Choosing a culture medium for SCNT and iSCNT reconstructed embryos: from domestic to wildlife species

  • Over the past decades, in vitro culture media have been developed to successfully support IVF embryo growth in a variety of species. Advanced reproductive technologies, such as somatic cell nuclear transfer (SCNT), challenge us with a new type of embryo, with special nutritional requirements and altered physiology under in vitro conditions. Numerous studies have successfully reconstructed cloned embryos of domestic animals for biomedical research and livestock production. However, studies evaluating suitable culture conditions for SCNT embryos in wildlife species are scarce (for both intra- and interspecies SCNT). Most of the existing studies derive from previous IVF work done in conventional domestic species. Extrapolation to non-domestic species presents significant challenges since we lack information on reproductive processes and embryo development in most wildlife species. Given the challenges in adapting culture media and conditions from IVF to SCNT embryos, developmental competence of SCNT embryos remains low. This review summarizes research efforts to tailor culture media to SCNT embryos and explore the different outcomes in diverse species. It will also consider how these culture media protocols have been extrapolated to wildlife species, most particularly using SCNT as a cutting-edge technical resource to assist in the preservation of endangered species.

수질개선과 생태서식환경을 고려한 수변생태구역 너비 결정 방법 - 진위천 적용을 중심으로 - (Determining widths of riparian ecosystem zone for water quality and ecosystem conservation - A case study for the Jinwee stream)

  • Riparian management has become important as stream water quality as well as riparian ecosystem gain more public attentions. The objective of this study was to determine riparian widths based on the functions of nutrient removal and wildlife habitat protection and to apply for the Jinwee stream area as a preliminary case. Nitrogen and phosphorus filtration efficiencies were considered in water quality aspect, while the habitat radii of amphibian and reptiles were used for wildlife conservation purpose. In addition, observation of endangered species and human impact on wildlife disturbance in riparian area were also taken into account in determining riparian widths. The stream confluence zone was emphasized by doubling the riparian widths as the focal point for wildlife habitat conservation. As the results, three different levels of riparian widths were proposed depending on the major riparian functions and applied to the Jinwee stream section as the case study. The proposed method can be used to determine riparian width in other stream areas based on different functional focus, ie, water quality or riparian conservation purposes.

2011년 환경부 멸종위기종 등록절차 및 대상 멸종위기종 식물 목록 재고-과연 현재 국가 멸종위기종 관리가 최선의 방안인가? - (Critiques of 'The Endangered and Protected Wild Species List in Korea' Proposed by Korea Ministry of Environment and Listing Process - Is This the Best Process for the Current National Management of Endangered Wildlife and Plants in Korea? -)

  • 환경부는 2005년 야생동 식물법을 제정 공포후, 2011년 6월에 멸종위기야생동 식물 지정 해제 목록과 함께 지정관리 기준안을 발표하였다. 고등식물의 경우 64종중 멸종위기 I, II등급 유지 및 등급 변경종은 40종, 멸종 위기 해제종(안)은 19종, 해제예정종(안)은 5종, 신규지정종은 29종, 지정후보종은 3종으로 본 연구에서는 모두 96종을 IUCN의 적색목록의 범주와 평가기준 ver. 3.1에 의해 재평가를 함과 동시에, 환경부에서 제시한 멸종위기야생동식물 지정관리 기준 및 절차와 시스템의 문제점을 검토하였다. 변경된 환경부 목록의 40종의 평가 결과를 보면 남한을 경계로 한 평가로 인하여 전세계 수준의 평가인 IUCN 적색목록으로 등재될 개느삼, 미선나무, 한계령풀, 솜다리, 노랑무늬붓꽃, 히어리가 모두 환경부 목록에서 해제되었다. 추가 신규제정도 이와 유사하며 지정후보종인 섬국수 나무는 기존 연구에서 인가목조팝의 이명으로 보는 견해와, 비합법적으로 발표한 학명인 참골담초가 포함되어 문제의 소지가 있다. 환경부에서 제시하는 IUCN적색목록을 개선하였다는 내용은 IUCN 적색목록을 수정한 것이 아니라 평가 방법을 잘못 번역함과 동시에 적색목록의 범주를 단순히 참고하는 수준이다. 특히, 관련용어의 정의에 대한 오역과 이해를 못하고 번역해서 임의로 평가기준을 혼용하고 있어 평가 자체를 혼란과 왜곡으로 유도하는 결과물로 수용하기 어렵다. 환경부의 멸종위기종 목록을 개선하는 데에는 고유종에 대한 매우 좁은 종의 개념대신 주변 국가에서 넓은 개념으로 보는 분류학적 소견을 수용할 필요가 있으며, 남한이나 북한의 국토 개념의 평가가 아니라 최 소 한반도를 기준으로 전세계 수준의 평가를 할 필요가 있다.

Developing habitat suitability index for habitat evaluation of Nannophya koreana Bae (Odonata: Libellulidae)

  • Background: The Korean scarlet dwarf, Nannophya koreana Bae (Odonata: Libellulidae), is anendangered dragonfly with an increasing risk of extinction owing to rapid climate changes and human activities. To prevent extinction, the N. koreana population and their habitat should be protected. Therefore, suitable habitat evaluation is important to build the N. koreana restoration project. The habitat suitability index model (HSI) has been widely used for habitat evaluation in diverse organisms. Results: To build a suitable HSI model for N. koreana, 16 factors were examined by seven experienced researchers. A field survey for N. koreana observed sites and spatial analysis were conducted to improve the model. Five factors were finally selected by this procedure (crown density, open water surface, water depth, pioneer plant cover, and type of water source). Finally, the N. koreana HSI model was generated with the five adjusted factors based on interview, field survey, and spatial analysis. This model was validated by a current N. koreana habitat in 2021. With this model, 46 sites in Uljin-gun, Korea, were surveyed for N. koreana habitats; five sites were identified as core habitats and seven as potential core habitats. Conclusions: This model will serve as a strong foundation for the N. koreana restoration project and as a reference for future studies on N. koreana and other endangered insect populations. Further analysis and long-term data will improve the efficacy of this model and restore endangered wildlife.

Growth environment characteristics of the habitat of Epilobium hirsutum L., a class II endangered wildlife species

  • Background: As wildlife habitats are being destroyed and growth environments are changing, the survival of animals and plants is under threat. Epilobium hirsutum L., a species that inhabits wetlands, has held legally protected status since 2012. However, no specific measures are currently in place to protect its habitat, leading to a decline in remaining populations as a result of land use change and human activities. Results: The growth environment (including location, climate, land use, soil, and vegetation) of the five habitat sites (Samcheok, Taebaek1, Taebaek2, Cheongsong, Ulleung) of E. hirsutum L. was investigated and analyzed. These habitats were predominantly situated in flat areas with gentle south-facing slopes, at an average altitude of 452.7 m (8-726 m) above sea level in Gangwon-do and Gyeongsangbuk-do. The average annual temperature ranged 11.5℃ (9.2℃-12.9℃), whereas the average annual precipitation ranged 1,304.5 mm (1,062.7-1,590.7 mm). The surrounding land use status was mainly characterized by mountainous areas, and human interference, such as agricultural land and roads, was commonly found in proximity to these natural habitats. Soil physicochemical analysis revealed that the soil was predominantly sandy loam with a slightly high sand content. The average pH measured 7.64, indicating an alkaline environment, and electrical conductivity (EC) averaged 0.33 dS/m. Organic matter (OM) content averaged 66.44 g/kg, available phosphoric acid (P2O5) content averaged 115.73 mg/kg, and cation exchange capacity (CEC) averaged 23.43 cmolc/kg. The exchangeable cations ranged 0.09-0.43 cmol+/kg for potassium (K), 10.23-16.21 cmol+/kg for calcium (Ca), 0.67-4.94 cmol+/kg for magnesium (Mg), and 0.05-0.74 cmol+/kg for sodium (Na). The vegetation type was categorized as E. hirsutum community with high numbers of E. hirsutum L., Persicaria thunbergii (Siebold & Zucc.) H. Gross, Phragmites japonica Steud., Humulus japonicus (Siebold & Zucc.), and Bidens frondosa L.. An ecological flora analysis, including the proportion of lianas, naturalized plants, and annual herbaceous plants, revealed that the native habitat of E. hirsutum L. was ecologically unstable. Conclusions: Analysis of the habitat of E. hirsutum L., a class II endangered wildlife species, provided essential data for local conservation and restoration efforts.

  • This study aimed to develop the HSI (Habitat Suitability Index) model of Cypripedium guttatum. and to verify this model by applying to the candidate sites for replacement habitat. The development of HSI and SI (Suitability Index) model was conducted based on the existing literature, field surveys, and expert opinions for information on ecological habitat characteristics. Seven variables were selected as habitat variables including mean maximum temperature in Jul.-Aug., lighting, slope, altitude, effective soil depth, soil texture, and artificial overexploitation (i.e. protected areas). HSI model was developed for C. guttaum based on these variables. This HSI model showed high applicability to selection and evaluation of replacement habitats for C. guttaum. Our findings could provide the basic information on habitat assessment to prevent the extinction of endangered C. guttatum. However, since there is a limitation that the survey data were insufficient, further field surveys should be conducted on several habitat types to improve the accuracy of the HSI model.

들소(Bos javanicus)로 밝혀진 호랑이 기념품: DNA 염기서열 분석에 의한 종 확인 (Souvenir made of tiger part revealed to be Banteng (Bos javanicus): species identification using DNA sequence analysis)

  • Tigers are one of the most endangered species over the world and protected internationally or locally. However, they are still traded illegally for the Traditional East Asia Medicine or souvenirs / mementos as well as fake products. In the present study, to identify the species of the specimen known as the tiger's genitalia that was sold for mementos in China, two approaches of genetic tools and morphological comparisons were applied. On the basis of the entire sequences of mitochondrial cytochrome b gene (1,140 bp), the result of nucleotide comparisons showed that the specimen examined here is matched with banteng, Bos javanicus (Identities = 1,138/1,140; 99%). In addition, the examination of external morphological characters revealed that the genitalia of the specimen is much more similar to the one of cows, not to tigers. The outcomes highlighted that there are still some fake products of tigers traded in market and molecular genetic tools could help identifying species of wildlife products.

야생동물사고사 자료 수집 현황 및 문제점에 관한 연구 (A Study on the Present Condition of Roadkill Research Datum Collection and Problems)

  • As more highways are constructed and more cars are running on them, toll of roadkill steadily increases. To make the problem more serious, roadkill include endangered species. This study compared two sets of roadkill data of selected Korean highways. One set of data were produced, as a routine job of everyday, by the Korea Highway Corporation (KHC), and the other by a team of wildlife researchers for a limited period of time for research purposes. The aims of the comparison were to identify if the species of the animal, location of the roadkill, time, and other information were kept reasonably, especially in the forms provided to the patrols of KHC, to be used for research purposes later. Major findings and suggestions are; 1) KHC patrols are not wildlife professionals, which is understandable, and could not identify some animal species killed on the highways. As they also are not aware of the importance of the accurate data for the wildlife protection, education to promote their awareness is needed. 2) Current form used by the patrols is far from enough to produce good data for research. A new and detailed form needs to be designed and provided to the patrols so that they can keep all the necessary data on roadkills for monitoring and research purposes. If these issues are addressed, the data collected daily by KHC will become good inputs for roadkill researches in Korea.

Estimating Population Density of Leopard Cat (Prionailurus bengalensis) from Camera Traps in Maekdo Riparian Park, South Korea

  • Although camera traps have been widely used to understand the abundance of wildlife in recent decades, the effort has been restricted to small sub-set of wildlife which can mark-and-recapture. The Random Encounter Model shows an alternative approach to estimate the absolute abundance from camera trap detection rate for any animals without the need for individual recognition. Our study aims to examine the feasibility and validity of the Random Encounter Model for the density estimation of endangered leopard cats (Prionailurus bengalensis) in Maekdo riparian park, Busan, South Korea. According to the model, the estimated leopard cat density was $1.76km^{-2}$ (CI 95%, 0.74-3.49), which indicated 2.46 leopard cats in $1.4km^2$ of our study area. This estimate was not statistically different from the previous leopard cat population count ($2.33{\pm}0.58$) in the same area. As follows, our research demonstrated the application and usefulness of the Random Encounter Model in density estimation of unmarked wildlife which helps to manage and protect the target species with a better understanding of their status.

시·도보호 야생생물 조례 지정 현황 조사 및 분석 (A Study on Survey and Analysis of Designated Status of Wildlife Protected by City/Do Ordinance)

  • 본 연구는 야생동 식물 보전 및 관리를 위해 제정된 지자체 야생동식물 보호 조례의 지정 현황 등을 조사 분석하고 이를 통해 문제점 및 개선방향을 도출하기 위하여 이루어졌다. 이를 위해 17개 광역지방자치단체(특별시 광역시 특별자치시 도 특별자치도)의 야생생물보호 관련 조례와 각 조항의 내용을 비교 분석하였고, 각 지자체의 보호야생생물의 지정현황을 파악 후 지침 및 법률을 바탕으로 적정성을 파악하여 보았다. 시 도보호 야생생물관련 조례는 모든 광역지자체에서 제정되어, 일반적으로 선정기준, 보호대책, 행위제한 등의 조항이 공통적으로 반영되어 있었다. 또한 세부적으로는 깃대종, 복원 추진계획에 있는 생물을 선정기준으로 추가하거나, 서식지역에 대한 보호 및 복원에 대한 조항이 포함되어 있는 등 각 지자체의 특성에 따라 조항이 상이하였다. 시 도보호 야생생물 지정 및 보호 지침이 마련된 2006년 이후로 시 도보호 야생생물이 급격하게 증가하였으며 최근 1~2년 이내에 새로 지정한 광역지자체도 존재하였다. 일부 지자체의 경우 지정된 지 10년이 넘은 곳도 있었으며, 선정기준에 부합하지 않은 곳도 다수 있는 것으로 나타났다. 이러한 문제점을 해결하기 위해서는 시 도보호 야생생물의 목적에 맞는 종 선정을 위한 보다 과학적이고 객관적인 선정기준 및 방법이 구축되어야 할 것이라 판단된다.
