Capsaicin 및 그 대사산물에 의한 in vitro에서의 니트로소 화합물 생성 저해효과 (Inhibition of in vitro Nitrosation by Capsaicin and Its Metabolites)

  • Capsaicin(8-methyl-N-vanillyl-6-nonenamide: CAP) known well as a major compound of not taste in hot pepper, was investigated for the inhibition effect on in vitro nitrosation. CAP(100$\mu$mol) inhibited the formation of N-nitrosoproline(NPRO) and N-nitrosothioproline(NTPRO) by 56% and 26%, respectively. Vanillyl alcohol inhibited the nitrosation of proline by a concentration-dependent manner, and vanillic acid and vanillin were less effective in blocking the nitrosation of proline compared to CAP and anillyl alcohol. The inhibitory effect of NPRO formation by CAP was evaluated to similar with alpha-tocopherol, and vanillyl alcohol was more effective than alpha-tocopherol in blocking the nitrosation of proline. Our results suggested that CAP and its metabolites such as vanillyl alcohol could inhibit endogenous nitrosation in hydrophobic biological environment.

  • Spraque-Dawley계 응성흰쥐에 계통분획한 cornsilk의 각 분획물을 경구투여하고 혈청 Ethanol농도와 간의 Alcohol dehydrogenase활서에 미치는 효과를 검색 추적한 결과 알코올대사를 촉진시키는 성분은 주로 에탄올가용부에, 억제시키는 성분은 에탄올불용부에 주로 존재함을 추정할 수 있었다.

Influence of preparation parameters on rheological behavior and microstructure of aqueous mixtures of hyaluronic acid/poly(vinyl alcohol)

  • Aqueous mixtures of hyaluronic acid and poly(vinyl alcohol) system and hydrogels thereof were introduced to obtain new bioartificial materials that have excellent mechanical properties, biocompatibility and enhanced rheological properties. The interactions between hyaluronic acid and poly(vinyl alcohol) and/or borax were investigated by rheological measurements. Preparation parameters of the aqueous mixtures were mixture composition, the degree of hydrolysis of poly(vinyl alcohol) and borax concentration. From the rheological behavior, it could be deduced that the key factor of the interaction between hyaluronic acid and poly(vinyl alcohol) was the hydrogen bonding between them and the effect was pronounced with borax. Enhanced viscosity was observed at the composition of $20wt\%$ of hyaluronic acid solution and $80wt\%$ of poly(vinyl alcohol) and borax solution. Rheological properties were influenced by the degree of hydrolysis of poly(vinyl alcohol) and borax concentration. As the degree of hydrolysis and borax concentration increased, rheological properties increased due to the increased hydrogen bonding and networking of hyaluronate aggregates. Physical hydrogels from hyaluronic acid and poly(vinyl alcohol) were prepared and the composition dependence of the gels was rheologically investigated as well.

페피노 추출액이 흰쥐의 알코올 대사에 미치는 영향 (Effect of Pepino Extract on Alcohol Metabolism in Rats)

  • 본 연구에서는 페피노(Solanum muricatum Ait.) 과실추출액이 흰쥐의 알코올 대사에 미치는 영향을 알아보기 위하여 페피노 추출액이 알코올 투여 쥐의 혈 중 알코올과 아세트알데히드 농도, GOT와 GPT에 미치는 영향에 대하여 알아보았다. 페피노 추출액($11.4^{\circ}$ Brix) 투여 시, 혈 중 알코올농도는 대조군에 비하여 유의적으로 31.5% 감소하였으며 혈 중 아세트알데히드 농도는 유의적 차이는 없었다. 페피노 추출액을 농도별로 알코올 투여 30분 전에 투여 시, 혈 중 알코올 농도는 $1^{\circ}$ Brix 페피노 추출액 투여군이 대조군보다 56.4% 유의적으로 낮았으며, 혈 중 아세트알데히드 농도는 유의적 차이는 없었다. 혈중 알코올 농도 감소효과는 페피노 추출액 농도가 $1^{\circ}$ Brix일 때 가장 좋았다. $1^{\circ}$ Brix 페피노 추출액을 투여하여 시간 경과에 따른 혈 중 알코올 농도는 알코올 투여 1, 3, 5시간 후 대조군보다 유의적으로 각각 19.9%, 45.4%, 60.7% 감소하였다. 혈 중 아세트알데히드 농도는 알코올 투여 5시간 후, 대조군보다 약 19% 낮게 나타났다. GOT, GPT 활성은 모두 정상범위에 속해 알코올의 1회 투여로는 간 손상을 일으키지 않았다. 페피노 추출액은 알코올을 투여한 흰쥐의 혈중 알코올을 낮추어 흰쥐의 알코올 대사에 긍정적으로 영향을 미치는 것으로 사료된다.

알코올이 노르에피네프린에 의한 혈관 수축에 미치는 영향 (Effect of alcohol on norepinephrine-induced vasoconstriction)

  • Purpose: Norepinephrine (NE) is a neurotransmitter of the sympathetic nervous system. It is used for treating hypotension on distributive shock, central nervous system injury, or sepsis. There are several reports that state that alcohol suppresses vasoconstriction by NE. Thus, our hypothesis is that the effect of NE is reduced in alcohol-drinking patients with distributive shock. We investigated whether alcohol suppresses NE-induced contraction and aimed at finding a solution to this problem. Methods: For this study, we used the aorta from male Sprague-Dawley rats (9-11 weeks) and an isometric contraction system. Results: Our results showed that alcohol suppresses NE contraction and does not affect epinephrine induced a contraction. Moreover, in the presence of alcohol, a 7:3 mixture of NE and epinephrine induced a contractile force similar to that induced by NE under normal conditions. Conclusion: We found that the vasoconstrictive force of NE decreased in the blood vessels in which alcohol was present, which was not because endothelial cells. The reduced contractile force was most similar to that induced by a 7:3 mixture of NE and epinephrine.

한국 성인남자에 있어서 알콜섭취와 혈중지질농도와의 관계 (The Effect of Alcohol on the Blood Lipid Level of Korean Adult Men)

  • This study was conducted to evaluate the relations between alcohol intake and blood lipid level in a group of 1,138 Korean adult men, ages 20 to 69 years. Total number of each of a variety of drinks in the previous two weeks was obtained by questionnaire and converted into grams of alcohol consumed in a week. The levels of blood lipid such as Cholesterol, HDL-Cholesterol, Triglyceride and LDL-Cholesterol were examined by enzyme method. We also observed the effects of various variables such as age, body mass index, smoking, exercise and blood pressure on blood lipid level. The results obtained were as follows : 1. Average weekly alcohol intake was $129.0{\pm}167.4gm$ and that of 30-39 age group was the highest as $149.3{\pm}170.4gm$. 2. Levels of Cholesterol Triglyceride and LDL-Cholesterol tended to increase with increasing age, but level of HDL-Cholesterol showed no significant relationship with age. 3. The positive linear regressions of alcohol intake on HDL-Cholesterol and Triglyceride were noted statistically significant. 4. Multiple regression analysis demonstrated that the effect of alcohol on HDL-Cholesterol and Triglyceride was statistically significant and the contribution rates were 5.0% and 0.8%, respectively. And, in the case of HDL-Cholesterol, the alcohol intake was the most significant independent variable.

  • 국내외 해양사고 통계에 의하면, 해양사고의 8U내외가 직접 또는 간접적으로 인적요인에 기인한 것으로 보고되고 있다. 이러한 인적요인 가운데, 해상승무원의 피로요인은 장시간 소요되는 선박의 운항과정에서 발생할 수 있는 사고의 주된 요인이다. 해상승무원의 피로를 유발하는 요인으로는 수면시간. 운항일정, 업무량, 휴식시간, 알코올 섭취, 건강상태, 근로 및 승선여건 및 각종 스트레스 둥이 포함된다. 본 연구에서는 이러한 피로유발 요인들 중에서 알코올의 영향이 선박운항능력에 미치는 영향을 알아보기 위해, 선박운항 시뮬레이터를 이용한 실험을 수행하였다. 실험결과를 분석한 결과, 알코을의 섭취가 선박운항능력을 상당히 저하시키는 것으로 나타났다. 이러한 분석결과는 앞으로 선박운항자의 피로에 의한 해양사고 발생을 근본적으로 줄이기 위한 기초자료로 활용할 수 있을 것이다.

흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과 (Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat)

  • 본 연구에서는 숙취해소에 좋은 것으로 알려진 식품 소재와 황칠나무 추출물을 복합하여 in vitro 및 in vivo에서 알코올 유도된 간 손상에 대한 보호효과를 검토하였다. HepG2세포에서 300 mM 알코올과 SBJ 혼합물을 처리하였을 때 농도 의존적으로 LDH 수치가 감소하였으나 황칠나무 추출물을 제외한 복합물인 SBJ-복합물에서는 그 효과가 유의적으로 낮아지는 것을 관찰하였다. In vivo에서 SBJ 혼합물의 보호효과를 확인하고자 흰쥐에 알코올과 SBJ 혼합물을 투여한 결과, 알코올 투여 후 1시간까지 급격하게 혈중 알코올 수치가 EtOH군에서 증가하는 것이 관찰되었으며, SBJ 혼합물 투여군은 유의적인 차이로 감소되었음이 관찰되었으며 또한 농도의존적인 경향을 확인하였다. ADH 및 ALDH 활성의 증가는 SBJ 혼합물의 알코올 분해 및 대사산물의 제거 활성에 기여할 것으로 예상된다. 뿐만 아니라, 알코올에 의하여 증가한 LDH의 농도가 대조군과 유사한 수준으로 유지하는 것을 확인하였으며, GST, SOD, GPx 및 reduced glutathione와 같은 항산화 인자 및 효소의 활성은 대조군보다 EtOH군이 유의적으로 감소했으며, SBJ 혼합물에 의해 개선되는 경향을 관찰하였다. 이를 통해 SBJ 혼합물은 ADH, ALDH 활성 증가 및 항산화 방어계를 향상시킴으로써 산화적 스트레스 감소를 통한 간보호 효과가 있는 것으로 생각되며, 이러한 간보호에 미치는 주요한 추출물은 황칠나무 추출물임을 알 수 있었다. 이러한 결과를 토대로 향후 숙취해소 작용을 갖는 신규 식품소재로 활용될 수 있을 것으로 기대된다.

Pine Needle Oil and Korean Medicinal Herb Complex Protect Hyperlipidemia and Liver Cell Damage Induced by Alcohol

  • The effect of treatment with pine needle oil complex (complex of pine needle oil and Korean medicinal herbs) upon rat hepatocytes exposed to alcohol was investigated. We compared body weight gain and ratios of liver and kidney to body weight and the serum biochemistry of rats administered both alcohol and Pine needle oil complex to control rats treated with alcohol alone. Pine needle oil complex treatment resulted in a significant reduction in the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and triglycerides (TG) compared to the control rats. These data suggest that Pine needle oil complex represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.

생약 조성물의 알코올 분해 및 항염증 효과 (Effect of Herbal Composition on Alcohol Degradation and Anti-inflammatory Activity in Mice)

  • These studies were conducted to investigate the effects an herbal extract composition(MHE) containing $Hovenia$ $dulcis$ T., $Artemisa$ $capillaris$ T., $Pueraria$ $thunbergiana$ B., $Polygonatum$ $falcatum$ A., $Agastache$ $rugosa$ O., $Silybum$ $marianum$ L. and $Glycyrrhiza$ $uralensis$ F. in alcohol administered mice. Prophylactic administration of different doses of MHE(20~100 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice. In addition, intraperitoneal administration of the MHE showed anti-inflammatory effects in inhibition tests of vascular permeability produced by acetic acid. MHE also reduced the concentrations of nitric oxide(NO) and tumor necrosis factor(TNF)-${\alpha}$ in macrophages that were activated by LPS. These results demonstrate that MHE possesses the potential to stimulate alcohol degradation and inhibit inflammatory effects in mice.
