A study on the Influence of Ginseng Components On cAMP-cGMP Regulation Mechanism (cAMP-cGMP 조절 메카니즘에 미치는 인삼 성분의 영향에 관한 연구)

  • The effect of ginsenosides on the adenylate cyclase and guanylate cyclase of rat brain has been studied. We have found that Rbl, Rc and one unknown ginsenoside (probably Ra) exerted reciprocal effects on adenylate cyclase and guanylate cyclase. This dual effect of ginsenosides leads us to speculate that some ginsenosides may act as regulatory agents and modulate the activities of these two enzyme systems.

PD184352 Releases the Regular Hypoxic Reversible DNA Replication Arrest in T24 Cells

  • The oxygen dependent regulation of DNA replication is an essential property of proliferating mammalian cells. In human T24 bladder cancer cells, several hours of hypoxia leads to reversible DNA replication arrest and re-entry of oxygen induces a burst of replication initiation. This short communication provides strong evidence that PD184352 initiates DNA replication in living hypoxic cells without elevating the oxygen level. PD184352 releases the regular hypoxic replicon arrest, however, at a low intensity compared to the effect of reoxygenation. Moreover, PD184352 shows no effect on normoxically incubated as well as reoxygenated T24 cells.

  • It has been observed that white loght can suppress both cell growth and protein secretion in Baker's yeast. This effect was explored in batch liquid fermentations. Possible applications of this phenomenon are (a) use as a tool for pre-concentrating excreted enzymes prior to subsequent purification and (b) an engineering variable for regulation yeast fermentations.

Effect of Apoptosis Induction of Ailanthus altissima on Human Lung Carcinoma Cells

  • We investigated the inhibitory effects of solvent extracts from Ailanthus altissima in A549 human lung cancer cell. A. altissima has been recognized as a traditional healthy food due to its various biological activities against hypertension, strokes, fever, pain, neuralgia, inflammation, and cancer effects. Recently, it has been reported that the extracts of various wild vegetables show strong anti-cancer properties by induction of apoptosis. However, the mechanisms of their cytotoxicity in human lung cancer cells have been poorly understood. The present study was investigated the effects of solvent extracts from A. altissima on cell growth and apoptosis on A549 human lung cancer cells. A treatment of A. altissima inhibited the growth of A549 cells in a dose-dependent manner by inducing apoptosis. Especially, the chloroform fraction showed the highest anti-cancer effect among five kinds of fractions. And also, induction of apoptosis by chloroform fraction were associated with down-regulation of Bcl-2, and up-regulation of pro-apoptotic Bax expression. From these results, A. altissima may have a therapeutic potential in human lung cancer cells and as a functional food.

Potentiation of Anticancer Effect of Artesunate by Combination with Nonsteroidal Anti-inflammatory Drugs on Human Cancer Cells (Artesunate와 비스테로이드소염제 NSAID의 병용 처리에 의한 항암 활성 증강)

  • The purpose of present study is to investigate the role of artesunate (ART) in enhancing anticancer effect of nonsteroidal anti-inflammatory drug (NSAID) on human cancer cells, and we elucidate a possible molecular mechanism of this combination effect. We showed that the combined effect of ART with NSAID such as celecoxib (CCB) or dimethyl-CCB (DMC) in various type of human cancer cells. After ART treatment, the expression of p62, nuclear factor erythroid 2-like 2 (NRF2) and cancer stemness (CS)-related proteins including CD44, CD133, aldehyde dehydrogenase 1 (ALDH1), octamer-binding transcription factor 4 (Oct4), mutated p53 (mutp53) and c-Myc was down-regulated. ART induced autophagy as reduction of the autophagy receptor p62, which was associated with up-regulation of activating transcription factor 4 (ATF4) and C/EBP homologous protein (CHOP), and simultaneous down-regulation of NRF2 and CS-related proteins was occurred in the human cancer cells. These results indicate a possibility that ART activates autophagy through ATF4-CHOP cascade leading to down-regulation of CS-related proteins and subsequently eradicated cancer stem cells. In addition, co-treatment with ART and imatinib was more effective than either drug alone on growth inhibition and apoptosis induction of cancer cells. In conclusion, induction of autophagy-dependent cell death by ART might play a critical role in mediating the synergistic effect of drug combination (ART/NSAID and ART/imatinib). Therefore, ART could be a promising candidate as a chemosensitizer to enhance the anticancer effects of NSAID and imatinib.

Air Pollution by Airplane Engines (항공기 엔진에 의한 대기오염)

  • Air pollutant emissions from airplane engines are estimated about 2 to 4 % of mobile source of USA and European countries which is not a large portion of current air pollution. But the passengers and airfreights are continuously increasing 5 to 7% annually and potential demands of air transportation services come to present, it could effect air pollutant emissions of USA will increase within 15 years. In case of our country, there has been continuous increase of air transportation service due to considerable economic growth in recent years and increase of air pollutant emissions of major international airports has followed. Rapid increase of air transportation due to launching of Inchon International Airport could effect air pollution dominantly. By this circumstance environmental specialist as well as mass communication raised necessity of air pollutant emission regulation from airplane engines. It is estimated that air pollutant emissions from airplane engines in our country is 2.7% of automobile sources, 10,809 ton, which is the same level as USA and European countries. It is increased by 12,2% compared to air pollutant emissions during 1996 and it will be increased more than a half of current air pollutant emission within 15 years due to our country's economic condition. Therefore implementation of airplane engine emissions regulation as well as test standards and accumulation of technology about characteristics of airplane engine emission and reduction method are needed. And continuous estimation of air pollutant emission from airplane engines and monitoring of increment as well as development of countermeasures by long term are necessary.

In Vitro Anti-Neuroblastoma Activity of Thymoquinone Against Neuro-2a Cells via Cell-cycle Arrest

  • We have recently shown that thymoquinone (TQ) has a potent cytotoxic effect and induces apoptosis via caspase-3 activation with down-regulation of XIAP in mouse neuroblastoma (Neuro-2a) cells. Interestingly, our results showed that TQ was significantly more cytotoxic towards Neuro-2a cells when compared with primary normal neuronal cells. In this study, the effects of TQ on cell-cycle regulation and the mechanisms that contribute to this effect were investigated using Neuro-2a cells. Cell-cycle analysis performed by flow cytometry revealed cell-cycle arrest at G2/M phase and a significant increase in the accumulation of TQ-treated cells at sub-G1 phase, indicating induction of apoptosis by the compound. Moreover, TQ increased the expression of p53, p21 mRNA and protein levels, whereas it decreased the protein expression of PCNA, cyclin B1 and Cdc2 in a dose-dependent manner. Our finding suggests that TQ could suppress cell growth and cell survival via arresting the cell-cycle in the G2/M phase and inducing apoptosis of neuroblastoma cells.

Effect of Amygdalin from Armeniacae Semen on Ion Currents Changed by Lipopolysaccharide in Rat Periaqueductal Gray Neurons

  • Background : Amygdalin is abundant in Armeniacae semen, and it is recently reported to treat cancers and relieve pain. But modus operandi of amygdalin at the level of neuron has not been reported, yet. Objective : This study aimed to find out the effect of amygdalin on glycine- and glutamate-induced ion currents in periaqueductal gray (PAG) neurons. And it was investigated that amygdalin participates in the regulation of the descending pain control system in the level of PAG neurons. Method : We investigated that the changes of glycine- and glutamate-induced ion currents in PAG neurons through application of lipopolysaccharides (LPS) and application of amygdalin with LPS by using the nystatin-perforated patch clamp method. Result : Application of LPS on PAG neurons resulted in increased glycine-induced ion current, and in decreased glutamate-induced ion current. In contrast, application of amygdalin with LPS resulted in decreased glycine-induced ion current increased by LPS, and increased glutamate-induced ion current decreased by LPS. Conclusion : Amygdalin from Armeniacae semen controls glycine- and glutamate-induced ion current by LPS in PAG neurons, and it is suggested that amygdalin participates in the regulation of the descending pain control system in the level of PAG neurons.

Inhibitory Effect of Fructus Ligustri Lucidi on Tyrosinase and MITF Expressions (여정자 추출물의 Tyrosinase 및 MITF 발현 억제 효과)

  • The purpose of this study was to investigate the mechanism of Hexane extract of Fructus Ligustri Lucidi (HFLL)-induced regulation of melanogenesis. An apparent down-regulatory effect of tyrosinase activity was observed when B16F10 cells were incubated with HFLL. Interestingly, HFLL did not inhibit the catalytic activity of cell-free tyrosinase from B16F10 cells, whereas kojic acid directly inhibited tyrosinase activity. Regarding protein levels of melanogenic enzymes, the amounts of tyrosinase and tyrosinase-related protein 1 (TRP-1) were decreased by HFLL, while the amount of tyrosinase-related protein 2 (TRP-2) slightly was reduced after incubation with HFLL. Treatment with HFLL was found to down-regulate microphthalmia-associated transcription factor (MITF). These results suggest that HFLL is an effective inhibitor of pigmentation caused by down regulation via MITF, tyrosinase, and TRP-1 expressions.
