카드뮴 내성 효모, Hansenula anomala B-7 세포에 축적된 카드뮴의 세포내 분포와 단백질 합성에 미치는 카드뮴의 영향 등에 대하여 연구하였다. 세포내 축적된 카드뮴의 84.9는 cytosol 등인 가용성 분획에 존재했다. 세포내 단백질 함량은 카드뮴(1,000$\mu g$0/ml)에 의하여 감소되었으나, 유안(30-75 포화)에 의하여 침전되는 가용성 단백질의 함량은 카드뮴에 의하여 증가되었다. 더욱이 카드뮴(1,00$\mu g$0/ml)의 첨가배지에서 배양된 세포에는 고분자 가용성 단백질이 카드뮴 무첨가 배지에서 배양된 세포에서 보다 증가되었으나, 저분자 단백질은 감소되었다. 이상의 결과로 단백질의 합성은 카드뮴에 의하여 저해되나, 유안(30-75 포화)에 의하여 침전되는 고분자 단백질의 합성은 카드뮴에 의하여 촉진된다.
This study was performed to investigate the effect of dietary protein and calcium levels on cadmium detoxication in rats. Seventy male Sprague-Dawley rats weighing 208 $\pm$ 19 g were blocked into 10 groups of 7 animals according to body weight. Five groups were fed 15% protein-0.6% calcium diet with 100ppm cadmium in drinking water for first 15days and the other 5groups fed same diet without cadmium in drinking water for same period and served as controls. After this 15-day intoxication period, each one of cadmium intoxication and control groups were fed each of 4 kinds of detoxifying diets different with protein(40%, 15%) and calcium(1.3%, 0.6%) levels without cadmimum in drinking water for following 15 days of detoxifying period. Results were summarized as follows: 1) Food intake, body weight gain, F.E.R. and weights of liver, kidney and femur were increased by detoxifying diets and high protein diet was most effective in weight gains of liver and kidney. 2) When cadmium and metallothionein contents of initial intoxication group and those of all detoxication groups were compared, cadmium and metallothionein contents in the liver were not changed, but those in kidney increased, and those in intestine decreased markedly. 3) Only dietary protein level affected cadmium and metallothionein distribution among organs, and cadmium contents of whole blood, liver, kidney and femur were lower in high protein diet, but metallothionein contents in liver and kidney were higher in high protein diet. 4) Gel filtration chromatogram showed that most of cadmium in the cytosol was bound to metallothionein fractions in high protein-high calcium group. Results obtained indicated that high protein diet was effective in cadmium detoxication by increasing the induction of metallothionein synthesis. But high calcium diet did not play a role in cadmium detoxication.
The changes of growth and protein synthesis pattern by aluminum (Al), cadmium (Cd), zinc (Zn) treatments were studied in Cyanobacterium synechocystis sp. PCC 6803. When exposed to Al from 5ppm to 3oppm, synechocystis grows normally. But more than that retard the growth of algae notably. The 0.05ppm Cd additions had no effect on the growth of algae. 0.1, 0.2, and 0.5ppm Cd inhibited growth. Under 1 and 2ppm Cd stress, growth was greatly diminished. Zn had dual effects. The growth of algae in media containing 5ppm was stimulated. As concentration increases more than l5ppm, growth inbition increases. Under 25ppm Zn stress, growth was greatly diminished. According to logistic theory, r and K values of each heavy metal-treated groups were estimated. Correlation analysis of r and K values with metal concentration shows that there is negative correlation between K and concentration in Cd and Zn treatments. Critical concentration which shows lethal or sublethal effect was estimated by t-test of each r and K value. The cells cultured in 10, 20, 30, 40 and 5oppm of Al, 1 and 2ppm of Cd, and 10, 15, 20, 25 and 30ppm of Zn for 4 days was used for protein analysis. Analysis of protein synthesis with SDS-PACE showed alterations of protein synthesis pattern. The synthesis of protein about 220kD increased markedly. In this study, it showed that resistance mechanism against Al, Cd, and Zn is K selection and that metal stress induced the change of protein synthesis in Cyanobacterium synechocystis sp. Garlic occupies a special position among the many foods of vegetable origin because it is the sole food for Koreans during the their lives. And vitamin A has been ingested by forms of food or additives. Cadmium has been described as one of the most dangerous trace elements in the food and environment of man and livestocks. Since the de novo synthesis of stress proteins can be detected early after exposure to some agents, analysis of cadmium-induced changes in gene expression , ie. alterations in patterns of protein synthesis, may be useful to develop as biomarkers of exposure and damage for food hygiene. He acute and chronic combine effects of cadmium (Cd, CdCl2 20mg/kg), garlic oil(Dds: diallyl disulfide 50mg/kg, 3 times a week) and vitamin A(Ra: retinol acetate 50,000 IU/kg, 3 times a week) on Wistar male rats were evaluated concerning cadmium contents, tissues enzyme activity, HSP expression histopathological and electron microscopical examinations. The results of the study are as follows ; 1. Less cadmium was absorbed through the digestive tracts, but the ratio of contents in tissue were not changed by the simultaneous adminstration of diallyl disufide or retinol acetate. 2. ALT(alanine aminotransferase) , AST(aspartate aminotransferase), glucose, BUN (blood urea nitrogen), creatinine, the key indices of the clinical changes in hepatic and renal function were significantly hanged by the cadmium treatment after 1 week in liver, after 4 weeks in kidney. 3. Histopathological changes in cadmium treated rats were appeared at 8 weeks age treatment in kidneys. Homogenous eosinophilic material was accumulated in cortical and collecting tubular lumens at 16 weeks. Degenerated or necrotized tubular cells were observed in cortex and medulla. Degenerated seminiferous tubules and homogeneous eosinophilic material was seen in interstitial tissue of rat treated with cadmium for 16 weeks. Calcium deposits were seen in degenerated seminiferous tubules and the tubules showed severe calcification of rat treated with cadmium for 16 weeks. Electron microscope changes in kidney were observed in rats treated with CdCl2 20 mg/kg. Proximal convoluted tubule cells showed selling of cytoplasm and narrow lumen. Capillary endothelial cells showed cytoplasmic vacuoles and swelling. Degenerated epithelial cells were accumulated in tubular lumen of kidney. 4. Enhanced synthesis of 70 KDa relateve molecular mass proteins were detected in 2 hours after cadmium, exposure, with maximum activity occurring at 8~48 hours. Induction of HSP 70 was evident at proximal tubules and glomeruli in kidney. Testicular cells produced enough HSP to be detected normally. From the above results, it could be concluded that HSP70 induction by the cadmium treatment was a rapid reaction to indicated the exposure of xenobiotics, and retinol acetate reduced the cadmium induced nephrotoxicity.
The uptake of cadmium in animals is mainly accumulated in and affected to the liver and kidney by binding with red blood cells and serum albumin. The process accounts for more than 50% of the total accumulated cadmium in the body. The kidneys may be damaged without regarding the pathway uptake of cadmium. In a group of rats on supplements of 1% chlorella and 40 ppm cadmium, the concentration of cadmium in urine greatly decreased by 66% compared to control group, and the total synthesis of metallothionein decreased by 48.6% compared to control group. However, no previous study has assessed the protective effect on kidney damage induced by cadmium uptake through supplementation with chlorella. This study analyzed the biochemical marker for kidney damage in the rats after uptake of 40 ppm $CdCl_2$ and supplementation of the diet of Sprague Dawley (SD) rats with 1%, 5%, and 10% chlorella during 4 weeks. In a group of SD rats on supplementation with 1% chlorella and uptake of 40 ppm $CdCl_2,\;\beta_2$ microglobulin in the urine was found to be $3.1\pm0.6\;{\mu}g/L$, a decrease of 58% compared to a group of Sp rats on uptake of $CdCl_2$ only, in which the $\beta_2$ microglobulin was found to be $4.9\pm0.7\;{\mu}g/L$. According to the results of histopathological observation, the accumulation of mild and localized chronic inflammatory cells in kidney tissues was observed in 50% of the SD rats on uptake of cadmium only. In contrast, only 30% of the SD rats on supplementation with 1% chlorella and uptake of 40ppm $CdCl_2$, representing a histopathological abnormality, and there were no histopathological abnormalities at all in groups of SD rats on supplementation with 5% or 10% chlorella and uptake of 40 ppm $CdCl_2$. In conclusion, protein, calcium, and iron, which account for more than 50% of the total dried chlorella composition, may contribute to the reduction nephrotoxicity by stimulating both inhibited absorption of cadium and increased excretion of accumulated cadmium in kidneys.

Cadmium (Cd) is of particular concern because of its widespread occurrence and high toxicity and may cause serious morpho-physiological and molecular abnormalities in in plants. The present study was performed to explore Cd-induced morpho-physiological alterations and their potentiality associated mechanisms in Sorghum bicolor leaves at the protein level. Ten-day-old sorghum seedlings were exposed to different concentrations (0, 100, and $150{\mu}M$) of $CdCl_2$, and different morpho-physiological responses were recorded. The effects of Cd exposure on protein expression patterns in S. bicolor were investigated using two-dimensional gel electrophoresis (2-DE) in samples derived from the leaves of both control and Cd-treated seedlings. The observed morphological changes revealed that the plants treated with Cd displayed dramatically altered shoot lengths, fresh weights, and relative water content. In addition, the concentration of Cd was markedly increased by treatment with Cd, and the amount of Cd taken up by the shoots was significantly and directly correlated with the applied level of Cd. Using the 2-DE method, a total of 33 differentially expressed protein spots were analyzed using MALDI-TOF/TOF MS. Of these, treatment with Cd resulted in significant increases in 15 proteins and decreases in 18 proteins. Significant changes were absorbed in the levels of proteins known to be involved in carbohydrate metabolism, transcriptional regulation, translation and stress responses. Proteomic results revealed that Cd stress had an inhibitory effect on carbon fixation, ATP production and the regulation of protein synthesis. In addition, the up-regulation of glutathione S-transferase and cytochrome P450 may play a significant role in Cd-related toxicity and stress responses. Our study provides insights into the integrated molecular mechanisms involved in response to Cd and the effects of Cd on the growth and physiological characteristics of sorghum seedlings. The upregulation of these stress-related genes may be candidates for further research and use in genetic manipulation of sorghum tolerance to Cd stress.
담배 유식물의 생장, 엽록소 및 광합성 효소에 대한 카드뮴과 질산염의 영향에 대해 연구하였다. 카드뮴에 의한 억제된 생장은 질산염에 의해 회복되지 않았다. 엽록소 a와 b의 함량은 카드뮴에 의해 감소하였으며, 카드뮴과 질산염 혼합처리구는 카드뮴 처리구보다 엽록소 함량이 감소하였다. 카드윰 처리구에서의 rubisco의 활성과 함량은 대조구에서의 함량보다 현저히 작았다. 이는 rubisco의 활성이 함량과 연관되어 있으며, rubisco의 활성화와 합성이 카드뮴에 의해 저해된다는 것을 의미한다. 카드뮴에 의해 감소된 rubisco의 활성과 함량은 질산염에 의해 더욱 더 감소되었다. 이와 비슷한 변화가 rubisco activase의 활성과 함량에서도 관찰되었다. 이 결과들은 카드뮴과 질산염에 의해 유도되는 rubisco의 변화가 rubisco activase와 서로 연관되어 있음을 의미하며, 질산염은 직접적으로 rubisco의 활성화와 합성에 관여 할 뿐만 아니라 rubisco activase를 통해서도 rubisco의 수준에 관여하는 것으로 해석된다.
3주간 생장한 닭의장풀을 Hoagland용액 $(\pm{100\mu{M}\;Cd^{2+}$ 또는 $100\mu{M}\;Cd^{2+}+100\mu{M\; SA)$에서 4일 동안 수경 재배한 후 $Cd^{2+}$ 및 살리실릭산(SA) 처리시, 닭의장풀의 뿌리와 분리 표피에서의 $Cd^{2+}$ 농도와 nonprotein-SH 합성을 측정하였다. $Cd^{2+}$ 처리구에서는 $98\mu{M}\;Cd^{2+}/g.fr.wt$이 검출되었다. 이는 대조구에 비해 약 6배 증가한 것이다. $Cd^{2+}$을 SA와 같이 처리한 경우$Cd^{2+}$만 처리한 경우보다 뿌리에서 약 1.2배의 $Cd^{2+}$이 증가하였다. $Cd^{2+}$처리는 nonprotein-SH농도를 약 2배 이상 증가시켰다. $Cd^{2+}$을 SA와 같이 처리한 경우 $Cd^{2+}$만 처리한 경우보다 뿌리에서 nonprotein-SH농도는 약 2배 증가되었다. 분리표피에서 $Cd^{2+}$+SA 처리는 $Cd^{2+}$ 처리구에 비해 $Cd^{2+}$농도를 약 1.1배 증가시켰으며, 뿌리에서는 약 1.2배 증가시켰다. Nonprotein-SH도 합성도 뿌리와 비슷하게 $Cd^{2+}$농도에 의존하며 증가되었으며, $Cd^{2+}$+SA처리는 nonprotein-SH합성을 $Cd^{2+}$처리구에 비해 약 2배 증가시켰다. 위 결과로부터 신호전달물질인 SA처리는 $Cd^{2+}$ 흡수를 촉진시키며, 아울러 non-protein-SH 합성 과정에도 영향을 주는 것으로 사료된다.
