Evaluation of endocrine disrupting effect of the herbicide Alachlor on Japanese Medaka using short term reproduction assay (단기번식독성시험법을 이용한 제초제 Alachlor의 송사리 내분비계 영향 조사)

  • Acute toxicity, water resolvability and short term reproduction test on Japanese medaka (Oriyzias latipes) for evaluating alachlor susceptibility to endocrine system were studied. Alachlor is known for suspected endocrine distruptors. As the results of tests, $LC_{50}$ (Median lethal concentration) was determined as 2.36 (1.994~2.805) mg/L, and test water replaced at 7 day intervals as its water resolvability was less than 20% in 7 days. The short term reproduction tests on Japanese medaka (Oriyzias latipes) were performed with a solvent control group, a treated group (alachlor concentrations of 0.02, 0.04, 0.11, 0.27, 0.68 ppm) and a positive control group (17 ${\beta}$ estradiol, 0.01, 0.1, 0.5 ppb). The number of spawning and embryo rates were declined in a alachlor-dose dependent manner, and the number of unfertilized eggs rates were in contrast increased depending on the concentrations. Further study should be needed to confirm whether the adverse effects may be effected by the concentrations. Additionally, alachlor was evaluated as a non-vitellogenin by the result of a test of significance of the vitellogenin content test for determination of the effect of estrogen among the endocrine disruptors.

A Study on the Antioxidative Effect of Orostachys Japonicus A. Berger Ethyl Acetate Fraction (와송 에틸아세테이트 분획물의 항산화 효능에 관한 연구)

  • In this study Orostachys japonica A. Berger used is a medicinal herb that has long been used as a folk remedy for cancer treatment. In this study, the antioxidant efficacy of the ethyl acetate fraction of Orostachys japonica A. Berger was confirmed. The results of the Orostachys japonica A. Berge ethyl acetate fraction of antioxidant activity assays showed Antioxidant effect of Orostachys japonica A. Berger EtOAc fraction extract at 0.10 mg/mL was showed a DPPH radical scavenging rate of 78.54% and ABTS+ radical scavenging rate of 73.48%. Also, the toxicity result of Orostachys japonica A. Berger EtOAc fraction extracts using alternative experimental animal model zebrafish, confirmed a 100% the survival of the zebrafish embryo was shown that there was no coagulation and no hatching delay at all concentrations. also ROS generation induced by UV-B irradiation was confirmed that the fluorescence intensity decreased as a whole in all larvae treated with Orostachys japonica A. Berger EtOAc fraction extracts. In particular, it was confirmed that ROS generation was effectively suppressed by showing a 35.7% reduction rate compared to the positive control at a concentration of 3 ㎍/mL. These results were confirmed that Orostachys japonica A. Berger EtOAc fraction extracts has the possibility of application in the cosmetics field as a natural antioxidant.

Effect of $Cd^{2+}$ on the Oocyte Maturation and Developmental Stages of Brown Frog Embryo, Rana dybowskii in vitro ($Cd^{2+}$이 북방산개구리의 난자성숙과 배아발달에 미치는 영향)

  • This study investigates the toxic effects of $Cd^{2+}$on frog (Rana dybowskii) by the determination of oocyte maturation and development of embryo exposure to different concentrations of the toxicant. The results show that $Cd^{2+}$ concentration of 0.1ppm suppressed the maturation of the oocytes. To examine the reversibility of the inhibitory effects, the oocytes were exposed to the $Cd^{2+}$ only for 3 hours, and then transferred to plain medium and cultured further for 17 hours. The oocytes were recovered from the toxic effect of the $Cd^{2+}$ when they were exposed to 1ppm, but not to 2.5ppm of the $Cd^{2+}$. The development of 2 cell embryos to 32 cell was completely suppressed at 0.1ppm and the longer the embryos were exposed to the $Cd^{2+}$, the more damage appeared to the embryos and the cytolysis of the 32 cell was induced by $Cd^{2+}$ at 0.1ppm. On the other hand, the embryos of blastula stage were cultured 96 hours in presence of the $Cd^{2+}$ at various concentrations and were examined. The rates of mortality and malformed larvae were investigated by probit analysis. From the results of LC$_{50}$ of 0.1ppm and EC$_{50}$ of 0.08ppm, Tl of 5.0 was derived, which indicates $Cd^{2+}$ is to be considered a teratogenic compound. Such specific malformations occurred in 14.3% as spine deformations at the 0.05ppm, in 75.0% as tail deformations at the 0.1ppm, in 66.7% as abdominal deformations at the 0.01ppm and in 26.0% as profound deformations at the 0.1ppm of $Cd^{2+}$ concentration which living embryos were exposed to. $Cd^{2+}$ suppressed growth to head-tail length at 0.1ppm. In conclusion, The study results reveal that $Cd^{2+}$ must be considered highly toxic effect to oocyte maturation and embryonic development.

Embryotoxicity and Teratogenicity of Excess Zinc on Xenopus laevis (과량의 아연에 의한 아프리카 발톱개구리 (Xenopus laevis)의 배발생 이상과 독성)

  • Concentrated releases of zinc into water usually results from discharges associated with industrial purpose. The released zinc into soil is corroded and released into water. In aquatic environment, exess zinc is toxic to the organisms and causes the growth inhibition and malformation of them as a heavy metal. In this study, excess zinc toxicity was tested by FETAX (frog embryo teratogenetic assay with Xenopus)as in vivo system. Xenopus embryos at st.9 were exposed to $100{\sim}900\;{\mu}M$ of zinc for 7 days and 81% of individuals were survived in 100 ${\mu}M$, and 25% were survived in 1000M of zinc solution. In external malformations, swelled belly and intestinal dysplasia were common, and all of tested individuals showed these malformations in 200 ${\mu}M$ or higher concentration of zinc. In 400 ${\mu}M$ or higher concentration, all of tested tadpoles showed faded heart. Also, hypo-pigmentation, lens hernia and loose digestive track were very frequently found in 100 ${\mu}M$ of zinc. The histological study with paraffin section of zinc treated tadpoles showed following abnormalities; regeneration of photoreceptor on retina, reduced vitreous chamber in eye, reduction of red blood cells in heart, abnormal liver, swelling of pronephric cell, muscle dysplasia and palatal papilloma. These abnormalities may be caused by the degeneration of mitochondria, inhibition of cell adhesion, and the formation of leghemoglobin by zinc due to the substitution of $Ca^{2+}$ by $Zn^{2+}$. The body length was reduced due to the excess zinc. From a statistical result, body lengths of 300 ${\mu}M$ or higher concentrative g개ups was significantly reduced comparing that of control group. Recently, many spontaneous malformations and reduction of amphibians are reported, From the results of present study, excess zinc mi호t be a factor of amphibian reduction, and the control of zinc discharges is very important.

Developmental Abnormality in Agricultural Region and Toxicity of the Fungicide Benomyl on Korea salamander, Hynobius leechii (한국산 도롱뇽(Hynobius leechii)의 농경지에서의 배 발생 이상과 살균제 Benomyl의 독성효과)

  • A numerical variation and abnormalities were studied on egg bags and embryos of Korean salamander, Hynobius leechii from agricultural habitat. The teratogenic and toxic effects of fungicide benomyl were also investigated with early embryos from non-agricultural habitat. We collected 144 egg bags from agricultural region, and 3418 of early embryos were contained. The lengths of egg bags were varied from 10 to 23 cm and the most frequent length was 19 cm. The number of embryos was varied from 7 to 43, and the most frequent range was 22 to 26. Spontaneous abnormalities were occurred in 406 embryos among 116 egg bags, and 24 kinds of external abnormalities were found. Individuals showing severe external defect were histologically studied and they showed optic dyspalsia, thyroid carcinoma, somatic muscular dysplasia, partial biaxial structure, decrease of red blood cells in the heart, cephalic degeneration and intestinal dysplasia. 385 embryos from non-agricultural region were exposed to 200 nM${\sim}$ 1 ${\mu}$M of benomyl at blastula or gastrula for 12 days. All embryo were dead in the concentration of 1 ${\mu}$M (LD$_{100}$) and 75% of embryos were dead in 800nM of benomyl. Speciflc effect due to benomyl was acrania or cephalic dysplasia and this restult suggests that the benomyl inhibit stongly to the development of neural tissue. These abnormal developments may be caused by antimitotic action, inhibition of tubulin complex, destruction of microtubule, inhibitions of neurulation and closing of neural fold, and by the inhibition of the movement of neural crest cells.

Toxic effects of antifouling agents (diuron and irgarol) on fertilization and normal embryogenesis rates in the sea urchin (Mesocentrotus nudus) (둥근성게(Mesocentrotus nudus)의 수정 및 배아 발생률에 미치는 신방오도료(Diuron, Irgarol)의 독성영향)

  • Toxic assessment of antifouling agents (diuron and irgarol) was conducted using the fertilization and the normal embryogenesis rates of the sea urchin, Mesocentrotus nudus. Bioassessment began with male and female reproductive cell induction. White or cream-colored male gametes(sperm) and yellow or orange-colored female gametes (eggs) were acquired and fully washed, separately. Then, the fertilization and normal embryogenesis rates were measured after 10 min and 48 h of exposure to the toxicants, respectively. The fertilization and embryo development rates were greater than 90% in the control, validating the suitability of both endpoints. The normal embryogenesis rates were significantly decreased with increasing concentrations of diuron and irgarol, but no changes in the fertilization rates were observed in concentrations ranging from 0 to 40 mg L-1. The EC50 values of diuron and irgarol for the normal embryogenesis rates were 20.07 mg L-1 and 22.45 mg L-1, respectively. The no observed effect concentrations (NOEC) were <1.25 mg L-1 and the lowest observed effect concentrations (LOEC) were 1.25 mg L-1 and 2.5 mg L-1, respectively. From these results, concentrations of diuron and irgarol over 1.25 mg L-1 and 2.5 mg L-1, respectively, can be considered to have toxic effects on invertebrates, including M. nudus. The ecotoxicological bioassay in this study using the noted fertilization and normal embryogenesis rates of M. nudus can be used as baseline data for the continued establishment of environmental quality standards for the effects of antifouling agents(especially diuron and irgarol) in a marine environment.
