Association of Endocrine Factors (Insulin-Like Growth Factor-II and Binding Protein-3) with Litter Size in Pigs

  • Litter size has been one of the important economic traits in porcine reproduction. The insulin-like growth factor (IGF) system has been shown to mediate actions of the steroid hormone or to synergize with other endocrine factors so that it consequently plays roles in reproductive processes, including ovulation, implantation, maintenance of pregnancy, and fetal development. However, the effect of the serum IGF system on porcine litter size has not been deeply studied. Therefore, this study was conducted to relate serum IFG-II concentration and IGF binding protein-3 (IGFBP-3) expression with porcine litter size. Moreover, the possible association of those with estrogen receptor (ER) as a candidate gene for litter size was investigated. Swine were separated into two groups showing high and low litter sizes, and sera were collected from sows in the estrous cycle to postnatal growth of their female progeny. Serum IFG-II concentration was measured by radioimmunoassay and IGFBP-3 expression was detected by Western ligand blotting. During the estrous cycle, IGFBP-3 expression in both groups decreased moderately from metestrus to estrus, but IFG-II concentration showed a reverse pattern. Also, IFG-II concentration and IGFBP-3 expression decreased gradually as pregnancy proceeded. Unlike IGFBP-3, IFG-II decreased moderately as newborn pigs grew. Significant differences in serum IFG-II amount between the two groups were detected at 60 (p<0.01), 75, 90, and 105 d (p<0.05) of pregnancy and at 60 (p<0.01), 45, and 105 d (p<0.05) of postnatal growth. Furthermore, based on ER genotypes, a high litter size group with genotypes AB and BB showed lower IFG-II concentration than a low litter size group with a genotype AA during pregnancy. Taken together, the results indicate that the serum IFG-II and IGFBP-3 are correlated with the litter size in pigs.

전남 농공단지 폐수처리장 방류수의 화학적/생물학적 독성평가 (Toxic Assessment on Effluents of Argo/Industrial Wastewater Treatment Plants in Jeonnam using Chemical and Biological Method)

  • 본 연구는 영산강유역 농공폐수처리장 방류수중의 유기오염물질 분포를 관찰하였다. 시료채취 지점은 영산강 유역에 존재하는 4지점의 농공단지 폐수처리장을 선정하였다. 본 연구에서 GC-Ion trap MS로 300종의 일반적인 화학물질을 ppt수준에서 검출할 수 있는 방법을 이용하여 분석하였다. 농공폐수처리방류수에서 검출된 주요 유기오염물질은 농약류, CH구조의 방향족, CHO구조의 프탈레이트류, 그리고 CHO(N) 구조의 아로메틱 아민류였다. 또한 diethylphthalate를 포함한 17종의 내분비계장애물질이 검출되었다. XTT assay를 이용한 세포독성 결과는 TV로 나타냈으며, 그들의 세포독성은 A지점에서 27.2, D지점에서 24.4로 가장 높게 나타났다. 한편 화학적 분석 결과와 생물학적 독성도와는 일치하지 않는 것으로 나타났다.

Emerging Genomics Technologies in Nutritional Sciences: Applications to obesity and hypertension research

  • While the sequencing of several genomes was underway, several advanced techniques in genetics, molecular biology and protein chemistry emerged. Within the nutritional sciences, while the focus on nutrition education, epidemiology and public health aspects remains essential; it is crucial to incorporate the new advances in gene and protein discovery in nutritional studies. Nutrition is a discipline that has always integrated social, biochemical and physiological sciences from the studies at the molecule level to studies at the population level. For this reason, nutritionists are in a prime position to readily incorporate the current genomics approaches in nutrition research, All the available analytical techniques can and should be used in modern nutritional sciences. These include genetics, genomics, proteomics and metabolomics which also require integration and use of bioinformatics and computational methods for data analysis and management. These applications will be briefly reviewed with a primary focus on what the genomics and genetics approaches offer to nutritionists. We will use one of our research focus areas to illustrate uses of some of these applications in obesity-hypertension research. Our central hypothesis is that adipose tissue is an endocrine organ that plays a major role in obesity and related hypertension. We are primarily studying the renin angiotensin system (RAS). We provide evidence from our own studies and others for the paracrine as well as endocrine role of adipocyte-derived angiotensin II in adipocyte gene expression, adiposity and blood pressure regulation. Both cell culture studies as well as knockout and transgenic mice models are used to test our hypothesis. Genomics and proteomics technologies are currently developed to complement our physiological and molecular studies on the RAS and for a fine analysis of this system and its function in health and disease.

혼합된 고급산화공정(AOPs)을 이용한 내분비계장애물질(DEP, NP)의 분해특성 연구 (Degradation Characteristic of Endocrine Disruptors (DEP, NP) Using Combined Advanced Oxidation Processes (AOPs))

  • Diethyl phthalate (DEP) and nonylphenol (NP) are widely spread in the natural environment as an endocrine disruption chemicals (EDs). Therefore, in this study, ultrasound (US) and ultraviolet (UVC), including $TiO_2$, as advanced oxidation processes (AOPs) were applied to a DEP and NP contaminated solution. When only the application of US, the optimum frequency for significant DEP degradation and a high rate of hydrogen peroxide ($H_2O_2$) formation was 283 kHz. We know that the main mechanism of DEP degradation is radical reaction and, NP can be affected by both of radical reaction and pyrolysis through only US (sonolysis) process and combined US+UVC (sonophotolysis) process. At combined AOPs (sonophotolysis/sonophotocatalysis) such as US+UVC and US+UVC+$TiO_2$, significant degradation of DEP and NP were observed. Enhancement effect of sonophotolysis and sonophotocatalysis system of DEP and NP were 1.68/1.38 and 0.99/1.17, respectively. From these results, combined sonophotocatalytic process could be more efficient system to obtain a significant DEP and NP degradation.

TERT mRNA Expression is Up-Regulated in MCF-7 Cells and a Mouse Mammary Organ Culture (MMOC) System by Endosulfan Treatment

  • Endosulfan is one of the organochlorine pesticides, which are well-known endocrine disruptors (EDs), and it acts as an estrogen agonist. Estrogen is a group of hormones that play an important role in mammary gland function and are implicated in mammary carcinogenesis. In the present study, we studied the effects of endosulfan on nodule like alveolar lesion (NLAL) formation in mouse mammary gland development using a mouse mammary gland organ culture (MMOC) system. Although endosulfan-treated mammary glands did not form NLALs, more alveolar buds were formed in this group than in the negative control (vehicle-treated) group. In addition, telomerase reverse transcriptase (TERT) mRNA expression levels were increased in endosulfan-treated mammary glands in a dose-dependent manner. Telomerase can be activated by estrogen, therefore, we examined the effects of endosulfan on telomerase activity, and found that the telomerase activity in estrogen receptor-positive MCF-7 cells was up-regulated by endosulfan treatment. Moreover, this activation was accompanied by the up­regulation of the TERT mRNA expression. Also, transient expression assays using CAT reporter plasm ids containing various fragments of the TERT promoter showed that this imperfect palindromic estrogen-responsive element is almost certainly responsible for the transcriptional activation by endosulfan. These results may help elucidate the endocrine disrupting mechanism of endosulfan.

  • Background: 2-Ethylhexyl diphenyl phosphate (EHDPP) is widely used as a flame-retardant plasticizer in the production of polyvinyl chloride, adhesives, and food packaging. This chemical has been frequently detected in water, sediment, and indoor environments, and its lipophilicity raises concerns about bioaccumulation. Objectives: In this study, the effects of EHDPP on the development, behavioral changes, and growth hormone (GH) endocrine system of zebrafish larvae were investigated. Methods: Fertilized embryos were exposed to various concentrations (control, solvent control, 0.07, 0.7, 7, 70, and 700 ㎍/L) of EHDPP for 96 h. Developmental toxicity endpoints were observed daily. Behavioral changes under light-dark-light conditions and changes in hormones and genes related to GH/insulin-like growth factors (IGFs) axis were determined. Results: Significant decreases in survival, body length and moving distance were observed in zebrafish larvae exposed to 70 and 700 ㎍/L EHDPP. The concentrations of GH and IGF-1 were significantly decreased in zebrafish larvae exposed to 70 and 700 ㎍/L EHDPP. This change was well supported by changes in the transcription of genes involved in GH, IGF, IGF receptors, and IGF binding proteins. Conclusions: Our observations showed that exposure to 70 and 700 ㎍/L EHDPP could disrupt the feedback circuits of the GH/IGFs axis, ultimately leading to developmental toxicity, hypoactivity, and mortality.

The Consequences of Mutations in the Reproductive Endocrine System

  • The reproductive activity in male mammals is well known to be regulated by the hypothalamus-pituitary-gonad axis. The hypothalamic neurons secreting gonadotropin releasing hormone (GnRH) govern the reproductive neuroendocrine system by integrating all the exogenous information impinging on themselves. The GnRH synthesized and released from the hypothalamus arrives at the anterior pituitary through the portal vessels, provoking the production of the gonadotropins(follicle-stimulating hormone (FSH) and luteinizing hormone (LH)) at the same time. The gonadotropins affect the gonads to promote spermatogenesis and to secret testosterone. Testosterone acts on the GnRH neurons by a feedback loop through the circulatory system, resulting in the balance of all the hormones by regulating reproductive activities. These hormones exert their effects by acting on their own receptors, which are included in the signal transduction pathways as well. Unexpected aberrants are arised during this course of action of each hormone. This review summarizes these abnormal phenomena, including various mutations of molecules and their actions related to the reproductive function.

The Health Effects of Whole Body Vibration

  • Objective: The purpose of this study is to review the hazards of whole body vibration(WBV) and prevent human injuries due to WBV. Background: Vibrations that shake the whole body in vehicles and vessels is WBV. Vibration at a specific frequency, and intensity in the human body can act as a hazard. But, it has not yet done enough research about effects of WBV. Method: We analyzed and reviewed occupational or ergonomic papers published past 30 years. Results: Exposure to whole body vibration can cause permanent physical injury or damage of the nervous system. In addition, it may cause problem in the cardiovascular system, digestive system, musculoskeletal system, endocrine system, reproductive system, and psychological system. The effect of vibration depends on its acceleration, duration, frequency, and direction. Conclusion: WBV-exposed human body experience a high risk of some illnesses such as lumbar spinal disorders. Some workers(bus drivers, truck drivers and heavy equipment operators) are the high risk population exposed to WBV. Application: The results of this study might help to judge and prevent occupational disease caused by WBV.

Effects of Prenatal and Neonatal Exposure to Bisphenol A on the Development of the Central Nervous System

  • Bisphenol A (BPA) is one of the most common endocrine disrupters. In the last decade, the number of studies concerning the effects of chronic treatment with BPA on the development of the central nervous system (CNS) has increased. However, little is known about the effects of chronic exposure to BPA on higher brain functions such as memory or psychomotor functions. Here, we report our following findings: (1) Prenatal and neonatal exposure to BPA enhances psychostimulant-induced rewarding effects, results in the up- or downregulation of dopamine receptors, causes memory impairment, and decreases choline acetyltransferase (ChAT) activity. (2) BPA activates astrocytes in vivo and in vitro. These findings suggest that prenatal and neonatal exposure to BPA affects the development of the CNS.
