Effects of Alcohol on Neurocognitive Function, Psychomotor Performance and Subjective Response in Koreans with Different ALDH2 Genotypes (한국인에서 Aldehyde Dehydrogenase 2 유전자 변이가 알코올의 신경인지 기능, 정신운동성 수행 및 주관적 반응에 미치는 영향)

  • Objective : The purpose of this study was to evaluate the effects of alcohol on neurocognitive function, psychomotor performance and subjective response in healthy Korean adults with different ALDH2 genotypes. Method : A total of 24 males, half with active $ALDH2^*1/2^*1$ and the other with inactive $ALDH2^*1/2^*2$, was selected through genotyping using restriction fragment length polymorphism. In a double-blind, placebo-controlled cross-over design, each subject consumed 0.5g/kg dose of alcohol, given as a mixture of 40% vodka and orange juice, and placebo(orange juice) on two separate occasions on an average of weekly intervals. The blood alcohol concentrations(BACs) were measured using a breath analyzer at baseline and at 30, 60 minutes after drinking. P300s were measured at baseline and at 30 minutes after alcohol and placebo intake. Vital signs and psychomotor performance[Critical Flicker Fusion Threshold(CFFT), Choice Reaction Time (CRT), Digit Symbol Substitution(DSS)] were measured at baseline and at 60 minutes after alcohol and placebo intake. Subjective responses were measured at the end of the study. The statistical analysis focused on whether there were any differences between groups with different ALDH2 genotypes. Results : The major results are as follows. 1) BACs in the inactive group were overall equivalent to those in the active group. Only in terms of time, BACs were significantly higher overall at 30 minutes than at 60 minutes after alcohol intake. 2) Pulse rates were significantly increased after alcohol intake compared with placebo, and the increase was greater in the inactive than in the active group. 3) P300 latencies in leads Fz(frontal), Cz(cental) and Pz(parietal) were significantly increased after alcohol intake compared to placebo, and the increase was greater in the inactive than in the active group. P300 amplitudes in leads Cz and Pz were significantly decreased overall after alcohol intake compared to placebo. 4) Compared with placebo, alcohol produced significant effect on the psychomotor performance : impairment in the inactive group, improvement in the active group. 5) Compared with placebo, alcohol significantly induced a negative or an intense effect on the subjective responses in the inactive group, but little negative and even a somewhat positive effect in the active group. Conclusions : These results suggest that ALDH isozyme variance might be an important factor to determine the effects of acute dose of alcohol on the various psychobehavioural functions and also to determine the alcohol use pattern and to predict the future development of alcohol overuse and/or abuse.

  • The use of alcohol is associated with the development and worsening of sleep disorder. Alcohol is generally known to have a sedative effect, but it has an arousal or sedative effect depending on the timing and drinking dose and directly affects REM sleep physiology. Alcohol acts on the central nervous system (CNS) to interfere with the sleep-wake cycle and to affect sleep-related hormone secretion. In addition, the ingestion of alcohol pre-sleep is associated with deterioration and development of sleep related breathing disorders (SBD). The increase in resistance of the upper respiratory tract and the decrease in sensitivity of the CNS respiratory center and the respiratory muscles are major mechanisms of alcohol-induced SBD, and result in snoring or apnea in healthy men or aggravating apnea in patients with OSA. Sleep-related restless leg syndrome and circadian rhythm disorders are common in alcohol use disorder patients. This review provides an assessment of scientific studies that investigated on the impact of alcohol ingestion on nocturnal sleep physiology and sleep disorders.

Citrus junos Fractions Decrease Alcohol-induced Liver Damage and Influence Lipid Metabolism in Alcohol-fed Rats

  • The effect of treatment with Citrus junos fractions (citron 3W, citron 3H, citron 4W and citron 4H) upon rat hepatocytes exposed to alcohol was investigated. We compared the serum biochemistry of rats administered both alcohol and Citrus junos fractions to control rats treated with alcohol alone. The effects of Alanine amino transferase (ALT) were significantly lower in the citron 3H extract group compared with the negative control group (p<0.05) and other experimental groups were not significantly low but a little low compared to negative control group. The levels of triglyceride (TG) were significantly low in all experimental groups compared with negative control group. Especially triglyceride level of citron 3H was lowest near to normal control group. The concentration of total cholesterol was significantly high in negative groups compared with normal control group but in all experimental groups, the concentration of total cholesterol was similar to that of negative control group. Total cholesterol of the citron 4W group was somewhat low compared with negative control group. In contrast, activities of alcohol dehydrogenase (ADH) were significantly higher in all experimental groups compared with the negative control (p<0.05) group. These data suggest that Citrus junos fractions may represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.

Effect of Liuweidihuang-tang on Alcohol-induced Decrease in New Cell Formation in Rat Dentate Gyrus

  • Liuweidihuang-tang has been traditionally used for the treatment of delayed mental and physical development in children, complications of diabetes. and glomerulonephritis. In the present study, the effect of Liuweidihuang-tang on cell proliferation in the dentate gyrus of alcohol-intoxicated Sprague-Dawley rats was investigated via 5-bromo-2' -deoxyuridine (BrdU) immunohistochemistry. Alcohol administration was shown to decrease the number of BrdU-positive cells in the dentate gyrus, while Liuweidihuang-tang treatment increased new cell formation in the dentate gyrus under normal conditions and alcohol intoxication. It was thus demonstrated that aqueous extract of Liuweidihuang-tang exerts a protective effect against alcohol-induced decrease in new cell formation.

Effects of Alcohol Exposure on Ship Operational Ability (알코올이 선박운항능력에 미치는 영향에 관한 기초연구(I))

  • It has been estimated that up to $80\%$ of all marine accidents have human elements as a cause. Out of these, human elements of crew fatigue is one of the main causes of marine accident. Crew fatigue involves sleep, schedule, workload, off-time, alcohol, health and stress. With a ship handling simulator, the effect of alcohol on ship operational performance is studied for each 0.05 and 0.08 $\%$ blood alcohol concentrations(BAC). The main effect of alcohol was found that ship operational performance was significantly impaired by alcohol compared to that in the non-alcohol condition. The results of this research can be used as basic data to minimize marine accidents.

Antioxidant Activities and the Effect of Reducing Serum Alcohol Concentration of Lentinus edodes (표고버섯의 항산화능과 알코올분해능에 미치는 영향)

  • Objectives : The purpose of this study is to evaluate antioxidant activities and reducing serum alcohol concentration of extract of Lentinus edodes on the alcohol administered rats. Methods : Antioxidant effect was measured by total phenolic compound and DPPH-radical scavenging activity of extract of Lentinus edodes in vitro. Blood alcohol concentration, aldehyde concentration, malondialdehyde concentration, glutathion concentration were measured in vivo. Results : The extract of Lentinus edodes increased DPPH-radical scavenging activity dose-dependently. The water extract with boiling water showed lower antioxidant activity and phenolic content than 70% ethanol extract in vitro. Blood alcohol concentration was significantly reduced by pre-treatment of ethanol extract of Lentinus edodes. The effect was more significant than commercial product used as a positive control. Conclusions : This study suggest that Lentinus edodes can be a potential nature resource for the management of ethanol toxicity although the mechanism of action involved in the treatment remains to be explored.

Effect of Cyclohexane Treatment on the Liver Cytochrome P-450 Dependent Aniline Hydroxylase Activity in Alcohol-pretreated Rats (알코올 전처치한 흰쥐에 Cyclohexane 투여가 Cytochrome P-450 dependent aniline hydroxylase 활성에 미치는 영향)

  • To evaluate the effect of cyclohexane(CH) treatment on the liver cytochrome P-450 dependent aniline hydroxylase(CYPdAH) activity in alcohol-pretreated animals, CH(1.56 g/kg body weight) was intraperitoneally administered to Sprague-Dawley male rats, which had been drunk 15% alcohol in distilled water for 1,3 and 6 weeks. CH was injected to rats 4 times every other day and the animals were sacrificed at 24 hours after injection of CH. In the alcohol-pretreated rats, liver injuries were not demonstrated on the basis of the liver weight per body weight, the levels of serum alanine aminotransferase and hepatic microsomal glucose-6-phosphatase activities. By the CH treatment, alcohol-pretreated animals showed the significantly increased activity of hepatic microsomal CYPdAH. Concomitantly $V_{max}$ value in CYPdAH was more increased, whereas $K_{M}$ value more decreased in alcohol-pretreated animals by the treatment of CH. In conclusion, the increasing cause of microsomal CYPdAH in CH-treated rats pretreated with alcohol may be due to induction of enzyme protein in rat liver.r.r.

  • Alcohol is a major risk factor for several diseases and excessive, long-term alcohol consumption are caues physical alteration-fatty liver, hepatitis, cirrhosis, breaking down, Wernicke-karsakoff's syndrome, weight loss, and poor immunity-in virtually all organ and tissue. This study was observed that liver, kidney, and stomach were altered in mouse by the effect of chronic alcohol administration. The mouse were sacrificed to obtain the tissue after mouse were orally injected with 25 % ethanol $18m{\ell}/kg/day$ for 120days. The tissue were stained by hematoxylin and eosin and then observed by light microscope. The results of this study were as follows : 1. The congestion was appeared in liver after 120days alcohol admistration. 2. The destruction of glomerulus were increased and the parietal cell of Bowman's capsule were swelled such as cuboidal cell after 120days alcohol administration. The congestion was appeared in alcohol administrated group. 3. The mucosa and gastric pit were destructed and the ulceration was appeared in stomach after 120days administration. The parietal cells and chief cells were damaged. Above results were shown that the tissue were damaged by chronic alcohol administration.

Moderate Alcohol Consumption Does Not Prevent the Hypertension among Korean: the 2001 Korean National Health and Nutrition Examination Survey (한국인 성인남녀의 알코올 섭취가 혈압에 미치는 영향 : 2001 국민건강.영양조사자료를 이용하여)

  • The purpose of this study was to test whether moderate alcohol consumption has any positive effect on lowering blood pressure among Koreans. Study subjects were Korean adults 20 years or older (n=5,234) who participated in the 2001 Korean National Health and Nutrition Examination Survey (KNHANES). Analysis of variance and analysis of covariance were used to construct univariate and multivariate models relating alcohol consumption to blood pressure for the analysis. After adjustment for possible covariates, drinkers (regardless of drinking level) had significantly higher systolic and diastolic blood pressure, compared with never-drinker for male subjects. Diastolic and systolic blood pressures were also significantly elevated with the drinking frequency and amount of alcohol intake among male subjects. For the female subjects, only diastolic blood pressure was significantly associated with the alcohol consumption at multivariate model. however, low level alcohol consumption did not show any sign of lowing effects on blood pressure. The result implies that moderate alcohol consumption did not have any positive effect on lowering blood pressure among Koreans for either sex.

A Meta-Analysis of Factors Related to Adolescent Problem Drinking (청소년 문제음주와 관련한 영향요인에 대한 메타분석)

  • Objectives: This study examined the trend of alcohol-related research, with a specific focus on adolescent problem drinking, to provide evidence for prevention and intervention for adolescent alcohol problems and alcohol policy in Korea. Methods: A total of 17 studies examining adolescent problem drinking were included for meta-analysis. Using CMA (Comprehensive Meta-Analysis) 2.0, the effect sizes of correlation coefficients of variables related to adolescent problem drinking were analyzed. Results: 176 variables were extracted under 4 higher categories and 20 sub-categories. Variables were extracted using the four higher categories, namely individual, family, environmental, and demographics, as the theoretical framework. Results showed that most studies on adolescent problem drinking focused mostly on individual factors followed by family factors. Variables including drinking motivation, drinking expectancy, and peer relations showed relatively higher effect size. Conclusions: Study findings suggest the importance of balance between individual and environmental factors in examining problem drinking among adolescents. In order to provide evidence for alcohol policy, more attention should be given to macro environment that are known to influence adolescent drinking.
