Studies on potato virus disease in Korea (한국산 감자바이러스병에 관하여)

  • The potato viruses, as possible potato virus X(PVX), potato virus Y(PVY), potato virus S(PVS) are isolated from potato tuber, which collected from eleven areas (Table 1) in Korea. These viruses are isolated by single lesion isolation, Aphid transmission and inoculating methods through the many species of the different plants. The PVX is identified by host range, symptoms, physical properties, serological reaction and electron micrography. The other two viruses are identified by the first two methods mentioned above. The results of the above experiments are as follows. The total value of these viruses infection is 81%. The value of PVX infection is higher than the other two viruses. The properties of PVX are marked as local lesions on Comphrena globosa. The dilution end point is $10^{-6}$, the thermal inactivation point is $70^{\circ}C$ and the size of virus particles is around 13 x 60 $m\mu$.

EFFECTS OF GAS PRESSURES ON GRANULAR STRUCTURE'S FOR MATION OF ALUMINUM FILMS PREPARED BY PVD PROCESS

  • In order to investigate the influence of gas pressure in PVD deposition conditions, aluminum films were prepared by vacuum evaporation and ion plating. The crystal orientation and morphology of the films affected by argon gas pressures were characterized by using X-ray diffraction (XRD) and scanning electron micrography (SEM) respectively. With the increasing of argon gas pressure, the preferred orientation of aluminum films exhibited (200) and the diffraction peaks of the films became less sharp and broadened. Film's morphology changed from columnar structure to granular structure with the increase of gas pressure. And the properties of these films on corrosion behaviors were estimated by measuring anodic polarization curves in deaerated 3% NaCl solution. The aluminum films which exhibited granular structure with (200) preferred orientation showed good corrosion resistance.

  • Incorporation of polar functional moieties into polyethylene (PE) film has been achieved by graft copolymerization of polar monomers such as methacrylic acid (MAAc) and acrylamide (AAm) on to PE film, preirradiated with ${\gamma}$-rays from $^{60}Co$ source, using benzoyl peroxide (BPO) as initiator in aqueous medium. Percentage of grafting of MAAc and AAm was determined as a function of irradiation dose, monomer and initiator concentration, temperature, reaction time and amount of water. Maximum percentage of grafting of MAAc (1453%) and AAm (21.28%) was obtained at [MAAc] = $235.3{\times}10^{-2}$ mol/L, [AAm] = $23.4{\times}10^{-2}$ mol/L, [BPO] = $5.5{\times}10^{-2}$ mol/L and $16.5{\times}10^{-2}$ mol/L at $80^{\circ}C$, $90^{\circ}C$ in 180 min and 90 min respectively. The grafted PE films were characterized by FTIR, Thermogravimetric analysis (TGA) Scanning Electron Micrography (SEM) and X-ray diffraction methods. Some selective properties of grafted films such as swelling behavior, ion and metal uptake have been carried out. The biodegradation studies of the grafted PE films have also been investigated. The grafted films developed superior swelling behavior with maximum swelling (480%) in water as compared to pristine PE (13.55%), better thermal stability and ion and metal uptake studies showed promising results that can be effectively used for desalination of brackish water and separation of metals from the industrial effluents.

A Study on the Photosynthesis of Chlorella Symbiotic with Paramecium bursaria (Paramecium bursaria와 공생하는 Chlorella의광합성에 관한 연구)

  • The relationship of Paramecium bursaria with its symbiiotic Chlorella was studied in the view-point of photosynthesis and its product movement. The following results were obtained. In 1/10 dilution of bacterized medium green Paramecium grew and survived better than colorless Paramecium. P. bursaria was competitively related to its symbiotic algae as far as the change of total chlorophyll contents was concerned. Therefore, in continuous darkness total chlorophyll contents of green Paramecia decreased proportionally to the time course. Under the condition, on the other hand, that the bacterized medium was not replenished over 10 days, total chlorophyll contents in P. bursaria increased by over 2 times as much as those in normal condition. By means of Oxygen Diometer it was assured that green Paramecium was able to utilize thoitic the great part of oxygen evolved photosynthetically by its symbiotic algae. Also when was given many starch grains were found in symbiotic chlorella through electron micrography. These data suggested that P. bursaria was photosynthetically given lots of benifit by its symbiotic chlorella.

Formation Mechanism and Corrosion-Resistance of Magnesium Film by Physical Vapour Deposition Process (물리증착법에 의해 제작한 마그네슘 박막의 형성기구와 내식특성)

  • Mg thin films were prepared on SPCC(cold-rolled steel) substrates by vasuum evapoaration and ion-plating. The influence of argon gas pressure and substrates bias voltage on the crystal orientation and morphology of the film was determined by using X-ray diffraction and scanning electron micrography (SEM), respectively. And the effect of crystal orientation and morphology of the Mg thin films on corrosion behavior was estimated by measuring the anodic polarization curves in deaerated 3% NaCl solution. The crystal orientation of the Mg films deposited at high argon gas pressure exhibited a (002) preferred orientation, regardless of the substrate bias voltage. Film morphology changed from a columnar to a granular structure with the increase of argon gas pressure. The morphology of the films depended not only on argon gas pressure but also bias voltage ; i.e., the effect of increasing bias voltage was similar to that of decreasing argon gas pressure. The influences of argon gas pressure and bias voltage were explained by applying the adsorption inhibitor theory and the sputter theory. And also, this showed that the corrosion resistance of the Mg thin films can be changed by controlling the crystal orientaton and morphology.

  • This study was conducted to examine mineralogical aspects on anthractite and bituminous coal-fired power-plant ashes as a source of mineral fertilizer. Fly ashes contain dominant amounts of silica and alumina and considerable quantitites of potassium and boron. However, potassium and silica present in unavailable forms for plant growth. X-ray, DTA, and IR analysis of ash particles indicated the formation of new mineral, mullite with shape of which were spherical in the surface morphologies of SEM. Detailed SEM investigation showed the presence of imbedded blocky shape silicate material.

The Formation Behavior and Thermal Stability of $E_6$ Eutectic Phase in Unidirectionally Solidified Al-Fe Alloys (일방향 응고시킨 Al-Fe계 합금에서 $E_6$ 공정상의 생성특성과 열적안정성)

  • Eutectic and off-eutectic Al-Fe alloys were unidirectionally solidified at the solidification rate of $1{\sim}50\;mm/min$ under the temperature gradients $75{\sim}80^{\circ}C/cm$. The investigation has been carried out for the microstructural variation, phase transition, mechanical properties by means of detailed analyses of stress-strain, micro-Vickers hardness and scanning electron micrography. The thermal stability at elevated temperature has been studied on $Al-Al_6Fe$ eutectic alloy held at $600^{\circ}C$ for $0{\sim}150$ hours. When the solidification rate was less than 10mm/min, the X-ray diffraction and EDS analysis showed the presence of $Al_3Fe$ compound. As the solidification rate more than 20 mm/min, $Al-Al_3Fe$ eutectic phase was transfered into $Al-Al_6Fe$ eutectic phase. The mechanical properties of unidirectionally solidified off-eutectic Al-Fe alloy is better than those of unidirectionally solidified eutecic Al-Fe alloy Maximum ultimate tensile strength was obtained in Al-2.25% Fe alloy which was unidirectionally solidified at the solidification rate of 20 mm/min. The metastable $Al-Al_6Fe$ phase was transferred into stable $Al-Al_3Fe$ phase at $600^{\circ}C$ held for 150 hours.

Fundamental Studies on Sinyewon Clays (新禮院粘土에 관한 基礎的 硏究)

  • Four kinds of clays from Sinyewon mine which is located in Chungchoungnam-Do district and two kinds of Japanese clays (Gairome, Kibushi) were selected and studied on the fundamental properties of these clays and those which are refined by the elutriation method. Chemical analysis, grading analysis such as refractoriness, specific gravity, color, fired color and plasticity, particle size distribution, various thermal tests, X-ray diffraction analysis and electron microscopic survey were carried out on these clays. The comparison of domestic clays with Japanese clays was made. 1. In chemical composition and physical properties, these four kinds of Sinyewon clays are almost similar to each other and also to Gairome and Kibushi clays. In refractoriness, Gairome and Kibushi clays have relatively high value such as SK 34 and 35, while Sinyewon clays have the value of about SK 32. Especially Sinyewon clay B has the low refractoriness (SK 18) and plasticity. Refined clays have the tendency of somewhat higher value than crude ones in refractoriness. 2. When burnt up to $900^{\circ}C$, the coefficients of contraction of Sinyewon clays were under 1.5%, Gairome under 1.3%, Kibushi under 3.0%. Only Sinyewon clay B showed expansion (1.0%) instead of contraction. 3. X-ray diffraction analysis and electron micrography showed that all of four Sinyewon clays and two Japanese clays are mainly composed of poorly crystallized kaolin, containing small amount of halloysite, sericite and considerable amount of quartz and feldspar. Sinyewon clay B contains especially large amount of quartz and feldspar. In general, it has been found out that the refined clays were free from large amount of iron oxide, quartz and feldspar.

  • Overlaid weld metals of austenitic stainless steel in a pressure vessel of power reactor are usually post-weld heated for a long period of time after welding. The PWHT is considered as a kind of sensitizing and it is important to check the soundness of the weld metal after PWHT, especially about the precipitation of carbides. The purpose of this report is to obtain information on the relation between the change of microstructure and Post-Weld Heat Treatment in the overlaid weld metals. Metallurgical aspects of the problem on austenitic stainless steel heated at $625^{\circ}C$, $670^{\circ}C$, $720^{\circ}C$ and $760^{\circ}C$ for 3, 10, 30, 100 and 300 hours have been investigated by means of optical-micrography, micro-hardness measurement, scanning electron microscope and electron-probe micro analysis. From the results obtained, the following conclusions are drawn; 1) The PWHT above $625^{\circ}C$ for a long time causes a diffusion of carbon atoms from low alloy steel into stainless steel, and consequently carbon is highly concentrated at the boundary layer of stainless steel. 2) C in ferritic steel migrated to austenitic steel and carbides precipitated in austenitic steel along fusion line. At higher temperatures, the ferrite grains coarsened in the decarburized zone. 3) In the change of microstructure of stainless steel overlaid weld metal, the width of carbides precipitated zone and decarburized zone increased with increase of PWHT temperature and time. 4) At about $625^{\circ}C$ to $760^{\circ}C$, chromium carbides, mainly $M_{23} C_6$, precipitate very closely in the carburized layer with remarkable hardening. 5) Precipitation of delta ferrite from molten weld metal depends on solidification phenomenon. There was a small of ferrite near the bond in which the local solidification time was short, comparing with after parts of weld metal. Shape and amount of ferrite were not changed by Post-Weld Heat Treatment after solidification.

  • Human periodontal ligament fibroblasts (hPDLF) are very important for curing the periodontal tissue because they can be differentiated into various cells. A tissue engineering approach using a cell-scaffold is essential for comprehending today's periodontal tissue regeneration procedure. This study examined the possibility of using an acellular dermal matrix as a scaffold for human periodontalligament fibroblast (hPDLF). The hPDLF was isolated from the middle third of the root of periodontally healthy teeth extracted for orthodontic reasons. The cells were cultured in a medium containing Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum at $37^{\circ}C$ in humidified air with 5% $CO_2$. The acellular dermal matrix(ADM) was provided by the US tissue banks(USA). Second passage cells were used in this study. The hPDLF cells were cultured with the acellular dermal matrix for 2 days, and the dermal matrix cultured by the hPDLF was transferred to a new petri dish and used as the experimental group. The control group was cultured without the acellular dermal matrix, The control and experimental cells were cultured for six weeks. The hPDLF cultured on the acellular dermal matrix was observed by Transmission Electron microscopy (TEM). Electron micrography shows that the hPDLF was proliferated on the acellular dermal matrix. This study suggests that the acellular dermal matrix can be used as a scaffold for hPDLF.
