Study on the Antioxidant and Human Neutrophil Elastase Inhibitory Activities of Mushroom Ramaria formosa Extracts (붉은싸리버섯 추출물의 항산화 및 Human Neutrophil Elastase 저해활성)

  • In searching for novel agents for skin anti-aging from natural resources, we found that the extract of the fruiting bodies of Ramaria formosa (R. formosa) had significant antioxidant and human neutrophil elastase (HNE) inhibitory activities. R. formosa extract exhibited a considerable DPPH radical scavenging activity with an antioxidant content of 117.0mg/mL (ascorbic acid equivalents) at the concentration of $500{\mu}g/mL$. The capacity of R. formosa extract to scavenge peroxy radicals measured by ORAC assay also showed dose-dependent antioxidant effect with $ORAC_{Roo}$ (trolox equivalents, $1{\mu}M$) values of 0.8, 5.2, and 7.8 at the concentrations of 1, 10, and $20{\mu}g/mL$. The cellular antioxidant capacity of R. formosa extract was investigated by assaying the cellular fluorescence intensity using dichlorodihydrofluorescein (DCF). The cellular oxidative stress induced by AAPH, $Cu^{2+}$ or $H_2O_2$ in HepG2 cells was significantly attenuated by more than 30% at $20{\mu}g/mL$ of R. formosa extract. HNE activity was reduced by treatment with R. formosa extract in a dose-dependent manner, and the $ED_{50}$ value for the ethanol extract of R. formosa was $42.9{\mu}g/mL$. R. formosa extract did not exhibited antimicrobial activity against four microorganisms including Bacillus subtilis (B. subtilis), Escherichia coli (E. coli), Candida albicans (C. albicans), Aspergillus oryzae (A. oryzae). Furthermore, the extract did not affect the inflammatory cytokine production of interleukin-10 and interferon-${\gamma}$ in NK92 cells. From the above results, we found that R. formosa extract has considerable antioxidant and elastase inhibitory effects, and does not stimulate immune cells. These findings suggest that R. formosa extract may be used as a bioactive component in cosmetic composition.

Beauty Food Activities of Isolated Phenolic Compounds from Ulmus pumila (유근피(Ulmus pumila)로부터 분리한 페놀성물질의 미용식품활성)

  • Phenolic compounds of $17.9{\pm}1.0mg/g$ were extracted from Ulmus pumila with 70% ethanol. The elastase inhibitory activity related with forming wrinkle was shown an excellent wrinkle improvement effect in 70% ethanol extracts as 55.5-69.5% at phenolic concentration of $50-200{\mu}g/mL$. The tyrosinase inhibitory activity related with skin-whitening was 24% in 70% ethanol extracts at phenolic concentration of $200{\mu}g/mL$. The astringent activity of 70% ethanol extracts was shown activity of 71% at phenolic concentration of $200{\mu}g/mL$ therefore it is judged that there is a high effect on pores reduction of the skin. The hyaluronidase inhibitory activity of U. pumila extracts was confirmed anti-inflammation effect of 80% at phenolic concentration of $50{\mu}g/mL$. Antimicrobial activity of U. pumila water extracts was shown each 8.7, 10.0, 11.1 and 11.8 mm clear zones on Propionebacterium acnes at phenolic concentration of $50-200{\mu}g/mL$. The stability of the multi-functional cosmetic (lotion) added U. pumila extracts was very stable for 28 days without changing of pH and viscosity also it's stable on temperature and sun lights. As the concentration of extracts was increased, the color of lotion was getting dark, but the sensory evaluation was high at score of 8.5.

Effect of Phellinus igniarius Quel Extract on the Anti-inflammatory, Anti allergy, Anti-oxidant, Anti-wrinkle (상황버섯 추출물이 항염증 및 항알레르기, 항산화, 주름제거에 미치는 실험적 연구)

  • Objectives : This study was carried out to investigate the effects of Phellinus igniarius Quel extract on the Anti-inflammatory, Anti allergy, Anti-oxidant and Anti-wrinkle. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH method. In the next experiment, to investigate anti-inflammatory test, the RAW 264.7 macrophage cells was cultured using DMEM including the 10% FBS. To study anti-allergic effect, we blended cultured Human Mast Cells(HMC-1), and then observe TNF-$\alpha$, IL-8 by ELISA Results : Phellinus igniarius Quel extract has the effects of anti-inflammation and anti-allergy, which may be due to its inhibitory potential on the macrophage activation. Furthermore, Phellinus igniarius Quel extract has the anti-wrinkle effects through the inhibitory potential on the collagnease, elastase and gelatinase activities. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

In vitro screening of anti-skin aging and antioxidant properties of aqueous/solvent extracts from distinctive stages of silkworm (Bombyx mori L.) pupae

  • Silkworm pupae, a by-product of the silk industry are known to be valuable resource of nutrients for humans as well as animals besides encompassing diverse bioactive constituents. However, there is a paucity of knowledge on their role in amelioration of oxidative stress and anti-skin aging properties. In the present study, we evaluated the inhibitory effect of aqueous as well as ethanolic (30% and 70%) extracts from distinct stages of male and female silkworm pupae belonging to two silkworm varieties on skin aging-related enzymes. The activities of collagenase, elastase and tyrosinase were effectively inhibited by 70% ethanolic silkworm pupal extracts (SPE), followed by 30% with aqueous extracts exhibiting meager inhibitory potential. SPE were also investigated for their antioxidant activity in oxidative-stressed murine fibroblasts (L929). The intracellular ROS and lipid peroxidation induced by tert-butyl hydroperoxide (t-BHP) in fibroblasts was better attenuated by pre treatment with ethanolic (30%) and aqueous extracts, respectively. The safety of the extracts was determined by studying their effect on fibroblast cell viability and it was found that none of the extracts were cytotoxic. Our findings indicate the potential utility of SPE as anti-aging components in cosmeceuticals.

Mass Production of Chaff-vinegar and Its Effect of Anti-Aging and Whitening (왕겨초액의 대량생산과 항노화 및 미백 효과)

  • Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.

Enzymological Evaluation of Oral Inflammation inhibitory activity by Aloe vera peel extract (Aloe vera peel 추출물에 의한 구강염증 저해 효과의 효소학적 평가)

  • To evaluate antiinflammation of Aloe vera peel, antiimflammation substances were extracted from Aloe vera peel and identified, and we investigated the effect of the its substance the inhibitory effect on the activity of hyaluoronidase, elastase, collagenase and prostaglandin endoperoxide synthase. The water extract from Aloe vera peel were successfully purified with solvent fractionation, silica gel column chromatography, preparative thin layer chromatography and UV spectrometer. Two purified active substances were identified as aloe-emodin and barbaloin by Mass Spectrometer, $^1H-NMR$ and FT-IR. Aloe-emodin and barbaloin. $IC_{50}$ values of aloe-emodin and barbaloin against hyaluronidase activity were 40 and $70\;{\mu}g/mL$, respectively. Leuckocyte elastase, which is related to the destruction of various tissue, $IC_{50}$ values of them were 50 and $60\;{\mu}g/mL$, respectively. $IC_{50}$ values of aloe-emodin and barbaloin against collagenase activity were 40 and $60\;{\mu}g/mL$, respectively. and $IC_{50}$ values of aloe-emodin and barbaloin aganist the prostaglandin endoperoxide synthase, which play an important role in inflammatory reactions, were 40 and $70\;{\mu}g/mL$, respectively. Inhibitory effects of aloe-emodin, barbaloin and aspirin against carrageenan paw edema were 74.9, 52.9 and 51.9% as inhibiton percentage, respectively, at dose of 100 mg/kg and that of indomethancin was 49.7 at dose of 10 mg/kg. Cell cytotoxicity of barbaloin against human gingival cells was lower than that of aloe-emodin. Aloe-emodin and barbaloin did not show cytotoxicity against human gingival cells at concentration of 1.0 and $5.0\;{\mu}g/mL$, However, aloe-emodin and barbaloin showed less cytotoxicity than chlorhexidine, which usually have been used as the agent of anticaries and antiinflammation. These results suggested that aloe-emodin and barbaloin from Aloe vera peel have the effect of anticaries and antiinflammation.

Antioxidative Activity and Antiaging Effects of Tetrapanax papyriferum extract (통초추출물의 항산화 활성 및 항노화효과)

  • In this study, we evaluated the anti-oxidative, whitening and anti-wrinkle effect of Tetrapanax papyriferum extract. Tetrapanax papyriferum was extracted by two different solvents which were n-Hexane, ethyl acetate. The anti-oxidant activity was measured by free radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl radical). And the inhibitory activities of tyrosinase for whitening effect and collagenase, elastase for anti-wrinkle were investigated. For anti-oxidant activity, ethyl acetate fraction of Tetrapanax papyriferum extract showed more significant activity than n-Hexane fraction of Tetrapanax papyriferum extract. For whitening activity, n-Hexane fraction of Tetrapanax papyriferum extract exhibited strong inhibition effects compared with reference. Therefore, Tetrapanax papyriferum extract may be useful as a new antioxidant and whitening agent.

Biological activities of Rosa multiflora Ethanol Extract as Cosmetic Material

  • The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity as a functional ingredient for cosmetic products have not yet been studied. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. The antioxidant activities were determined by DPPH. Additionally, the contents of total phenols and flavonoids were analyzed. Also, the phenolic compounds were detected using HPLC. The melanogenesis regulatory effect was evaluated using melanin content and cellular tyrosinase activity in B16F10 melanoma cells. The elastase inhibitory activity assay was performed for anti-wrinkle effect. The antimicrobial activity was assessed using the disc diffusion assay. The DPPH radical scavenging ability, denoted by the $SC_{50}$ value was found to be $123.1{\mu}g/ml$, whereas that of positive control (ascorbic acid) was $27.5{\mu}g/mL$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In addition, astragalin and gallic acid were identified in the extract. Also, the ethanol extract significantly inhibited ${\alpha}$-MSH-induced melanogenesis in B16F10 cells. For anti-wrinkle effect, elastase inhibition activity of the ethanol extract was 53.2% at a concentration of $100{\mu}g/ml$. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 0.5 - 5%, and no significant activity was noted against C. albicans. Therefore, the ethanol extract of Rosa multiflora can be used effectively for development of functional cosmetic materials.

Effects of Dendrobii herba and Punica granatum Extract on the Anti-oxidant, Anti-inflammatory, Anti-wrinkle and Whitening (석곡(石斛), 석류(石榴)의 항산화, 항염증, 주름, 미백에 미치는 영향)

  • Objective : The aim of this study is to determine the effects of Dendrobii herba extract and Punica granatum extract on skin disease and skin beauty. Methods : To investigate in vitro anti-oxidant activity assay, ethanol extracts of medicinal plants tested by DPPH radical, xanthine oxidase activity. In the next experiment, to investigate anti-inflammatory activity assay, examined by relations in NO synthesis, IL-$1{\beta}$, IL-6, TNF-${\alpha}$, NF-${\kappa}B$, COX-2, MAP kinase. To study Skin wrinkle formation effect, we were examined by tyrosinase activities, melanin synthesis in MNT-1 cell. Results : 1. In an anti-oxidant test, Dendrobii and Punica granatum extract showed high radical scavenging activity. 2. In an anti-inflammatory test, Dendrobii herba and Punica granatum extract weakly inhibited the lipopolysaccharide(LPS)-induced nitric oxide(NO) release from RAW 246.7 macrophage cells. Dendrobii herba and Punica granatum extract also inhibited LPS-induced IL-$1{\beta}$ and COX-2 expressions. The inhibitory effect of Dendrobii herba and Punica granatum extract on macrophage activation were via the inhibition of NF-${\kappa}B$, evidenced by transient transfection assay. however, Dendrobii herba and Punica granatum extract did not have any effects about activation of Jun-N-terminal kinase(JNK) and inhibition of p38 MAP kinase in RAW 264.7 cells. 3. In the skin wrinkle formation assay, Dendrobii herba and Punica granatum extract weakly inhibited collagenase and elastase, however it was not statistically significant. 4. In the skin whitening assay, Dendrobii herba and Punica granatum extract weakly inhibited tyrosinase activity, however, it was not statistically significant. They did not have any effect on melanin synthesis, indicating that they could not be applicable for skin whitening. Conclusion : Dendrobii herba extract and Punica granatum extract may play a significant role in skin disease and skin beauty.

Antioxidative and Biological Activity of Hot Water and Ethanol Extracts from Phellinus linteus (상황버섯 열수 및 에탄올 추출물의 항산화 효과 및 생리활성)

  • To investigate the applicability of hot water extract (PLW) and ethanol extract (PLE) from Phellinus linteus as functional food and cosmeceutical materials, its total flavonoids content, total phenolics content, electron donating ability (EDA), nitrite-scavenging ability (NSA), SOD-like activity, inhibitory effect of tyrosinase and elastase were examined. Total flavonoids contents of PLW and PLE were 17.31 mg/g and 42.61 mg/g, respectively, and total phenolics contents were estimated as 149.92 mg/g for PLW and 432.42 mg/g for PLE. The EDA of PLW and PLE were $6.49{\sim}92.98%$ and $22.61{\sim}94.28%$. The EDA and total phenolics contents had a high correlation of 0.83. The NSA was pH dependent, and was highest at pH 1.2 and lowest at pH 6.0. The NSA of PLE was higher than that of PLW. The SOD-like activities of PLW and PLE were $14.36{\sim}35.21%$ and $17.27{\sim}81.84%$, respectively, and the activity was dependent on the sample concentration. The tyrosinase inhibitory activity was the highest in PLE ($10.51{\sim}80.93%$) while that of PLW was $4.77{\sim}43.69%$. Finally, the elastase inhibitory activity was $10.01{\sim}76.02%$ at PLE. Based on the above results, we deemed that the ethanol extract of Phellinus linteus was the most pertinent for use as functional food and cosmeceutical materials.
