Glucocorticoids (GCs) are steroid hormones regulated through responses to stress to maintain diverse metabolic and homeostatic functions. GCs act on the glucocorticoid receptor (GR), a member of the nuclear receptor family. This study identified and characterized the GR gene from the big-belly seahorse Hippocampus abdominalis designating it HaGR. The open reading frame of the HaGR cDNA was 2,346 bp in length, encoding a 782-amino-acid polypeptide with a theoretical isoelectric point of 6.26 and predicted molecular mass of 86.8 kDa. Nuclear receptors share a common structural organization, comprising an N-terminal transactivation domain, DNA-binding domain, and C-terminal ligand-binding domain. The tissue-specific mRNA expression profile of HaGR was analyzed in healthy seahorses using a qPCR technique. HaGR mRNA was expressed ubiquitously in all of the tissues examined, with the highest expression levels in kidney, intestine, stomach, and gill tissues. The mRNA expression in response to immune challenge with lipopolysaccharide (LPS), polyinosinic:polycytidylic acid (poly I:C), Edwardsiella tarda, and Streptococcus iniae revealed that it is inducible in response to pathogen infection. These results suggest that HaGR is involved in the immune response of the big-belly seahorse.
Stimulator of interferon gene (STING) is induced by various inflammatory agents, such as lipopolysaccharide and microbial pathogens, including virus and bacteria. In this study, we obtained a full-length cDNA of a STING homolog from olive flounder using rapid amplification of cDNA ends PCR technique. The full-length cDNA of Paralichthys olivaceus STING (PoSTING) was 1442 bp in length and contained a 1209-bp open reading frame that translated into 402 amino acids. The theoretical molecular mass of the predicted protein sequence was 45.09 kDa. In the PoSTING protein, three transmembrane domains and the STING superfamily domain were identified as characteristic features. Quantitative real-time PCR revealed that PoSTING expressed in all the tissues analyzed, but showed the highest level in the spleen. Temporal expression analysis examined the significantly upregulated expression of PoSTING mRNA after viral hemorrhagic septicemia virus (VHSV) stimulation. In contrast, no significant changes in the PoSTING expression were detected in Edwardsiella tarda-challenged group compared to the un-injected control. The expression of P. olivaceus type I interferon (PoIFN-I) was also highly upregulated upon VHSV challenge. These results suggest that STING might be involved in the essential immune defense against viral infection together with the activation of IFN-I in olive flounder.

This experiment was conducted to evaluate the effect of two different organic acid products as antibiotic replacement in olive flounder paralichthys olivaceus. Fish averaging $3.5{\pm}0.05$ g($mean{\pm}SD$) were fed one of the ten semi-purified diets : Diet 1 ; Control, Diet 2 ; add antibiotics - 50mg OTC/kg body weight/day(OTC), Diet 3 ; Add organic acid bland A(OABA) - 4g/kg diet, Diet 4 ; add organic acid bland B(OABB) - 4g/kg diet for 10 weeks. Total gut microflora counts were significantly higher in the control group compared to the OTC and organic acid groups(P<0.05). Fish fed OABA, OABB and OTC had lower gut Vibrio counts compared to the control, but were not significantly different. Results from the challenge study indicate that mortality in the different treatment groups (50%) was significantly lower than those observed for the control group (100%). There were no differences in mortality between the OTC and organic acid groups. Overall findings from this study indicate that the organic acid blends A and B were as effective as oxytetracycline, an antibiotic, in regulating total gut bacterial numbers, Vibrio counts and providing protection against a pathogen such as Edwardsiella tarda.
The study was conducted to investigate the dietary supplementation of citrus by-product (CBP) and CBP fermented with Bacillus subtilis (BS) or B. pumilus (BP) on growth performance, feed utilization, innate immune responses and disease resistance of Korean rockfish Sebastes schlegelii. Triplicate groups of juvenile fish (initial weight of 22.9 g) were fed one of five experimental diets which were formulated to replace a synthetic vitamin C (L-ascorbyl-2-polyphosphate, LAPP) with CBP or fermented CBP. A basal diet without LAPP and CBP was used as the control diet (Con). At the end of the 13 weeks feeding trial, growth performance, feed utilization and survival of fish fed the Con were significantly lower than those of fish fed the LAPP, CBP and fermented CBP diets. Fish fed the Con were significantly lower in lysozyme activity than fish fed LAPP, CBP and fermented CBP diets. In a challenge test against Edwardsiella tarda, cumulative mortality of fish fed LAPP, CBP and fermented CBP diets was significantly lower (20-30%) than that of the Con (60%). The findings in this study indicate that the CBP and/or fermented CBP can be a promising vitamin C source for LAPP replacement in diet for Korean rockfish.
This study has been performed to investigate the potential effects of by-product discarded after probiotics production (BPPP) on growth performance, immune gene expression, innate-immunity status, and disease resistance of carp, Cyprinus carpio. For 3 weeks, carp were fed four diets containing different levels of BPPP at 0, 0.1, 0.2 and 0.5% per kg of normal diet. Every 7 days of feeding, immune-related gene expression, serum lysozyme activity and ACH50 were analyzed. Growth rates and challenge test with E. tarda were conducted after 3 weeks of BPPP feeding. Both lysozyme activity and ACH50 were significantly (p<0.05) increased in all BPPP supplemented groups compared to the control at every 7 day for 3 weeks of feeding trial. The gene expression of pro-inflammatory cytokines, IL-1β and TNF-α was significantly (p<0.05) up-regulated until 21 days of feeding in all groups except for 0.2% group on day 7 post feeding. The anti-inflammatory cytokine IL-10 gene expression was only significantly (p<0.05) increased in 0.1% group on day 7 and decreased (p<0.05) on day 14 in all BPPP supplemented groups. On day 21, the IL-10 gene expression was augmented (p<0.05) in all groups. SOD gene expression was significantly (p<0.05) increased compared to the control on day 14 and 21 post feeding, whereas no significant difference was observed on day 7. In challenging test, 0.2%, 0.1%, 0.5% and control group showed 80%, 70%, 60% and 40% of survival rate, respectively. Feed conversion rate was only improved in 0.5% group. In conclusion, the present study indicates that dietary BPPP suplementation improved growth performance, innate immune response and bactericidal activity in carp.
고삼 뿌리 열수 추출물을 농도별로 넙치 사료에 흡습시켜 12주 동안 어류에 급여 후 비특이적 면역 및 질병저항성에 대한 효과를 조사하였다. 고삼 추출물의 첨가는 어류 성장에는 아무런 영향을 미치지 않았다. 혈액화학적인 조사 결과에서도 고삼 첨가로 인한 넙치의 생리에는 아무런 영향을 나타내지 않았다. 라이소자임 활성은 0.05%의 고삼 추출물 농도로 흡습시킨 사료 그룹에서 라이소자임 활성이 증가됨을 알 수 있었다. 백혈구의 식세포 활성은 0.05%의 고삼 추출물로 흡습시킨 사료 그룹에서 유의적으로 증가되었다. 병리학적 결과를 통하여 고삼 추출물을 농도별로 넙치에 투여하였을때 병리조직학적 영향이 없음을 알 수 있었다. 에드와드균의 인위감염에 의한 질병저항성은 0.05%의 고삼 추출물로 흡습시킨 사료 그룹에서만 상대생존율이 높게 나타났다. 이상의 결과를 종합하여 볼 때 고삼뿌리 열수 추출물은 넙치에 대한 비특이적 면역력 증강 및 질병 저항성 증강에 효과가 있는 것으로 사료되었다.
본 연구는 치어기 조피볼락 Sebastes schlegeli 사료내 생균제 첨가가 넙치의 성장 및 면역반응에 미치는 영향을 평가하였다. 실험사료는 주 단백질원으로 북양어분(white fish meal), 콘글루텐밀(corn gluten meal), 탈피대두박(dehulled soybean meal)을 사용하였으며, 탄수화물원으로는 밀가루(wheat flour)를, 지질원으로는 고도불포화지방산(n-3 HUFA)이 다량 함유된 오징어간유(squid liver oil)를 사용하였다. 생균제의 첨가효과를 확인하기 위하여 B. polyfermenticus(BP), Bacillus licheniformis(BL) 및 복합종균(B. polyfermenticus+Saccharomyces cerevisiae; BP+SC)을 실험사료 내에 각각 $1.0{\times}10^7$ CFU/kg diet 수준으로 첨가하였다. 2주간의 예비사육 후, 평균무게 $12.0{\pm}0.1g(mean{\pm}SD)$인 조피볼락을 500 L 원형수조에 각 실험구 당 각각 20마리씩 3반복으로 무작위로 배치하였고, 실험사료는 1일 2회 어체중의 $1.7{\pm}0.6%$(오전 10시, 오후 4시)씩 12주간 공급 하였다. 증체율, 사료효율, 일간성장율 및 단백질 전환효율에 있어서 BP+SC를 공급한 실험구가 대조구(control)에 비하여 유의하게 높게 나타났지만(P<0.05), BP, BL 및 BP+SC를 공급한 실험구간에는 유의한 차이가 나타나지 않았다. 생존율에 있어서는 모든 실험구간에 있어 유의한 차이가 나타나지 않았다. 전어체 지방함량에 있어서 대조구가 다른 모든 실험구에 비해 높게 나타났으며, 전어체 단백질, 회분 및 수분함량에 있어서는 모든실험구간에 유의한 차이가 없었다. 혈액분석에 있어서 혈장내 glucose의 함량은 BP+SC를 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으나, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. Respiratory burst activity(NBT assay)에 있어서 BP+SC와 BL을 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으며, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. 혈청의 lysozyme 활성에 있어서는 BP와 BL을 공급한 실험구가 대조구에 비해 높게 나타났으며, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. 공격실험 결과, 폐사는 Edwardsiella tarda를 접종한지 1일 후부터 시작하여 11일째 종료 하였다. 생균제를 투여한 모든 실험구가 대조구에 비해 초기폐사율이 낮게 나타났다. 공격실험 후 8일째부터 종료시 까지는 BP+SC를 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으며, BP, BL및 BP+SC를 공급한 실험구간에는 유의한 차이가 나타나지 않았다. 상기 결과를 토대로, 조피볼락 사료내 B. polyfermenticus, B. licheniformis 및 B. polyfermenticus 와 S. cerevisiae의 혼합첨가는 조피볼락의 성장 및 사료효율 증진과 항산화능 및 특정질병저항성에 좋은 효과를 나타낼 수 있을 것으로 사료된다.
