Effects of Phthalate/Adipate Esters Exposure during Perinatal Period on Reproductive Function after Maturation in Rats (성숙한 랫트의 번식 기능에 있어 프탈레이트/아디페이트 에스테르의 주산기 노출의 영향)

  • Phthalate esters that are used as plasticizers and also found at low levels in foods such as dairy products are often mentioned as suspected endocrine disrupters. The purpose of the present study is to elucidate whether perinatal exposure to di-n-butyl phthalate(DBP), diisononyl phthalate (DINP) and di-2-ethylhexyl adipate (DEHA) affects several aspects of reproductive function in rats especially sexual differentiation of the brain. To this end, the dams were provided with pulverized soy-free diet containing 20, 200, 2,000 and 10,000 ppm of DBP, 40, 400, 4,000 and 20,000 ppm of DINP, or 480, 2,400 and 12,000 ppm of DEHA from gestational day (GD) 15 to postnatal day (PDN) 21, the day of weaning, and serum sex steroid hormone, gonadotropin levels and sexual behaviors after maturation were assessed. At Postnatal week (PNW) 20-21, serum levels of sex steroids and gonadotropins in both male and female rats, as well as estrous cyclicity in females, were not changed by perinatal exposure to DBP, DINP and DEHA, indicating that these chemicals did not affect sexual differentiation of the brain controlling the endocrine system of hypothalamo-pituitary-gonadal (HPG) axis. On the other hand, inhibitory influences on sexual behaviors, especially on ejaculation in males and lordosis in females, were observed by perinatal exposure to these chemicals. These results suggest that these chemicals may act directly on discrete regions of the hypothalamus regulating sexual behaviors, but not regulating gonadotropin secretion, thereby affect sexual differentiation of the brain with a resultant decrease in sex-specific behaviors in adulthood.

Toxicologic Effects of BPA and DEHP on Semen Characteristics in Pig (BPA와 DEHP가 돼지 정액 성상에 미치는 독성 영향)

  • The objective of this study was to investigate the toxic effects of Bisphenol A(BPA) and di-2 ethyhexyl phthalate (DEHP) as endocrine disrupters on sperm motility, viability, membrane integrity and abnormality during in vitro incubation of boar semen. Semen were randomly divide into 24 groups and healed with different concentrations of BPA md DEHP($1{\sim}100{\mu}M$) for 3, 6 and 9 hrs, respectively. The percentages of sperm motility and viability decreased by treatment time with both BPA and DEHP, and obiously differ from the controls. The percentages of sperm motility and viability significantly decreased by incubation with both $100{\mu}M$ of BPA and DEHP compared to control and other treatment groups(p<0.05). Sperm membrane integrity was significantly reduced by incubation with 10 and $100{\mu}M$ of BPA and DEHP, respectively(p<0.05), but sperm abnormality were not significantly affect both BPA and DEHP. These results indicate that high concentration of BPA and DEHP($>10{\mu}M$ can affect noxiously the sperm characteristics.

  • In recent years, dioxins which were designated as persistent organic pollutants and endocrine disrupters are treated as substance of environmental pollution and studied about human health risk assessment, emission pollutants estimation, analytical methods and so on. It is easy that dioxins are accumulated to soil because of the atmosphere circulation of burning up the waste. This is the comparative studies on the distribution relationship of dioxin isomers in exhausted gas of industrial waste and urban waste incinerators, ambient air and soil. A basis of PCDDs and PCDFs based on OCDD was drawn up to the curve and they correspond to dioxin isomers in exhausted gas of industrial waste and urban waste incinerators and ambient air. On comparing these results, It was found that the ambient air and exhausted gas of industrial waste incinerators were very similar in curve and ratio. Consequently, environmental by exposed dioxin depends on the exhausted gas of industrial waste incinerators than urban waste incinerators. In case of soil, even though we can not completely rule out the possibility of pollution source bring on pesticide and other factors, and naturally biological dissociations, the curve shape is very similar to exhausted gas of industrial waste incinerators and ambient air. So, we inform here that it was mainly caused by these environmental factors.

  • Objectives : The aim was to investigate the characteristics of low dose effect and non monotonic dose responses(NMDRs) and to predict the influence it might have on the health and medicine, traditional Korean medicine. Methods : By investigating recently published major literatures related to low dose effect and NMDRs, the definition, mechanism, major related fields, and the influence on public health and medicine has been categorized and summarized. Results : Although there are still heated debates about the definition of low dose, it implies the biological responses in environmental exposure level and the NDMDRs means the nonlinear relationship between the dose-response in the slope sign change. Also, it implies the new form of the curve showing a U, reverse U shape, and the slop sign constantly changing showing various forms. This mechanism is because the two acceptor shows opposing effects to toxic materials and the affinity is different along with the numerical value that increase and decrease being different. These characteristics generally appear in endocrine disrupters such as bisphenol A, agricultural pesticides, metal, and radiation. The research field in the public health and medical treatment is obesity, problems in metabolism, growth hormone treatment, climacteric treatment, breast cancer, intake of Korean traditional medicine for pregnancy, menopause and phytoestrogen. Conclusions : As a result of discussing implications, NMDRs is a particular effect in low dose and heated debates surround this response, research is being conducted surrounding the field of obesity, problems in metabolism, growth hormone treatment, climacteric treatment, breast cancer, intake of Korean traditional medicine for pregnancy, menopause, and phytoestrogen. More research and interest in needed as it can have a massive influence in the public health and medicine.

  • To evaluate the estrogenic activities of di-ethyl hexyl phthalate (DEHP) and di-butyl phthalate (DBP), two phthalates known as endocrine disrupters, we used MCF-7 human breast cancer cell line. As results, DBP and DEHP had estrogenic effects. In brief, the concentration of maximal MCF-7 cell proliferation was $10^{-7}M\;and\;10^{-8}M$ for DEHP and DBP, respectively. The ratio of maximal cell yield of the test compounds to that of $17{\beta}-estradiol$ was 87.5% for DEHP and 73.4% for DBP. In summary, both DEHP and DBP had cell proliferation potencies in the MCF-7 cell. Potencies ranged from approximately 10 to 100 times less than 17beta-estradiol. DBP was stronger than DEHP in the concentration of maximal efficacy. However, DEHP was stronger than DBP in the MCF-7 cell proliferation. Results from this study suggested that DEHP and DBP may play an important role in the estrogenic activity. Therefore, it is suggested that DEHP and DBP are estrogenic.

Protective Effect of the Stem Bark of Syringa velutina on Bisphenol-A in the Human Breast Cancer Cell Line and Immature Rat (사람의 유방암 세포주와 미성숙 랫드에서 정향피의 비스페놀 A 독성방어 효과)

  • The inhibitory activity against bisphenol-A (BPA), one of well-known endocrine disrupters was examined with the water extracts prepared from the Stem Bark of Syringa velutina (SBS). In this study, we have investigated the effect of SBS on the toxicity caused by BPA in human breast cancer cell line, MCF-7 cells and immature Sprague-Dawley rats. In the estrogen receptor-mediated proliferation assay using MCF-7 cells, BPA (16 ng/ml) induced the cell proliferation, but the water extract of SBS inhibited BPA-induced cell proliferation in a dose-dependent manner. These results are associated with PARP degradation and specific cleavage of anti-apoptotic protein Bcl-2 of apoptotic regulatory factors. Additionally, the BPA (400 mg/100 g) significantly induced the increase of the uterine and virginal weights, while SBS (50 mg/100 g) showed the inhibitory action against BPA, i.e. caused the increase of estrogen-related organ weights in immature rat uterotrophic assay. Taken together, the present data suggest that SBS may have anti-toxicity activities against BPA in vitro and in vivo systems. SBS may be capable of inhibiting adverse effects of BPA such as reproductive disorder.

Effect of Phyto-Extract Fermented Mixture (MP119) on the Sexual Functions and on the Toxicities of Cadmium (식물추출복합발효물(MP119)이 성기능에 미치는 영향 및 카드뮴 독성에 대한 효과)

  • This study was aimed to investigate the effect of phyto-extract fermented mixture (MP119) on the male sexual functions. The MP119 was evaluated for anti-impotency and anti-hypertensive effects via ACE (angiotensin converting enzyme) or PDE (phosphodiesterase) inhibition assay. $IC_{50}$ values of MP119 against ACE and PDE were 241.3${\pm}$35.5 ppm and 372.2${\pm}$33.8 ppm, respectively. To investigate the effect of testosterone expression by MP119, we performed cell media test using mouse Leydig-derived TM3 cells. Production of testosterone in TM3 cell was increased by MP119. Also, NO (nitric oxide) production of HUVEC (human umbilical vein endothelial cell) was increased when MP119 was added to the cultures. Forty male ICR mice were divided into 4 groups. MP119 was orally intubated for 7 days to group 1 and 3, and same volume of vehicle to group 2 and 4 as controls. After that, group 3 and 4 were intraperitoneally injected cadmium chloride at a single dose of 2 mg/kg. On the 8th experimental day, weights of testis, epididymis and seminal vesicle, number of sperm, concentrations of serum testosterone and cGMP were determined. The number of sperm, the concentrations of testosterone and cGMP were significantly increased in two experimental groups (group 1, 3). These results suggest that MP119 enhanced the sexual function of male mice, and could protect the sexual organs from the cadmium chloride as one of the endocrine disrupters.

Effects of Nonylphenol and Diethylhexyl Phthalate on the Population Growth of Freshwater HABs, Microcystis and Stephanodiscus (내분비교란물질이 조류의 성장에 미치는 영향)

  • Effects of two endocrine disrupters (EDs) , nonylphenol (NP) and diethylhexyl phthalate (DEHP), on the population growth and morphology of two freshwater HABs (harmful algal blooms) , Microcystis aeruginosa and Stephanodiscus hantzschii, which frequently evoked the hazard bloom in an eutrophic lakes and reservoirs worldwide, were examined with seven different concentrations of EDs (0.01, 0.05, 0.1, 1,2, 2.5 and 3 ppm). Even at concentration below 0.01 ppm, NP strongly inhibited the algal growth of both M. aeruginosa and S. hantzschii, regardless of the algal growth phase. Morphologically, the algal cell treated with NP gradually lost green color in cytoplasm, became smaller in cell size, and then, was hardly seen in microscopic field. On the other hand, DEHP employed did not affect two algae at all concentrations, and rather stimulated the growth by about 10% with a treatment at 3.00 ppm compared to control. These results indicate that the continuous input of EDs, DEHP into the natural water system plays a crucial role to enhance or help an outbreak of algal bloom in eutrophic waters.

Effects of Perinatal Exposure to Phthalate/Adipate Esters on Sex Steroid Levels and Hypothalamic Gene Expression during Early Postnatal Periods in Rats

  • Our previous research has identified granulin (grn) and p130 genes as sex steroid-inducible genes in the rat hypothalamus, which might be involved in sexual differentiation of the brain. Phthalate esters that are used as plasticizers and also found at low levels in foods such as dairy products are often mentioned as suspected endocrine disrupters. The purpose of the present study is to elucidate whether perinatal exposure to di-n-butyl phthalate (DBP), diisononyl phthalate (DINP) and di-2-ethylhexyl adipate (DEHA) affects hypothalamic sex steroid-inducible genes. The present study assessed the effects of perinatal exposure to DBP, DINP and DEHA on sex steroid hormones levels and hypothalamic gm and p130 mRNA expressions at postnatal day (PND) 3 and 7. Pregnant rats were fed a soy-free diet containing 20, 200, 2,000 and 10,000 ppm of DBP, 40, 400, 4,000 and 20,000 ppm of DINP, or 480, 2,400 and 12,000 ppm of DEHA from gestational day (GD) 15 to GD 3 or 7. At PND 3 and 7, perinatal exposure to these chemicals did not substantially affect serum concentrations of testosterone and estradiol. At PND 3, the expression of grn mRNA levels in males was decreased by DEHA, and that of p130 was decreased by DBP, DINP and DEHA, though the effects were not dose-dependent. At PND 7, the expression of gm gene in female pups was increased by higher doses of DBP and all the doses, except for 4,000 ppm, of DINP, while that in male pups decreased by 480 and 12,000 ppm of DEHA. Hypothalamic expression of p130 mRNA in males was increased by lower doses of DBP and all the doses of DINP, whereas that of females was decreased by 480 and 2,400 ppm of DEHA. These results suggest that these chemicals may affect the expression of gm and p130 genes by directly acting on the hypothalamus, thus leading to inappropriate expression of these genes.

Effects of Plant-mineral Composites (PMC) on the Water Quality, Plankton Community and Microcystin-LR in Eutrophic Waters (식물-광물 혼합제가 부영양 수체의 수질, 플랑크톤 및 microcystin-LR에 미치는 영향)

  • We examined two reservoirs (Inkyung res. and Joongang res.) and two streams (Kyungan str. and Jecheon str.), all of which were eutrophic, during the 2010 warm season, to evaluate the water quality improvement activity (WQIA) of plant-mineral composite (PMC), which was previously developed to control suspended solids, including cyanobacterial bloom (Kim et al., 2010). We simultaneously measured both solid (S-MCLR) and dissolved microcystin-LR (D-MCLR), before and after PMC treatment, in the Joongang reservoir. Taking water body size and volume into account, we conducted the whole-scale experiment in the Inkyung reservoir, and mesocosm-scale experiments in the other three systems. The WQIAs of PMC were found to be comparatively high in SS (70~81%), TP (75~91%), BOD (65~91%), Chl-a (88~98%), phytoplankton (84~92%) and zooplankton (68~88%), except for the Kyungan stream, which was below 45% in all parameters. After PMC treatment, the concentrations of both SMCLR (47%) and D-MCLR (96%) decreased within two days, suggesting a mitigation possibility of hazardous chemicals such as agrochemicals and endocrine disrupters in the aquatic ecosystem. Our results collectively indicate that PMC is a useful agent to control suspended solids, including nuisance cyanobacterial bloom and their exudates, in an undisturbed water system with a long residence time.
