A CYTOTOXIC ACTIVITY OF EXTRACT OF PANAX GINSENG ROOT AGAINST SOME CANCER CELLS IN VITRO AND IN VIVO

  • This study was devised to observe the cytotoxic activity of extracts of Panax ginseng root against some cancer cells and to purify the crude extract. Three kinds of cancer cells(leukemic cells L5178Y, HeLa cells and Sarcoma 180 cells) and mouse embryo cells (as normal cells) were used for this study. The ginseng roots were extracted with petroleum ether in soxhlet apparatus, and the crude extracts were purified by the silicic acid column chromatography and thin-layer chromatography methods. The results obtained are summarized as follows; 1. Eight to ten mg of the petroleum ether extract (crude extract) were obtained from 1 g of Panax ginseng root, and its activities per mg were about 1,000 units. 2. Doubling time of the L5178Y cells was increased to two fold by 24 hours incubation in culture medium containing about one ${\mu}g$ of extract per ml, and eight and ten folds higher concentration of ginseng extract were required for the Sarcoma 180 cells and HeLa cells, respectively, than for the leukemic cells(L5178Y) to inhibit the cellular growth to the same degree. 3. When the L5178Y cells were exposed to medium containing various concentration of the extract for 24 hours before initiation of the soft agar cloning procedure, about $99\%$ of the L5178Y cells were killed at concentration of 8 units per ml. 4. The growth rate of mouse embryo cell (as normal cell) was not affected by the culture with media containing various amounts (1.45 to 30.0 ${\mu}g/ml$) of the extract. 5. The crude extract could be purified about four times by silicic acid column chromatography using several solvent systems, and one spot of active compound could be obtained on the thin-layer chromatogram. 6. In the Swiss mice inoculated with Sarcoma 180 cells, a survival time of the experimental group (injection group of active compound) was extended more. 1.5 to 2.0 times than the control group's(no injection group).

아프리카발톱개구리의 시각계 발생에 관여하는 식물추출물 3종에 관한 연구 (Study of Effects of Crude Extracts of Three Plants Concerned on Optic Development of African Clawed Frog, Xenopus laevis)

  • 본 연구는 한약재로 널리 시용되는 원지 (Polygala tenuifolia), 구기자(Lycium chinensis), 산수유(Cornus officinalis) 추출물을 아프리카발톱개구리(Xenopus laevis)의 초기 배시기에 1, 10, $100{\mu}g/ml$의 농도로 처리하여 96시간 $24^{\circ}C{\pm}0.5^{\circ}C$에서 배양하였을 때 배 발생 중 나타나는 전반적인 기관 이상을 조사하였으며 특히 시각계의 발생이상에 초점을 맞추었다. 각 식물 추출물의 농도가 증가할수록 성장이 저해되고 시각계의 발생이상, 체축의 뒤틀림, 수종, 소화관의 꼬임이상, 체색이상 등의 기형의 발생률이 증가하였다. 시각계 이상이 가장 뚜렷이 나타난 군은 $100{\mu}g/ml$ 농도로 구기자 추출물을 처리한 것이었으며 각 실험군의 150개체 중 27% 정도가 시각계 발생이상을 나타내었다. 조직학적 연구에서는 뇌의 뇌실은 비정상적으로 확대되었고 눈에서는 유리체방의 형성부전이 나타났으며 망막세포층의 구분이 뚜렷하지 않았다.

Phosphorylation of a 66 kDa Protein, a Putative Protein Kinase C Substrate, is Related to Chondrogenesis of Chick Embryo Mesenchymes In Vitro

  • To understand the role of protein kinase C (PKC) in the regulation of chondrogenesis, we examined proteins which are phosphorylated by PKC. Stage 23/24 chick embryo wing mesenchymes were micromass-cultured to induce chondrogenesis and cell extracts were phosphorylated in a condition that activates PKC. Several proteins including 63 and 66 kDa proteins were phosphorylated. The 66 kDa protein was phosphorylated only in the presence of phorbol 12-myristate 13-acetate (PMA) and phosphatidylserine CPS), and the phosphorylation was almost completely diminished by bisindolylmaleimide, a PKC inhibitor. In addition, partially purified PKC increased the phosphorylation of the 66 kDa protein. Treatment of cultures with lysophosphatidylcholine (LPC) promoted chondrogenesis and phosphorylation of 66 kDa protein, while PMA and thymeleatoxin inhibited both of the two events. Our results suggest that the 66 kDa protein is a putative substrate of PKC, and phosphorylation of the 66 kDa protein, probably by $PKC\alpha$ is required for chondrogenesis.

다양한 부위별 연 추출물의 항혈전 활성 (In-vitro Anti-thrombosis Activities of Different Parts of Nelumbo nucifera Gaertner)

  • 연(Nelumbo nucifera Gaertner)으로부터 신규의 유용 생리활성을 확인하기 위해, 연잎, 연자방, 연자육, 연자심, 우절 및 연근으로부터 각각 ethanol 추출물을 제조하고, 현재까지 보고되지 않은 항혈전 활성을 평가하였다. 그 결과, 연근, 연잎 및 연자심 추출물은 항응고 활성이 인정되지 않았으나, 우절, 연자방 및 연자육 추출물에서는 강력한 TT, PT, aPTT 연장효과를 나타내었으며, aspirin 보다 강력한 항응고 활성을 나타내었다. 또한 혈소판 응집저해 활성 평가 결과, 우절, 연잎, 연자방, 연자육 추출물에서 응집저해가 나타났으며, 가장 강력한 응집저해는 연자방과 연자육에서 확인된 바, 이는 아스피린에 필적하는 강력한 활성이었다. 상기 활성 추출물은 1.0 mg/ml 농도까지 인간 적혈구 용혈활성이 없음을 확인하여, 우절, 연자방 및 연자육 추출물이 신규의 항혈전제로 사용 가능함을 제시하였다.

  • Biomphalaria glabrata와 Indoplanorbis exustus의 알에 대한 식물성 살패제 Brassaia actinophylla (Araliaceae)의 메탄올 추출물을 아치사 농도로 처리하였을 때 효과를 평가하였다. 그 결과 B. actinophylla를 10 mg/1 농도로 처리하였을 때, 두 종류의 패류의 배(embryo)에서 모두 변화를 보였다. Niclosamide를 0.004mg/1로 처리하였을 경우 역시 B. glabrata와 I. exustus의 배 발생을 억제하였다. 따라서 B. actinophylla는 주혈흡충증의 중간숙주는 패류 관리에 있어 세포치사제로 이용될 수 있을 것이다.

Saponin-SOD가 SNUC-4 Cell에 의해 유발편 대장암 Nude Mice의 암 성장 억제 효과 (Tumor-Suppressing Effects of Saponin-SOD in Nude Mice)

  • This study was performed to measure the antioxidant effects of red ginseng extracts which antioxidation had been promoted through enzyme hydrolysis. In order to observe their tumor-suppressing effects, an anti-cancer medicine and Saponin-SOD, which was a highly antioxidant beverage made from red ginseng saponin adding SOD-like rice (with embryo buds) extracts, were administered to nude mice with large intestine cancer induced. There was a significant increase in the content of phenolic compounds as the enzyme was added. The red ginseng extracts showed a high electron-donating ability with the passage of time. The electron-donating ability was particularly high in the enzyme-treated red ginseng extract, and also observed as high in Saponin-SOD. The lipid-peroxide generation was inhibited depending on the concentration of Saponin-SOD added; the addition of 0.625% Saponin-SOD served to decrease the inhibition level up to 65% compared with the case of no addition (100%). As a result, it could be assumed that Saponin-SOD would strongly inhibit the oxidation of ghost membrane. After the cancer was induced in nude mice through the injection of SNUC-4 cell, there was a significant inhibition in the growth of tumors in nude mice into which Saponin-SOD were injected; the growth of tumors was gradually decreasing with the passage of time after the cancer induction. In particular, when Saponin-SOD was administered together with an anti-cancer medicine, the synergic effect was observed. In conclusion, Saponin-SOD, when used with an anti-cancer medicine, is expected to reduce the amount of free radical and lipid peroxide, which are known to cause harmful effects occurring from the internal application of medicine.

Inhibition of Angiogenesis by Propolis

  • Propolis, obtained from honeybee hives, has been used in Oriental folk medicine as an anti-inflammatory, anti-carcinogenic, and immunomodulatory agent. There is considerable evidence suggesting that angiogenesis and chronic inflammation are codependent. Blockage of angiogenesis results in an anti-inflammatory effect. Ethanol (EEP) and ether extracts of propolis (REP), and caffeic acid phenethyl ester (CAPE), an active component of propolis, were examined for their anti-angiogenic activities using the chick embryo chorioallantoic membrane (CAM), and the calf pulmonary arterial endothelial (CPAE) cell proliferation, assays. The presence of EEP, REP and CAPE inhibited angiogenesis in the CAM assay and the proliferation of CPAE cells. The results suggest that anti-angiogenic activities of EEP, REP and CAPE are also responsible for their anti-inflammatory effect.

돼지 단위 발생 난자의 체외 발달에 있어서 피라칸타 추출액의 처리 효과 (Pyracantha Extract Acts as an Antioxidant Agent to Support Porcine Parthenogenetic Embryo Development In Vitro)

  • Pyracantha is a genus of thorny evergreen large shrubs in the family of Rosaceae, with common names Firethorn or Pyracantha. It's extract has also been used in cosmetics as a skin-whitening agent and functioning through tyrosinase inhibition. Recent studies have shown that pyracantha extract possesses antioxidant activities and may significantly improve lipoprotein metabolism in rats. Although the mode of action of Pyracantha extract is not fully understood, a strong relationship was observed between antioxidant and apoptosis in some types of cells. Thus, the aim of this study was to evaluated the effect of pyracantha extract on blastocysts formation and their quality of the porcine parthenogenetic embryos. After parthenogenetic activation by chemicals, presumptive porcine parthenogenetic embryos were cultured in PZM-3 medium supplemented with extracts of pyracantha leaf, stalk and root for 6 day (1, 5 and $10{\mu}g/ml$, respectively). In our results, the frequency of blastocyst formation in pyracantha root extract ($5{\mu}g/ml$) treated group had increased that of other groups. Furthermore, blastocysts derived from pyracantha root extract ($5{\mu}g/ml$) treated group had increased the total cell numbers and reduced apoptotic index. Blastocyst development was significantly improved in the pyracantha root extract ($5{\mu}g/ml$) treated group when compared with the $H_2O_2$ treated group (p<0.05). Subsequent evaluation of the intracellular levels of ROS in pyracantha root extract ($5{\mu}g/ml$) treated groups under $H_2O_2$ induced oxidative stress were decreased (p<0.05). In conclusion, our results indicate that treatment of pyracantha root extract may improve in vitro development of porcine parthenogenetic embryos through its antioxidative and antiapoptotic effects.

Native plants (Phellodendron amurense and Humulus japonicus) extracts act as antioxidants to support developmental competence of bovine blastocysts

  • Objective: Phellodendron amurense (P. amurense) and Humulus japonicus (H. japonicus) are closely involved in anti-oxidative response and increasing antioxidant enzymes activities. However, the effects of their extracts on development of preimplantation bovine embryos have not been investigated. Therefore, we investigated the effects of P. amurense and H. japonicus extracts on developmental competence and quality of preimplantation bovine embryos. Methods: After in vitro fertilization, bovine embryos were cultured for 7 days in Charles Rosenkrans amino acid medium supplemented with P. amurense ($0.01{\mu}g/mL$) and H. japonicus ($0.01{\mu}g/mL$). The effect of this supplementation during in vitro culture on development competence and antioxidant was investigated. Results: We observed that the blastocysts rate was significantly increased (p<0.05) in P. amurense ($28.9%{\pm}2.9%$), H. japonicus ($30.9%{\pm}1.5%$), and a mixture of P. amurense and H. japonicus ($34.8%{\pm}2.1%$) treated groups compared with the control group ($25.4%{\pm}1.6%$). We next confirmed that the intracellular levels of reactive oxygen species (ROS) were significantly decreased (p<0.01) in P. amurense and/or H. japonicus extract treated groups when compared with the control group. Our results also showed that expression of cleaved caspase-3 and apoptotic cells of blastocysts were significantly decreased (p<0.05) in bovine blastocysts derived from both P. amurense and H. japonicus extract treated embryos. Conclusion: These results suggest that proper treatment with P. amurense and H. japonicus extracts in the development of preimplantation bovine embryos improves the quality of blastocysts, which may be related to the reduction of ROS level and apoptosis.

四君子湯.四君子湯加黃기 煎湯液이 生肌作用에 미치는 影響 (Effects of Sa Gun Ja Tang(四君子湯) extract and Sa Gun Ja Tang added Radix Astragali(黃기) extract on the Granulation tissues)

  • This study was under taken to investigate the healing of Sa Gun Ja Tang extract (SE) and Sa Gun Ja Tang added Radix Astragali extract (SHE) on the artificial wound on rabbit skin. The granulation tissues were observed by microscope at the five day interval for twenty days. In order to understand the healing mechanism chick embryo culture was carried out in the presence of the extracts and the growth ratio and fusion index were counted. The results obtained were as follows: 1. The healing power of SE treated group on wound was more significant than the control group. 2. The healing power on wound was more effective at the SHE than the SE. 3. The growth ratio and fusion index were higher at the SHE treated groups than the SE treated groups. From the above result, SHE was more effective than the SE on wound healing activity by the mechanism of stimulating the cell growth ratio and fusion index.
