss (p<0.01) and citrate synthase activity (p<0.001). Shear force value of fresh meat was higher than freeze-thawed 1 and 2 cycles (F-T 1 cycle and F-T 2 cycles). Freeze-thawed cycles affected muscle characteristics. Muscle fiber area and muscle fiber diameter decreased with an increasing number of freeze-thawed cycles (p<0.001), while the thickness of endomysium and perimysium were increased (p<0.001). Conclusion: Repeated freeze-thawed cycles degraded muscle fiber structure and deteriorated pork quality.

어유식이가 본태성고혈압쥐(SHR)혈압 및 지질대사에 미치는 영향 -혈청지질상태를 중심으로- (Effect of Fish Oil Diet on Blood Pressure and Lipid Metabolism in Spontaneously Hypertensive Rat -Changes in Serum Lipid Status-)

  • $C_{20}$이상의 ${\omega}3$ 다불포화지방산을 많이 함유한 어유가 본태성 고혈압쥐(SHR)의 혈압과 혈청 지질상태에 미치는 영향을 조사하기 위해 고등 어유, 대두유, 쇠기름을 식이에 5% 함유시킨 분말식이를 각각 조제하여, 생후 4주된 SHR과 정상혈압 Wistar쥐 숫컷에게 8주간 섭취시켰다. Wistar종의 성장은 세 군 모두에서 8주간 지속적이었으며 고등어유군의 성장율이 5주 이후 에는 다른 군보다 다소 높았다. SHR의 성장은 6주까지는 원만하였으나 그 후 거의 정지상태였고 식이지방군 간의 차이는 거의 없었다. 수축기혈압은 실험초기부터 SHR이 높아서 6주부터는 150mmHg이상의 고혈압을 나타내었다. 식이 8주의 혈압은 SHR에서는 고등어유군이 가장 낮았고, Wistar에서는 세 군 사이에 차이가 없었다. 심장과 신장무게가 SHR에서 컸으며, 현미경관찰에 의하여 SHR 심장조직 endo mysium 과 신장조직의 urinary space 가 축소되었음을 볼 수 있었으나 식이에 따른 조직무게나 구조상 차이는 없었다. 고등어유는 Wistar종과 SHR 모두에게서 혈청 중성지방을 낮추는 효과가 있었고, SHR의 중성지방값이 Wistar종보다 대체로 낮았다. 혈청 총 콜레스테롤과 HDL-콜레스테롤값은 식이군간에 차이는 없었으나 SHR의 HDL-콜레스테롤값이 Wistar종에서 보다 낮았다. 혈청지단백질 분획비를 보면 일반적으로 SHR이 VLDL이 높고 HDL이 낮았고, 고등어유군의 $HDL_1HDL_{2+3}$ 비율이 타군 보다 낮았다. 세 식이지방의 주요 지방산이 혈청지질에 반영되어 혈청 인지질과 중성지질 구성지방산은 세 군이 각각 특색있는 지방산 구성 양상을 나타내었는데 대두유군의 $C_{18:2}({\omega}_6)$의 반영도가 제일 뚜렷하였으나 SHR 에서는 Wistar 종에서 보다 그 정도가 낮았고 고등어유의 $C_{22:6}({\omega}_3)$는 중성지방에서만 주로 관찰되었다. 이러한 지질 구성지방산의 차이가 혈청지질 패턴변화에 중요원인으로 사료된다.
