The tadpole larva of the ascidian Halocynthia roretzi has two sensory pigment cells in its brain vesicle. To elucidate the temporal requirement for FGF signaling in formation of the pigment cells, embryos were treated with an FGF receptor 1 inhibitor, SU5402, or an MEK inhibitor, U0126 during various embryonic stages. In the present study, it is shown that the embryos treated with SU5402 from the 16-cell stage to the early gastrula stage do not form pigment cells, whereas those treated after the early gastrula stage form pigment cells. In pigment cell formation, embryos suddenly exhibited the sensitivity to SU5402 only for 1 h at the neural plate stage(-4 h after the beginning of gastrulation). When U0126 treatment was carried out at various stages between the 8-cell and late neurula stages, the embryos scarcely formed pigment cells. Pigment cell formation occurred when the embryos were placed in U0126 at early tail bud stage. These results indicate that FGF signaling is involved in pigment cell formation at two separate processes during ascidian embryogenesis, whereas more prolonged period is required for MEK signaling.
Growth factors (GFs) produced by the embryo or by the maternal reproductive tract have been reported to regulate the embryonic development and differentiation. Among GFs, EGF as a mitogen plays a role in mitosis and functional differentiation of trophectoderm cells in mouse. The present study was carried out to investigate the effect of EGF on development of mouse embryos and to localize EGF in the mouse oocytes and embryos, which has been reported to be detected in the reproductive tract in mammals. To investigate the effect of EGF on the development of the embryo, mouse 2-cell embryos were cultured to blastocysts stage in Ham's F10 medium, treated with EGF(10-50 ng/ml) for 72 hrs. Immunocytochemistry was performed from oocyte to blastocyst stage with anti-EGF and anti-Mouse IgG, in order to determine the stage which EGF would be expressed in mouse. Exogenous EGF (more than 10 ng/ml) in the culture medium improved the developmental and hatching rates in the mouse embryos. As a result of immunocytochemistry, the embryonic EGF was expressed after the late 4-cell stage. EGF is thought to enhance preimplantation embryonic development and hatching. Exogenous EGF in the culture medium is thought to activate EGF receptor in the late 4-cell embryos and to enhance blastulation and hatching in mouse embryos. It is concluded that EGF enhances the developmental and hatching rates in the mouse embryos.
세포자기사(programmed cell death)는 배자발생과정에서 중요한 역할을 하는 정상적인 생리적현상으로 알려져왔다. 생쥐 배자에서 세포자기사에 관한 많은 연구가 있었지만 초기 형태발생기동안의 전반적이고 구체적인 세포자기사 양상에 관한 보고는 없었다. 이에 본 연구에서는 발생 4.5일$\sim$11.5일째의 Balb/c생쥐배자에서 생쥐발생초기의 세포자기사가 일어나는 양상을 알아보았다. 세포자기사가 일어나는 양상은 배자 조직절편과 온전한 배자에 in situ terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) 반응을 시켜 알아보았다. 주머니포배시기 (발생 4.5일째)에는 세포자기사 과정에 있는 세포가 극히 드물게 속세포명이에서 관찰되었다. Egg cylinder시기 초기(발생 $5.0\sim5.5$일째)에는 소수의 세포자기사 세포가 배자외배엽과 배자내배엽, 원시양막공간에서 나타났다. Egg cylinder시기 후반부(발생 $5.5\sim6.5$일)에는 세포자기사 세포가 배자외배엽, 배자내배엽, 원시양막공간 뿐만 아니라 바깥배자외배엽과 바깥배자내배엽 에서도 관찰되었다. Streak 시기 (발생 $6.75\sim7.75$일)에는 많은 세포자기사 세포가 ectoplacental cone부위에서 관찰된 반면에, 배자바깥 부위에서는 융모막과 바깥배자내배엽에서 매우 적은 수의 세포자기사 세포가 관찰되었고, 배자부위에서는 소수의 세포자기사 세포가 무작위적으로 배자외배엽과 중배엽, 그리고 배자내배엽 부위에서 관찰되었다. 체절형성기 초기 (발생 $8.0\sim8.5$일)에는 많은 수의 세포자기사 세포가 신경주름(neural fold)의 가장 앞쪽(머리쪽)에 주로 분포하는 것이 관찰되었다. 체절형성기 중기(발생 $9.0\sim9.5$일)에 귀기원판(otic placode)부위에서 처음으로 세포자기사 세포가 관찰되었고, 적은 수의 세포자기사 세포들이 또한 눈기원판(optic placode)과 아가미궁에서도 관찰되기 시작하였다. 발생 $9.5\sim9.75$일째에 크게 세가지 흐름의 세포자기사 세포들이 머리쪽 부위에서 관찰되었다. 발생 10.5일째에는 세포자기사 세포들이 발생중인 눈 부위와 안쪽코돌기와 바깥쪽코돌기 그리고 상악돌기가 만나는 부위, 아가미궁의 가쪽부위, 양쪽 하악돌기가 만나는 중앙부위, 그리고 팔다리싹의 꼭대기외배엽능선에 국한되어 관찰되었다. 발생 11.5일째에는 세포자기사 세포가 발생중인 팔다리부위와 꼬리부위를 제외한 나머지 부위에서는 현저히 감소되어 나타났다. 본 연구에서는 생쥐 발생초기 동안에 나타나는 세포자기사 세포가 발생시기에 따라 어느 부위에 발현되는 가는 알아보았는데, 이러한 연구 결과는 배자의 형태발생을 연구하는데 기초적인 자료로 활용될 수 있을 것으로 사료된다.

We determined the morphologic characteristics (body weight and degree of abdomen inflation) of the red spotted grouper, Epinephelus akaara, mother fish producing healthy eggs. Experimental fish were chosen from fish reared in a sea cage. The fish were divided into four size groups by body weight: 400~600, 600~800, 800~1,000, and 1,000~1,200 g and four stages (I~IV) of the degree of abdomen inflation. After hormone treatment, we observed the amount of ovulation-induced eggs, and rates of buoyancy, fertilization, embryonic survival, and hatching. As a result, mother fish with a body weight of 600 g or more spawned, and the fertilization rate, embryonic survival rate, and hatching rate were high in the 800~1,000 g range, thus showing effective ovulation induction. As a result of dividing the degree of abdomen inflation based on the anal fin of the mother fish into I-IV stages and determining hormone treatment time, the GSI was $0.9{\pm}0.2%$ at stage I, $2.3{\pm}0.2%$ at stage II, $5.6{\pm0.2%$ at stage III, and $7.9{\pm}0.9%$ at stage IV. The flotation rate and hatching rate were highest at stage III, and the fertilization rate and embryonic survival rate were highest at stage IV. Therefore, in terms of egg quality, the amount of eggs collected per mother fish, maturation, and histology were different depending on the degree of abdomen inflation. At stage III, where the abdomen inflation degree of the mother fish was based on the basal part of the dorsal fin relative to the height of the anal fin was 1, the egg quality was highest.
This study was undertaken to investigate the effects of ITS and EGF on embryonic development of in vitro matured and fertilized bovine oocytes in culture medium su, pp.ementing with or without calf serum. When fertilized oocytes were cultured in TCM-199 containing 0, 0.1, 0.5 and 1.0\ulcorner/ml ITS with 5% calf serum, the rates of development to blastocyst stage and the cell number of blastocysts were not significantly different among all treatments. And also monolayer of cumulus/granulosa cells prepared in containing calf serum and ITS were no beneficial effects of embryonic development. On the other hand, when EGF was su, pp.imented to TCM-199 containing calf serum or calf serum free, embryonic development rates(24.0 2.8% to 29.2 1.7% or 8% to 9%) and cell number of blastocysts(p<0.05) were significantly increased compared with EGF-free(22.1 2.1 or 1.0%, p<0.05). But when fertilized oocytes were cultured with cumulus/granurosa cells in TCM-199 containing EGF and calf serum, the rate of embryos development to the blastocyst stage and cell number of blastocysts were not significantly different compared with EGF-free and any concentrations. These results showed that ITS and EGF was not improved the development of bovine embryo in vitro matured and in vitro fertilized with calf serum and/or monolayer of cumulus/granulosa cells.

Parthenogenesis is maternally uniparental reproduction through the embryonic development of oocytes without fertilization. Artificial activation of mature oocytes could generate homozygous haploid embryos with the extrusion of the second polar body. However, the haploid embryos showed low embryo development in preimplantation embryos. In this study, we investigated whether the electronic fusion of the haploid embryos could enhance embryo development and ESC establishment in mice. Haploid embryos showed the developmental delay from 4-cell to the blastocyst stage. The haploid blastomeres of the 2-cell stage were fused electronically, resulting in that the fused embryos showed a significantly higher rate of blastocysts compared to non-fused haploid embryos (55% vs. 37%). Further, the embryonic stem cells (ESCs) derived from the fused embryos were confirmed to be diploid. The rate of ESC establishment in fused embryos was significantly higher compared to non-fused ones. Based on the results, we concluded that the electronic fusion of haploid embryos could be efficient to generate homozygous ESCs.
Beta-catenin is very important in early development including involvement in cell adhesion, cell signaling, and developmental fate specification. Cell-cell interaction is an important process during mammalian embryonic development. In preimplantation embryos, embryonic compaction is the process of increased cellular flattening and adhesion of junctional complexes and results in a polarized distribution. Beta-catenin is associated with embryonic compaction in mammals. Here, we examined the relationship between beta-catenin expression and compaction in porcine embryos derived from in vitro fertilization. First of all, we investigated beta-catenin expression in each embryonic developmental stage and also focused on expression pattern according to full, partial and non-compaction at morula stage. We used the immunocyto-chemical method in this research. To confirm compaction affects on the embryonic development, we compared between compaction and developmental rates to the blastocyst. The result showed that compaction and non-compaction rates were 14.6% and 63.8% at 4 days after IVF, respectively The developmental rates to the blastocyst and their total cell number were 50.9% vs 36.4% and 41.4$\pm$11.5 vs 26.8$\pm$12.7 in compaction and non-compaction groups. Although no difference was detected in the ratio of ICM to total cells between two groups, total cell number of the blastocysts in compaction group was superior to that of the blastocysts in non-compaction group (P<0.05). Expression of beta-catenin appeared in the boundary of membrane surface between blastomeres in 2- and 4-cell stage, and observed irregular pattern from 8-cell to blastocyst stage. We also investigated beta-catenin expression pattern according to the degree of compaction in the 3 groups; full, partial (>50%) and non-compaction. The expression signal in fully compacted embryos was stronger than those of partial and non-compacted embryos. Especially, beta-catenin expression appeared various patterns in morula stage suggesting the aberrant distribution of beta-catenin is affected by compaction patterns. Our results suggest that abnormal beta-catenin expression was affected by embryo quality and further development in porcine embryos in vitro.
We have launched an investigation for Embryonic Development, Larvae and Juvenile Morphology, of Buenos aires tetra in order to build basic data of Characidae and fish seeding production. We brought 50 couples of Characidae from Bizidduck aquarium in Yeosu-si, Jeollanamdo, from Korea on March of 2015. We put them in the tetragonal glass aquarium ($50{\times}50{\times}30cm$). Breeding water temperature was $27.5{\sim}28.5^{\circ}C$ (mean $28.0{\pm}0.05^{\circ}C$) and being maintained. The shape of fertilized egg was round shape, and it was adhesive demersal egg. The egg size was 0.63~0.91 mm (mean $0.74{\pm}0.07mm$, n=20). After getting fertilized egg, the developmental stage was gastrula stage, and embryo covered almost two-thirds of Yolk. Incubation was happened after 16 hours 13 minutes from gastrula stage, and the tail of juvenile came out first with tearing egg capsule. Immediately after the incubation, prelarvae had 3.78~3.88 mm length (mean $3.84{\pm}0.04mm$, n=5), and it had no mouth and anus yet. 34 days after hatching from the incubation, juvenile had 8.63~13.1 mm (mean $10.9{\pm}1.66mm$), and it had similar silver-colored body shape with its mother.
Embryos and fetuses are more sensitive to various environmental agents than adults of children biological effects following the exposure, such as intrauterin, malformation, have intimate conception with the prenatal exposure. There have been many studies on radiation and other agent. However, imformation about the ultrasound effects is limited. It is very important to study the effect of ultrasound with these kinds of fatera in consideration of ultrasound protection and safty. In this study, embryonic and fefal effects of ICR mouse embryos irradiated on 24, 48, 12 and 192 hpc of preimplantation and organogenesis period at the intensity of $0.5{\sim}3\;W/cm^2$ were investigated. Many type of external malformation observed in mouse irradiated on 72 hpc and 192 hpc. However, the embryos irradiated on 24 hpc and 48 hpc, at witch embryos had less then 6 cells and were pre-compaction stage, had no sensitivity for external malformation. The threshold doses of external malformation in mouse irradiated on 72 hpc and 192 hpc, at which embryos were consisted of $16{\sim}32$ cells and neural formation stage, were $1\;W/cm^2$ and $0.5\;W/cm^2$.

The purpose of this study was to evaluate some factors in the bovine embryonic development from one-cell to blastocyst using modified synthetic oviduct fluid medium (mSOFM), after maturation and in vitro fertilization of the oocytes. The embryonic development to the blastocyst stage was assessed at 7-10 days after in vitro fertilization, and the total cells in the blastocysts were counted by staining nuclei with fluorochrome. Some commercial calf sera (CS) and a superovulated cow serum had different effects on the embryonic development to the blastocyst stage (8.6-21.4%), dependent upon their product lots, although the development might not be affected at least by serum progesterone levels. ${\beta}$-Mercaptoethanol (${\beta}$-ME) supplemented into mSOFM was effective to the embryonic development (27.8%), as well as the co-culture system with cumulus cells (19.5%). In a serum- and feeder cell-free culture using mSOFM containing several growth factors and ${\beta}$-ME instead of CS plus co-cultured cumulus cells, bovine serum albumin (BSA, fraction V), but not polyvinyl alcohol (PVA), was highly effective in embryonic development to the blastocyst stage, almost comparable to CS in the serum-contained culture (CS, BSA and PVA; 27.8, 19.5 and 5.7%, respectively). However, fatty acid free BSA rather reduced the number of developed blastocysts, compared with fraction V BSA (7.3 vs 29.4%). In the serum- and feeder cell-free culture, supplement of glucose to the medium (final 2.0 mM) stimulated the cell proliferation of developing embryos 120 hr after in vitro fertilization. These results indicated that a serum-free medium supplemented with ${\beta}$-ME could successfully support the development of bovine one-cell embryos to the blastocyst stage. Moreover, supplement of glucose and fatty acids to the medium might support preferably the development and cell proliferation of embryos.
